Optimization of Embryo Density and the Volume of Culture Medium for an Improvement of Mouse Parthenogenetic Embryo Development

  • Autocrine or paracrine mediators released by the early embryo are implicated in the support of embryonic development. Their mechanisms and optimal embryo density in the medium, however, are uncertain. This study was conducted to establish the optimal embryo density and culture medium volume in mouse parthenogenetic embryo culture. In experiment 1, culture of parthenogenetirally activated oocytes at a concentration of $2{\~}4$ embryos/${\mu}L$ significantly improved development to the blastoryst stage ($72{\%}{\leq}$) compared with culture at the lower ($0.2{\~}1$e mbryos/${\mu}L,\;0\~37.5\%$) and the higher ($5{\~}6$ embryos/${\mu}L,\;30\~53\%$) concentration for 120 h when the oocytes were cultured in a 5 ${\mu}L$ drop under mineral oil In experiment 2, the embryos cultured at a concentration of $2{\~}4$ embryos/${\mu}L$ in a 10 ${\mu}L$ drop ($81.1{\%}$) showed significantly higher blastocyst rates than those in a 5 ${\mu}L$ drop ($68.5{\%}$). This study optimizes in vitro culture condition by modifying embryo density and the volume of culture medium It may give appropriate level of autocrine and/or paracrine factors to enhance viability and subsequent normal development of mouse parthenogenetic embryos in vitro.

  • The stability of liquid membrane in the extraction process was investigated through the extraction of the Zn component by using W/O/W emulsion type liquid surfactant membrane which was $D_2EHPA-Kerosene-Span$ $80-H_2SO_4$ system. The highest stability for liquid membrane through the Zn extraction process was obtained under the following conditions. That conditions were that span 80 concentration, as surfactant, of 2~3 vol.%;$D_2EHPA$ concentration, as extractant, of 5~7 vol.%;paraffin oil concentration, as membrane strengthening agent, of 10 vol.%;emulsion volume ratio to the external aqueous phase volume of 0.1, and internal aqueous phase volume ratio to the organic phase volume of 1.0.

Effect of Initial Volume of Hard Water and Contact Time on the Reduction of Calcium Ion Concentration using High Voltage Impulse Technique (고전압 임펄스 기술을 활용한 경도 제거에서 경수의 초기부피와 인가시간이 칼슘이온 농도에 미치는 영향)

  • Recently high voltage impulse (HVI) technique has been extensively studied for desalting processes to control the $CaCO_3$ scale formation in industrial water practices such as power plant, boiler, and heat exchange operations. Investigation of the operational parameters for the HVI is important, however, those had not been reported yet. In this study, the effect of initial feed volume and contact time on reduction of calcium ion concentration by the HVI technique was investigated. Initial feed volumes of artificial hard water which contained 100 mg/L of $Ca^{2+}$, were set to 1, 2, and 3 L respectively. After 24hr of HVI contact with 12kV, $Ca^{2+}$ ion was reduced to 50, 29 and 19 % of their initial concentration, indicating that calcium removal increased as initial feed volume decreased. This implies the applied HVI pulse energy per unit mass of calcium is important parameter determining overall desalting efficiency. A series of extended operations of HVI up to 30 days verified the long term stability of the HVI system. The calcium ion declined to 40 mg/L after 2~3 days, and further reduction of calcium was not achieved, indicating that optimum operation time could be 2~3 days under these experimental conditions. Consequently, it was confirmed that the important operational parameter of HVI technique is initial feed volume and contact time as well as the applied voltage that was already proven in the previous study.

