Stability of Antioxidant Properties and Essential Amino Acids in Squid Viscera Hydrolysate Produced using Subcritical Water

  • Subcritical water hydrolysis was carried out in a batch reactor to produce valuable materials, such as low-molecular weight (MW) peptides and essential amino acids with antioxidant properties, from heat-dried squid viscera. Hydrolysis of squid viscera was performed at 160 to $280^{\circ}C$ for 3 min. The yield was increased by increasing the temperature and pressure, while the protein content of squid viscera hydrolysate decreased with increasing temperature. Low-MW peptides were detected in all hydrolysates by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The highest yields of free and structural amino acids in heat-dried squid viscera hydrolysate were at $160^{\circ}C$ and were $411.95{\pm}1.15$ and $346.62{\pm}1.25$ mg/100 g, respectively. All essential amino acids were detected in viscera hydrolysates; leucine was the most abundant. Antioxidant activities of hydrolysates were highest at $220^{\circ}C$. Greater than $98{\pm}0.26%$ of the ABTS antioxidant activity was retained in hydrolysates after long-term heat treatment.

가다랭이 내장 단백질 가수분해물의 이용 (Utilization of the Protein Hydrolysates of Skipjack Tuna Viscera)

  • 본 연구는 영양적으로 양호하고 가격면에서도 저렴한 단백질 자원을 개발하기 위하여 가다랭이 내장을 에틸 알코올 및 단백질 가수분해 효소인 펩신으로 처리함으로써 식품소재로서의 이용가능성을 검토한 것이다. 이 가다랭이 내장 단백질 가수분해물의 성분과 가수분해도 및 용해도 등의 변화를 조사하고, 모조간장 또는 식빵 제조시 첨가하여 관능검사를 통하여 영양적 가치와 기호성 등을 확인하였던 바, 가다랭이 내장으로부터의 단백질 농축물은 40% 정도의 용해도를 나타내었으나, 단백질 가수분해물은 90% 정도까지 향상되었다. 한편 필수아미노산 함량면에서 단백질 가수분해물의 경우 전체 아미노산의 63.2%로 우수한 단백질 소재임을 알 수 있었고, 타우린의 함량이 9.4%로 상당히 높은 특징을 보였다. 또한 내장 단백질 가수분해물을 첨가하여 모조간장 및 식빵을 제조한 다음 관능평가 결과, 모조간장의 경우는 물 100ml에 다른 원료들과 함께 내장 단백질 가수분해물을 10.0g 첨가했을 때 제품의 색깔, 맛, 냄새 등에서 좋은 평점을 얻었으며, 식빵의 경우는 $3{\sim}5%$ 수준의 내장 단백질 가수분해물 첨가시 제품의 맛, 색깔, 냄새, 조직 및 촉감 등의 품질면에서 손색이 없었다.

Effect of Maillard reaction with xylose, yeast extract and methionine on volatile components and potent odorants of tuna viscera hydrolysate

  • The aim of this research was to enhance the flavor of visceral extracts from skipjack tuna. Flavor precursors and the optimum condition for the Maillard reaction were determined. The flavor extract was prepared from the tuna viscera using Endo/Exo Protease controlled in 3 factors; temperature, enzyme amounts and incubation time. The optimal condition for producing tuna viscera protein hydrolysate (TVPH) was 60℃, 0.5% enzyme (w/w) and 4-hour incubation time. TVPH were further processed to tuna viscera flavor enhancer (TVFE) with Maillard reaction. The Maillard reactions of TVFE were conducted with or without supplements such as xylose, yeast extract and methionine. The Maillard volatile components were analyzed with gas chromatography-mass spectrometry. Sixteen volatiles such as 2-methylpropanal, methylpyrazine, 2,5-dimethylpyrazine, dimethyl disulfide and 2-acetylthaizone were newly formed via Maillard reaction and the similarity of volatile contents from TVPH and TVFE were virtualized using Pearson's correlation integrated with heat-map and principal component analysis. To virtualize aromagram of TVPH and TVFE, odor activity value and odor impact spectrum (OIS) techniques were applied. According to OIS results, 3-methylbutanal, 2-methylbutanal, 1-octen-3-ol 2,5-dimethylpyrazine, methional and dimethyl trisulfide were the potent odorants contributed to the meaty, creamy, and toasted aroma in TVFE.
