The Actual Management State of Trial Contact Lenses and Lens Care Products in Local Optical Shops (안경원의 시험착용 콘택트렌즈 및 관리용품 관리 실태)

  • Purpose: In the present study, the actual management state of trial contact lenses and lens care products in local optical shops was surveyed and analyzed to reduce the risk of lens complication possibly induced by neglecting lens care. Methods: The feeling of contact lens wearers during the wear of trial contact lenses was surveyed. Futhermore, the actual management state of trial contact lenses such as cosmetic lens and RGP lens and lens care products was also investigated by surveying opticians who trade contact lenses in local optical shops. Results: It was found that consumers trusted the sanitary conditions of the lens since trial cosmetic contact lens and RGP lens were cleaned before and after trails by over 98% of opticians in local optical shops. For trial cosmetic lens, cleaning with normal saline, multipurpose solution for soft lens and combination of saline and multipurpose solution were 38.5%, 40.5% and 21%, respectively, before trials. After trials, cosmetic lenses were cleaned with normal saline, multipurpose solution for soft lens and a combination of saline and multipurpose solution were 13%, 75%, and 12%, respectively. On the other hand, cleaning with normal saline, multipurpose solution for RGP lens and combination of saline and multipurpose solution were 28.5%, 38.5% and 33%, respectively, before trying trial RGP lens. After trials, RGP lenses were cleaned with normal saline, multipurpose solution for RGP lens and a combination of saline and multipurpose solution were 2.5%, 70%, and 27.5%, respectively, indicating that relatively many opticians followed the lens cleaning regimen. In local optical shops, the cleaning trial cosmetic lens was mainly conducted at every 10 days or a month and the washing cycle of cosmetic lens case was in a month or 2~3 months. The cleaning interval of trial RGP lens was primarily in a month or 2~3 months. For those lens cases, more than 75% of opticians washed them with a surfactant and then rinsed with cold water. The storing periods of lens care products were primarily in a week for saline and in a month and 2~3 months indicating that storing period of lens care products was relatively well-kept in local optical shops. Conclusions: It is thought that the concern about any microbial infection is not that high since trial contact lenses and lens care products were generally well-managed by opticians in local optical shops from the results above. However, better public eye health and better public confidence in opticians may be possible if further strengthen in avoidance of lens cleaning with saline, keep of cleaning cycles within 2 weeks and rinsing of lens cases with hot water happens.

The Optimal Condition of Performing MTT Assay for the Determination of Radiation Sensitivity (방사선 감수성 측정법으로서 MTT 법 시행 시의 최적 조건에 대한 연구)

  • Purpose : The measurement of radiation survival using a clonogenic assay, the established standard, can be difficult and time consuming. In this study, We have used the MTT assay, based on the reduction of a tetrazolium salt to a purple formazan precipitate by living cells, as a substitution for clonogenic assay and have examined the optimal condition for performing this assay in determination of radiation sensitivity. Materials and Methods : Four human cancer cell lines - PCI-1, SNU-1066, NCI-H630 and RKO cells have been used. For each cell line, a clonogenic assay and a MTT assay using Premix WST-1 solution, which is one of the tetrazolium salts and does not require washing or solubilization of the precipitate were carried out after irradiation of 0, 2, 4, 6, 8, 10 Gy. For clonogenic assay, cells in $25\;cm^2$ flasks were irradiated after overnight incubation and the resultant colonies containing more than 50 cells were scored after culturing the cells for $10\~14$ days. For MTT assay, the relationship between absorbance and cell number, optimal seeding cell number, and optimal timing of assay was determined. Then, MTT assay was performed when the irradiated cells had regained exponential growth or when the non-irradiated cells had undergone four or more doubling times. Results : There was minimal variation in the values gained from these two methods with the standard deviation generally less than $5\%$, and there were no statistically significant differences between two methods according to t-test in low radiation dose (below 6 Gy). The regression analyses showed high linear correlation with the $R^2$ value of $0.975\~0.992$ between data from the two different methods. The optimal cell numbers for MTT assay were found to be dependent on plating efficiency of used cell line. Less than 300 cells/well were appropriate for cells with high plating efficiency (more than $30\%$). For cells with low plating efficiency (less than $30\%$), 500 cells/well or more were appropriate for assay. The optimal time for MTT assay was after 6 doubling times for the results compatible with those of clonogenic assay, at least after 4 doubling times was required for valid results. In consideration of practical limits of assay (12 days, in this study) cells with doubling time more than 3 days were inappropriate for application. Conclusion : In conclusion, it is found that MTT assay can successfully replace clonogenic assay of tested cancer cell lines after irradiation only if MTT assay was undertaken with optimal assay conditions that included plating efficiency of each cell line and doubling time at least.

