Use of Chemical Treatments to Reduce Tannins and Trypsin Inhibitor Contents in Salseed (Shorea robusta) Meal

  • This study investigated the effect of chemical treatments on tannins (condensed and hydrolysable) and on the trypsin inhibitor (TI) activity in salseed meal. Triplicate samples of ground salseed meal (1 kg) were mixed with 820 ml of either distilled water (pH 5.3), 0.67 M acetic acid (pH 2.4), 0.67 M sodium bicarbonate (pH 8.2) or 2% polyvinyl-pyrrolidone (PVP) solution. The material was placed in airtight plastic containers and incubated at $37^{\circ}C$ for 0, 3, 6, 12, 24, 48 and 72 h. Samples of untreated salseed meal which had not been subjected to soaking or incubation were run through the analysis to serve as control. Addition of water, acetic acid, sodium bicarbonate and PVP solutions to salseed meal and subsequent anaerobic incubation at $37^{\circ}C$ significantly reduced chemically detectable tannins. At each incubation time, alkali solution was more effective than its counterparts. The effect of acidic solution on hydrolysable tannin was least among the treatments. All the treatments reduced TI activity of salseed meal. The reduction in TI activity by these treatments was similar and ranged between 80-84%. Treatment time effected a decrease in the contents of antinutritional substances. However, the effect of the treatment with the reagents, even for zero incubation time, was quite pronounced. It may be concluded from the present results that the treatment of salseed meal with sodium bicarbonate (0.67 M) is more effective in reducing hydrolysable and condensed tannin contents than PVP, water and acid solutions. Treatment with sodium bicarbonate solution is more economical and easier to handle than acid and PVP treatments. Incubation of the treated material for 12 h is reasonably effective, economical and safe from any mould growth.

Study on New Extraction Method of Microcystins from Cyanobacteria (남조류로부터 마이크로시스틴을 추출하는 새로운 추출법 연구)

  • A new analytical method of cyanobacterial toxins, i, e, microcysins was deveeloped using supercritical fluid extraction(SFE). The microcystins mcluded in the study are sparsely soluble in neat supercritical fluid CO$_2$ However, the microcystins were successfully extracted with a temary mixture(90% CO$_2$,9.0% methanol 1.0% water) at 40$^{\circ}$C and 250 atm. The SFE method developed in this study has several advantages over solid-phase extraction(SPE) sample preparation for the analysis of microcystins. Sample handling steps are minimized thus reducing possible losses of analytes and saving analysis time. No clean-up steps are employed in this SFE method. Althouhgh many methods have been described for microcystim RR and LR, the method using solid-phase extraction with ODS cartridges is the most commonly used. However, the adsorbing power of ODS caridges for microcystins is weak, so we have attempted to use a more polar CN cartridge, to increase the adsorbing power for microcystins. Lyophilized cells(100mg) were wxtracted with 5% (v/v) acetic acid. The extract was centrifuged and then the supernatant was applied to a CN cartridge. The cartridge which contained microcystins was rinsed with 5 ml of water and 5 ml of 0.5 M acetic acid. followed by 5 ml of 5% acetonitrile in water , and were determined by HPLC. Better recoveries and chromatogram were observed than with ODS cartridge.

Effects of Added Buckwheat(Fagopyrum esculentum) Extract on Starch Hydrolysis In Vitro and Glucose Responses in Healthy Subjects (메밀추출물 첨가가 In Vitro 전분가수분해율 및 정상성인의 혈당반응에 미치는 영향)

  • This study determined the effects of added buckwheat extract on the rate of corn starch hydrolysis in vitro as well as blood glucose responses through its supplementation in healthy subjects. The rate of corn starch hydrolysis in the presence or absence of various buckwheat extracts was determined in an in vitro enzyme/dialysis system for 2 hr. The buckwheat was extracted by water, ethanol(40%, 70%, 100%) and methanol(40%, 70%, 100%), respectively. Twenty percent(w/w) additions of the ethanol, methanol and water buckwheat extract to corn starch solution significantly reduced the starch hydrolysis at every minute for 2 hr(p<0.05). The calculated hydrolysis indices of the buckwheat extracts were in the order of 100% ethanol extract(50), 100% methanol(54), 40% ethanol(58), 40% methanol(62), 70% methanol(64), 70% ethanol(68), water (82). For the blood glucose response study, groups of 12 volunteers were given 50 g of boiled rice with or without buckwheat extract(10% and 20% of starch weight) using the 100%, 70%, and 40% ethanol extracts, respectively. The addition of each buckwheat ethanol extract significantly reduced blood glucose concentrations at three or more points during 2 hr and also reduced the mean peak rise and area under the blood glucose curve(p<0.05). The calculated glycemic index(GI) values for all ethanol buckwheat extract groups were significantly decreased compared to the control(rice). At the concentrations of 20%, the buckwheat 100% ethanol extracts lowered the GI by 68%. The 100% ethanol extract was more effective than the 70% and 40% extracts for reducing GI. Therefore, the 100% ethanol buckwheat extract would be the most therapeutically useful in modifying postprandial hyperglycemia.

