Human and Animal Study on the Natural Food for Obesity and Metabolic Syndrome Risk Factors (비만 및 대사성증후군 위험인자에 대한 천연물 식품의 인체 및 동물 효능연구)

  • In this study a natural composition containing oriental herbs, KSH28, for reducing obesity and metabolic syndrome was constructed and its efficacy was evaluated in animal and human. To investigate the anti-obesity effect of KSH28, animal study was conducted using high fat diet-induced obese mice. KSH28 significantly decreased body weight and adipose tissue in high fat diet-fed obese mice. The mean size of fat cells in adipose tissue was significantly reduced. Glucose and triglyceride levels were also significantly decreased. To elucidate its efficacy in human, a natural food containing KSH28 with grains, vegetables, vitamins, minerals and dietary fibers was constructed and 40 subjects (8 male and 32 female) were tested for the change of body composition, blood pressure and blood lipid profile. All subjects had 2 pack (309 each) of natural food per day for 4 weeks. Compared to the baseline value, body fat was significantly reduced, however, water, protein and mineral contents in the body were not changed, suggesting selective reduction of fat tissue. Blood pressure and serum lipid profile were significantly decreased to reduce risk for metabolic syndrome. Serum GPT, a liver function indicator, was not changed and no significant side effects were detected. Therefore, it was shown that the KSH28 is a safe and effective composition for reducing obesity and metabolic syndrome.

Antioxidant activity of extracts with extraction methods from Phellinus linteus mycelium on Mori ramulus (추출 방법에 따른 상지에 배양한 상황버섯 균사체 추출물의 항산화 활성)

  • This study was conducted to examine the antioxidant activities of extracts derived with different extraction methods (HE, hot water extraction; UE, ultrasonic extraction; and PE, pressured extraction) from Phellinuslinteus mycelium in Mori ramulus. The extraction yield of PE (7.73%) was higher than that of UE (6.03%) and of HE (5.44%). The total phenolic and flavonoid contents of PE were 9.87 g/100 g and 1.90 g/100 g, respectively. The oxygen radical absorbance capacity (ORAC) of PE ($769.63{\mu}M/g$ FW) was higher than that of HE ($622.96{\mu}M/g$ FW) and of UE ($249.06{\mu}M/g$ FW). The DPPH and ABTS radical scavenging activities of PE at $1,000{\mu}g/mL$ were 35.85% and 51.42%, respectively. The superoxide radical scavenging activity of PE, 29.16-93.89%, was higher than that of other extracts. The ferric-reducing antioxidant power and the reducing power of PE were $180.60-607.93{\mu}M$ and 0.14-0.51, respectively. The tyrosinase inhibition activity of PE (9.66-24.05%) improved with an increase in the treatment concentration. The antioxidant and tyrosinase inhibition activities of PE were significantly higher than those of the other extracts. In conclusion, we provided experimental evidence that Phellinus linteus mycelium extracts from Mori ramulus have potential as functional materials.

Antioxidant Properties of Cultured Wild Ginseng Roots Extracts (산삼배양근 추출물의 항산화 특성)

  • We obtained hot-water extracts (HWE) and 70% (v/v) ethanol extracts (EE) from cultured wild ginseng roots (CWGR) and determined the saponin and total polyphenol contents, and antioxidant activities. The yields of freeze-dried powder from the HWE and EE were 27.86% and 18.33% (both w/w), respectively. The total polyphenol content of the EE (22.63 mg/g) was higher than that of the HWE (17.90 mg/g). Ginsenoside-Rb1 and -Rg1 contents of hot-air-dried CWGR were 17.90 mg/g and 22.63 mg/g, respectively. The electron-donating ability of HWE and EE were 2.82-60.58% and 3.88?70.88%, respectively, and the reducing powers ($OD_{700}$) were 0.02-0.17 and 0.07-1.90, respectively, at concentrations of 1-20 mg/mL. Thus, the HWE reducing power was markedly lower than that of the EE, but the SOD-like activity of the EE was significantly higher than that of the HWE. The nitrite-scavenging activities of HWE and EE were 9.25-19.18% and 11.94-53.49%, respectively, at concentrations of 1-20 mg/mL. Additionally, the TBARS (Thiobarbituric acid reactive substances, % value) of the EE (1-20 mg/mL) was 9.18-66.59%, thus 1.9-2.8-fold greater than that of the HWE (4.74-24.88%). In conclusion, we provide experimental evidence that extracts of CWGR may be natural antioxidants.