  • Numerical study was performed to investigate the convective heat transfer of Al2O3/water nanofluid flowing through the concentric double pipe counterflow heat exchangers. Hot fluid flowing through the inner pipe transfers its heat to cooling fluid flowing in the outer pipe. Effects of important parameters such as hot and cold volume flow rates, fluid type in the outer and inner pipes, and nanoparticles concentration on the heat transfer and flow characteristics are investigated. The results indicated that the heat transfer performance increases with increasing the hot and cold volume flow rates, as well as the particle concentrations. When both outer and inner pipes are nanofluids with 8% nanoparticle volume concentration, nanofluids showed up to 17% better heat transfer rate than basic fluids. Also, the average heat transfer coefficient of the base fluid for annulus-side improved by 31%. Approximately 20% enhancement in the heat exchanger effectiveness can be achieved with the addition of 8% alumina particles in base fluid. But, addition of nanoparticles to the base fluid enhanced friction factor by about 196%.

  • Emulsion Liquid Membrane (ELM) is a prominent technique for the separation of heavy metal ions from wastewater due to the fast extraction and is a single-stage operation of stripping-extraction. The selection of the components (Surfactant and Carrier) of ELM is a very significant step for its preparation. In the ELM technique, the primary water- in-oil (W/O) emulsion is emulsified in water to produce water-in-oil-in-water (W/O/W) emulsion. The water in oil emulsion was prepared by mixing the membrane phase and internal phase. To prepare the membrane phase, the extractant D2EHPA (di-2-ethylhexylphosphoric acid) was used as a mobile carrier, Span-80 as a surfactant, and Paraffin as a diluent. Moreover, the internal (receiving) phase was prepared by dissolving sulphuric acid in water. Di-(2- ethylhexyl) phosphoric acid such as surfactant concentration, carrier concentration, sulphuric acid concentration in the receiving (internal) phase, agitation time (emulsion phase and feed phase), the volume ratio of the membrane phase to the receiving phase, the volume ratio of the external feed phase to the primary water-in-oil emulsion and pH of feed were studied on the percentage extraction of metal ions at 20℃. The results show that it is possible to remove 78% for As(V), 98% for Cd(II), and 99% for Pb(II). Effects of vasopressin administration in the oral cavity on cardiac function and hemodynamics in rats

  • Background: The vasoconstrictive effect of epinephrine in local anesthetics affects the heart, which leads to hesitation among dentists in injecting local anesthetics into patients with cardiovascular disease. Due to its vasoconstrictive effects, the present study investigated the effects of vasopressin administration on cardiac function in rats. Methods: Experiment 1 aimed to determine the vasopressin concentration that could affect cardiac function. An arterial catheter was inserted into the male Wistar rats. Next, 0.03, 0.3, and 3.0 U/mL arginine vasopressin (AVP) (0.03V, 0.3V, and 3.0V) was injected into the tongue, and the blood pressure was measured. The control group received normal saline only. In Experiment 2, following anesthesia infiltration, a pressure-volume catheter was placed in the left ventricle. Baseline values of end-systolic elastance, end-diastolic volume, end-systolic pressure, stroke work, stroke volume, and end-systolic elastance were recorded. Next, normal saline and 3.0V AVP were injected into the tongue to measure their effect on hemodynamic and cardiac function. Results: After 3.0V administration, systolic blood pressures at 10 and 15 min were higher than those of the control group; they increased at 10 min compared with those at baseline. The diastolic blood pressures at 5-15 min were higher than those of the control group; they increased at 5 and 10 min compared with those at baseline. The preload decreased at 5 and 10 min compared to that at baseline. However, the afterload increased from 5 to 15 min compared with that of the control group; it increased at 10 min compared with that at baseline. Stroke volume decreased at 10 and 15 min compared with that of the control group; it decreased from 5 to 15 min compared with that at baseline. Stroke work decreased from 5 to 15 min compared with that of the control group; it decreased from 5 to 15 min compared with that at baseline. Conclusion: Our results showed that 3.0 U/mL concentration of vasopressin resulted in increased blood pressure, decreased stroke volume and stoke work, decreased preload and increased afterload, without any effect on myocardial contractility.

