  • This study was conducted to investigate the supplementary effects of vitamin C and E to moist pellet on growth and body composition in juvenile olive flounder (Paralichthys olivaceus). Four groups of diet made : moist pellet (MP) diet group was used as the control group where the ratio of raw feed and commercial compound meal is 5:5 $(CP\;30\%,\;CL\;17\%),$ vitamin C added group (VC), vitamin I added group (VE) and vitamin C and I added group (VCE). The supplementary amount of vitamin C and E corresponded to 1000 mg/kg (dry wt.) and 220mg/kg (dry wt.), respectively. Vitamin C was destroyed $50\%$ and vitamin E was destroyed $20\%$ for manufacturing process. After 8 weeks feeding trial, weight gain was $121.9\%$ in MP group, while it ranged from 180.5 to $184.9\%$ in the VC, VE and VCE group. Feed efficiency was $71.\6%$ in MP group, whereas it ranged from 78.7 to $80.6\%$ in the VC, VE and VCE group, weight gain and feed efficiency of fish fed vitamin supplemented diets were significantly higher than control group and no significant difference among the vitamin added groups was observed (p>0.05). There were no significant differences in moisture, crude protein, crude lipid, crude ash, condition factor (CF), hepatosomatic index (HSI), and visceraweight index (VWI) of body composition. Hemoglobin (Hb) was $3.79\%$ in MP group, whereas $4.99\%$ in VC group, $4.55\%$ in VE group and $5.07\%$ in VCE group; Hb was significantly higher in the vitamin added groups than the control group (p<0.05). Vitamin C retention in liver was 18.1, 20.7, 46.6, 54.0 mg/kg in MP, VE, VC and VCE groups, respectively. Vitamin E retention in liver was 25.9, 53.8, 88.2, 124.5 mg/kg in MP, VC, VE and VCE groups, respectively. These results indicate that supplementation of vitamin C and E to moist pellet diet significantly improved growth and feed efficiency of olive flounder, and the mixed addition of vitamin C (1,000 mg/kg dry wt.) and vitamin E (220 mg/kg dry wt.) did not lead to the synergy effect.

  • Encapsulation of L-ascorbic acid (vitamin C) within a bio-compatible layered inorganic material was achieved by coprecipitation reaction, in which the layered inorganic lattice and its intercalate of vitamin C are simultaneously formed. The nano-meter sized powders of vitamin C intercalate thus prepared was again encapsulated with silica nano-sol to form a nanoporous shell structure. This ternary nanohybrid of vitamin Clayered inorganic core-$SiO_2$ shell exhibited an enhanced storage stability and a sustained releasing of vitamin C. Furthermore, the nano-encapsulation of vitamin C with inorganic mineral was very helpful in delivering vitamin C molecules into skin through stratum corneum, facilitating transdermal penetration of vitamin C in topical application.

  • The purpose of this study was to investigate the effects of vitamin E on antioxidative defense system of liver in acute cadmium poisoned rats. Sprague-Dawley male rats weighing 100$\pm$10gm were randomly assigned to one control and three cadmium injected groups. Cadmium injected groups were fed vitamin E free diet(OE-Cd group), 40mg vitamin E per kg diet(40E-Cd group) or 400 mg vitamin E per kg diet(400E-Cd group). Vitamin E level of normal group was 40mg per kg diet. Animals were injected intraperitoneally with 2.0mg Cd$^2$$\^$+//kg bw for 4 days after the rats were fed diets with three different levels of vitamin E for 2 and 4weeks. Activities of superoxide dismutase(SOD), glutathione peroxidase(GSHPx) and glutathione S-transferase(GST) were decreased in cadmium injected groups but those were significantly improved by dietary vitamin I supplementations. Vitamin E contents reduced glutathione(GSH) in the live were decreased in cadmium injected groups, but we., not significantly different among three groups with different levels of vitamin E supplementations. Contents of liver thiobarbituric acid reactive substance (TBARS) of 0E-Cd group were higher than those of 400E-Cd and 400E-Cd groups, but those were markedly alleviated according to vitamin E supplementations. These results indicate that cadmium poisoning in rats causes decreasing antioxidative defense system and increasing peroxidative damage in liver, however can be restored by vitamin E supplements. • Four experiments were conducted to investigate the effect of fat-soluble vitamin administration to sows or newborn pigs on plasma vitamin status. In Exp. 1 and 2, a total of 24 and 43 newborn pigs were allotted to control and vitamin treatments (vitamin $D_3$ with variable addition of vitamins A and E) orally or by i.m. injection. In Exp. 3, pigs from Exp. 2 were allotted to 2 treatments (${\alpha}$vitamins $D_3$ and E in drinking water) for 14 d postweaning. In Exp. 4, twenty-four gestating sows were used for 2 treatments (${\pm}injection$ of a vitamin $D_3$/A/E product 2 wk prepartum). In Exp. 1 and 2, when vitamin $D_3$ was administrated orally or by i.m. injection on d 1 of age, pigs had increased plasma 25-hydroxycholecalciferol (25-OH $D_3$) concentration 10 d after administration compared with control pigs (p<0.05). The injectable administration with vitamin $D_3$ and E was able to achieve higher plasma 25-OH $D_3$ (p<0.05) and ${\alpha}$-tocopherol (p<0.05) concentrations than oral administration. At weaning, the pigs in the injection group had higher plasma 25-OH $D_3$ concentration than those in the other groups in both studies (p<0.05). In Exp. 3, water supplementation of vitamin $D_3$ and E postweaning increased plasma 25-OH $D_3$ and ${\alpha}$-tocopherol concentrations at d 14 postweaning (p<0.01). In Exp. 4, when sows were injected with the vitamin $D_3$ product prepartum, serum 25-OH $D_3$ concentrations of sows at farrowing (p<0.01), and in their progeny at birth (p<0.01) and weaning (p<0.05) were increased. These results demonstrated that fat-soluble vitamin administration to newborn pigs increased plasma 25-OH $D_3$ concentration regardless of administration routes and ${\alpha}$-tocopherol concentration by the injectable route, and that water supplementation of vitamin $D_3$ and E to nursery pigs increased plasma 25-OH $D_3$ and ${\alpha}$-tocopherol concentrations. Additionally, injecting sows with vitamin $D_3$ prepartum increased 25-OH $D_3$ in sows and their offspring. If continued research demonstrates that the serum levels of 25-OH $D_3$ are critical in weanling pigs, a variety of means to increase those levels are available to swine producers.

