The purpose of this study was to estimate the microbial and physicochemical quality of some raw vegetables and suggest safer methods of sanitization and storage for foodservice operations. Three sanitization methods were utilized during pre-preparation (tap water, chlorine water and electrolyzed water). Leek and Chicory were monitored as ingredient, with different storage temperatures $(3,\;10^{\circ}C)$ and periods (1, 2, 3, 4 and 7 days). The largest reduction in the microbial counts was shown with the electrolyzed water and for the case before immersion in chlorine water, performing a first washing was more effective in reducing the microbial counts than with no washing. The results showed that the storage temperature, pH, moisture content and microbial loads were important factors affecting the quality of vegetables.
Several tests were conducted to determine the optimum operational conditions of soil washing techniques for floe-forming arsenic-contaminated soils, collected from D abandoned Iron-mine in Korea. The optimum cut-off size was 0.15 mm $(sieve\;\#100)$, about $94\%$ of the mass of soils. Both sodium hydroxide and hydrochloric acid were effective to remove arsenic and the optimum mixing ratio (soil [g] : washing solution [mL]) was 1:5 for both washing agents. Arsenic concentrations, determined by KST Methods, for the dried floe solids obtained from flocculation at pH 5-6 were $990\~1,086\;mg/kg$ dry solids, which were higher concentrations than at the other pH values. Therefore, batch tests for sequential washings with or without removing floc were conducted to find the enhancement of washing efficiencies. After removing floe with 0.2 M HCl, sequential washings of 1 M HCl followed by 1 M NaOH showed the best results (15 mg/kg dry soil). The arsenic concentrations of washing effluent from each washing step were about $2\~3\;mg/L$. However, when these acidic and basic effluents were mixed together, arsenic concentration was decreased to be less than $50\;{\mu}g/L$, due to the pH condition of coagulation followed by precipitation for arsenic removal.
This study used gas chromatography combined with the microelectron capture detection method to determine the most effective washing and cooking methods for removing isoprothiolane from rice. The initial isoprothiolane concentrations in brown and polished rice, before washing, were 17.03 mg/kg and 1.67 mg/kg, respectively. Residual concentrations declined with more washing cycles (19.3-59.3% for brown rice; 43.1-66.5% for polished rice); and by increasing the temperature of the washing water from 5℃ to 40℃ (56.6-63.1% for brown rice; 67.1-74.9% for polished rice). Hand-washing samples using gentle stirring or harsh rubbing reduced pesticide concentrations by 63.1-71.6% for brown rice, versus 75.4-87.4% for polished rice. Reduction in isoprothiolane concentrations varied based on the rice cooker type and whether the rice was pre-soaked. Immediate cooking using an electric- or pressure-cooker showed 78.5% and 78.4% reduction in brown rice, compared with 94.0% and 94.0% for polished rice, respectively. Pre-cooking immersion for 30 min showed similar reductions of 83.4% and 83.4% in brown rice, versus 95.8% and 95.8% in polished rice. The results of this study suggest that the most effective method for removing residual isoprothiolane from both brown rice and polished rice was to wash six times (with vigorous rubbing during the 2nd and 3rd washing) in 7-fold water at 40℃, followed by immersion for 30 min before cooking. Regardless of the type of rice cooker, heating is sufficient to remove an average of 83.4% and 95.8% of isoprothiolane from brown rice and polished rice, respectively.
Objectives: This study was performed to investigate hygienic behavior of food handlers and general population focusing on awareness of hand-washing and the microbial load of their hands. Methods: A questionnaire survey and microbiological analysis were carried out for sixty-four people each. Samples for microbiological analysis were collected through the glove-juice method from the hands, and were analyzed for the presence of aerobic plate counts, total coliform, fecal coliform, Escherichia coli, Staphylococcus aureus, and Salmonella spp. according to the Food Code of Korea. Results: In the survey, significant differences between the food handlers and general population (p < 0.01) were found in hand-washing frequency, duration, use of hand-washing agents, washing parts of hands, hand-drying method, and method of turning off water. In eight different situations among the ten particular situations in their daily life, more food handlers responded to wash their hands than general population (p < 0.05). Bacterial load on hands with general population was consistently higher than with food handlers (p < 0.05), however, percentages of positive hands of S. aureus and Salmonella spp. were not. Conclusions: Poor hand hygiene practices were indicated by the positive results for E. coli, S. aureus, and Salmonella spp. on the hands of some respondents in both groups. This study reveals that there is the need for programs or campaigns to increase hand-washing practices of both groups.
