  • We try to look for optimum conditions of pre-reductants like L-Cysteine, KI and $FeSO_4$ when analyzing inorganic arsenic by using hydride generation-atomic absorption spectrometry, and run a comparative study of effect in the analysis of them. Also, we separated and analyzed only inorganic arsenic by using $H_2SO_4$-trap to eliminate organic arsenic which are MMA(monomethylarsonate) and DMA(dimethylarsinate). Under the conditions of mixture acid of 1.8 M HCl and 0.08 M $HNO_3$, arsenic standard solution of 20 ppb have more higher absorbance than without adding acid. In case of L-Cysteine, As(V) completely reduces into As(III) when 0.5 g of L-Cysteine is reacted more than 30 mins. in weak acid condition of approximately 0.07 M $HNO_3$ or HCl. In the event of KI, As(V) completely reduces into As(III) when 3 g of KI is reacted more than 1hour in acid condition of 0.8 M $HNO_3$. On the occasion of $FeSO_4$, the inside of tube is blocked by precipitation by mixture reaction of $NaBH_4$ and $Fe^{2+}$, therefore, comparing to other pre-reductants, reproducibility of efficiency of reducing As(V) to As(III) is low. To evaluate the accuracy of the analytical results, we use NIST SRM 1643C Trace Elements in Water ($82.1{\pm}1.2ng/mL$). The results are satisfactory.

  • In this study, the model enhancing the safety of reservoirs and reducing the downstream flood damage by reservoirs system operation during abnormal flood was developed. Linear programming was used for the optimal reservoirs system operation during an abnormal flood and fuzzy inference system was introduced to solve the uncertainty problem which is included in hydrological factors like inflow, water level and inflow variation of reservoir operation. The linear programming model determined the optimal reservoir system operation rules and could be used in situation where water demands varies rapidly during the abnormal flood events using fuzzy control technique. In this study, the optimal reservoirs system operation for Andong and Imha reservoirs located in the upper basin of Nakdong river was performed in order that the design flood discharge at Andong city would not be exceeded for the design flood of 100 year and PMF(Probable Maximum Flood). And the model that determines the release according to the downstream flow discharge, the reservoir storage, the inflow and the inflow variation of each reservoir was developed using the optimal system operation result and fuzzy control technique. The developed model consisted of 224 fuzzy rules according to the conditions of Andong reservoir, Imha reservoir and Andong city. And the release from each reservoir could be determined when the current data are used as input data through the developed GUI.

  • Perchlorate ($ClO_4^-$) is a contaminant found in surface water and soil/ground water. Microbial removal of perchlorate is the method of choice since microorganisms can reduce perchlorate into harmless end-products. Such microorganisms require an electron donor to reduce perchlorate. Conventional perchlorate-removal techniques employ heterotrophic perchlorate-reducing bacteria that use organic compounds as electron donors to reduce perchlorate. Since continuous removal of perchlorate requires a continuous supply of organic compounds, heterotrophic perchlorate removal is an expensive process. Feasibility of autotrophic perchlorate-removal using elemental sulfur granules and activated sludge was examined in this study. Granular sulfur is relatively inexpensive and activated sludge is easily available from wastewater treatment plants. Batch tests showed that activated sludge microorganisms could successfully degrade perchlorate in the presence of granular sulfur as an electron donor. Perchlorate biodegradation was confirmed by molar yield of $Cl^-$ as the perchlorate was degraded. Scanning electron microscope revealed that rod-shaped microorganisms on the surface of sulfur particles were used for the autotrophic perchlorate-removal, suggesting that sulfur particles could serve as supporting media for the formation of biofilm as well. DGGE analyses revealed that microbial profile of the inoculum (activated sludge) was different from that of the biofilm sample obtained from enrichment culture that used sulfur particles for $ClO_4^-$-degradation.