Physicochemical components of Cynanchi wilfordii Radix fermented with Rhizopus delemar (Rhizopus delemar 발효 백수오의 이화학적 성분 분석)

  • This study carried out to determine the physicochemical components of Cynanchi wilfordii Radix (CWR) fermented with Rhizopus delemar. The physicochemical components such as changes on moisture content, pH, total acidity, reducing sugars, and the contents of free sugars, free amino acid, cynandione A were investigated. The moisture content of fermented CWR was increased after 4 days of fermentation. The pH value of fermented CWR was decreased after 4 days of fermentation. The reducing sugars contents were in the range of 1.42-1.69%. The major free sugars were identified as rhamnose, glucose, fructose and sucrose. The content of free sugars was decreased during fermentation process of CWR while the glucose contents in 80% ethanol and water extracts were increased. In addition, the free amino acid increased significantly during fermentation. Finally, The content of cynandion A of the fermented CWR was highest at the level of 14.09 mg/g on the $8^{th}$ day. These results are indicated that fermentation with Rhizopus strains could be used to increase the bioactivity of CWR and the fermented CWR can be used as a functional material and edible resource in the food industry.

Effects of Regulate in Feed Intakes on Performance and Meat Quality in Old Laying Hens (산란성계에서 사료 급이량 조절이 생산성과 계육품질에 미치는 영향)

  • This study aimed to investigate the effects of reducing feed intake on performance and meat quality in old laying hens. A total of 200 Hy-Line Brown laying hens (100 weeks old) were randomly allotted to five dietary treatments: control (100% daily feed intake), 90%, 60%, 50%, and 20% daily feed intake. Each treatment was replicated four times with 10 birds per replication and two birds per cage. Ten-bird units were arranged according to a randomized block design. The feeding trial lasted for 4 weeks under a 16L:8D lighting regimen. The results indicated that the daily feed intake correlated with hen-day egg production and feed conversion ratios (P<0.05). The carcass yields and partial ratios were also correlated with daily feed intake (P<0.05). The levels of leukocytes (without basophils) were higher in the 50% and 20% daily feed intake groups than in the other groups. The concentrations of dry matter, crude ash, crude fat, and crude protein, water holding capacity, cooking loss, and fatty acids in the breast meat did not decrease as the daily feed intake decreased. In conclusion, reducing daily feed intake decreased laying performance and carcass yield but had no effect on breast meat quality.

Effects of Mole Crickets (Gryllotalpa orientalis) Extracts on Anti-oxidant and Anti-inflammatory Activities. (땅강아지(Gryllotalpa orientalis) 추출물의 항산화 및 항염증 활성)

  • Tremendous natural product extracts were used as a herb medicine remedy or therapy for centuries. Because these extracts have various biological activities, we examined the effects of Gryllotalpa orientalis extract for anti-oxidant and anti-inflammation activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing ability of plasma (FRAP), hydroxy radical scarvenging and cyclooxygenase-2 promoter assays. Gryllotalpa orientalis extracts were prepared from solvents such as distilled water (DW), dimethyl sulfoxide (DMSO), ethanol and methanol. The results showed that Gryllotalpa orientalis extracts have potent DPPH (methanol extract), FRAP (DW extract) and hydroxy radical scavenging (DW and methanol extracts) activity than any other extracts used. A significant inhibition of cyclooxygenase-2 (Cox-2) promoter activity was detected in the presence of DMSO extract or ethanol extract. Collectively, the present results suggested that Gryllotalpa orientalis extract could be used for anti-oxidant and/or anti-inflammation agent for human or agricultural purposes.

