Association between the Polymorphism of Glutathione S-transferase Genes and Autoimmune Diseases in Asian Population: a Meta-analysis (아시아인종에서 자가면역질환과 GST 유전자 다형성의 메타분석)

  • To verify the association between GSTM1/GSTT1 gene polymorphisms and susceptibility to autoimmune diseases in Asian population. 18 published reports cited in EMBASE, Google, KISS, MEDLINE and PubMed up to December 2015 were collected for a meta-analysis. The GSTM1/GSTT1 polymorphism for null and present type were analysed separately. The significant association was found between the GST polymorphism and autoimmune diseases in an overall population (GSTM1, OR=1.334, 95% CI=1.137-1.567, p=0.000; GSTT1, OR=1.212, 95% CI=1.012-1.452, p=0.037). Asian population showed the significant association of GSTM1 in the autoimmune diseases, especially vitiligo and atopic dermatitis but non-significant association of GSTT1 in RA and SLE. The GSTM1 null and the GSTT1 present type showed the association with autoimmune diseases in Asian population. The null type frequency of the combination of GSTM1-GSTT1 polymorphism in autoimmune diseases in Asian population was higher than that of the control group. This result indicated that null type of GSTM1-GSTT1 combination can be a risk factor of autoimmune diseases in Asian population.

Adipose-Derived Stem Cells Improve Efficacy of Melanocyte Transplantation in Animal Skin

  • Vitiligo is a pigmentary disorder induced by a loss of melanocytes. In addition to replacement of pure melanocytes, cocultures of melanocytes with keratinocytes have been used to improve the repigmentation outcome in vitiligo treatment. We previously identified by in vitro studies, that adipose-derived stem cells (ADSCs) could be a potential substitute for keratinocytes in cocultures with melanocytes. In this study, the efficacy of pigmentation including durability of grafted melanocytes and short-term safety was examined in the nude mouse and Sprague-Dawley rat after grafting of primary cultured human melanocytes, with or without different ratios of primary cultured human ADSCs. Simultaneous grafting of melanocytes and ADSCs, which were separately cultured and mixed on grafting at the ratios of 1:1, 1:2, or 1:3, showed better efficacy than that of pure melanocytes. Grafting of melanocytes cocultured with ADSCs resulted in a similar outcome as the grafting of cell mixtures. Skin pigmentation by melanocytes : ADSCs at the ratios of 1:1 and 1:2 was better than at 1:3. No significant difference was observed between the 1-week and 2-week durations in coculturing. Time-course microscopic examination showed that the grafted melanocytes remained a little longer than 6-week post-grafting. No inflammatory cell infiltration was observed in the grafted skin and no melanocytes were detectable in other organs. Collectively, grafting of melanocytes and ADSCs was equally safe and more effective than grafting of melanocytes alone. Despite the absence of significant differences in efficacy between the group of 1:1 and that of 1:2 ratio, 1:2 ratio for 1-week coculturing may be better for clinical use from the cost-benefit viewpoint.

  • INTRODUCTION : Psoralens are planar tricyclic furocoumarins present in numerous plants and fungi found throughout the world.' Naturally occurring and synthesized psoralen derivatives(see Figure 1) are photosensitizers of UVA especially from 320 nm to 400 nm, a range at which cellular nucleic acids and proteins are weakly absorbing if any at all. Because of their skinphotosensitizing properties, these compounds have been used in the photochemotherapy of psoriasis and vitiligo. However, undesirable side effects such as carcinoma development in hairless mice as well as possible liver damage from the use of 8-methoxypsoralen(8-MOP) have been reported. The other photobiological effects include inactivation of DNA viruses, killing and mutagenesis of bacteria, inhibition of tumor transmitting capacity of various cells, and hyperpigmentation on human and guinea pig skin. PUVA(psoralen+UVA) photochemotherapy is in fact thousands of years old, having been used in Egypt and India since B.C. 1200-2000. Photochemotherapy for a common disfiguring disease, vitiligo, was practiced in the ancient world by physicians and herbalists who used boiled extracts of the fruits of certain umbelliferous plants, e.g. Ammi majus Linnaeus in Egypt or the leguminous plants, Psoralea corylifolia L. in India. It was first described by Kuske in 19388 that photosensitization of skin by plants was related to the presence of psoralen. He identified natural psoralens in plants as photosensitizers and isolated bergapten(5methoxypsoralen) from the oil of bergamot.   • In this paper, we introduce a digital cross-polarization and fluorescent color imaging system for quantitative evaluation of skin lesions. We describe the characterization of the imaging systems and the quantitative image analysis methods to show the feasibility for quantitative evaluation of skin lesions. The polarization color image was used to compute erythema and melanin index image which are useful for quantitative evaluation of pigmentation and vascular skin lesions, respectively. The fluorescent color image was used to quantitatively evaluate "sebum" and "vitiligo". In quantitative evaluation of various skin lesions, we confirmed the clinical efficacy of the imaging systems for dermatological applications. Finally, we sure that the imaging systems can be utilized as important assistant tools for the evaluation of skin lesions by providing reproducible quantitative result for widely distributed skin lesions.

