Effect of Chitinase-Producing Paenibacillus illinoisensis KJA-424 on Egg Hatching of Root-Knot Nematode (Meloidogyne incognita)

  • A bacterium having strong chitinolytic activity on $0.2\%$ colloidal chitin-containing agar medium was isolated from coastal soil in Korea. Based on the nucleotide sequence of conserved segment of a 165 rRNA gene, the bacterium was identified as Paenibacillus illinoisensis KJA-424. The population of P. illinoisensis KJA-424 and chitinase activity significantly increased for the first 2 days of incubation. On SDS-PACE analysis with $0.01\%$ glycol chitin, three protein bands (63, 54, and 38 kDa) with chitinolytic activity were detected tooted. The effect of P illinoisensis KJA-424 on the egg hatch of root-knot nematode (Meloidogyne incognita) was investigated. After 7 days of incubation with the chitinase-producing P. illinoisensis KJA-424, none of the eggs hatched, whereas a $39.8\%$ egg hatching rate was observed in the water control. Inverted and scanning electron microscopic observations demonstrated that P. illinoisensis KJA-424 deformed and destroyed the eggshell of M. incognita. In conclusion, chitinase-produced by p. illinoisensis KJA-424 caused the lysis of M. incognita eggshell and resulted in the inhibition of egg hatching in vitro.

Tower-based Flux Measurement Using the Eddy Covariance Method at Ieodo Ocean Research Station (이어도해양과학기지에서의 에디 공분산 방법을 이용한 플럭스 관측)

  • Surface energy and $CO_2$ fluxes have been measured over an ocean at Ieodo Ocean Research Station of KORDI since May 2003. Eddy covariance technique, which is a direct flux measurement, is used to quantitatively understand the interaction between the ocean surface and the atmospheric boundary layer. Although fluxes were continuously measured during the period from May 2003 to February 2004, the quality control of these data yielded <20% of data retrieval. The atmospheric stability did not show any distinct dirunal patterns and remained near-neutral to stable from May to June but mostly unstable during fall and winter in 2003. Sensible heat flux showed a good correlation with the difference between the sea water temperature and the air temperature. The maximum fluxes of sensible heat and latent heat were $120Wm^{-2}$ and $350Wm^{-2}$ respectively, with an averaged Bowen ratio of 0.2. The ocean around the tower absorbed $CO_2$ from the atmosphere and the uptake rates showed seasonal variations. Based our preliminary results, the daytime $CO_2$ flux was steady with an average of $-0.1 mgCO_2m^{-2}s^{-1}$ in summer and increased in winter. The nighttime $CO_2$ uptake was greater and fluctuating, reaching up to $-0.1 mgCO_2m^{-2}s^{-1}$ but these data require further examination due to weak turbulent mixing at nighttime. The magnitude of $CO_2$ flux was positively correlated with the half hourly changes in horizontal mean wind speed. Due to the paucity of quality data, further data collection is needed for more detailed analyses and interpretation.

  • The East gate station area is 205M long and 24m deep which is located 13 meter in front of cultural treasure east gate. The area to be excavated by blasting is composed of granite rocks from 10M depth to 25M. Surface earth extends to up 10M depth. This job site has in involves heavy traffic congestion such as over 10,000 cars passing in rush hour where clossing No 1 lint of subway running 3 minitues head way. This east gate station construction is to be executed for the provent of the setting down of underground level and blasting vibration effects to cultural treasure east gate. Therefore, the caltural treasure committee approved this execution subject to the following condition. 1. Subway gelogical foundation and measured natural frequency 2. Execution of water tight wall 3. Sellection and test of damping material for wall and under rail 4. Measurement of monitoring system during the execution 5. Measurement of histogram system The above two projects was carried out by Dr. Kwang team in KAIST and prof, Han in Hanyang University under accadamic study contract. In the blasting work, for the pourpose of reduced vibration and low explosion velocity such as CCR, Kovex slurry. The 2nd, used electrical caps shall be delay cap and M/S caps in multi delay. The 3rd, drilling pattern is bench cut in open cut and applied control blasting in tunnelling and also shall drill anti-vibration holes as line drilling.

Soluble extract of soybean fermented with Aspergillus oryzae GB107 inhibits fat accumulation in cultured 3T3-L1 adipocytes

  • BACKGROUND/OBJECTIVES: This study was conducted to investigate the effects of fermented soybean (FS) extract on adipocyte differentiation and fat accumulation using cultured 3T3-L1 adipocytes. MATERIALS/METHODS: 3T3-L1 adipocytes were treated with FS and nonfermented soybean (NFS) extract during differentiation for 10 days in vitro. Oil red O staining was performed and glycerol-3-phosphate dehydrogenase (GPDH) activity was measured for analysis of fat accumulation. Expressions of adipogenic genes were measured. RESULTS: Soluble extract of soybean fermented with Aspergillus oryzae GB107 contained higher levels of low-molecular-weight protein than conventional soybean protein did. FS extract ($50{\mu}g/ml$) inhibited adipocyte differentiation and fat accumulation during differentiation of 3T3-L1 preadipocytes for 10 days in vitro. Significantly lower GPDH activity was observed in differentiated adipocytes treated with the FS extract than those treated with NFS extract. Treatment with FS extract resulted in decreased expression levels of leptin, adiponectin, and adipogenin genes, which are associated with adipogenesis. CONCLUSIONS: This report is the first to demonstrate that the water-soluble extract from FS inhibits fat accumulation and lipid storage in 3T3-L1 adipocytes. Thus, the soybean extract fermented with A. oryzae GB107 could be used to control lipid accumulation in adipocytes.

