Effect of Antioxidant and Irradiation Treatment under Freezing Temperature Conditions on Physicochemical and Sensory Properties of Tarakjuk (Milk Porridge) (항산화제 첨가 및 냉동 방사선 조사 병용처리가 타락죽의 이화학적 및 관능적 품질에 미치는 영향)

  • This study evaluated the effect of antioxidants treatment; (Vit. C and E) on the physicochemical and sensory properties of Tarakjuk treated with gamma-irradiation under two different temperature conditions, e.g. room and frozen temperatures. In samples of Tarakjuk irradiated at room temperature, Vit. C gradually decreased the pH level as the concentration increased (p<0.05), whereas Vit. E did not. Addition of both Vit. C and E to Tarakjuk decreased TBA (2-thiobarbituric acid) value while also increasing viscosity. However, these antioxidants did not cause any change in Hunter's color values. Sensory properties showed that Vit. C and E significantly improved the overall acceptability of Tarakjuk, and the addition of 0.1% each Vit. C and E was best treatment. In samples of Tarakjuk subjected to combined treatment with antioxidants and irradiation under frozen temperature conditions, Vit. C and E were effective in decreasing TBA value, improving viscosity, and maintaining sensory quality. Especially, co-treatment with 0.1% Vit. C and E augmented the observed effect (p<0.05). Therefore, combined treatment with antioxidants and irradiation under frozen temperature conditions could improve the final sensory quality of gamma-irradiated Tarakjuk.

  • This study was conducted to examine the effects of recombinant Bovine Somatotropin (rBST), vitamin E(Vit. E) and selenium (Se) administration on the blood chemical values and hormone concentrations of serum in Hanwoo sires. Hanwoo sires were randomly assigned to five groups; 1) control, 2) rBST,0.09mg/kg body weight (BW), 3) Vit E, 1,500IU/kg BW, 4) Se 0.1mg/kg BW, 5) Vit E, 1,500IU plus Se 0.1mg/kg BW. rBST, Vit. E and Se for each experimental group were given 6 times at 15 days interval by intermuscular injection. Blood samples were collected ten times for experimental periods and separated the serum by centrifugation and stored into deep freezer. The concentration of albumin was the highest in Se group than those of any other groups (P<0.05) and Vit. E plus Se group was significantly higher than in the control and rBST groups (P<0.05). The concentrations of blood urea nitrogen (BUN) and creatinine were significantly higher in rBST group than any other groups (P<0.05). The concentration of total protein in rBST, Se and Vit. E plus Se groups were significantly higher than in control group (P<0.05). The concentrations of calcium, cholesterol,, glucose, inorganic phosphorus and triglycerides in serum were not difference in all experimental groups (P>0.05). The concentration of estradiol was slightly higher in Se and Vit. E plus Se groups than those of any other groups, but not significantly difference among the experimental groups (P<0.05). The concentration of testosterone was not affected by the administration of rBST, Vit. E and Se.

The Effect of Butanol Fraction of Polygonatum odoratum with Vitamin E on Blood Glucose Levels and Lipid Peroxidations in Streptozotocin-Induced Diabetic Rats (둥굴레(Polygonatum odoratum)분획물과 비타민 E 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당과 지질과산화에 미치는 영향)