Effects of Purifying Rice Paddy in Reducing Nutrient Loadings from Rice Paddy fields area using Free Range Ducks and Rice Bran (정화논에 의한 벼 친환경재배단지 발생 영양염류 저감효과)

  • To manage the effluent nutrients amounts from rice paddy fields using free range ducks or rice bran, we evaluated the effects of a purifying paddy field which is no fertilizer, no pesticides, and dense rice seedling having a concept of constructed wetland. The experimental paddy field was located at downstream in the watershed of ducks using rice culture area in Milyang. The purifying paddy of land design were treated with seeding method, and vegetation type. As land design, direct seeding on plane, and direct seeding on high-ridge field in 2007. Planting rice only, and planting rice with water hyacinth were treated as vegetation type in purifying paddy in 2008. The purifying paddy fields were effective to reduce amount of T-N and T-P contents in effluent to 33.2~45.3%, and 53.1~55.4%, respectively. The direct seeding on high-ridge treatment, having long residence time of effluent water was more effective than plane plot as T-N 0.29 $g^{-1}m^{-2}d^{-1}$, and T-P 0.031 $g^{-1}m^{-2}d^{-1}$. The planting rice with water hyacinth treatment was effective than planting rice only as T-N 0.23 $g^{-1}m^{-2}d^{-1}$, and T-P 0.049 $g^{-1}m^{-2}d^{-1}$. The optimum area of purifying paddy field to treats all effluent were found out 3.2~4.7% of rice culture area using free range ducks, and rice bran at upper stream.

Changes in Physicochemical Properties of Steamed Rice for Soong-Neung during Roasting (숭늉제조용 찐백미의 볶음조건에 따른 이화학적 특성변화)

  • Changes in physicochemical characteristics of steamed rice during roasting were investigated. Steamed rice was roasted at various temperature $(150{\sim}220^{\circ}C)$ for various time $(10{\sim}30min)$. L and b Hunter color values of steamed rice were decreased, but E value was increased by roasting. The water absorption index and the water solubility index were gradually with increasing roasting temperature and time up to $200^{\circ}C$ for 30min, but decreased at $220^{\circ}C$ for 20min. The yield of water extracts from steamed and roasted rice were sharply increased with increasing roasting temperature and time, however, precipitates and turbidity were decreased. The content of reducing sugar was slowly increased up to $200^{\circ}C$ and remarkably increased above it. The contents of amino acids were decreased by roasting. Among them, cystine, lysine and serin were significantly reduced with increasing roasting temperature.

  • An essential measure to prevent healthcare-associated infections (HAI) is to develop a consistent system of surveillance, thereby promoting a reliable situation diagnosis to perform efficient control for the problem. Patient-to-patient transmission of pathogens within the hospital plays a substantial role in the epidemiology of HAIs. Contamination of healthcare environments commonly occurs, including facilities surfaces (e.g., bed rails, bedside tables), drinking water, cooling tower water, endoscopic instruments, food, airborne, endotoxin test, sterile test and medical equipment, with pathogenic organisms. In addition, epidemiological analysis is performed by multi locus sequence tying, pulsed-field gel electrophoresis for active surveillance. Therefore, an environmental surveillance culture test for prevention improves patient safety and blocks infection agents. Effective infection control and increased safety are possible by controlling the national infection control system. In conclusion, this study contributes to an effective infection control system through the standardization of active surveillance culture laboratory and secure expertise as infection control specialist. The primary objective of the standardization is to improve the safety of the nation's healthcare system by reducing the rates of HAIs.

