Influence of Amino Acid and Vitamin Addition on the Growth and Metabolism of a Hybridoma in Batch Culture (하이브리도마의 회분식배양에서 아미노산과 비타민의 첨가에 따른 세포성장과 대사의 변화)

  • The effects of various step-fortifications of the initial medium with amino acids, glucose, and vitamines on the growth and metabolism of a hybridoma cell line in batch cultures were quantified. Comparisons between the metabolic rates of the various cultivations were made for the exponential growth phase. Fortification of the basal medium resulted in higher cell densities through a prolonged growth phase, but the maximum specific growth rate was not affected. The uptake rate of glutamine increased with the addition of amino acids but did not change upon the addition of glucose or vitamines. The specific glucose consumption decreased slightly with the addition of amino acids but increased production of lactate and {{{{ { NH}`_{4 } ^{ +} }}}}. A reciprocal relationship between the yields of {{{{ { NH}`_{4 } ^{+ } }}}} and lactate indicated a joint regulation of glycolysis and glutaminolysis.

Medium Fortification based on the Analysis of Amino Acids and Wastes in Hybridoma Culture (하이브리도마 배양에서 아미노산과 노폐물의 조성 분석에 기초한 배지의 선택적 강화)

  • The cell growth and amino acid metabolism of a hybridoma cell line in T-flasks, spinner flasks, and a 2L bioreactor were compared. Similar growth and metabolic behaviour were observed for spinner flask and bioreactor cultivations, while those in T-flasks differed significantly. Through a detailed analysis of nutrients and wastes, 7 amino acids were found to be consumed to a much higher extent than the rest of the amino acids. Supplementing the based medium with selected amino acids, glucose, and vitamines increased the cell density by 70%. The addition of vitamines was found to increase the metabolic rates of glucose and lactate.

Contents of Minerals and Vitamines in Soybean Sprouts (콩나물의 무기이온 및 비타민 함량)

  • Soy sprouts cultured in a lab with five soybean cultivars and three commercial soy sprouts purchased in a market were investigated to compare the mineral and vitamine contents. In case of minerals, K, Ca, Mg, and Na were found as major minerals and a small quantity of Fe, Zn, Mn, and Cu was also analyzed in soy sprout. The all minerals were distributed in the cotyledon and the hypocotyl but their concentration orders were different between in cotyledon and in hypocotyl. After boiling, much of minerals eluted in water, the pattern of release of the minerals from the soy sprout to water was similar in all soy sprouts. The vitamin C was concentrated a great amount in hypocotyl than in cotyledon whereas vitamin B was found in the cotyledon generally. And their contents were reduced after boiling owing to heat-mediated degrading. The vitamin C and B contents were different according to cultivars, which shows chemical composition of soybean seed was important to soy sprout characters. The present results provide a basis for attempt to improve seed quality in soybean breeding program for soy sprout.

  • Mohan's aniline procedure for the quantitative determination of niacin in feed additives containing various vitamines and minerals was compared with microbiological procedure utilizing growth rate of Lactobacillus arabinosus 17-5. Microbioassay was more sensitive than chemical procedure to the detection of standard niacin and was applicable to the determination of minute amounts of niacin. while both microbioassay and chemical proce dure were discovered to be unsatisfactory by the recovery test for the determination of niacin in feed additives containing much interfering substances. But the possibility of the determination of niacin in feed additives on chemical prccedure together with microbioassay was proved.

  • This study was to investigate current regulatory status of nutrition labeling in advanced countries, such as US and Japan. In US, the mandatory and voluntary components and the order in which they must appear are total calories, total fat, cholesterol, sodium, potassium, total carbohydrate, protein vitamin and iron. The amount of each nutrient must be reported on the basis of the serving size except vitamines and minerals. In Japan, new regulatation on nutrition labeling was made in 1995. For nutrition labeling on processed food, a standard must be appeared and it is mandatory. The union of Europe and Codex also newly regulated on nutrition labeling. It is time to make new regulation on nutrition labeling for being advanced country.

  • Medium components for lactic acid production were optimized with a strain of Lactobacillus sp., isolated by our Lab. Nitrogen source was the key component and manganese ion was also important for lactic acid production in this strain. Optimal concentration of manganese ion was 0.03 g/l as MnSO$_{4}$ 4 - 5 H$_{2}$O base. Other mineral elements, however, had little effect on it. Among the nitrogen sources we examined, yeast extract showed the highest productivity. Yeast extract, the exellent but very expensive medium component, could be partially replaced by soytone until 60% dry base with higher productivity, or by tryptone enforced with vitamines and nucleic acids. In order to replace yeast extract completely, we examined several inexpensive nitrogen sources and their enzymatic hydrolyzates. The hydrolyzate of vital wheat gluten was the best of them.