  • The hypoglycemic effects of butanol(BuOH) fraction of Polygonatum odoratum with vitamin E in streptozotocin(STZ)-induced diabetic rats were investigated. Sprague-Dawley rats weighing 200-230g were devided into five groups, and four groups induced diabetes mellitus by the STZ injection(45mg/kg b.w.) into the tail vein : Normal, diabetic-control, and three diabetic experimental groups(p. odoratum group, P. od-vit. E group and Vit. E group). All groups were fed on a AIN-76 diet, and the experimental groups were orally administered with the BuOH fraction of Polygonatum odoratum(500mg/kg b.w.) and vitamin E(10mg/kg b.w.) for 14 days. The body weight, diet intake and organ weights were monitored. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol and aspartate and alanine aminotransferase activities were analyzed. The levels of glycogen in liver and muscle, cholesterol in liver were determined. The malondialdehyde(MDA) levels in liver, kidney and lung were assayed. The body weight loss was seen in P. odoratum group, P. od-vit. E group, Vit. E group and diabetic control group, while the loss in P. odoratum group was much less than that in the diabetic control group. The plasma glucose levels were significantly lowered in P. odoratum group compared to diabetic control group. The plasma insulin levels were noticeably higher in P. odoratum and Vit. E groups. The rats in P. odoratum and P. od-vit. E group showed higher liver glycogen levels than in the diabetic control group. The MDA levels in liver, kidney and lung were also significantly reduced in P. od-vit. E and Vit. E groups compared to the diabetic control group. The results suggest that the administration of BuOH fraction of Polygonatum odoratum along with vitamin E reduced blood glucose levels and peroxidative tissue damage in STZ-induced diabetic rats, showing the possibility of preventive and therapeutic use of the wild edible plant to the diabetes mellitus. Curcumin and Vit. E Alleviate Alone or Synergetically Hydrogen Peroxide Induced-Oxidative Stress on Boar Sperm Characteristics during In Vitro Storage

  • Antioxidants partially ameliorated the detrimental effects of reactive oxygen species (ROS) on sperm characteristics during in vitro storage. The objective of the present study was to investigate the single or synergetic antioxidative effect of curcumin and Vit. E on the characteristics of fresh boar sperm during in vitro storage. The sperm viability in curcumin, Vit. E supplementation and curcumin+Vit. $E+H_2O_2$ groups remained over 85.0% in 3 hr incubation period, but in 6 hr incubation period, curcumin+Vit. $E+H_2O_2$ groups was sharply dropped than those of curcumin and Vit. E group. The membrane intergrity in all evaluated groups except for $H_2O_2$ group did not significantly difference in 3 hr incubation period. The viability in curcumin or Vit. E supplementation were significantly increased than in curcumin+$H_2O_2$ and Vit. $E+H_2O_2$ group in 6 hr incubation period. The percentage of mitochondrial activity and acrosome intergrity obtained similar trends within same incubation periods irrespective of treatment. The lipid peroxidation of spermatozoal plasma membrane ranged from $11.6{\sim}17.5\;nM/l{\times}10^6$ and $14.0{\sim}19.0\;nM/l{\times}10^6$ in 3 hr and 6 hr incubation periods. In conclusion, curcumin or Vit. E surpplementation alone or cooperatively improved sperm viability index (motility, membrane intergrity, viability and survival rates) and fertility index (mitochondria activity, acrosome intergrity and lipid peroxidation) of fresh boar sperm, indicating that curcumin and Vit. E have a antioxidative properties through its scavenging activity against hydrogen peroxide.

  • The objectives of this study were to investigate 1) the effects of Selenium(Se), Vitamin E (Vit. E) or recombinant Bovine Somatotropin(rBST) administration on fresh and frozen/thawed semen characteristics and 2) the effect of taurine on frozen/thawed semen characteristics in Hanwoo sires Hanwoo sires were randomly assigned to five groups (1. control, 2. rBST, 0.09mg/kg body weight (BW), 3. Vito E 1,500IU/kg BW, 4. Se 0.l mg/kg BW, 5. Vit. E 1,500IU plus Se 0.1 mg/kg BW). The administration of Se, Vit. E and rBST for each experimental group were given 6 times at 15 days interval by intramuscular injection. The administration of Se, Vit. E or rBST in Hanwoo sires didn't affect semen volume and pH values, but sperm viability was significantly increased comparing to the control group. Also, frozen/thawed semen analysis showed that the sperm viability increased, but any other effects were not found in total sperm :lumber, motility and abnormality among treatments. The addition of taurine in semen freezing extender had a beneficial effects on frozen/thawecl semen characteristics in all groups. The administrations of rBST, Vit. E and Se did not affect the sperm capacitation and acrosome reaction, either the ratio of F pattern(uncapacitated and acrosome intact sperm) or AR pattern(capacitated and acrosome-reacted sperm), but the ratio of B patten(capacitated and acrosome intact sperm) of treatment groups was significantly higher than that of control group, These results indicated that the viability, motility and quality of semen in Hanwoo sires were slightly increased by the injection of rBST, Vit. E and Se, and the addition of taurine in semen freezing extender were also increased the semen characteristics after thawing.