The system of washing and shrouding a dead body is a series of procedures for encoffining in funeral rites, which are taken after death indentification by using CHOKWANG and praying for the resurrection of a dead body by calling a soul departed from the body. The washing and shrouding a dead body (hereinafter reffered to as "YEOMSEUP") is the broad concept which contains the procedures of washing, clothing and shrouding the body. The clothing includes washing, putting rices into the mouth. and then dressing while the shrouding includes first wrapping, second-wrapping and encoffining. The clothes for YEOMSEUP (all clothes and articles for funeral rites are designated) will be put in the coffin along with a dead body according to the YEOMSEUP procedures. Dasan, a SILHAK scholar in the late Yi Dynasty, pointed out some problems of the funeral rites observed in those days. At the same time, he found that the problems had arisen from the misinterpretation of the original descriptions in Chinese characters. and had tried to correct them. The books written by Dasan basically emphasized his ideology reshaping the whole procedures by trimming and removing meaningless formalities which are too much luxurious and wasteful and making them as a part of the original principles of the funeral rites. His Intents are clearly shown in his wrings on practical ettiquttes, such as SANGRYESAJON and SANGUIJEOLYO. In its attempt, this study aims at reformulating the DASAN\\`s SANGRYESAJON in terms of YEOMSEUP methods. A lot of virtues of the Dasan, like WOOSU, SIMUI, DAEDAE, MO, SORYUMHYO and DAERYUMHYO could also be found accordingly. The merits and characteristic in funeral rites are an obedience in filial duty, the pursuit of frugality. and the efficiency of practical functions. It could be said that the resurvey of DASAN′s theory on YEOMSEUP procedures is a meaningful work today when the original meaning of funeral rites fades out. Furthermore, discussions of refined burial service and encouraging cremation designed to improve land use, which are widely spread among people, could weaken the basic philosophy of YEOMSEUP and more likely propagate the atmosphere of despising the dignity of human beings.
Background: The main goal of this study was to evaluate the diagnostic efficacy of reverse transcription-nested polymerase chain reaction (RT-nested PCR) in bronchial washing fluid with MAGE A1-6 common primers for the detection of lung cancers invisible by bronchoscopy. Methods: To determine the expression of MAGE A1-6 gene in 189 lung cancers diagnosed by conventional fluoroscopy-guided lung biopsy and 89 cancer-free controls, RT-nested PCR was performed in bronchial washing specimens. We analyzed MAGE A1-6 RT-nested PCR data according to tumor histology, stage, size, and compared them with cytological data. Results: 189 patients (111 cases in adenocarcinoma, 47 cases in squamous cell carcinoma, 22 cases in small cell lung carcinoma, and 9 cases in other cancers) and 89 benign patients were investigated. The expression of MAGE was performed by nested RT-PCR using common MAGE primer. Among 189 cancer patients, the expression rate of MAGE was 49.2%, and the positive predictive value was 89.4%. However, the expression rate of MAGE in patients with benign lesions was 12.4%. In peripheral lung cancer, the positive rate of MAGE expression was 57.4% in squamous cell carcinoma, 44.1% in adenocarcinoma and 59.1% in small cell lung cancer. Whereas the expression rate of bronchial washing cytology in peripheral lung cancer was 9.0% (p=0.011). Conclusion: MAGE RT-PCR in bronchial washing fluid gave us promising data for the detection of peripheral lung cancer. It could be a useful method for selecting diagnostic tools for peripheral lesions.

To investigate the manufacturing methods of surimi-like materials (SLM) from breast muscle of spent hen, the muscles were diced, chopped and washed with distilled water or sodium chloride solution at 0.1, 0.5 and 1% level and then washed with distilled water to extract myofibrillar protein. When used only distilled water to extract myofibrillar protein, washing was repeated 3 times followed by homogenization and centrifugation of breast muscle after each washing (CM; conventional method). Whereas, to extract myofibrillar protein using sodium chloride solution had sufficient to do 2 times washing by distilled water after 1 time washing by sodium chloride followed by homogenization and centrifugation of breast muscle after each washing (NM; new method). The both batter and cooked SLM gel from NM had significantly (p<0.05) lower redness compared with CM. Again, SDS-PAGE with sarcoplasmic protein fractions showed that the bands of phosphorylase had increased staining intensity in NM compared with CM. These results indicated that the brightness was related to sarcoplasmic protein fractions. SDS-PAGE with myofibrillar protein showed that the bands of myosin had increased staining intensity in NM compared with CM. Data implied that myofibrillar protein extraction with sodium chloride solution had the better adaptability for the breast muscle of spent hen then the commonly used distilled water method.

