Anti-oxidative Activities of Extracts from Chionanthus retusus leaves, Fruits and FLower (이팝나무 잎과 열매, 꽃 (Chionanthus retusus) 추출물의 항산화 효과)

  • The aim of this research was to investigate the anti-oxidantive activities of extracts from Chionanthus retusus leaves, fruits and flower. The content of phenolics were 20.8 mg/g in water extracts and 32.2 mg/g in 90% ethanol extracts from flower. The DPPH free radical scavenging activity of extracts from Chionanthus retusus was above 70% at phenolic concentration $100{\mu}g/mL$. The ABTS radical decolorization activities of water and ethanol extracts were both above 80% at $100{\mu}g/mL$ phenolic concentration, respectively. The antioxidant protection factor(PF) of water and 80% ethanol extracts from leaves was the highest as 2.27 PF and 1.70 PF at $50{\mu}g/mL$ phenolic concentration. The TBARs inhibition rate of the Chionanthus retusus extracts, was above 70% at $200{\mu}g/mL$ phenolic concentration. The anti-oxidantive activities of extracts from leaves were shown more active than BHT as a positive control except TBARs. These results confirmed that the extracts from Chionanthus retusus leaves, fruits and flower was shown the high anti-oxidant activity. The results can be expected isolated phenolic compounds from Chionanthus retusus to use as functional beauty food resource.

Antioxidant Activities of Extracts from Medicinal Plants (약용식물 추출물의 항산화 활성)

  • In this study, the optimal conditions for phenolic-compound extraction from medicinal plants were found to be 24 h and about 50% ethanol. The electron-donating scavenging activities (DPPH), ABTS radical-cation decolorization (ABTS), antioxidant protection factor (PF), and thiobarbituric acid reaction substance (TBAR) were measured to determine the antioxidant activities of the extracts of Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L. The total phenolic contents of the extracts of medical plants were determined to be 0.45-3.00 mg/g in the water extracts and 0.33-3.15 mg/g in the 50% ethanol extracts. The electron-donating abilities (EDA) of the water and ethanol extracts were both above 85% at the $50{\mu}g/ml$ concentration. The ABTS radical-cation decolorization was above 80% at the $100{\mu}g/ml$ concentration in all the extracts of various medicinal plants. The antioxidant protection factor (PF) of the Melia azedarach L. extracts was found to be $1.65{\pm}0.40$ PF in the water extracts at the $100{\mu}g/ml$ concentration, and was higher than those of the other medicinal-plant extracts. The TBAR inhibition rates of all the medicinal-plant extracts, except Asparagus cochinchinensis Merr., were above 85% at the $100{\mu}g/ml$ concentration. These results confirmed that the various oriental medicinal plants (Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L.) that were included in this study are useful functional-food sources.

Preparation and Characterization of Nanofiltration Membrane for Recycling Alcoholic Organic Solvent (알코올성 유기용매 재활용을 위한 나노여과막의 제조와 특성평가)

  • The organic solvent robust polybenzimidazole (PBI) membranes were prepared as organic solvent nanofiltration (OSN) membrane for the recycling of alcoholic solvents using non-solvent induced phase separation with different dope solution concentration and coagulant composition of water/ethanol mixtures to control the membrane morphology and permeation performance. Investigation on crosslinking of polybenzimidazole indicated that the membrane crosslinked with dibromoxylene (DBX) had enough mechanical strength and solvent resistance to be applied as a OSN membranes. The crosslinked PBI membrane prepared by more than 20wt% dope concentration coagulated in water showed a rejection of > 90% to Congo Red (MW of 696.66 g/mol) while pure ethanol permeances was more than 22.5 LMH/bar at 5 bar. Investigation on coagulant composition indicated that ethanol permeance through crosslinked PBI OSN membrane increased with increasing of ethanol concentration in water/ethanol mixture coagulants.

Evaluation of a Waterless, Scrubless Chlorhexidine Gluconate/Ethanol Surgical Scrub and Povidone-Iodine for Antimicrobial Efficacy (물과 솔 없이 사용하는 외과적 손 소독제 Chlorhexidine/Ethanol 혼합제와 Povidone-iodine의 소독 효과)

  • Purpose: The purpose of this study was to compare 1% chlorhexidine-gluconate/61% ethanol (CHG/Ethanol) emollient and 7.5% povidone-iodine (PVI) scrub for antimicrobial, residual effect, and skin condition. Method: CHG/Ethanol emollient hand hygiene was performed waterless, and brushless by operating doctors and nurses (N=20). PVI hand washing was performed with water and a brush (N=20) for 5 min. The subjects were asked to press their left hand in hand-shaped agar before a surgical scrub, immediately after a surgical scrub and after the operation. The amount of isolated microorganisms were calculated by counting the number of divided areas($1{\times}1cm$, 160 cell) which were culture positive in the hand culture plate. The skin condition was evaluated. Result: The antimicrobial count of CHG/Ethanol emollient and PVI immediately post surgical scrub was 0.0 vs. 4.1 (p>.05), and after the operation was 0.1 vs. 37.8 (p>.05)respectively. The Residual effect of CHG/Ethanol emollient immediately post surgical scrub and after the operation were 0.0 vs. 0.1 (p>.05), and PVI were 4.1 vs. 37.8 (p>.05)respectively. The skin condition and satisfaction of CHG/Ethanol emollient was higher than PVI (p<.05). Conclusion: The antimicrobial effect between CHG/Ethanol emollient and PVI were the same. Considering skin condition, satisfaction and allergic reaction CHG/Ethanol emollient for surgical scrub is recommended in Korea.