Fluid intake, hydration status and its association with cognitive function among adolescents in Petaling Perdana, Selangor, Malaysia

  • BACKGROUND/OBJECTIVES: A cross-sectional study was undertaken to evaluate fluid intake and hydration status in association with cognitive function among 230 adolescents (10-14 years of age) in Petaling Perdana, Selangor, Malaysia. SUBJECTS/METHODS: Urine color was used to measure hydration status, while fluid intake was assessed using the 15-item beverage intake questionnaire. Cognitive function was assessed using the Wechsler Intelligence Scale for Children, Fourth Edition. RESULTS: More than half of the adolescents were mildly or moderately dehydrated (59.6%) and only one-third (33.0%) were well hydrated. Among the daily fluid types, intakes of soft drinks (r = -0.180; P = 0.006), sweetened tea (r = -0.184; P = 0.005) and total sugar-sweetened beverages (SSBs) (r = -0.199; P = 0.002) were negatively correlated with cognitive function. In terms of hydration status, cognitive function score was significantly higher (F-ratio = 4.102; P = 0.018) among hydrated adolescents (100.38 ± 12.01) than in dehydrated (92.00 ± 13.63) counterparts. Hierarchical multiple linear regression analysis, after adjusting for socio-demographic factors, showed that soft drinks (β = -0.009; P < 0.05) and sweetened tea (β = -0.019; P < 0.05) negatively predicted cognitive function (ΔR2 = 0.044). When further control for sources of fluid, hydration status (β = -2.839; P < 0.05) was shown to negatively predict cognitive function (ΔR2 = 0.021). The above variables contributed 20.1% of the variance in cognitive function. CONCLUSIONS: The results highlight the links between fluid intake (soft drinks, sweetened tea, total SSBs) and hydration status with cognitive function in adolescents. Interventions aimed at decreasing the consumption of SSBs and increasing hydration status through healthy fluid choices, such as water, could improve cognitive performance in adolescents.

Effects of galactagogue herbal tea containing Chrysanthemum indicum as the main component on milk production in postpartum rats (감국을 주성분으로 하는 허브차의 투여가 출산 후 쥐의 유즙 생성 및 관련 호르몬 분비에 미치는 영향)

  • Purpose: Breastfeeding is the optimal method for feeding a newborn. But insufficient breast milk is the major reason why mothers give up breastfeeding. Herbal galactagogues have been used increasingly to treat postpartum hypogalactia. This study examined the effect of an herbal tea containing Chrysanthemum indicum, as the main ingredient, on milk production in lactating rats. Methods: The herbal tea contained C. indicum (27%), Foeniculum vulgare Mill (21%), Pimpinella anisum (18.2%), Carum carvi (16.1%), Urtica dioica (14.7%), and Gardenia jasminoides (3%). Sixteen lactating Sprague-Dawley (SD) rats were divided randomly into 2 groups, the normal control group (NC; n = 8), and the galactagogue herbal tea group (GHT; n = 8) for 7 days. Lactating rats were administered the decoction of an herbal galactagogue mixture by oral gavage or the same amount of distilled water and milk production was assessed by measuring the pups' weights during the suckling period. The blood concentrations of prolactin, cortisol, oxytocin levels and mammary gland tissues were examined to assess the effects of the galactagogue. Results: Milk production was 9.2% higher in the GHT group given the herbal tea than in the NC group and the difference was statistically significant. The cortisol level in the GHT group was 17.2% higher than the NC group. The herbal tea containing C. indicum increased the size of the alveoli epithelium cells and the mammary lobe. Conclusion: The present study revealed the potential of herbal tea containing C. indicum to enhance milk production in postpartum SD rats.

