Background: Panax ginseng root is used in traditional oriental medicine for human health. Its main active components such as saponins and polysaccharides have been widely evaluated for treating diseases, but secondary active components such as oligosaccharides have been rarely studied. This study aimed to assess the impact of water-soluble ginseng oligosaccharides (WGOS), which were isolated from the warm-water extract of Panax ginseng root, on scopolamine-induced cognitive impairment in mice and its antineuroinflammatory mechanisms. Methods: We investigated the impact of WGOS on scopolamine-induced cognitive impairment in mice by using Morris water maze and novel object recognition task. We also analyzed the impact of WGOS on scopolamine-induced inflammatory response (e.g., the hyperexpression of proinflammatory cytokines IL-$1{\beta}$ and IL-6 and astrocyte activation) by quantitative real-time polymerase chain reaction and glial fibrillary acid protein (GFAP) immunohistochemical staining. Results: WGOS pretreatment protected against scopolamine-induced learning and memory deficits in the Morris water maze and in the novel object recognition task. Furthermore, WGOS pretreatment downregulated scopolamine-induced hyperexpression of proinflammatory cytokines interleukin (IL)-$1{\beta}$ and IL-6 mRNA and astrocyte activation in the hippocampus. These results indicate that WGOS can protect against scopolamine-induced alterations in learning and memory and inflammatory response. Conclusion: Our data suggest that WGOS may be beneficial as a medicine or functional food supplement to treat disorders with cognitive deficits and increased inflammation.
During two years, four samples per year were collected in Vietnam from rice bran, cassava residue, brewer's grain, tofu residue, soybean meal, coconut cake, sweet potato vines and water spinach for chemical analysis and assessment of water holding capacity (WHC). The selected feedstuffs represent fibre-rich plant sources and agro-industry co-products commonly used in pig feeding in Vietnam. The content (g/kg DM) of crude protein (CP), ether extract (EE) and non-starch polysaccharides (NSP) varied between feedstuffs and ranged from 21 to 506 for CP, from 14 to 118 for EE and from 197 to 572 for NSP. Cassava residue had a high starch content of 563 g/kg DM, while sweet potato vines, water spinach, coconut cake and soybean meal had a high content of sugars (63-71 g/kg DM). The content of individual neutral sugars varied between feed ingredients, with the highest content of arabinose, galactose and glucose in tofu residue, the highest content of xylose in brewer's grain and the highest content of mannose in coconut cake. The content of uronic acid was high for cassava residue, tofu residue, sweet potato vines and water spinach (57-88 g/kg DM). The content of soluble non-cellulosic polysaccharides (S-NCP) was positively correlated ($r^2$ = 0.82) to the WHC. The content (g/kg DM) of CP, NDF, neutral sugars, total NSP, total NCP, S-NCP and total dietary fibre in tofu residue, water spinach and coconut cake varied (p<0.05) between years. In conclusion, diet formulation to pigs can be improved if the variation in chemical composition of the fibre fraction and in WHC between potential feed ingredients is taken into account.
해양으로부터 polysaccharide를 생산하는 세균을 분리하여 동정한 결과 Zoogloea sp.(KCCM 10036) 로 밝혀졌으며, 이 해양유래 Zoogloea sp.는 셔로 다 른 두종류의 다당류인 수용성다당류(WSP)와 세포 결합다당류(CBP)를 통시에 생산하여 지금까지 보 고된 관련 미생물과는 다른 특이성을 보였다. 이들 중 WSP는 원심분리 시킨 배양액의 상등액으로부터 acetone 침전법에 의해 분리하였고, CBP는 침전물 로부터 acetone 빛 cetylpyridinium chloride 처 리 에 의 해 분리하였다. WSP와 CBP는 모두 단백 질을 함유하였으나, 이 중 CBP가 보다 높은 단백질 함유 율을 나타내었다. WSP를 다량 생산하기 위한 최척 탄소원은 fructose였고, CBP의 경우는 glucose였 다. Polysaccharide 생산촉진인자로 biotin, ampicillin, 그리고 surf actant를 각각 배지에 첨가하였을 경우, 각각의 농도가 $0.2mg/\ell, 0.4mg/\ell,0.6mg/\ell,$에서 생산량이 가장 높은 값을 보였다. 1%의 glucose를 탄소원으로 하여 fermentor 배양하였을 때, $9.1g/\ell$ 의 WSP와 $2.5g/\ell$ 의 CBP가 생산되었다.