Quality Characteristics of Kwamegi (Semi-dried Coloabis saira) During Cold Air Drying after Washing with Various Washing Solutions (살균소독수로 세척한 후 냉풍건조한 꽁치과메기의 품질특성)

  • In this study, the microbial control effect after treatment of washing solutions (tap water, electrolyzed water, chlorine dioxide solution) and quality changes were investigated when Kwamegi is produced by means of the cold air drying. The initial moisture rate of the sample was 56.62% before drying. At the beginning of drying period, moisture rate sharply fell down but as the experiment progressed, moisture reducing rate was smoothly decreased. The color difference of Kwamegi before drying was 42.40, but it was reduced depending on the increment of drying temperature and time. TBA value of the initial sample was 0.219, rancidity were increased continuously when drying progressed. Total amino acid content was showed the highest value at $25^{\circ}C$ for 36h and the lowest at $40^{\circ}C$ for 12h. From the fatty acids analysis, major fatty acids were consisted of the 14:0, 16:0 and 18:1 (18.1520.96%, 28.0632.51% and 17.0619.81%, respectively). The microbial control effect was biggest when Kwamegi was washed with chlorine dioxide 100ppm for 60s. The microbe of the Kwamegi, Pseudomonas sp. and Pseudomonas putida were identified.

Antimicrobial and Antioxidant activity of the Fruit of Prunus avium L (체리(Prunus avium L.)의 항균 및 항산화 활성)

  • The fruits of sweet cherry are highly appreciated by the consumer due to their precocity and quality, such as their sweetness, color and sourness. In this study, the hot-water extract and its sequential organic solvent fractions were prepared from domestic Napoleon sweet cherry (Prunus avium L.) to investigate antimicrobial and antioxidant activity. The hot-water extract contained about 40% sugars, and the solvent fraction yields for hexane, ethylacetate (EA), butanol, and water residue were 0.01%, 3.45%, 16.30%, and 80.24%, respectively. Contents of total polyphenol and total flavonoid of the fractions were 1.24~5.24%, and 0~3.76%, respectively. Among the fractions, EA fraction showed the highest total polyphenol and total flavonoid concentrations. Evaluation of antimicrobial activity of the extract and fractions revealed that EA fraction and butanol fraction contained strong antibacterial activity against Listeria monocytogenes, Staphylococcus aureus, and Salmonella typhimurium with minimal inhibitory concentration (MIC) of 0.5~1.0 mg/mL. But the extract and fractions tested were not active to Pseudomonas aeruginosa. In a while, only hexane fraction showed anti-Candida activity with 0.5~1.0 mg/mL of MIC. The fraction showed strong activity against different multi-antibiotics resistant strains such as C. albicans CCARM 14020. Antioxidative activity assay showed that EA fraction has a strong DPPH scavenging activity and a reducing power. The $IC_{50}s$ of vitamin E and EA fraction were 15.5 and $195.5\;{\mu}g/mL$, respectively. Our results suggest that the fruit of P. avium L. has high potentials with anti-Candida and antioxidative activity.

  • On-site Algae Collecting System (OACS) is one of urgent countermeasures to clean the raw water when the algae blooms severely. Rapid reaction capability, high efficiency and large capacity are required when applying OACS to large water areas. The total performance of OACS are always determined by unit process named Flotation, Trapping and Collection. The working efficiency and daily treatment quantity of OACS can be increased when it runs automatically. As the rapid development of OACS technology, in the first place, equipment are miniaturized and simple. And in the second place, automation process from Trapping to Collection are advanced. So, They produce results higher working efficiency, smaller residual sludge on treated water, system's advanced environmental friendly features and the increased amount of sludge by Collection process to achieve large capacity. Now OACS has overcome the algae multiplication rate to ensured the amount of removal algae. In another aspect, it is high economically feasible because of reducing operation cost against the large capacity.

A Study on the Removal of Low-concentration Fluoride-ion by Modified Alumina (변형 알루미나를 이용한 저농도 불소이온 제거 연구)

  • The typical treatment method for fluoride polluted water is the flocculation and precipitation method which usually is capable of reducing the fluoride concentration down to the level of about 10 ppm. However, this method is no longer effective for the treatment of contaminated water having less than 10 ppm of fluorides. To remove fluorides in polluted water from the fluoride concentration between 1 to 10 ppm, several adsorbents were prepared mainly based on an activated alumina and the fluoride removal efficiencies of the adsorbents were analyzed. The best fluoride removal efficiency was obtained when the activated alumina treated by sulfuric acid was used as the adsorbent. A proper calcination temperature for the sulfuric acid contained activated alumina was found to be about $500^{\circ}C$. An adsorption isotherm for the adsorbent was also obtained by using Freundlich model. The values of the constants in Freundlich isotherm model were calculated to be K=6.63 and 1/n=0.29 based on the results obtained from the series of batch type adsorption experiments.