Studies on Electrostatic Propensity of Fabrics (직물대전성에 관한 연구)

  • This studies has been carried out how to effect on electrostafic propensity of synthetic fabrics by coating with 0.5% acrylic polymer solution which was previously developed by the author to improve anticrease nature of silk. The work conditions are: (A) Applied synthetic polymer was acrylic polymer 525, developed by the author. (B) Electrostatic voltage for various fabrics were carried out by Korea standard abrasion partner with Korea standard (KS K 0905) cotton, nylon, polyester and the self sample fabric. (C) Applied fabrics for the investigations were carried out by using abrasion partner with Korea standard (KS K 0905) cotton, nylon, polyester and the self sample fabric. (D) Electrostatic propensity investigations were carried out by use of sample as silk, nylon, polyester and acrylic fabrics, seperating before finish or after finish. (E) Washing after the finish or the original fabric was carried out by Korea standard method, KS K 0465. Through the investigations, he happened to find many interesting matters and the obtained results are as followings. 1. Electrostatic voltage for the finished fabrics increased more than their original silk, nylon, acrylic fabrics except polyester fabric. (See Table 5) 2. Electrostatic voltage for the finished polyester against K.S. polyester decreased remarkably than the original fabric test. 3. In spite of no problem on electrostatic propensity of silk, it showed high electrostatic voltage between the same nature fabric abrasion, because silk is very weak against abrasion and because the test method had been developed to be useful for only synthetic fabrics. 4. Electrostatic voltage increased more in case of abrasion between different nature of fabrics than the same nature of fabrics. 5. Electrostatic voltage of each fabric increased by repeat of wash. 6. Many investigation data were followed with Contact Electrification Series Principle, another word, the farther each other located fabric on the series abrasion was, the higher electrostatic voltage. (See Fig. 6) 7. Such investigation gives warning of use on the mix fiber spinning service as far as concern with electrification. 8. It may also call attention for such increase of electrification in case any finishing of silk textile.

Determination of Greening and Shelf Life of Potato Based on Washing and Storage Temperature Conditions (감자의 세척유무 및 저장온도에 따른 녹화 및 상품성 유지 기간 구명)

  • Potato greening determines the shelf life and affects the marketability of this tuber. Various stresses during handling and storage interact to affect the tuber's physiological status and can affect the rate of greening. This study investigated the effects of storage temperature on tuber greening and shelf life in unwashed and washed potatoes of the cultivar Superior. Physiological and biochemical changes were examined during 15 days at room temperature ($23{\pm}2^{\circ}C$) under cool-white fluorescent light after storage for 1 month at different temperatures ($4^{\circ}C$, $20^{\circ}C$). Hunter a values were negative (-) for washed potatoes after 3 days (-0.8) and 15 days (-2.5) at room temperature following 1 month of storage at $4^{\circ}C$ while positive (+) values were observed for unwashed potatoes after 15 days at room temperature. The Hunter ${\Delta}E$ values of washed potatoes previously stored at $4^{\circ}C$ for 1 month increased after 3 days at room temperature compared with those of unwashed potatoes. The total chlorophyll content of washed potatoes was higher than that of unwashed potatoes. The highest correlation was observed between the Hunter ${\Delta}E$ value and Hunter a value (-0.93506), while a positive correlation coefficient (0.89806) was observed between greening criteria and Hunter ${\Delta}E$ value by using colorimetry. We conclude, therefore, that there is a biosynthetic link between temperature-induced chlorophyll accumulation and tuber greening in storage.