  • This study was designed to investigate antioxidant activities of pomegranate endocarp extracts. Pomegranate endocarp extract contains the highest antioxidant function compared to pomegranate extracts from other parts. Pomegranate endocarp extract was fractionated with hexane, dichloromethane, ethyl acetate, butanol extract, and water, followed by evaluation for antioxidant activity. During this experiment, various antioxidant tests such as nitrite scavenging activity, reducing power, superoxide anion scavenging activity, $ABTS^+$ scavenging activity, SOD like-activity, and DPPH radical scavenging activity were conducted on the $CH_2Cl_2$, EtOAc, BuOH, $H_2O$ fractions of pomegranate endocarp extract. Results showed that the ethyl acetate fraction contained the highest DPPH radical scavenging activity among the samples. For SOD like-activity, the dichloromethane fraction had the highest antioxidant activity. For superoxide anion scavenging activity, the ethyl acetate fraction had the highest antioxidant activity. In the $ABTS^+$ scavenging activity test, the ethyl acetate fraction $IC_{50}$ was 39.26 ${\mu}g$/mL, whereas that of the butanol fraction was 40.95 ${\mu}g$/mL. In testing reduction power at 0.1 mg/mL, the O.D. of the ethyl acetate fraction was highest at 1.404 and showed higher activity than ascorbic acid at 1.332. The results of the nitrite scavenging activity test were very similar to those of the SOD like-activity test. These results suggest that pomegranate endocarp extract may have value as a natural antioxidant.

  • The quality characteristics of Campbell grape juice with different extraction processes (AE; autoclave extraction, HWE: hot water extraction, and EE: enzyme extraction) were investigated. The juice yields of the AE, HWE, and EE juices were 79.8%, 82.3% and 92.6%, respectively. The titratable acidity and soluble solids of the EE juice were significantly higher than those of the other extracts. There was no significant difference in the $L^*$ values of the juices, but the $a^*$ and $b^*$ values of the EE juice were higher than those of the other extracts. The viscosity of the HWE and EE juices was higher than that of the AE juice. The major free sugars in the grape juice were identified as glucose and fructose, and the highest organic acid content was found in the EE juice. The total polyphenol content of the EE juice was 55.7 mg% and was higher than those of the AE and HWE juices, which were 31.3 mg% and 39.5 mg%, respectively. Especially, the anthocyanin content of the EE juice was 22.1 mg%, which was two to four times higher than those of the AE and HWE juices, which were 4.5 mg% and 10.5 mg%, respectively. The EE juice showed the highest antioxidant effect, measured from the DPPH free radical scavenging activity and reducing power. In conclusion, we suggest that the enzyme treatment in the grape extraction was more effective than the autoclave and hot water methods.

  • This study was conducted to investigate the quality characteristics and antioxidant activity of Kochujang following the addition of deodeok (Condonopsis lanceolata) powder. To accomplish this, 1%, 3% and 5% deodeok powder was added to Kochujang and the samples were then incubated at $30^{\circ}C$. during which time the general ingredients, water content, acidity, salinity, reducing sugar content, amino-nitrogen content and DPPH were analyzed at intervals of about two weeks. The water crude protein, crude fat and ash content were 3.61%, 8.44%, 4.62% and 8.85%, respectively. The initial acidity was 3.23~3.97%, and this increased to 4.39~44.86%. highlight please clarify this, I cannot infer your intended meaning; however, this can likely be deleted. The salinity was 7.42~7.73%, and 5% with deodeok powder producing the lowest content and showing a tendency to decrease during the storage period. The early amino-nitrogen content was 274.0~333.1%, with higher nitrogen contents being associated with large amounts of deodeok powder. The amino-nitrogen content during the storage period increased to the 4th week, then decreased. Additionally, the a value decreased sharply during the 4-week storage period. Addition of large amount of deodeok powder significantly improved the free radical scavenging activity. The free radical scavenging activity of 1%, 3% and 5% deodeok powder was higher than that of Kochujang throughout the maturation period. Overall, the results of this study indicate that there is the potential to develop functional foods by the addition of deodeok.

  • Ammonium perchlorate (AP) is primarily derived from the process of liquid incineration treatment when dismantling a solid rocket propellant. A series of batch dead-end nanofiltration (NF) and reverse osmosis (RO) membrane experiments were conducted to explore the retention mechanisms of AP under various hydrodynamic and solution conditions. Low levels of silicate type of siloxane had been detected through the GC/MS and FTIR analysis of liquid solutions extracted from solid ammonium perchlorate composite propellant (APCP). It is indicated that NF/RO membranes fouling in the presence of APCP was mainly attributed to the AP interactions because the concentration of silicate type of siloxane was negligible compared to that of AP. The osmotic pressure of AP was presumably resulted in the flux declines ranging from 13 to 17% in the case of the application of low-pressure (551 and 896 kPa for NF and RO) compared to those in application of high-pressure. The retention of AP by NF/RO membranes significantly varied from approximately 10 to 70% for NF and 26 to 87% for RO, depending on the operating and solution water chemistry conditions. The results suggested that retention efficiency of AP was fairly increased by reducing concentration polarization (i.e. application of low-pressure and stirring speed of 600 rpm) and increasing the pH of a solution. The result of this study was also consistent with the previous modeling of 'solute mass transfer of NF/RO membranes' and demonstrated that hydrodynamic and solution water chemistry conditions are to be a key factor in the retention of AP by NF/RO membranes.

