The bursa of Fabricius (BF) is a central humoral immune organ unique to birds. Four bursal peptides (BP-I, BP-II, BP-III, and BP-IV) have been isolated and identified from the BF. In this study, the immunoadjuvant activities of BPs I to IV were examined in mice immunized with H9N2 avian influenza virus (AIV) vaccine. The results suggested that BP-I effectively enhanced cell-mediated immune responses, increased the secretion of Th1 (interferon gamma)- and Th2 (interleukin-4)-type cytokines, and induced an improved cytotoxic T-lymphocyte (CTL) response to the H9N2 virus. BP-II mainly elevated specific antibody production, especially neutralizing antibodies, and increased Th1- and Th2-type cytokine secretion. BP-III had no significant effect on antibody production or cell-mediated immune responses compared to those in the control group. A strong immune response at both the humoral and cellular levels was induced by BP-IV. Furthermore, a virus challenge experiment followed by H&E staining revealed that BP-I and BP-II promoted removal of the virus and conferred protection in mouse lungs. BP-IV significantly reduced viral titers and histopathological changes and contributed to protection against H9N2 AIV challenge in mouse lungs. This study further elucidated the immunoadjuvant activities of BPs I to IV, providing a novel insight into immunoadjuvants for use in vaccine design.

Hepatitis E virus (HEV), previously referred to as enterically transmitted non-A, non-B hepatitis, is responsible for sporadic infections as well as large epidemics of acute viral hepatitis in developing countries. The disease generally affects young adults and reportedly has a mortality rate of up to 20% in infected pregnant women. HEV was once considered to be a member of the family Caliciviridae, but the unique genomic organization of HEV has led to the removal of HEV from the family and it was provisionally classified in an unassigned family of HEV-like viruses. In situ hybridization provides any cellular detail and histological architecture.[1] However, use of in situ hybridization is largely restricted to the laboratories because this technique is the greater technical complexity and expense compared with immunohistochemistry. Therefore, the objective of this study is to develop the immunohistochemistry for the detection of swine HEV from formalin-fixed, paraffin-embedded hepatic tissues. (omitted)
Validation of viral safety is essential in ensuring the safety of mammalian cell culture-derived biopharmaceuticals, because numerous adventitious viruses have been contaminated during the manufacture of the products. Mammalian cells are highly susceptible to minute virus of mice(MVM), and there are several reports of MVM contamination during the manufacture of biopharmaceuticals. In order to establish the validation system for the MVM safety, a real-time PCR method was developed for quantitative detection of MVM in cell lines, raw materials, manufacturing processes, and final products as well as MVM clearance validation. Specific primers for amplification of MVM DNA was selected, and MVM DNA was quantified by use of SYBR Green I. The sensitivity of the assay was calculated to be $6{\times}10^{-2}TCID_{50}/mL$. The real-time PCR method was proven to be reproducible and very specific to MVM. The established real-time PCR assay was successfully applied to the validation of Chinese hamster ovary (CHO) cell artificially infected with MVM. MVM DNA could be Quantified in CHO cell as well as culture supernatant. When the real-time PCR assay was applied to the validation of virus removal during a virus filtration process, the result was similar to that of virus infectivity assay. Therefore, it was concluded that this rapid, specific, sensitive, and robust assay could replace infectivity assay for detection and clearance validation of MVM.
A bench-scale test has recently been proposed as a predictive tool to minimize the scope of pilot-scale testing or to optimize the operation of full-scale membrane filtration systems. Consequently, a bench-scale testing unit was developed for this purpose and systematically evaluated in this study. This unit was capable of accommodating commercially available, low pressure, hollow fiber (LPHF) membranes with various configurations for testing under conditions comparable to real-world applications. Reproducibility of this unit in assessing membrane fouling and microbial removal efficiency of LPHF membranes was tested and statistically comparable results were obtained. This unit serves as a useful apparatus for academic researchers and utilities to evaluate the performance of LPHF membranes used for water treatment.
The synthesis of 5-hyterocyclimethyl-2'-deoxyuridines (4a-f) has been accomplished by displacement reaction of 5-(bromomethyl)-3', 5'-di-O-acetyl-2'-deoxyuridine with heterocyclic compounds, followed by removal of acetyl protecting group with methanolic ammonia. The compoudns synthesized were evaluated the inhibitory effects on L1210 cell probiferation and antiviral activities against Herpes simplex virus type 1 (HSV-1) None of the compounds exhibited sufficient biological activities.
Recently coastal seawater has been contaminated by enteric viruses such as the norovirus via untreated groundwater globally. Accordingly, the consumption of molluscan shellfish from seawater that has been contaminated with fecal material has become an important issues. The levels of enteric viruses in seawater are low and recovery and concentration of the virus from large volumes of water is difficult. We compared the effectiveness of two representative method of concentrating virus using negatively and positively charged filters. The mean retention of seeded FCV by HAMF and NCCF was 48% and 78%, respectively. Overall, the recovery of NCCF was 43.3$\pm$11% better than that of HAMF. However, the eluate obtained by using beef extract solution in the NCCF procedure caused an inhibitory effect on the RT-PCR; therefore, it was necessary to employ a PCR inhibitor removal procedure. The HAMF eluate contained no PCR inhibitors, but HAMF was not an effective method of concentrating the virus from large volumes of natural seawater due to clogging.

In Japan, the membrane filtration is becoming a common technology for municipal water supply system especially for small plant. 6 years before (1991), the national research project of membrane filtration for small plant has started. The project was named as "MAC 21", MEMBRANE AQUA CENTURY 21. In the project the Ministry of Health and Welfare, 8 universities and 18 water treatment plant companies have been involved. This was the first attempt to research a common theme in joint with government, universities and private companies. After three years, the guide line for membrane filtration application for small plant has been established. This has promoted to install some actual plant. And also, another joint research for "RESEARCH OF MEMBRANE FILTRATION FOR ADVANCED WATER TREATMENT" has started in 1994 and completed in March, 1997. The project was named as MAC21. In the former project the main objectives were removal of turbidity and bacteria from water. However, in new project the objective was establishment of the further advanced membrane filtration technology which would be applicable for trace chemical components removal such as tri-halo-methane pre-courser, agricultural chemicals removal, offensive smell and taste removal and virus removal. For the objectives, application of nanofiltration and hybrid-system, a combination of micro-filtration ultra-filtration with biological, ozone and activated carbon treatment process have been studied. In addition, application of membrane filtration for treatment of back-wash waste water originated from membrane filters and conventional sand filters has.been studied. At the end of March of this year, about 30 membrane filtration plants are actually supplying the water, the total treatment capacity is about 6,000 m$^{3}$/day and another 20 will be installed within one year.led within one year.
To improve the efficiency of removal of bacteria and virus with DynaSand Filters used for treatment of recycling wastewater from aquaculture, effect of biofilm formation on bacterial transport through coated sand was estimated. At the neutral pH (pH 7.0), the coated sand was positive of zeta potential (surface charge). Column experiments were also carried out to test the effect of uncoated sand as well as coated sand with Al and Fe. The coated sand influenced more significantly the surface properties, adsorption and transport than the uncoated sand. The leaching batch system investigated for synthetic water showed concentrations of 7.47, 4.80, 20.89 and 7.23 mg/L for the uncoated sand, coated sand with Al, Fe and Al+Fe, respectively. Hence there are significant differences among the tested coatings with reference to bacterial transport and surface properties.