Effect of Cryoprotectants on Quality Properties of Chicken Breast Surimi Manufactured by pH Adjustment (냉동변성방지제 조건에 따른 닭가슴살 수리미의 저장특성)

  • This study was conducted to determine the effect of pH adjustment and the addition of cryoprotectants on the storage properties of chicken breast surimi. Surimi as a control was prepared with Alaska pollack by two times of washing treatment and addition of cryoprotectants(4% sugar, 5% sorbitol and 0.3% polyphosphate). Three types of surimi were manufactured by different cryoprotectant conditions(T1: 5% sorbitol+0.3% polyphosphate, T2: 4% sugar+5% sorbitol+0.3% polyphosphate, T3: 2% salt+4% sugar+5% sorbitol+0.3% polyphosphate) and frozen after adjusting the surimi to pH 11.0. The amino acid and saturated fatty acid composition were significantly higher in the control. EAA(essential amino acid), FAA(amino acid in relation to flavor), SAAA(amino acid in relation to saccarinity) and FRAA (fragrant amino acid) were significantly higher in the control. The TBARS(thiobarbituric acid reactive substances) and VBN(volatile basic nitrogen) were increased with storage times. TBARS and VBN values were significantly higher in the control and T3 than the other treatments. The VBN was significantly higher in control and T3 than other surimi samples. Chicken breast surimi adjusted to pH 11.0 had higher in microorganisms than the control, and T1 sample had the highest total plate count. Sensory evaluations were significantly higher in the control and T3 than the other samples. Especially, the overall acceptability of T3 was similar to the control.

Treatment of Garbage Leachate with Two-phase Anaerobic Digestion Coupled with Ultra Filtration (막결합형 2상 혐기성 소화 공정을 이용한 음식물 탈리액 처리)

  • Organic removal efficiency and methane production rate, a feasibility of power generation from biogas, and the optimum conditions for membrane operation were evaluated for the pilot scale (5 tons/day) two-phase anaerobic digestion coupled with ultra filtration (TPADUF) system fed with garbage leachate. The TPADUF system is consisted of a thermophilic acidogenic reactor, a mesophilic methanogenic reactor, and an UF membrane. When garbage leachate with 150 g/L of TCOD was fed to the TPADUF up to organic loading rate (OLR) of 11.1 g COD/L/d, the effluent TCOD was lower than 6 g/L and the average removal efficiencies of TCOD and SCOD were higher than 95%. The methane composition of the gas was 65%, and the methane yield was 39 $m^3/m^3$ garbage leachatefed, 260 $m^3$/tons $COD_{added}$, or 270 $m^3$/tons $COD_{removed}$, even there was some gas leak. The power production per consumed gas was 0.96 kWh/$m^3$ gas or 1.49 kWh/$m^3$ methane. This lower power production efficiency mainly due to the small capacity of gas engine (15 kW class). The membrane was operated at the average flux of 10 L/$m^2$/hr. When the flux decreased, washing with water and chemical (NaOCl) was conducted to restore the flux. In the TPADUF system, optimum pH could be maintained without alkali addition by recycling the membrane concentrate or mixed liquor of the methanogenic digester to the acidogenic reactor. Also, partial production of methane in the acidogenic reactor had a positive effect on lowering the OLR of the methanogenic reactor.