  • This study focused on the physiological change of the shrimp, Litopenaeus vannamei postlarvae 15 stages, under different acclimation methods up to the endpoint of 4 practical salinity unit (psu). Besides using sea water as the control, two acclimation methods, fast acclimation (50% salinity reduction every 8 hours) and slow acclimation (50% salinity reduction every day), were adapted. Results show that the survival rate, glucose and blood uric nitrogen of each group were not significantly different. However, the ion profile differed according to the acclimation methods. Magnesium and sodium of shrimps acclimated to low salinity in both the methods, showed lower concentration than shrimps at 32 psu sea water. Especially, $Na^+$ concentration, which directly influences the osmolality of shrimp, decreased sharply in the fast acclimated group during the first eight hours (from 32 psu to 16 psu). To reduce acclimation stress, it is recommended to take more than eight hours during the first step for reducing the salinity.

  • Objectives: This study evaluated the effects of Guibi-tang (GBT) on neuronal apoptosis and cognitive impairment induced by beta amyloid ($A{\beta}$), (1-42) injection in the hippocampus of ICR mice. Methods: $A{\beta}$ (1-42) was injected unilaterally into the lateral ventricle using a Hamilton syringe and micropump ($2{\mu}g/3{\mu}{\ell}$, $0.6{\mu}{\ell}/min$). Water extract of GBT was administered orally once a day (500 mg/kg) for 3 weeks after the $A{\beta}$ (1-42) injection. Acquisition of learning and retention of memory were tested using the Morris water maze. Neuronal damage and $A{\beta}$ accumulation in the hippocampus was observed using cresyl violet and Congo red staining. The anti-apoptotic effect of GBT was evaluated using TUNEL labeling in the hippocampus. Results: GBT significantly shortened the escape latencies during acquisition training trials. GBT significantly increased the number of target headings to the platform site, the swimming time spent in the target quadrant, and significantly shortened the time for the 1st target heading during the retention test trial. GBT significantly attenuated the reduction in thickness and number of CA1 neurons, and $A{\beta}$ accumulation in the hippocampus produced by $A{\beta}$ (1-42) injection. GBT significantly reduced the number of TUNEL-labeled neurons in the hippocampus. Conclusion: These results suggest that GBT improved cognitive impairment by reducing neuronal apoptosis and $A{\beta}$ accumulation in the hippocampus. GBT may be a beneficial herbal formulation in treating cognitive impairment including Alzheimer's disease.

  • Intensity and stability of the blue color of phospho-molybdate complex in P analysis are known to be influenced by the matrix, reducing regent and acidity of the extractants. Objective of this research was to compare the efficiency of the color-developing reagents for P concentrations in distilled water and extracts of Lancaster, Mehlich II, and Mehlich III methods. Efficiencies on which to base this study were evaluated by the optimum ranges of P, reproducibility of calibration curve and stability of the developed color. Color-developing reagents employed were ammonium molybdate-1,2,4- aminonaphtholsulfonic acid (ANS), ammonium molybdate-ascorbic acid-bismuth subcarbonate (AB), and ammonium molybdate-ascorbic acid-antimony potassium tartarate (AA). The ANS revealed the lowest sensitivity but the widest ranges for P concentrations in color development. On the other hand, the AA bore the narrowest color-developing ranges and its sensitivity was similar to AB. However, at P concentrations lower than $0.5mg\;L^{-1}$, AA was more sensitive than AB. Based on the data on the ranges of calibration curve, stability of color and reproducibility of analytical data. AA reagent was considered to be suitable for the determination of P in distilled water and AB reagent was practically recommendable for soil P analysis in extracts by Lancaster, Mehlich II, and Mehlich III procedures.