  • Candida albcans growth inhibition experiments were carried out to determine the photoxicity of new monofunctional psoralen derivatives, PzPs and HMPzPs, as well as well known 8-MOP an d5-MOP. Although 8-MOP and 5-MOP showed distince photoxicity, PzPs and HMPzPs did not inhibit the growth of Candida albicans strain indicating that PzPs derivatives were not good candidate for the treatment of psoriasis. However, vitili해 could be treated by PzPs derivatives without severe phototosicity because psoralens are known to treat vitiligo by stimulating melanogenesis on skin.

Quantitative Analysis of Psoralen and Angelicin in the Psoraleae Semen and Processed Psoraleae Semen (보골지와 염초보골지의 Psoralen과 Angelicin 함량 분석)

  • Psoralen is used in the treatment of vitiligo and angelicin(isopsoralen) is a photosensitizing agent. A reversed-phase high performance chromatographic method was developed to determine the contents of psoralen and angelicin from Psoraleae Semen and processed Psoraleae Semen. The contents of psoralen and angelicin from Psoraleae Semen showed 0.39% and 0.34% respectively. Processing Psoraleae Semen showed that increased the contents of psoralen (0.52%) and angelicin (0.50%).

  • Freckle is defined that it is one sort of dermatoses which appears in face, neck and the back of the hand et and is due to excess pigmentation. Likewise, Vitiligo and Cloasma belongs to dyspigmentation. Although dermatitis it self is a s1ight illness, the extent of studying dermatitis is quite extensive because it is connected direct1y with the patient's appearance. However there has not been a certain way yet. After taking into consideration of previous orienta1 medical doctor's views about freckle by refering to ten kinds of books inc1uding "Je-Bung-Won-Hu-Ron(諸病源候論)", it obtained some opinions which is about a synthetic views, and the difference in disease to discriminate about several different names, cause of disease, symptom and treatment. The effcect of Gorynju and a Soju extract Psoraleae fructus on Melanin synthesis of B16 melanoma cells (보골지(補骨脂)의 고량주(高梁酒), 소주(燒酒) 추출물이 B16 melanoma 세포주의 멜라닌 합성에 미치는 영향)

  • Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3 ${\mu}g/ml$), in a Soju extract Psoraleae fructus, 1 ${\mu}g/ml$ were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4 ${\mu}g/ml$). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.

  • Vogt-Koyanagi-Harada(VKH) Disease is a bilateral granulomatous uveitis often associated with exudative retinal detachment and extraocular manifestations, such as pleocytosis in the cerebrospinal fluid and, in some case, vitiligo, poliosis, alopecia, and dysacusis. There has been no previous report for the disease in the field of Korean traditional medicine. The case was treated with conventional medical intervention and Korean traditional treatment including acupuncture and herbal medicine. The case was discharged with improvement in fundus photography and optical coherence tomography. The authors report the case to suggest that Korean medical approach combined with conventional treatment can be helpful for the disease.

  • Acquired pigmentary skin diseases such as abnormal melanogenesis, vitiligo, chloasma and inflammatory pigmentation are related to regulate the melanin production, In this study, an ethanol extract of Hovenia dulcis Thunb.(EHD) makedly inhibited melanin biosynthesis and suppressed, the protein expression of tyrosinase, tyrosinase-related protein 1(TRP-1), and tyrosinase-related protein 2(TRP-2) in B16F10 cells. On the other hand, EHD did not inhibit mushroom tyrosinase activity. These results indicate that EHD may contribute to the inhibition of melanin biosynthesis through regulating tyrosinase activity and expression, and serve as a new candidate in the design of new skin-whitening or therapeutic agents.