Flexible Bronchoscopic Findings and Therapeutic Effects in Atelectasis of Children (소아 무기폐에서 굴곡성 기관지내시경 소견 및 치료적 유용성)

  • Purpose : We evaluated the clinical manifestations, bronchoscopic findings and therapeutic effects of flexible fiberoptic bronchoscopy in atelectasis of children. Methods : Sixty six children who received bronchoscopy due to persistent atelectasis, acute severe atelectasis and incidental atelectasis on plain chest radiography were studied retrospectively. Results : The most common causative underlying disease was pneumonia(60.4%). Other underlying conditions were pulmonary tuberculosis, chronic lung disease, postoperative state, bronchial asthma and chest trauma. The most common abnormal findings were inflammatory changes such as bronchial stenosis(n=15), mucosal edema and large amount of secretion(n=14), granulation tissue( n=3) and mucus plug(n=3) although 39.4% showed normal airways. Other findings were congenital airway anomalies, endobronchial tuberculosis, extrinsic compression and obstruction by blood clot. In 32 children with pneumonia-associated atelectasis, 43.7% revealed normal airways, and the most common abnormal findings were also inflammatory changes. Eighteen out of 39 patients who received therapeutic intervention such as suctioning of secretion, bronchial washing and intrabronchial administration of N-acetylcysteine($Mucomyst^{(R)}$) had complete or partial resolution of their atelectasis. In 32 patients with pneumonia-associated atelectasis, 56.5% showed improvement by therapeutic intervention. Conclusion : In this study, atelectasis was mainly associated with inflammatory airway diseases such as pneumonia. The most common abnormal bronchoscopic findings were inflammatory changes such as mucosal edema and large amounts of secretion and bronchial stenosis, although about 40% revealed normal airway. Flexible bronchoscopy is helpful for either diagnosis or treatment, especially in pneumonia-associated atelectasis.

Changes in Quality of Carrot During Storage by Hydrocooling (냉수냉각에 의한 당근의 저장중 품질변화)

  • Quality changes in carrot during storage were studied to investigate the efficiency, cooling properties and the washing and storage effects of hydrocooling method. As a result of plotting the nondimensionalized carrot temperature versus cooling time, its cooling rate coefficient was shown $-0.0171min^{-1}{\sim}-0.0121min^{-1}\;(R^{2}=0.99{\sim}0.95)$, and $-0.141min^{-1}{\sim}-0.0038min^{-1}\;(R^{2}=0.98{\sim}0.92)$ in package condition7. Rate of weight loss and change in moisture content of carrot were not significantly different by treatment conditions during storage at $5^{\circ}C$. During storage at $15^{\circ}C$, however, weight loss of hydrocooled carrot was lower than that of non-treated carrot from the 30th to 40th day. Especially, PE was more effective than tray for packaging hydrocooled carrots. Carrot pretreated wish sterilizing agent, packed with PE vinyl film and with residual water removed after hydrocooling showed a lower decaying-rate than any other carrots. Changes in Hunter L and b values of hydrocooled carrot were slower than those of non-treated one. The carotenoids contents of stored carrot $(0.736{\sim}0.780mg%)$ decreased to $9{\sim}43%$ after 40 days of storage at $5^{\circ}C$ and before 20 days of storage at $15^{\circ}C$. It could be presumed that the addition of sterilizing agent reduced the initial level of overall microbial and coliform counts and their growth rate during storage.

