상황버섯에 대한 다양한 추출방법별 항산화 활성과 항돌연변이성, 세포독성 등의 생물학적 활성을 S. typhimurium과 인체암세포주들을 이용하여 측정하였다. DPPH free radical scavenging 활성을 통한 항산화 효과를 조사한 결과 열수로 추출한 것보다ethanol로 처리한 것이 높게 나타났으며, ethanol 추출과 열수 후 ethanol 추출 시 각각 17.14, $17.79\;{\mu}g/ml$로 매우 높은 항산화 활성을 나타내었다. S. typhimurium TA97과 TA100을 이용한 Ames test에서는 직접변이원 물질인 MNNG와 4NQO에 대한 돌연변이 억제효과를 조사한 결과는 2가지 변이원 물질에 대한 억제율은 추출물의 농도와 양이 증가함에 따라 비례적으로 증가함을 보여주었고, MNNG에 대해서는 ethanol 추출물이 70.7%로 가장 높은 억제율을 보였고, 열수후 ethanol 추출 시에도 50% 이상의 억제율을 보였다. 4NQO에 대해서는 ethanol 추출물이 62.9%, 열수후 ethanol 추출물이 52.9%로 50% 이상의 억제율을 나타내었다. 간접변이원 물질인 $B({\alpha})P$에 대한 항돌연변이 효과는 TA98과 TA100 두 균주 모두 ethanol 추출물이 가장 높은 억제율을 보였다. 즉 TA98 균주는 ethanol 추출물 61.5%, 열수후 ethanol 추출물이 50% 이상의 항돌연변이 효과를 나타내었으며, TA100 균주에서는 ethanol 추출물과 열수 후 ethanol 추출물이 50% 이상의 억제율을 보였다. 상황버섯 추출물에 의한 암세포 성장 억제효과를 사람의 결장암 세포인 HT-29와 간암세포인 HepG2에 미치는 영향으로 검토한 결과 각 추출물들이 추출농도에 따라 증가함을 보여주었고, ethanol 분획물이 열수 분획물보다 상대적으로 높은 억제율을 보였다. 반면 시료 0.5 mg/ml 투여 시 HT-29 세포에 대해서 ethanol 추출물 59%, 열수 추출물 54%, 열수후 ethanol 추출물 57%로 50% 이상의 억제율을 나타내었고, 특히 ethanol 추출물과 열수 후 ethanol 추출물에서는 0.25 mg/ml 농도에서 50% 이상의 암세포 증식 억제효과를 나타내었다. 그리고 간암세포 HepG2의 경우 ethanol 추출물이 30%로 결장암세포인 HT-29에 비해 비교적 낮은 활성을 나타내었다. Ethanol 추출물과 열수추출물은 다양한 pH 조건에서 아질산염 소거증을 보여주었으며, 대체로 ethanol 추출물이 열수추출물 보다 높은 효과를 보여주었다.
Antioxidant activities of 80% methanol, water, and 70% ethanol extracts of Lentinula edodes GNA01 were compared and estimated. The yield of Lentinula edodes GNA01 was identified to be in the following order: water>70% ethanol>80% methanol, but there was no significant difference between 80% methanol and 70% ethanol extracts. The highest total phenolic content (TPC) and total flavonoid content (TFC) were found in water extract, and TPC of 80% methanol extract was higher than that of 70% ethanol extract and TFC of 70% ethanol extract was higher than that of 80% methanol extract. Water extract exhibited the strongest DPPH, ABTS radicals, and nitrite scavenging activities, $Fe^{2+}$ chelating ability, and FRAP among the three extracts. In addition, antioxidant activity of 80% methanol extract was higher than that of 70% ethanol extract in most of the experiments. As a result, antioxidant activity of Lentinula edodes GNA01 showed a difference according to extraction solvent and concentration; nevertheless, water extract exhibiting high polarity had the strongest antioxidant effect. Consequently, water extract from Lentinula edodes GNA01 is anticipated to be useful for the development of a high value-added functional product.
The antimutagenic and cytotoxic activities of ethanol and water extracts from Rubus coreanum were investigated in this study. Their antimutagenic activities were measured by the Ames test and their cytotoxic activities were evaluated by the growth inhibition of cancer cells via the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay and the sulforhodamine B (SRB) assay. In the results, the inhibition rates of the ethanol and water extracts toward mutagenicity induced by 4-NQO were 95.0% and 93.6% at 5 mg/plate, respectively, while their inhibition rates against mutagenicity induced by sodium azide were 27.2% and 40.8%, respectively. According to MTT assay, the cytotoxicity values of the ethanol extract against Hep3B and HeLa cells were 67.2% and 68.5%, respectively, and the values for the water extract were 65.8% and 66.4%, respectively. In the SRB assay, the ethanol and water extracts inhibited over 60% of cancer cell growth. In conclusion, both the ethanol and water extracts of Rubus coreanum offer potentially good antimutagenic and anticancer effects.
Objectives : In this study, the antioxidant activities of the 80% ethanol and hot water extracts of Euphorbia supina Rafinesque were investigated. Methods : We measured total phenol contents, flavonoid contents, 2,2-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging assay. The production of reactive oxygen species was measured in LPS-induced RAW 264.7 cells using flow cytometry system. Results : The content of phenol in the hot water extract was $65.529{\pm}0.462mg/g$ and $126.932{\pm}1.894mg/g$ in the 80% ethanol extract, and that of flavonoid in the hot water extract was $16.063{\pm}0.471mg/g$ and $29.159{\pm}1.963mg/g$ in the ethanol extract. The 80% ethanol extract also showed higher DPPH and ABTS radical scavenging activities ($90.8{\pm}1.0%$ and $92.5{\pm}0.7%$) than the hot water extract ($81.5{\pm}0.5%$ and $91.5{\pm}0.2%$). The production of reactive oxygen species(ROS) was reduced dose-dependently by 80% ethanol and hot water extract at concentration of 1, 10 and $100{\mu}g/m{\ell}$ of RAW 264.7 cells. Conclusion : According to these results, the 80% ethanol extract of Euphorbia supina Rafinesque has a good anti-oxidative effects than the hot water extract. Thus, the 80% ethanol extract of Euphorbia supina Rafinesque may serve as useful natural antioxidants.