Palmul-tang, a Traditional Herbal Formula, Protects against Ethanol-induced Acute Gastric Injury in Rats

  • Objectives: Palmul-tang (hachimotsu-to in Japanese and bawu-tang in Chinese) is a mixture of eight herbs. It is traditionally used for the treatment of anemia, anorexia, general weakness, and female infertility in China, Japan, and Korea. In this study, we investigated the protective effects of Palmul-tang water extract (PTE) against ethanol-induced acute gastric injury in rats. Material and Methods: Acute gastric lesions were induced by intragastric administration of 5mL/kg body weight of absolute ethanol to each rat. Control group rats were given PBS orally and the ethanol group (EtOH group) received absolute ethanol (5mL/kg) by oral gavage. The positive control group and the PTE group were given oral doses of omeprazole (50mg/kg) or PTE (400mg/kg), respectively, 2 h prior to the administration of absolute ethanol. The stomach of each animal was excised and examined for gastric mucosal lesions. To confirm the protective effects of PTE, we evaluated the degree of lipid peroxidation, the level of reduced glutathione (GSH), and the activities of the antioxidant enzymes catalase, glutathione-S-transferase, glutathione peroxidase, and glutathione reductase in the stomach. Results: PTE reduced ethanol-induced hemorrhage and hyperemia in the gastric mucosa. PTE reduced the increase in lipid peroxidation associated with ethanol-induced acute gastric lesions and increased mucosal GSH content and the activities of antioxidant enzymes. Conclusion: These results indicate that PTE protects gastric mucosa against ethanol-induced acute gastric injury by increasing antioxidant status. We suggest that PTE could be developed as an effective drug for the treatment of acute gastric injury.

Effects of Ethanol Extract of Prunus mume on the Antioxidative System and Lipid Peroxidation on Ethanol-Induced Hepatotoxicity in Rat Liver (매실 추출물이 알코올 투여 흰쥐의 항산화계 및 지질과산화에 미치는 영향)

  • To investigate the antioxidative effects of Prunus mume ethanol extract on the ethanol-induced hepatotoxicity in rat liver, Sprague-Dawley rats weighing 120∼160 g were divided into 5 groups; normal group(NOR), Prunus mume ethanol extract 200mg/kg treated group(PME), ethanol(10 mL/kg, 35%) treated group(ETH), Prunus mume ethanol extract 200 mg/kg and ethanol treated group (PML) and Prunus mume ethanol extract 400 mg/kg, and ethanol treated group(PMF), respectively. The antioxidative activity in vitro was reduced in order of EtOAC>n-hexane>water> chloroform fraction. The growth rate and feed efficiency ratio decreased by ethanol administration were gradually increased to the adjacent level of NOR by administering Prunus mume ethanol extract. It was observed that activities of catalase, superoxide dismutase(SOD), xanthine oxidase and glutathione peroxidase(GSH-Px) of liver and alanine aminotransferase(ALT) and asparate aminotransferase(AST) of serum were elevated by ethanol administration. Besides, Prunus mume ethanol extract markedly decreased elevated activites of catalase, GSH-Px, ALT and AST, except in activites of SOD and xanthine oxidase compared to ETH. Also, the depleted content of GSH by ethanol was increased similar to NOR level by administering Prunus mume ethanol extract. These results suggested that Prunus mume ethanol extract has a possible protective effect on the ethanol-induced hepatotoxicity in rat liver.

Component Analysis and Comparison of Biological Activities of Salvia miltiorrhiza Bunge from Different Cultivation Regions (재배지역별 단삼의 성분 분석 및 생리활성 비교)

  • The purpose of this study was to compare the chemical components and biological activities of Salvia miltiorrhiza Bunge (SMB) from different cultivation regions (Gochang, Yeongyang, Jangheung, and China). Proximate compositions and contents of total polyphenols and flavonoids were measured. Depending on the cultivation region, proximate composition of SMB showed significant variations. Total polyphenol and total flavonoid contents were highest in Yeongyang SMB and Gochang SMB, respectively. We prepared hot water and 75% ethanol extracts of SMB from different cultivation regions and also investigated the biological effects of extracts on anti-oxidant, antibacterial, and anti-inflammatory activities. Extracts of Yeongyang SMB were the most effective at scavenging DPPH and ABTS free radicals, and hot water extract showed better scavenging activity than 75% ethanol extract. We observed that extracts of Yeongyang SMB exhibited the best antibacterial activity against Bacillus cereus, and 75% ethanol extract showed better antibacterial activity than water extract. Anti-inflammatory activities of SMB extracts from different cultivation regions were investigated through evaluation of their inhibitory effects on production of nitric oxide (NO) in lipopolysaccharides-induced RAW 264.7 cells. At $50{\mu}g/mL$, extract of China SMB showed the highest NO inhibitory activity of 34% among water extracts, and extract of Gochang SMB showed the highest NO inhibitory activity of 68% among 75% ethanol extracts. Salvianolic acid B level was the highest in extract of Yeongyang SMB with values of 110.59 mg/g for water extract and 111.93 mg/g for 75% ethanol extract.

