In this work, caffeine and some catechin compounds such as +C, EGC and EGCG were extracted from green tea using quercetin molecular imprinted polymers in solid-phase extraction. For synthesis of MIP, quercetin as the templates, MAA as the monomer, EGDMA as the crosslinker and AIBN as the initiator were used. For extraction of caffeine and catechin compounds from green tea, the solid-phase extractions of a load followed by wash and elution procedures were done with water, methanol and methanol:acetic acid=90:10 (vol.%) as the solvents, respectively. HPLC analysis (C18 column, 5 μm, 250×4.6 mm) with the mobile phase of methanol:water=40:60 (vol.%) at a flow rate of 0.5 ml/min was adopted for the quantitative determination. By solid-phase extraction, the resolutions of caffeine and some catechin compounds from green tea were increased. The quercetin-MIP had higher selectivity to +C compounds.
Antibiotic substances were produced by Bacillus subtilis YB-70, a potential biocontrol agent found to suppress root-rot of eggplant (Solanum melonggena L) caused by Fusarium solani, in a dextrose glutamate medium and isolated by isoelectric precipitation. Partial purification was performed by column chromatography on silica gel with two solvent systems: chloroform-methanol and methanol-chloroform-water as eluting solvents, This active fraction YBS-1 s contained antifungal activity were soluble in ethanol, methanol, and water, but were not soluble in other solvents including acetone, butanol, ethyl ether, dimethylformamide, propanol, and etc. High performance liquid chromatography and thin layer chromatographic separation of YBS-1s showed that they have been composed of three biological active bands that were named YBS-1A, -1B, and -1C. The substances were stable to heat and resistant to protease. YBS-1s were active against a wide range of plant pathogenic fungi but did not inhibit the growth of bacteria and yeasts. They were not only fungicidal but also fungistatic against chlamydospores of F. solani. The $ED_{50}$ values for the chlamydospore germination and the germ-tube growth of F. solani were $O.725\mu\textrm{m}/ml\;and\;O.562\mu\textrm{m}/ml$, respectively. Microscopic observations proved the substances restricted the growth of phytopathogenic fungus F. solani by spore burst followed by dissolving of its germ-tube, and caused abnormal hyphal swelling after application to chlamydospores or growing hyphae. Cultural filtrate of B; subtilis YB-70 also suppressed the development of root-rot of eggplant in pot tests.
A high performance liquid chromatograpic procedure is described for determining ginseng saponins such as ginsenoside-Rb1, -Rb2, -Rc, -Rd, -Re, -Rf, -Rg1, and-Rg2. Ginseng saponins extracted with 90% methanol and water-saturated butanol were compared with pure standard ginsenosides. The resolution of the saponins was satisfactory and detection limit for each saponin was about 5.mu.g. Separation of the saponins was accomplished using a .mu. Bondapak carbohydrate analysis column, mobile phase of acetonitrile-water-butanol (80:20:15) and differential refractive index (RI) detector. The reproducibility and the recovery were also studied. This method was applied for determining the saponin contents of several parts of leaf, fresh ginseng, white ginseng, and red ginseng.

The cytotoxic effects of lipid extracts from soybean products were studied using K562 human leukemia cell, Yac1 mouse leukemia cell and S 180 mouse sarcoma cell. Total lipids from soybean powder, soybean curd residue and doenjang were extracted with chloroform/methanol (2 : 1) and water saturated butanol, consecutively, and fractionated into acetone supernatants (AS fraction) and acetone precipitates (AP fraction) by adding excess acetone. AS fraction of doenjang lipids showed the strongest cytotoxic effects on K562, Yac1 and S180 cancer cells, whereas each lipid fraction of soybean curd residue also showed relatively weak cytotoxic effects on cancer cells but soybean powder did not. AS and AP fractions of doenjang contained more free fatty acids than those of soybean curd residue and soybean. And when lipid fractions were digested with 0.4 N KOH/methanol, doenjang lipid fractions showed to contain some alkali-stable substances which showed positive reaction with ninhydrin solution on silica TLC separation.
Bergenia ciliata (Family: Saxifragaceae) is a folklore remedy for the treatment of various ailments in Asian countries. Bergenin (1) has been isolated as an active constituent in many studies, however, the amount of bergenin has not been determined in all parts of the plant. A simple RP-HPLC method was developed to determine the amount of bergenin in methanol extracts of leaves, rhizomes and roots of the plant. Separation was achieved on an Agilent Eclipse XDB-C18 column maintained at 25 ℃ using isocratic solvent system (water: methanol: acetic acid; 62.5:37:0.5 v/v/v) adjusted at pH 2 0 at a flow rate of 1.0 mL/min. and detected at 275 nm. Correlation coefficient (0.9952) showed linearity of concentration (5-200 ㎍/mL) and response. The values of LOD (0.00947 ㎍/mL) and LOQ (0.02869 ㎍/mL) indicated that method was sensitive. The recovery of bergenin was 99.99-100% indicating accuracy of method. The methanol extract of rhizomes contained higher amount of bergenin (19.4%) than roots (9.2%) and leaves (6.9%). It is concluded that methanol extract of rhizomes is a better source of bergenin than other parts of the plant. The findings are useful for standardization of bergenin containing extracts and herbal preparations.