The present study was to investigate the antioxidant activity in ethanol and water extracts of root and stem of Sargassum coreanum. Antioxidant activities were evaluated by total polyphenol contents, DPPH radical scavenging activity, chelating effect, reducing power, and rancimat method. Total polyphenol contents of ethanol and water extracts were 32.79 mg/g and 15.55 mg/g, respectively. Ethanol extract showed higher DPPH radical scavenging activity than water extract and similar activity to BHT. Reducing power of extracts was increased in a concentration-dependent manner and ethanol extract had more reducing power than water extract. Ethanol and water extracts have little chelating effect at all concentrations. Antioxidant index (AI) of ethanol extract measured by Rancimat was higher than that of water extract, but their AI was lower than that of BHT. These results indicate that ethanol extract of S. coreanum root and stem has more potent antioxidant activity than water extract through DPPH radical scavenging and reducing power, and could potentially be used as a good source of natural antioxidants.
분리막 투과증발은 증류에 비하여 낮은 에너지를 요구하기 때문에 물/유기물 혼합물의 분리에 이용될 수 있으며 공비 혼합물이나 이성질체의 분리에 선호되는 방법이다. 제올라이트 분리막은 고분자 분리막에 비하여 열적, 기계적, 화학적 안정성이 우수하며 NaA 제올라이트 분리막은 큰 친수성을 나타내므로 물/유기물 혼합물로부터 효율적인 물 분리에 적합하다. 본 연구에서는 NaA 제올라이트 분리막을 이용한 투과증발 막분리 공정을 이용하여 물/에탄올 혼합물에서 선택적으로 물을 분리하고자 하였다. 에탄올의 공급 농도가 증가함에 따라 총투과플럭스는 감소하였으며 선택도는 증가한 후 높은 에탄올 농도에서 감소하는 것을 관찰할 수 있었다. 실험 온도의 증가에 따라 총투과플럭스는 증가하였으며 선택도 또한 증가하였지만 높은 에탄올 농도에서는 감소하였다. 160시간 이상의 장기간 실험을 통하여 총투과플럭스와 선택도가 안정적으로 일정함을 확인할 수 있었다. 또한, 본 연구를 통하여 얻어진 NaA 제올라이트 분리막은 증류 및 PVA 계열 고분자 분리막보다 우수한 성능을 나타냄을 알 수 있었다.
Alzheimer's disease (AD), most common form of dementia is characterized that memory deficit and loss of cognitive function. This study was evaluated cognitive enhancing effect of water and ethanol extracts of Codonopsis lanceolata and compared using Morris water maze and passive avoidance test. The water and 70% ethanol extracts (100, 300 and 500 mg/kg) were administered to mice. The neuroprotective effect on glutamate-induced cell death in HT22 cells was additionally investigated using MTT assay. Results showed 70% ethanol extract of Codonopsis lanceolata enhanced cognitive function than water extract, as shown by decrease in escape latency time in Morris water maze test. In passive avoidance test, 70% ethanol extract also increased the latency time compared to the water extract. Furthermore, 70% ethanol extract significantly protected neuronal cell against glutamate cytotoxicity and showed higher than neuroprotective effect of water extract. These results indicate that 70% ethanol extract more improve spatial cognitive ability and protected neuronal cells than water extract.
The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. In an effort to develop novel mushroom-derived anti-obesity nutraceuticals, water and ethanol extracts containing the lipaseinhibitory compound from Phellinus linteus were prepared, and their nutritional components were determined. The optimal conditions for the extraction of P. linteus lipase inhibitor involved the treatment of the fruiting bodies with distilled water at $80^{\circ}C$ for 72 hr and 80% ethanol at $100^{\circ}C$ for 60 hr, respectively. The distilled water extract and ethanol extract contained 10.9% and 6.11% of crude protein, and 0.96% and 15.86% of crude fat, respectively. Additionally, the distilled water extract contained a large quantity of minerals, including 239.5 mg of K, 39.3 mg of Mg, and 39.3 mg of Na. The free amino acid content of the distilled water extracts was also higher than that of the ethanol extracts, and in particular, the distilled water extracts contained 5,139 mg of asparagine, 3,891 mg of tryptophan, 2,598 mg of alanine, and 2,066 mg of serine in 100 g of the distilled water extracts. 100 g of the distilled water and ethanol extracts were found to contain 12.31 g and 8.16 g of malic acid, respectively.
It has been well known that mushroom polysaccharide extracts have superoxide and hydroxyl radical scavenging activities. So we studied the efficacy and anti-oxidative effect of mushroom(Auricularia auricula U.) in rats treated with water and ethanol extracts of Auricularia auricula U after inducing dryness of skin by the application of detergent. The results of the research for of the efficacy and anti-oxidative effect Auricularia auricula U. are as follows thats. Weight gain and feed intake in animals treated with water extracts and ethanol extracts are significantly higher(p<0.05) than animal treated with detergent. Animals treated with water extracts of Auricularia auricula U. compared to animals treated with ethanol extracts have the higher HDL-cholesterol levels in the blood and the lower LDL-cholesterol levels in the blood. SOD-like activity of the ethanol extracts of Auricularia auricula U. was $27.39{\pm}1.61%$ at 2000ppm.