Objectives: This study was designed to evaluate the changes in the particle filtration efficiencies of five cloth masks (4 plate types, 1 cup type) with an increasing number of machine washings and the degree of cloth expansion. Methods: NaCl aerosols were generated using an atomizer and passed through cloth masks in a dynamic aerosol chamber. Particle concentrations were measured both before and after for the cloth masks using an optical particle counter (OPC) in the size range of $0.3{\sim}10{\mu}m$. Results: In the original condition, the filtration efficiencies of the five cloth masks were A: 20.1%, B:30.9%, C: 25.0%, D: 26.5%, and E: 40.9%. As the number of washings increased in the order of one, two, and four times, the filtration efficiencies of cloth mask C increased. The filtration efficiency of A, D, and E increased after the first washing. With the exception of B, the filtration efficiency of cloth masks increased after the second washing and those of all cloth masks increased after the fourth washing. This might be caused by the fibers untangling from the yarn and being freed at one end. In this status, the packing density of the textile will not change, but the distances between fibers will increase, which might bring about an increase in filtration efficiency. When the cloth masks were extended by 10% and 20% in one direction, the filtration efficiencies of cloth masks B, D, and E decreased at 10% extension, and those of all cloth masks decreased at 20% extension. When the cloth masks were expanded by 10% and 20% in two directions, the filtration efficiencies of all cloth masks decreased by at least 34.7% at 10% extension, and by at least 60.9% at 20% extension Conclusions: The filtration efficiency of cloth masks could decrease after one to two machine washings, but will increase after four washings in comparison with their original performances. The filtration efficiency of cloth masks will decrease when they are expanded, such as when stretching over the nose during wearing status.
Objectives: This study was performed to investigate hand washing awareness of females and load of indicator bacteria on their hands. This study focused on the variation according to their age. Methods: A self-administered questionnaire survey and bacterial analysis of indicator bacteria were carried out for 100 Korean women in their age from 20 s to 60 s. Hand samples were collected through a modified glovejuice method. Results: In the survey, significant difference (p < 0.05) was found among the age groups in the use of hand washing agents. The levels of aerobic colony count (ACC) were the highest in both hands among the 20s (p < 0.05). The levels of Escherichia coli were higher in both hands in their 20s and 30s. No significant difference was found in the levels of Staphylococcus aureus and Salmonella spp. However, the positive rates of S. aureus (left hand, 37.5~47.1%; right hand, 58.5~62.5%) and Salmonella spp. (left hand, 25.0~52.9%; right hand, 37.5~64.7%) were higher in the hands of the 20s and 30s, and then showed decreasing trend according to increase of age. The effect of hand washing frequency on the ACC level of hands was significant (p < 0.001). Conclusions: These results indicate that there was no remarkable difference of hand hygiene awareness among female age groups. The detection of S. aureus and Salmonella spp. on the hands of some females in each age group revealed poor hand hygiene practices. The significant effect of hand washing frequency on the ACC level suggests that frequent hand washing is helpful to reduce hand contamination.
Purpose: The purpose of this study was to analyze effects of hand drying methods after hand-washing. Hand drying methods consisted of spontaneous evaporation, paper towel, hand dryer with rubbing, and hand dryer without rubbing. Methods: The research design was a crossover design. The subjects were eighteen university students and randomly assigned in four group. Each group had the schedule of treatment. The schedule tested one drying method per a day and proceeded for 4 days. The data collection was performed from July 27 to July 30, 2010. Data were analyzed by Kruskal-Wallis, Mann-Whitney and Wilcoxon test. Results: There was significant difference between the after CFU between natural dry, paper towel, air dryer with hand rubbing, and air dryer without hand rubbing(p=.006). The after CFUs of paper towel, natural dry and air dryer without hand rubbing groups were statistically less than air dryer with hand rubbing group. Conclusion: It was shown that the paper towel, spontaneous evaporation and without rubbing hand dryer are effective hand drying interventions on reducing CFU on hand. However additional researches with large sample and strict methodology are needed.