  • Objectives: The aim of this study was to evaluate the effect of three different drinks on the color parameters of four different restorative materials. Materials and Methods: Three different composites (Filtek Ultimate Universal Restorative, Filtek Ultimate Flowable, and Filtek Silorane, 3M ESPE) and a polyacid-modified composite resin material (Dyract XP, Dentsply DeTrey GmbH) were evaluated. Eighty-four disc-shaped specimens of 8 mm in diameter and 2 mm in thickness were prepared (n = 21 each). Color coordinates ($L^*a^*b^*$, ${\Delta}L^*$, ${\Delta}a^*$, ${\Delta}b^*$, and ${\Delta}E^*$) were measured using a $V{\dot{I}}TA$ Easyshade Compact ($V{\dot{I}}TA$ Zahnfabrik) after 24 hr of storage (baseline) and after 30 day of storage in three different beverages of black tea, Coca cola, or water (control) (n = 7). In each beverage, the specimens were stored three times a day, one hr each, for 30 day. The color changes (${\Delta}E$) were calculated and were analyzed by Kruskal-Wallis and Dunn multiple comparison test. Results: The color difference (${\Delta}E^*$) of the resin materials ranged between 1.31 and 15.28 after 30 day of immersion in the staining solutions. Dyract XP in Coca cola ($15.28{\pm}2.61$) and black tea ($12.22{\pm}2.73$) showed the highest mean ${\Delta}E^*$ value after 30 day, followed by Filtek Ultimate Universal Restorative ($5.99{\pm}1.25$) and Filtek Ultimate Flowable ($4.71{\pm}1.40$) in black tea (p < 0.05). Conclusions: The compomers displayed unacceptable color changes at the end of 30 day in all beverages. Among resin composites, the silorane based composite exhibited relatively good color stability than the others. Filtek Ultimate Universal Restorative and Filtek Flowable showed similar color changes in all beverages.

Performance of Humidifying Element Made of Cellulose and PET Composite (셀룰로오스와 PET 복합체로 만들어진 가습소자의 가습성능)

  • To control the indoor humidity, humidifier is installed in an air handling unit. Up to now, foreign product made of glasswool has been widely used as a humidifying element. In this study, a new humidifying element made of cellulose and PET was tested, and the performance was compared with Glasdek element, which is made of glasswool. Mass transfer rates and pressure drops were measured from the element which was installed at the entrance of the suction-type wind tunnel. Results show that, $j_m$ and f factors of the new element are 33%~39% larger and 0%~51% smaller than those of Glasdek. Large water absorption capacity and the smoothness of the new element appear to be responsible for the large $j_m$ and small f factor. The mass transfer effectivenesses ($j_m/f^{1/3}$) of the new element are 36%~63% larger than those of Glasdek.

Bacteriostatic Effect of Condensed Phosphate on the Growth of Bacteria (중합인산염의 항균효과에 관한 연구)

  • Condensed phosphates have been used in various meat products to enhance the water holding capacity, to improve texture and to prevent the development of off flavors and off odors. This study was intended to observe the effects of poly - and pyrophosphate on the growth of sanitary indicative bacteria and food poisoning bacteria. The bacteriostatic effect of poly - and pyrophosphates against Gram positive bacteria was much stronger than that of against Gram negative organisms. The effective inhibitory concentration of sodium polyphosphate on the growth of bacteria was varied by species such as $0.3\%$ to Staphylococcus aureus, $0.9\%$ to Salmonella, and more than $1.0\%$ to Escherichia coli in nutrient broth. When Staphylococcus aureus suspension was treated with $0.5\%$ sodium polyphosphate at $35^{\circ}C$ for 1 hour, the release of UV-absorbing substances from the organism was confirmed. However no significant effect was observed in Escherichia coli under the similar condition. When alaska pollack fillets were dipped in $ 3.5\%$ sodium polyphospahate at $2^{\circ}C$ for 1 min. prior to freezing, the viable cell count and coliform MPN's of the frozen product were decreased with the range of 30 to $50\%$ in comparison with those of control.

Bactericidal Efficacy of a Monopersulfate Compound against Vibrio harveyi and Toxicity to Litopenaeus vannamei (모노퍼설페이트 화합물의 Vibrio harveyi에 대한 살균력 및 횐다리새우(Litopenaeus vannamei)에서의 독성)

  • Vibrio harveyi is one of the most serious pathogens causing vibriosis in larval and grow-out shrimp culture. This study was performed to investigate the bactericidal effect of a monopersulfate compound against V. harveyi and its toxicity in Litopenaeus vannamei. The monopersulfate compound was prepared at 0, 0.15, 0.3, 0.6, 1.2, and 2.4 ppm for the bactericidal efficacy study, and then V. harveyi was added at a rate of 1×106 CFU/mL. Subsequently, five shrimps/dose were added to the water bath. The other groups of shrimp were exposed to monopersulfate, but not to the bacterium. None of the shrimps exposed to any of the monopersulfate treatment doses without bacteria died, and no changes in their movement were detected for 7 days. However, shrimps exposed to bacteria without monopersulfate showed decreased movement and lethargy, but no death. The total number of other bacteria and V. harveyi at the different concentrations decreased significantly by 4-6 log values compared to that in the bacterial control group. The monopersulfate compound completely inhibited V. harveyi growth beginning 1 h after exposure to 2.4 ppm. These results show that the monopersulfate compound is an efficient disinfectant against naturally occurring marine bacteria and V. harveyi without being toxic to shrimp.