  • The objective of this study were to investigate the effects of Se and Vit. E administration at last month or two month before parturition on the reproductive function of dams and developmental ability of calves. On developmental ability of calves obtained afier Se and Vit. E administration at the last month of pregnancing periods, the birth weight were significantly higher in administration groups than in control group(P<0.05). However, the weaning weight, average daily gain and weaning age were not significantly difference in all experimental groups. In the comparision of reproductive function in dams, estrus postpartum was faster in treatment groups than control groups(P>0.05). However, the number of A.1 service fur conception were lower groups injected than control group(P>0.05), there were not significantly differences in all experimental groups. The developmental ability of calves obtained after Se and Vit. E administration at two months before parturition were also examined. The birth weight, weaning weight, average daily gain and weaning age were higher in treatment groups than control group, but there were not significantly differences in all experimental groups. In reproductive function of dams, the days to 1 st estrus postpartum was slightly faster injected groups than control group(P>0.05). Number of A.I. service for conception in each groups were lower in treatment groups than control group.

Study on Suitable Semen Additives Incorporation into the Extender Stored at Refrigerated Temperature

  • The objective of this study was to compare the effect of Butylated Hydroxy Toluene (BHT), Pentoxifylline (PTX) and ${\alpha}$-tocopherol (Vit E) on semen quality parameters of Karan Fries bulls. The fortification of extender by various semen additives improves motility as well as fertility of spermatozoa. Split samples of 24 ejaculates of four Karan Fries bulls were extended in extender with or without various additives such as BHT, PTX and Vit E, and performance was evaluated at an interval of 0, 24, 48 and 72 h at refrigerated temperature (4-$7^{\circ}C$). Results of the present study revealed that addition of BHT, PTX and Vit E in extender improved sperm cell function, such as motility, viability, HOST, and acrosome integrity, as compared to the control during liquid storage up to 48 h of preservation at refrigerated temperature. There was no significant (p<0.05) difference between any of the additives up to 48 h of preservation. Overall, the results showed a significant (p<0.05) deterioration in motility after each storage interval. The results showed a significant deterioration in the acrosome integrity and plasma membrane integrity up to 48 h; subsequently, there was not much degradation of both the semen quality parameters. There was a significant increase in spermatozoal tail and total abnormality after each storage interval at refrigerator temperature (4 to $7^{\circ}C$); however, the head and mid-piece abnormalities were almost unaffected. Tail and total abnormality were least in extender fortified with BHT, PTX and Vit E at different hours of incubation as compared to the control. The addition of 1.5 mM BHT, 3.6 mM PTX and 1 mg/ml Vit E in the semen extender has more beneficial effect in terms of semen quality and preservability of spermatozoa.

  • The sows which had not returned estrus since 15 to 152 days after weaning were treated with PMSG or PGF2$\alpha$ to induce estrus and inseminated to conceive on that estrus. 1. Eight (83%) among 12 sows treated with PMSG came into estrus on average 5.3 days after treatment. 2. The one sow which didn't show any estrus by treatment of PMSG showed an estrus by intramuscular use of 10mg PGF2$\alpha$ and the other one showed an estrus by another intramuscular use of 1000 I.U. of PMSG at 15 days after first injection of PMSG. 3. The one sow injected with Vit. A.D.E. complex(Injacom) (Vit. A 500,000 I.U., Vit. D. 75,000 I.U., and Vit. E. 50 I.U. per ml) on 6 days before PMSG injection showed an estrus on 6 days after PMSG injection and farrowed 8 piglets. 4. Five among 10 sows showed an estrus by treatment of PMSG or PGF2$\alpha$ were pregnant and litter size of the four farrowed sows averaged 9.5.