  • There is a limitation to estimate vitamin $B_{12}$ intake due to a lack of data on vitamin $B_{12}$ content in many Korean foods. In this study, vitamin $B_{12}$ content was determined in some seaweeds, fish, and shellfish and their product that are consumed in Korea using a modified microbioassay with Lactobacillus delbruecki ATCC 7830. Dried laver and dried seasoned and toasted laver contained very high levels of vitamin $B_{12}$ (66.8 and $55.2-71.3\;{\mu}g$/100 g, respectively. Sea lettuce and seaweed fulvescene also contained high vitamin $B_{12}$ content of 5.47-9.41 and $6.46-7.20\;{\mu}g$/100 g, respectively, whereas sea mustard and sea tangle contained low levels of vitamin $B_{12}$; vitamin $B_{12}$ was not detected in seaweed fusifome. Pacific saury, trout, sea-bass, or squid contained 12.01, 2.00, 0.49 and $2.33\;{\mu}g$ vitamin $B_{12}$/100 g, respectively. Ochellatus octopus, and naked sand lance contained 0.72-1.43 and $3.68\;{\mu}g$ vitamin $B_{12}$/100 g, respectively. Dried Alaska pollack con-tained $0.19-2.64\;{\mu}g$ vitamin $B_{12}$/100 g. Shellfish such as little neck clam and small ark shellfish contained high levels of vitamin $B_{12}$ of $30.5-40.5\;{\mu}g$/100 g, and mussel and abalone contained 17.71 and $7.82\;{\mu}g$/100 g, respectively. Of unique Korean traditional fermented seafood products, salt-fermented products of squid ($2.91\;{\mu}g$/100 g), clams ($34.31\;{\mu}g$/100 g), Ala-ska pollack roe ($9.98-12.02\;{\mu}g$/100 g), hairtail guts ($4.58\;{\mu}g$/100 g) or small shrimp ($0.58-1.55\;{\mu}g$/100 g), and fish sauce from anchovies ($1.52-1.78\;{\mu}g$/100 mL), sand eel ($0.22-0.24\;{\mu}g$/100 mL) or small shrimp ($0.19-0.78\;{\mu}g$/100 mL) were analyzed. A few commercial brands of flying fish roe ($0.73-1.73\;{\mu}g$/100 g), canned tuna ($0.40\;{\mu}g$/100 g), and fried fish paste ($0.25-0.69\;{\mu}g$/100 g) were also analyzed. In conclusion, vitamin $B_{12}$ content in these foods, chosen considering the Korean food culture, should contribute to improve the present vitamin $B_{12}$ food database. It may be helpful to estimate vitamin $B_{12}$ intake more correctly than before, and provide additional information for dietary education related to vitamin $B_{12}$ and meal management.