Most, if not all, Basidiomycetes mushrooms have biologically active polysaccharides showing potent antitumor activity with immunomodulating properties. These polysaccharides have various chemical compositions and belong primarily to the $\beta$-glucan group. In this study, the crude water-soluble polysaccharide HEF-P, which was obtained from the fruiting body of Hericium erinaceus by hot water extraction and ethanol precipitation, was fractionated by DEAE-cellulose and Sepharose CL-6B column chromatographies. This process resulted in four polysaccharide fractions, named HEF-NP Fr I, HEF-NP Fr II, HEF-AP Fr I, and HEF-AP Fr II. Of these fractions, HEF-AP Fr II was able to upregulate the functional events mediated by activated macrophages, such as production of nitric oxide and expression ofcytokines (IL-1${\beta}$ and TNF-${\alpha}$). The molecular mass of HEF-AP Fr II was estimated by gel filtration to be 13 kDa. Its structural characteristics were investigated by a combination of chemical and instrumental analyses, including methylation, reductive cleavage, acetylation, Fourier transform infrared spectroscopy (FT-IR), and gas chromatography-mass spectrometry (GC-MS). Results indicate that HEF-AP Fr II is a low-molecular-mass polysaccharide with a laminarin-like triple helix conformation of a ${\beta}$-1,3-branched-${\beta}$-1,6-glucan.
The effects of addition of water extract of pine needle(WEPN) on texture and cell wall polysac-charides content of kimchi during fermentation at 1$0^{\circ}C$ were investigated. Textural properties of hardness, gumminess and cohesiveness of kimchi were higher for WEPN-added kimchi than for the control during the entire fermentation periods, while its adhesiveness was lower. Alcohol insoluble substance, among cell wall polysaccharide fractions of kimchi was higher in WEPN-added kimchi than in the control but water soluble materals was high in control during fermentation periods. The separation phenomenon of middle lamella of control kimchi tissue was observed at 14th days of fermentation but WEPN-added kimchi showed at 21th days fermentation. The vasular of kimchi tissue was more destroyed in control than in WEPN-added kimchi.
동양배 '신고'와 '사과배'의 저장기간 중 서로 다른 생리적 특성과 세포벽 특성 변화를 비교 검토하여 과실생리 및 저장기술의 기초자료로 얻고자 본 실험을 실시하였다. 저장 기간 중 '신고'의 품질변화는 중량감소가 큰 반면, '사과배'에서는 급격한 호흡급등현상과 에틸렌 생합성이 많았으며, 전분감소가 컸다. 수용성 펙틴함량은 '신고'가 '사과배'보다 높았으며 CDTA-가용성 펙틴은 두 품종간의 차이가 크지 않았다. $Na_2CO_3$-가용성 펙틴은 '사과배'가 '신고'보다 높았으며 수용성 펙틴과는 달리 저장 기간이 길어질수록 증가하였다. 4% 및 24% KOH 가용성 펙틴은 저장기간에 따른 변화 폭이 크지 않았다. 수용성 펙틴의 분획 결과,'신고'와 '사과배' 저장 중 거대 분자의 구조 변화에서 많은 차이를 보였고, '신고'와 '사과배' 모두 저장 기간이 길어질수록 펙틴의 가수분해 및 측쇄 사슬의 당 분해가 관찰되었다. $Na_2CO_3$-가용성 펙틴의 경우 당 분포는 저장 기간에 관계없이 일정하였지만 펙틴은 분자량 변화가 컸다. 4% 및 24% KOH 가용성 펙틴의 분획 결과 두 품종에 있어 헤미셀룰로스의 구조 변화가 서로 달라, '사과배'의 경우는 4% KOH 가용성 추출물의 분획에서 세포벽을 구성하고 있는 당들의 가수분해가 컸으며 '신고' 보다 '사과배'에서 노화 과정 중 헤미셀룰로스의 변화가 더 심하였다.
민긴뿌리버섯은 송이버섯과에 속하는 버섯으로 예로부터 식용은 물론 항암, 고혈압 및 진균감염증의 치료에 널리 이용해온 식의약용 버섯이다. 민긴뿌리버섯의 자실체로부터 중성염용액, 열수 및 메탄올을 이용하여 조다당류를 추출하여 Sarcoma 180에 접종된 ICR mice에 주사하여 수명연장 및 항암효과를 조사하였다. 세포독성 실험결과, 각각의 세포는 $10{\sim}1000\;{\mu}g/ml$ 추출물 농도에서 70% 내외의 생존율을 보여 세포독성을 나타내지 않았다. 각각의 조다당류가 투여된 실험군이 대조군에 비해 평균수명이 각각 $42.9{\sim}66.7%$ 연장되었다. 중성염용액 추출물은 B 임파구의 alkaline phosphatase 활성을 대조군과 LPS군에 비해 약 $1.4{\sim}3$배 내외의 증가율을 보였다. 총 복강 세포수도 대조군에 비하여 최고 3.5배 정도 증가하였으며, 혈액 중 백혈구의 수도 대조군에 비하여 약 2.5배 증가하였다. 그리고 면역에 관련된 장기인 간, 비장 및 흉선의 체중이 대조군에 비하여 증가된 것을 확인하였다.