D,L-tryptophans were separated by using $Chirobiotic^{(R)}$ T HPLC column. Mobile phases were the mixture of methanol and water(70:30, 80:20, 90:10, v/v). Experimental temperatures were adjusted as 25, 40 and $55^{\circ}C$ in order to compare retention times. Difference in D,L-tryptophan retention times was studied in terms of the interaction between stationary phase and tryptophans. Selectivity, resolution and efficiency of column were utilized to find an optimum separation condition. Retention times were shortened by increasing the amount of methanol in mobile phase and the temperature of column. The best selectivity and resolution was obtained with the temperature($25^{\circ}C$) and the ratio of mobilephase(70/30 v/v%).
Objecives : Aucklandiae Radix is a root of Aucklandia lappa which has been widely used for regulating the flow of vital energy, invigorating the spleen, alleviating pain. Aucklandiae Radix contains the costunolide which is the main ingredient. The substitutive Aucklandiae Radix are Inulae helenii Radix, Aristolchiae Radix, Vladimiriae Radix, and Inulae racemosi Radix in Korea and China. This paper is analysised and compared the costunolide and HPLC pattern in Aucklandiae Radix and substitute herbs. Methods : Chromatographic separation performed using C18 column(Luna 5 u, 250 mm ${\times}$ 4.6 mm) with a mixture of methanol and water(65:35)(v/v). The analyses detected at UV(210 nm). Results : Optimal extraction condition of costunolide was 100% methanol for 2hr. Costunolide was detected in Aucklandiae Radix and Vladimiriae Radix, but other herbs were not detected. In Korea herbal market, Aristolchiae Radix merchandise was identified as the imported Inulae helenii Radix. Conclusions : According to above results, this method was useful identified to Aucklandiae Radix and substitutive herbs. In Korea herbal market, Aristolchiae Radix was identified as Inulae helenii Radix.
To graft N-isopropylacrylamide (NIPAAm) onto alginate, varying dosages of ${\gamma}$-rays were irradiated onto alginate films in deionized water and methanol media, which are non-solvents of alginate. We investigated the hydrogels graft ratio, mechanical strength, swelling kinetics and ratio, and behavior with respect to drug release. The graft yield of NIPAAm increased upon increasing the irradiation dose. The use of the aqueous solution increased the graft yield relative to that obtained in methanol. The mechanical strength of the grafted hydrogels increased after grafting with NIPAAm. In a study of the swelling kinetics, we found that all hydrogels reached an equilibrium swollen state within 3 h. The equilibrium swelling ratio of the hydrogels decreased upon increasing the irradiation dose. The swelling ratio of the hydrogels decreased dramatically between 30 and 35$^{\circ}C$ because phase separation of NIPAAm occurred at 32$^{\circ}C$. The swelling process, with respect to the temperature change, was repeatable. An NIPAAm-grafted alginate containing a drug sustained its release rate until 3 h after an initial high drug release caused by a burst effect.
Small angle light scattering (SALS) and field emission scanning electron microscope (FE-SEM) have been used to investigate the light scattering patterns with time evolved during water vapor quenching (relative humidity of 53 (${\pm}3)%$ at $26^{\circ}C$ of polysulfone (PSf)/NMP/Alcohol and chlorinated poly(vinyl chloride) (CPVC)/THF/Alcohol, respective1y. Time dependence of the position of the light scattering maximum was observed at PSf dope solutions, confirming spinodal decomposition (SD), while CPVC dope solutions showed a decreased scattered light intensity with an increased q-value, indicating nucleation & growth (NG). For the each system, domain growth rate in the intermediate and late stage of phase separation decreased with increasing the number of carbon of alcohol used as additive (non-solvent). Also, in the early stage for SD, the scattering intensity with time was in accordance with Cahn's linear theory of spinodal decomposition, regardless of types of non-solvent additive. Also, the size scales obtained by SALS were mutually compared to domain sizes gained by FE-SEM measurement. These observations of scattering pattern were much clearly observed for the 20PSf/70NMP/10n-butanol (w/w%) and agreed with the theoretical predictions for scattering patterns of each stage like the early, the intermediate, and the late stage of SD type phase separation. As the scattering maximum was observed at the larger angles (larger q) in the order of n-butanol > n-propanol > methanol > no alcohol, the pore size of final morphology decreased.
The hydorgen ions in PVA/SSA membranes were substituted with monovalent metal ions, $Li^{+}$, $Na^{+}$, $K^{+}$, divalent metal ion forms, $Mg^{2+}$, $Ca^{2+}$, $Ba^{2+}$, trivalent metal ion forms, $Al^{+}$. The effect of exchange with metal ions was investigated through the swelling measurement and pervaporative experiments for water-ethanol and water-methanol mixtures at various operating conditions. In addition, ESCA analysis was carried out to study the substitution of the metal ions in membranes. The swelling ratio decerased in the sequence of $Li^{+}$, $Na^{+}$, $K^{+}$ and this might be due to the 'salting-out` effect while the swelling ratios for divalnet and trivalent ion-substituted membranes were affected by the combined effect of salting-out, electrostatic crosslinking and extent of metal ion substitution. For the pervaporation performance, PVA/SSA-$H^{+}$membrane showed the lowest flux and highest separation factor for all aqueous ethanol solutions. The typical results of the flux, 59 g/$m^{2}$hr and the separation factor, 44 were obtained at $50^{\circ}C$ for 90% ethanol aqueous mixture. For water-methanol solutions, the PVA/SSA membranes substituted with monovalent PVA/SSA membranes substituted with divalent and tribalent metal ions, both `salting-out` and electrostatic effects affected the pervaporative results.