  • A ginseng preparation (GP) consisting of ginseng ex., lycium fructus ex., four kinds of vitamines and caffein was evaluated for acute toxicity and general pharmacology. Average lethal doses $(LD_{50})$ of GP in male mice were 2,988mg/kg (i.p.) and more than 3g/kg (p.o.). In dosage of 300 and 900mg/kg (p.o.) showed no analgesic activity in both tests of the writhing method induced by acetic acid and of tail pressure method and no effect on the pentetrazole-induced convulsion. However, it appeared to have a hypothermic action only in dose of 900mg/kg. The duration of hypnosis induced by hexobarbital sodium in mice was shortened by GP, being probably due to caffein. GP Produced no marked contraction of isolated ileum and uterus in high concentrations of up to $1{\pm}10^{-3}g/ml$. These results suggested that GP did not show any considerable central nervous depressant activity and exhibited very weak actue toxicity in mice.

  • It has long teen known in herbal medicine that the mugwort not only exerts a strong hemostatic and parasiticidal actions but also has therapeutic effects for stomachache and diarrhea. In recent pharmaceutical botany the mugwort is known to have antipyretic and astringent actions also. Among the major principles which have been found in the leaves and stems of mugwort are inulin, alkaloids, vitamines, and esestial oil. It is well known that santonin, one of the well known parasiticides, is one of the glucosides extracted from the limited species of mugwort. The present study was undertaken to investigate effects of mugwort(Artemisia asiatica Nakai) on the motility of isolated rabbit duodenum. The results obtained are as follows: At does of 0.2%, 0.5% and 1. 0% AAJ(Artemisia asiatica juice) markedly enhanced contractility of isolated duodenum and tonus of the intestine was also augmented with doses of 0.5% and 1.0%. The augmentative effect of AAJ on intestinal motility was not affected by pretreatment with epinephrine and avil while it was completely abolished by atropine. Therefore it is strongly suggested that augmentative action of AAJ on duodenal motility was exerted solely through muscarinic cholinergic receptors.

  • As a basic study for the application of the spore-tearing lactic acid bacteria to foods, the effects of the sporulating conditions on the growth and sporogenesis were studied were observed. The results obtained are as follow. 1. All carbohydrates added to sporulation media except dextrin decreased the sporulation rate and the thermal resistance of spores. Dextrin stimulated the growth, however, there in no effect on the thermal resistance. 2. As nitrogen source, the protein hydrolysates such as peptone, casamino acid were effective to obtain were spores of the increased thermal resistance. 3. Ca$\^$++/, Mn$\^$++/ of the metal ions added to casamino acid containing medium validly increased the total growth, sporulation rate and thermal resistance. Its optimum concentration was 40 ppm each. 4. Biotin of vitamines had an effect on the total growth, sporulation and thermal resistance of spores. Its optimum concentration was 30${\gamma}$/ml. 5. The resistant spores required the adequate maturation period, more than 36 hours, sufficient aeration. and optimum temperature, 37∼45$^{\circ}C$.

In vitro infection of Cryptosporidium parvum to four different cell lines

  • To determine a suitable condition for in vitro infection model of cryptosporidium parvum, four different cell lines, AGS, MDCK, HCT-8 and Caco-2, were used as host cell lines which were cultured at various concentrations of added supplements. These supplement include fetal bovine serum (FBS), sodium choleate, ascorbic acid, folic acid, calcium pantothenate, para-aminobenzoic acid and pyruvate and their effects on the cell lines which were infected with C. parvum were evaluated. The results of this study showed that the AGS cell line was most susceptible to C. parvum whereas the Caco-2 cells appeared to be least susceptible to C. parvum. In regards to the serum condition, 10% FBS was suitable for the growth of AGS and HCT-8 cells, and 1% FBS was good for the growth of the MDCK cells when they were inoculated with C. parvum. Vitamines had a positive effect on the AGS cells, and pyruvate also showed positive effects on all of the cell lines except for Caco-2. Modified medium for each cell line was prepared by adding appropriate amounts of each supplement which resulted in the highest parasite infection number. Modified media increased the number of parasites infected on AGS cells to 2.3-fold higher when compared to the control media. In this study, we found that the AGS cell line was a suitable host model for evaluating C. parvum in vitro study and the media contents for the optimal infection conditions were suggested.