Studying of the Effects of Gamichungsangboha-tang water-extract and ethanol-extract on $IFN{\gamma}$, IL-4, T-bet, GATA-3, c-maf in Asthma Model (가미청상보하탕(加味淸上補下湯)의 물과 에탄올 추출법에 따른 항(抗)알레르기 효과(效果)에 대(對)한 연구(硏究))

  • Objective: This study aimed to identify the different effects of GMCSBHT water-extract and ethanol-extract on Th1/Th2 differentiation by monitoring Th1/Th2 specific cytokine secretion patterns and the transcriptional activities of T-bet, GATA-3, c-maf, $INF{\gamma}$ and IL-4. Materials and Methods: Spleen cells from eight week-old BALB/c mice were cultured in GMCSBHT extracts containing medium without activation for 24 hours and with activation for 48 hours. CD4+ T cells were isolated and mRNA expression levels of $INF{\gamma}$, IL-4, T-bet, GATA-3, c-maf by RT-PCR and secretion cytokines levels of $IFN{\gamma}$, IL-4 by ELISA were analyzed. Results: GMCSBHT extracts didn't have mitogenic effects on the unstimulated CD4+ T cells. In Th1 skewed condition, GMCSBAHT water extract had no significant effects on mRNA expression levels of $IFN{\gamma}$, T-bet and c-maf, but inhibited mRNA expression levels of IL-4, GATA-3. It showed significantly increased secretion cytokine levels of $IFN{\gamma}$, but had no significant effect on secretion cytokine levels of IL-4. In Th2 skewed condition, GMCSBHT ethanol extract inhibited mRNA expression levels of $INF{\gamma}$, IL-4, GATA-3 and c-maf significantly, but had no significant effects on mRNA expression levels of T-bet. It had no significant effects on secretion cytokine levels of $INF{\gamma}$, but showed remarkable inhibitory effects on secretion cytokine levels of IL-4. Conclusion: Results suggest that on Th1/Th2 deviation, GMCSBHT water extract has both amplifying effects on Th1 differentiation and inhibitory effects on Th2, but GMCSBHT ethanol extract has stronger inhibitory effects on Th2 differentiation than on Th1.

Inhibition of Adipogenesis in 3T3-Ll Adipocytes with Water and Ethanol Extracts of Cudrania tricuspidata Leaves (꾸지뽕잎(Cudrania tricuspidata) 추출물의 3TS-L1 세포 분화 억제)

  • The inhibitory effects of adipogenesis in 3T3-L1 adipocytes with water and ethanol extracts of Curdrania tricuspidata leaves were investigated. The lipid accumulation of ethanol extract from Curdrania tricuspidata leaves at a concentration of 5 mg/mL was reduced by 50% compared with control cells, whereas water extracts reduced by 86%. The survival rate of cell viability test showed 80% in the growth of cells at concentrations of 0.5-5 mg/mL. The contents of DNJ(1-deoxynojirimycin) and rutin were higher in leaves than those of stem. Moreover, DNJ contents were detected in large amounts in ethanol extract ($3921.3{\mu}g/g$) compared with water extract ($2906.3{\mu}g/g$). On the contrary, the rutin contents was detected in large amounts in water extracts compared with ethanol extract. These results indicate that Curdrania tricuspidata leaves extract is potential sources of natural antiobesity agent.

  • Objectives : The cosmeceutical activity of Cuacuta japonica Choisy were investigated for cosmetics industrial application. Methods : Cosmeceutical activities include antioxidant, tyrosinase inhibition effects and nitrite-scavenging ability were examined through the Cuacuta japonica Choisy Results : The result were obtained as follows : The electron donating ability of water and ethanol extract was 69.5%, 78.3% at 1,000ppm. In SOD-like activity, water extract showed low effect, but ethanol extract scored 63.4% at 1,000ppm. In inhibition activity of xanthine oxidase, water extract showed low effect, which is 27.2% at 500ppm, in contrast to ethanol extract high effect: 99.5% at 500ppm, in nitrite-scavenging ability test, 500ppm of C. japonica Choisy water extract showed the effect of 51.3%, while 500ppm of C. japonica Choisy ethanol extract showed the effect of 72.7%. From the test of inhibitory effect of tyrosinase, both water and ethanol extract showed insufficient inhibitory rate. Conclusion : According to all the test results that have been given, it is possible that the extract of C. japonica Choisy can be used as a new natural material of the cosmeceutical industry.