Evaluating Effective Volume and Hydrodynamic Behavior in a Full-Scale Ozone Contactor with CFD Simulation (전산유체역학을 이용한 실규모 오존 접촉에서의 수리거동과 유효 체적 평가에 관한 연구)

  • An Ozone reaction model combined with CFD(Computational Fluid Dynamics) technique was developed in this research, in the simulation of ozonation, hydrodynamic behavior as well as reaction model is important because ozone is supplied to treated water as gas ozone. In order to evaluate hydrodynamic behavior in an ozone contactor, CFD technique was applied. CFD technique elucidated hydrodynamic behavior in the selected ozone contactor, which consisted of three main chambers. Three back-mixing zones were found in the contactor. The higher velocities of water were observed in the second and third compartments than that in the first compartment. The flow of the opposite direction to the main flow was observed near the water surface. Based on the results of CFD simulation, the ozone contactor was divided into small compartments. Mass balance equations were established were established in each compartment with reaction terms. This reaction model was intended to predict dissolved ozone concentration, especially. We concluded that the model could predict favorably the mass balance of ozone, namely absorption efficiency of gaseous ozone, dissolved ozone concentration and ozone consumption. After establishing the model, we discussed the effect of concentration of gaseous ozone at inlet, temperature and organic compounds on dissolved ozone concentration.

  • In this study, we develop a methane sensor module that can detect low concentrations below 5,000 ppm and measure up to the detection limit of 50 ppm with the NDIR method, with a long lifetime and high accuracy. Methane ($CH_4$) is one of a representative greenhouse gas, which is very explosive. Thus, it is important to quickly and accurately measure methane concentration in the air. To adjust the methane sensor for industrial field applications, a NDIR-based small sensor was implemented and characterized, where its volume was $4cm{\times}4cm{\times}2cm$ and its response time ($T_{90}$) was less than 30 sec. These results demonstrate that the proposed sensor is commercially available for low-concentration measurement, low volume, and fast response application, such as IoT sensor nodes and portable devices.

Black Carbon Pollution Level at a Roadside of Seoul in Spring (봄철 서울 도로변의 검댕 오염 특성)

  • To understand particle pollution phenomena caused by vehicles, black carbon (BC) concentrations were continuously monitored using an aethalometer at a roadside in Seoul during the period of May 79 to 25, 2005. The BC concentration was highly fluctuated for a short duration, responding to the traffic situation on the road. The lowest BC concentration was observed between 2 and 5 a.m. The local highest BC concentrations were observed during the periods of both morning and evening rush hours. Change in traffic volume accounts for the trend of hourly averaged BC concentrations from the late evening to the morning. Particularly, the slower increase of BC concentration on Sunday seems to respond directly the lower traffic volume in the morning rush hours. From the comparison with a previous work, it is concluded that the BC concentration around midday hours might be dependent on the distance from a road.

A Study on the Characteristics of Urban Aerosol Concentration in the Size Range of 0.01~$1.0\mu\textrm{m}$ (도시 대기 Aerosol의 입자직경 0.01~$1.0\mu\textrm{m}$ 범위의 농도변화 특성)

  • Urban aerosol concentrations in the size range of $0.01 \sim 1.0 \mum$ have been measured by using an electrical aerosol analyzer from May through October, 1984. The total diurnal variation of the number concentration indicates that a minimum value is observed at 3 hr and a sharp increase is noticed early in the morning with a subsequent slow and continuous increase from around 7 hr until 20 hr. After that it is decreased to reach its minimum by dawn. However, both surface and volume concentrations have shown that their first maxima at 8 hr and their second at about 20 hr simultaneously. It is found that the aerosol number is mainly governed by the particles in the size range of $0.01 \sim 0.1 \mum$, while most volume is in $0.1 \sim 1.0 \mum$ size range. It is known fact that particles of $0.1 \sim 1.0 \mum$ size range affect the visibility reduction in the atmosphere. The monthly variation of aerosol concentration remarks its minimum in summer. The main factors influencing the aerosol concentration are emission of autoexhausts, various processes of production and removal, and meteorological parameters.