  • BACKGROUND/OBJECTIVES: Energy production and the rebuilding and repair of muscle tissue by physical activity require folate and vitamin $B_{12}$ as a cofactor. Thus, this study investigated the effects of regular moderate exercise training and durations of acute aerobic exercise on plasma folate and vitamin $B_{12}$ concentrations in moderate exercise trained rats. MATERIALS/METHODS: Fifty rats underwent non-exercise training (NT, n = 25) and regular exercise training (ET, n = 25) for 5 weeks. The ET group performed moderate exercise on a treadmill for 30 min/day, 5 days/week. At the end of week 5, each group was subdivided into 4 groups: non-exercise and 3 exercise groups. The non-exercise group (E0) was sacrificed without exercising and the 3 exercise groups were sacrificed immediately after exercising on a treadmill for 0.5 h (E0.5), 1 h (E1), and 2 h (E2). Blood samples were collected and plasma folate and vitamin $B_{12}$ were analyzed. RESULTS: After exercise training, plasma folate level was significantly lower and vitamin $B_{12}$ concentration was significantly higher in the ET group compared with the NT group (P < 0.05). No significant associations were observed between plasma folate and vitamin $B_{12}$ concentrations. In both the NT and ET groups, plasma folate and vitamin $B_{12}$ were not significantly changed by increasing duration of aerobic exercise. Plasma folate concentration of E0.5 was significantly lower in the ET group compared with that in the NT group. Significantly higher vitamin $B_{12}$ concentrations were observed in the E0 and E0.5 groups of the ET group compared to those of the NT group. CONCLUSION: Regular moderate exercise training decreased plasma folate and increased plasma vitamin $B_{12}$ levels. However, no significant changes in plasma folate and vitamin $B_{12}$ concentrations were observed by increasing duration of acute aerobic exercise.

  • This study was conducted to determine the effects of excess vitamin A on alkaline phosphatase (ALP) activity, contents of calcium-binding protein (CaBP), bone gla-protein (BGP) in culture medium and CaBP mRNA expression in chicken osteoblasts in vitro. Osteoblastic cells in the tibia from 1-day-old Arbor Acre broiler chickens were isolated using enzyme digestion. The subconfluenced cells were divided into eight treatments with six replicates in each treatment and cultured in a medium containing either vehicle or different levels of vitamin A (0, 0.2, 0.6, 1.0, 2.0, 5.0, 10.0 and $20.0\;{\mu}g$/ml), and the control received an equivalent volume of ethanol. The incubation lasted 48 h. The results showed that vitamin A down-regulated ALP activity in the culture medium as well as CaBP mRNA expression of osteoblasts in a linear dose-dependent manner (p = 0.124 and p<0.10, respectively), and suppressed the contents of BGP and CaBP in the culture medium in a quadratic dose-dependent manner (p<0.05 and p<0.10, respectively) with increasing addition of vitamin A. The addition of 0-$0.2\;{\mu}g$/ml vitamin A to the culture medium increased ALP activity, BGP and CaBP contents as well as CaBP mRNA expression compared with other groups, but positive effects of vitamin A tended to be suppressed when vitamin A was increased to $1.0\;{\mu}g$/ml, and adverse effects occurred when vitamin A was increased to 10.0-$20.0\;{\mu}g$/ml. These results implied that there was a threshold level of vitamin A inclusion beyond which inhibitory effects occurred, and the mechanism by which overdose of vitamin A reduced bone growth in chickens was probably reduced osteoblastic cell activity, and inhibited expression of CaBP mRNA and CaBP secretion.

  • The objective of this study was to develop a method to simultaneously quantify vitamins A and E in infant formula. To determine the vitamin A and E content, vitamin A and four different vitamin E isomers (${\alpha}$-, ${\beta}$-, ${\gamma}$-, and ${\delta}$-tocopherol) were separated by high performance liquid chromatography with a photodiode array detector using a Develosil RPAQUEOUS RP-$C_{30}$ column ($4.6{\times}250$ mm, 5 ${\mu}M$). The vitamin A and E contents in the certified reference material determined using this method were within the certified range of standard values. The limits of detection (LODs) and limits of quantitation (LOQs) for vitamin A were 0.02 and 0.06 ${\mu}g/L$, respectively. LODs and LOQs for the vitamin E isomers ranged from 0.20 to 0.55 and from 0.67 to 1.81 ${\mu}g/L$, respectively. Linear analyses indicated that the square of the correlation coefficient for the vitamin A and E isomers was 0.9997-0.9999. The recovery of vitamins ranged from 96.69 to 97.79%. The results demonstrate that this novel method could be used to reliably analyze vitamin A and E content in infant formula.

  • Vitamin K is a group of fat-soluble vitamins that naturally exist in phylloquinone (vitamin K1) and menaquinone (vitamin K2). In this study, the vitamin K content in different meats commonly consumed in Korea was analyzed using HPLC, and the analytical method was validated. Vitamin K1 was not detected in any of the meat samples. Vitamin K2 contents in different cuts of beef ranged from 0.00 to 5.87 ㎍/100 g, whereas the corresponding value in different parts of chicken ranged from 16.59 to 46.64 ㎍/100 g. In the case of pork, vitamin K2 contents varied from 4.33 to 22.90 ㎍/100 g. Among the different types of meat, the highest vitamin K2 content was found in boiled chicken meat and skin (46.64 ㎍/100 g). The analytical method was found to be reliable and had high accuracy. These results provide accurate nutritional information and contribute a food composition database for meat consumption.