표고버섯의 열수추출물과 발암원인 butter yellow를 6주동안 흰주에게 급여한 후 간 및 혈장의 지질성분 분석 및 간기능에 미치는 영향을 검토한 결과는 다음과 같다. Butter yellow 단독 투여군인 BO군에서는 기본식이만을 준 대조군(NO)에 비하여 식욕 감퇴와 체중이 감소한 반면, 표고버섯의 열수추출물(PS) 급여군인 NP군 및 BP군에서는 PS 비급여군인 NO군과 BO군에 비하여 체중 증가량이 현저히 높았다. 간의 총 지질, 총 콜레스테롤, 중성 지질 및 인지질의 함량은 PS투여군이 대조군에 비하여 유의적으로 낮았으나, butter yellow 첨가군(BO)에서는 NO군과 비교하여 인지질은 비슷한 수치였으며, 총 지질 및 총 콜레스테롤은 유의적으로 높았다. 혈장의 중성지질과 총 콜레스테롤 함량 또한 BO군에서 지질성분들의 수치가 유의적으로 높았고 PS의 투여로(BP군) 중성지질과 총 콜레스테롤 농도가 감소되는 효과가 있었다. 이상의 결과를 종합해 볼 때 표고버섯 열수추출 다당류(PS)는 화학적 발암불질인 butter yellow 투여시에 나타나는 지칠대사 이상으로 인한 지질의 상승을 방지하며 이러한 결과는 PS의 지질과 산화에 대한 방어 작용 또는 콜레스테롤 저하작용을 하는 성분에 의한 것으로 추측되며 이러한 혈장과 간의 중성 지질 및 콜레스테를 농도의 상승을 방지하는 작용의 물질은 동맥경화증을 예방할 수 있으며, 이는 지질과 산화로 인한 지단백질의 변화에 대하여 방어작용과 세포, 면역 수준에서의 부수적 효과를 배제할 수는 없는 것으로 사료된다. 또한 PS의 투여로 발암 물질을 투여하지 않았을 때와 유사한 수치를 나타남으로서, 이 분획물들 중에 함유되어 있는 유효 성분이 간장의 해독기구에 영향을 주어 간 기능을 활성화 하거나 또는 지방축적 및 고지질화에 완만한 개선작용을 나타내고 있음을 알 수 있었다.
마치현을 압출성형 및 효소분해 처리할 경우 원료에 비하여 수용성 고분자 다당류 및 아라비노갈락탄 함량이 증가하였다. 마치현 수용성 다당류 중 아라비노스와 갈락토오스의 함량이 원료 마치현보다 1.5배 증가하였으며, 람노스 함량도 2.6배 증가한 유의적인 결과를 보였다. 압출성형 처리효과로 고분자 분획(I)은 Ext I, Ext II 및 Ext III 시료에서 각각 37, 29 및 26% 정도 저분자 분획(II)으로 분자 재배열이 발생함과 동시에 66,000-74,000 Da범위의 분자량을 갖는 다당체로 구조변형 되었다. 특히, 저분자 분획의 분자량과 조성비에 있어서 압출성형 처리한 마치현은 처리하지 않은 원료에 비하여 9-13% 정도 증가하여 유의성이 있었다. 이같은 다당류의 붕괴 및 변형 정도는 압출성형 처리시 투입된 기계적 소모 에너지와 비례적인 상관성을 보였다. 압출성형 처리를 한 수용성 다당류의 경우 압출성형 처리온도 120$^{\circ}C$ 및 140$^{\circ}C$인 경우 자유 라디칼소거활성능이 압출성형 처리하지 않은 원료에 비하여 높게 증가하였다. 상기와 같은 마치현 유래 아라비노갈락탄의 항산화 활성 기능의 결과에 비추어볼 때 보다 폭 넓은 범위의 분자량을 갖는 분획물 제조 및 생리활성 평가실험을 지속적으로 추진한다면 새로운 기능성 식품소재로 활용할 가치가 있다고 기대된다.
장수버섯 자실체로부터 분리된 강력한 항보체 활성 다당체인 FF-AP1을 정제하고 이것의 화학적 특성을 구명하기 위해 본 실험을 수행한 결과는 다음과 같다. ConA-Sepharose 4B affinity chromatography에 의해 FF-AP1은 $FF-AP1{\alpha}$와 $FF-AP1{\beta}$로 더 분리되었으며 이 중 항보체 활성의 주 유효 다당체인 $FF-AP1{\beta}$는 $10\;{\mu}g/ml$ 농도에서도 약 70%의 항보체 활성도를 나타내었다. $FF-AP1{\beta}$는 약 9.6%의 단백질을 함유한 산성다당체이며 glutamic acid와 aspartic acid의 함량이 상대적으로 다른 아미노산들에 비해 높았다. 이것의 분자량은 gel filtration법에 의해 15,000인 것으로 추정되었고 적외선 분광 분석에서 $890cm^{-1}$ 부근의 소peak를 탐지함으로써 ${\beta}$ glucosidic 결합의 존재를 확인하였다. Gas chromatography에 의한 구성당 조성 분석 결과, 주 구성당이 glucose, mannose, 및 galactose인 galacto-manno-glucan이었고, 소량의 rhamnose, xylose, fucose와 uronic acid도 탐지되었다.