The Effect of Dietry Se Levels and Alcohol Administration on the Lipid Hyperoxidation and the Lipid Metabolism in the Rats (식이내 Se수준과 알콜 섭취가 흰쥐의 지질과산화와 지방대사에 미치는 영향)

  • The purposes of this studies were to investigate the effect of dietry Se levels and alcohol administration on the lipid hyperoxidation and the lipid metabolism in the rat. Seventy two male rats of Sprague-Dawley Strain weighting about 58~629 were divided into 12groups. The dietary Se levels were 10, 0.4 and Omg, and the dietary a-tocopherol levels were 150 and 0mg per kg diet, respectively. Alcohol-administrated groups received drinking water solution containing 10% of ethanol from the 3-weeks of experimental periods. The obtained experimental results are summarized as follows. 1. Food intake, body weight gain and food efficiency ratio were significantly lower in H-, L-and alcohol administrated groups(-A) by administrated Se and alcohol in diet. The weight of liver and spleen tended to be greater in H- and alcohol administrated groups. 2. The glutathione values in liver tend to be lower in alcohol or Vit. E administrated groups than nonadministrated groups. Also there were higher in H- and L- than C-groups, but the increasing range decreased due to administrated alcohol. The lipid peroxide values In liver were significantly higher in alcohol groups, and L- and tocopherol groups were higher values. Specially the increasing of lipid peroxide values were significantly effected by alcohol in low Se and Vit. E groups. 3. The contents of total glyceride in plasma were higher in alcohol groups, there were significantly higher values in alcohol administrated groups under low Se and Vit. E groups. The contents of total cholesterol and HDL-cholesterol In plasma were significantly higher in alcohol groups. 4. The contents of total lipid in liver were higher alcohol groups, and slightly higher values in low Se groups(L-groups) than other groups, also higher values in low Vit. E groups. Those of total glyceride in liver were significantly higher in alcohol groups, appeared highest values when alcohol was administrated in low Se and Vit. E groups. The increasing of total glyceride content was significantly effected by alcohol in low Se groups than that in C-groups.

Algae-based antioxidant containing selenium yeast (Economase®) enhanced the growth performance, oxidative stability, and meat quality of broiler chickens

  • Objective: An experiment was conducted to determine the effect of algae-based antioxidant containing Se yeast (EconomasE®) on the growth performance, visceral organ weight, meat quality, and oxidative stability of broiler chickens. Methods: Nine hundred sixty, day-old male broiler chickens (Cobb, 43.97±0.55 g) were divided into three dietary treatments and allocated into 12 deep litter pens in a completely randomized design giving 4 replicate cages for each treatment. Three dietary treatments were: i) control (CON, basal diet with sufficient nutrient), ii) vitamin E (VitE, basal diet supplemented with 100 IU VitE), and iii) Algae-based antioxidant containing Se yeast (EcoE, basal diet supplemented with 0.2% algae-based antioxidant containing Se yeast: EconomasE®). Maize soybean meal based basal diets were formulated to meet or exceed the nutrition requirement for broiler chickens. Chickens were fed ad-libitum experimental diets during the 42 days experiment period. On days 21 and 42, body weight and feed intake were measured to calculate the feed conversion ratio of the chickens. Intestine and visceral organs were measured together with meat quality and oxidative stability on days 14 and 42. Results: Chickens fed with EcoE showed improved (p<0.05) growth performance, meat quality, and higher (p<0.05) oxidative stability compared to the chicken fed on CON. Moreover, broiler chickens fed with EcoE showed similar (p<0.05) growth performance with better (p<0.05) meat quality and higher oxidative stability compared to the broiler chickens fed VitE (p<0.05). Conclusion: The algae-based antioxidant containing Se yeast can be supplemented into commercial broiler diets as a substitution of VitE while maintaining growth performance with enhancing meat quality and oxidative stability of the broiler chickens.