Antioxidative Activities of Pine Needles and Quality Characteristics of Korean Wheat Noodle with Pine Needle Powder (솔잎의 항산화활성 및 솔잎분말을 첨가한 우리밀국수의 품질특성)

  • This study examined antioxidative activities and quality characteristics of noodle produced with the pine needle powder as 0.5, 1.0 and 2.0% (v/v to korean wheat powder). DPPH radical scavenging activity, reducing power, antioxidative activities, tyrosinase inhibitory activity of the solvent fraction extracts increased in a dose-dependent manner. Especially, butanol fraction showed high DPPH radical scavenging activity, reducing power, antioxidative activities and tyrosinase inhibitory activity. The weight, volume, water absorption and turbidity of cooked noodles containing pine needle powder increased as pine needle powders were higher. When the amount of pine needle powder increased, the Hunter L (lightness) and a (redness) value of cooked noodles decreased, but b (yellowness) value increased. The texture of noodle containing pine needle powder have decreased than control using only Korean wheat powder in hardness, cohesiveness, springiness, gumminess and chewiness. The results of sensory evaluation of cooked noodles containing pine needle powder indicated that the cooked noodles with 1.0% pine needle powder showed the highest value.

Antioxidant and Acetylcholinesterase Inhibitory Effect of Aged Raw Garlic Extracts (생마늘 추출 숙성물의 항산화 및 아세틸콜린에스터라아제 저해 효과)

  • In vitro antioxidant activities and acetylcholinesterase (AChE) inhibitory effects of solvent fractions from aged raw garlic extracts were investigated. Total phenolics fractioned by hexane, chloroform, ethyl acetate and water from Aged raw garlic extracts were 3.70, 23.63, 31.27 and 2.35 mg/g, respectively. We found that ethyl acetate fractions had the highest in ABTS radical scavenging activities, ferric reducing antioxidant power and inhibitory effect on auto-oxidation of linoleic acid. Intracellular ROS accumulation resulting from $H_2O_2$ treatment of PC12 cells was significantly reduced when ethyl acetate fractions were present in the medium compared to PC12 cells treated with $H_2O_2$ only. In addition, we found that ethyl acetate fractions from aged raw garlic extracts resulted in a dose-dependent manner on AChE inhibition. Consequently, our results suggest that ethyl acetate fractions from aged raw garlic extracts may be useful as decreasing agents of oxidative stress and AChE inhibitors.

Ginsenoside Contents and Antioxidative Activities from Red Ginseng Treated with High Hydrostatic Pressure (초고압 처리한 홍삼의 ginsenoside 함량 및 항산화 활성)

  • This study was carried out to investigate the changes of ginsenoside contents and antioxidative activities from red ginseng after treated with high hydrostatic pressure (RGHHP). Crude saponin content in traditional red ginseng (TRG) and RGHHP were 21.93 and 27.29 mg/g, respectively. The contents of total phenolics, crude saponin and ginsenoside increased after treated with high hydrostatic pressure. TRG and RGHHP showed an increase 25.60% the highest content of Rb1 (14.10 mg/g and 17.71 mg/g). Also, Rg3 content compared with TRG and RGHHP increased 10.46%. The radical scavenging activity of hot water extract from red ginseng against the DPPH and ABTS radicals increased with the increasing amount of extract and RGHHP higher than TRG. The reducing power and ferric reducing antioxidant power (FRAP) assays of the red ginseng were increased in a dose dependent manner. The FRAP of TRG and RGHHP were 0.30 and 0.36 absorbance, respectively at a concentration of 10 mg/mL. The present results suggest that RGHHP would have the protective potential from oxidative stress induced by free radicals.

Antioxidant activities of ethanol extracts from barley sprouts (새싹보리 에탄올 농도별 추출물의 산화방지 활성)

  • Phenolic compounds and antioxidant activities of ethanol extracts from barley sprouts were evaluated in this study. Barley sprouts were extracted using water and ethanol in various concentration (25, 50, and 75%) using reflux extraction methods. Ultra performance liquid chromatography (UPLC) analysis showed that barley sprouts are mainly composed of rutin, gallic acid, ferulic acid, and ${\rho}$-coumaric acid. The 75% ethanol extracts had higher total polyphenol contents ($44.01{\pm}1.32mg/g$) and total flavonoid contents ($102.96{\pm}2.49mg/g$). 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity ($EC_{50}$ value: $1.65{\pm}0.02mg/mL$) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging activity ($EC_{50}$ value: $1.67{\pm}0.02mg/mL$) of the 75% ethanol extracts of barley sprouts were found to be the most effective. The 75% ethanol extracts of barley sprouts exhibited a strong reducing activity and ferric reducing antioxidant activity. As a result, the 75% ethanol extracts of barley sprouts showed stronger antioxidant activity than other extracts.