  • This study was conducted to observe seasonal and individual changes in semen characteristics and sperm freezability, and sperm penetration into zona-free hamster eggs in Korean native goats. Buck response and change in semen characteristics to electrical stimulations was evaluated for four seasons throughout 2 years and percentage of motile sperm and normal apical ridge acrosome was investigated after equilibration and thawing for 4 seasons with 5 bucks. Sperm penetration rate was evaluated for 4 bucks. 1. Probe insertion at depth of 7cm and repeated stimulation for 3 sec was more effective(P<0.05) in buck response and semen collection than those of other conditions. 2. Semen characteristics from electrojaculation was signficantly(P<0.005) higher in spring and fall for semen volume, in spring and summer for sperm concentration and in fall for sperm motility than those in other seasons, respectively. However, there were no differences in total sperm among seasons. 3. Buck response to electrical stimulation showed significant difference(P<0.05) among individuals in all 3 seasons except winter. Significant individual difference in semen volume was only in spring and summer, but there was no indivudual difference in sperm concentration and total sperm in all season. 4. Washing of semen before freezing treatment was greatly(P<0.05) beneficial to sperm motility after thawing, no matter whether ejaculates exhibit egg yolk coagulation or not. 5. Sperm motility after glycerol equilibration was significantly(P<0.05) low in summer semen and motility after thawing was greatly(P<0.05) higher in winter semen than in other seasons. Freezability of unwashed sperm was significantly difference among bucks, but a yearly freezability of washed sperm after chilling and thawing were no differences among bucks and percentage of normal apical ridge acrosome were not different among seasons and bucks. 6. There was no significant difference in sperm motility after thawing between egg yolk levels in summer, although 20% level gave more higher motility than 5% level. 7. In summer, 3.2% glycerol and 3-h equilibration gave greatest percentage(P<0.05) of sperm motility and normal apical ridge acrosome after thawing. 8. Sperm penetration rate into zona-free hamster eggs was not different between bucks and seasons. Overall, it is concluded that to obtain maximum sperm output and successive semen freezing by electrojaculation method, buck selection with good response in all season could be basically considered and that seasonal effect on sperm freezability was more greater than that of individual bucks.

Change of Particle Size of Magnesium Silicate According to Reaction Conditions and Evaluation of Its Polyol Purification Ability (반응 조건에 따른 규산마그네슘의 입도 변화 및 폴리올 정제 능력평가)

  • The efficiency of the synthetic magnesium silicate used in basic polyols and edible oil purification is evaluated by its purification ability and filtration rate and is affected by the particle size and surface area of magnesium silicate. In this study, it was investigated the change on the particle size of magnesium silicate was influenced by the reaction temperature, injection rate, injection order (Si, Mg) and Mg/Si reaction mole ratio. The synthesized magnesium silicate was compared and analyzed for the synthesis, grinding, and refining processes. In the synthesis process, the reaction temperature and feed rate did not affect the average particle size change of magnesium silicate, while the reaction molar ratio of Mg / Si and the order of injection acted as main factors for the change of average particle size. The average particle size of magnesium silicate increased by 8.7 ㎛ from 54.4 ㎛ to 63.1 ㎛ at Mg injection when Mg molar ratio increased from 0.125 to 0.500, and increased by about 4.8 ㎛ from 47.3 ㎛ to 52.1 ㎛ at Si injection. The average particle size according to the order of injection was 59.1 ㎛ for Mg injection and 48.4 ㎛ for Si injection and the difference was shown 10.7 ㎛, therefore the filtration rate was about 2 times faster under the condition of Mg injection. That is, as the particle size increases, the filtration time is shortened and washing filtration rate can be increased to improve the productivity of magnesium silicate. The cake form of separated magnesium silicate after filtration becomes a solid through drying process and is used as powdery adsorbent through the grinding process. As the physical strength of the dried magnesium silicate increased, the average particle size of the powder increased and it was confirmed that this strength was affected by the reaction molar ratio. As the reaction molar ratio of Mg / Si increased, the physical strength of magnesium silicate decreased and the average particle size after grinding decreased by about 40% compared to the average particle size after synthesis. This reduction of strength resulted in an improvement of the refining ability due to the decrease of the average particle size and the increase of the amount of fine particle after the pulverization, but it resulted in the decrease of the purification filtration rate. While the molar ratio of Mg/Si was increased from 0.125 to 0.5 at Mg injection, the refining ability increased about 1.3 times, but the purification filtration rate decreased about 1.5 times. Therefore, in order to improve the productivity of magnesium silicate, the reaction molar ratio of Mg / Si should be increased, but in order to increase the purification filtration rate of the polyol, the reaction molar ratio should be decreased. In the synthesis parameters of magnesium silicate, the order of injection and the reaction molar ratio of Mg / Si are important factors affecting the changes in average particle size after synthesis and the changes of particle size after grinding due to the changes of compressive strength, therefore the synthetic parameter is an important thing that determines productivity and refining capacity.
