  • 최근 위치 인식 기술과 휴대 장치의 발달로 인해 이동하는 객체를 기반으로 하는 위치 기반 서비스(Location Based Service, LBS)의 관심이 점점 증가하고 있고 그에 관련된 연구들이 활발하게 진행되고 있다. 이동 객체의 응용은 빈번하게 변경되는 이동객체의 위치정보를 효과적으로 처리할 수 있는 색인구조를 필요로 한다. 위치정보를 색인하기 위해 R-tree 기반의 색인들이 제안되었다. 하지만 R-tree는 변경보다는 검색 연산에 초점이 맞추어진 색인구조이기 때문에 잦은 변경을 다루어야 하는 이동객체 환경에 적합하지 못하다. 최근 이러한 객체의 빠른 위치 변경을 지원하는 그리드 기반의 색인 구조가 제안되었다. 하지만 셀의 객체 점유율에 따라 검색 속도가 저하되는 단점은 여전히 해결되지 못하고 있다. 이러한 단점은 객체들이 특정 영역에 몰리는 경우 또는 그리드의 해상도를 잘못 지정한 경우 더욱 부각된다. 본 논문에서는 이러한 단점을 해결하기 위해 가상 셀 기반의 색인 구조를 제안한다. 데이터 페이지에 객체의 점유율을 보장하기 위해 여러 개의 인접한 셀들의 데이터를 한 데이터 페이지에 함께 저장한다. 공간 채움 곡선을 기반으로 순서화된 셀들로 셀의 인접성을 결정한다. 또한 공간 채움 곡선의 차수를 동적으로 지정하여 객체가 집중된 셀에 대해서는 셀의 단위 크기를 작게 지정한다. 뿐만 아니라 셀을 표현하기 위한 식별자를 위해 비트를 이용한 표현식을 제안하였다. 이로 인해 노드의 팬아웃을 증가시켰고, 저장공간을 절약하였다. 실험을 통해서 제안하는 색인 기법의 우수성을 증명하였다.

Investigation of Conserved Gene in Microbial Genomes using in silico Analysis (미생물 유전체의 in silico분석에 의한 보존적 유전자 탐색)

  • Conserved genes are importantly used to understand the major function in survival and replication of living organism. This study was focused on identification of conserved genes in microbial species and measuring the degree of conservation. For this purpose, in silico analysis was performed to search conserved genes based on the conservation level within microbial species. The ortholog list of COGs (Clusters of Orthologous Groups of proteins) in NCBI was used and whole genomes of 43 microbial species were included in that list. The distance value, derived from CLUSTALW multiple alignment program, was used as a descriptor of the conservation level of orthologs. It was revealed that 43 microbial genomes hold 72 conserved orthologs in common. The majority(72.2%) of the conserved genes was related to "translation, ribosomal structure and biogenesis" functional category. A GTPase-translation elogation factor(COG0050) was the best conserved gene from the distance value analysis. The 72 conserved genes, found in this research, would be useful not only to study minimal function genes but also new drug target among pathogens and to make a model of the virtual cell.tual cell.

  • Communications for mobile sink groups such as rescue teams or platoons bring about a new challenging issue for handling group mobility in wireless sensor networks. Recently, a group region-based approach has been proposed to send data to a mobile sink group. However, it uses excessive flooding for 3 steps (group region discovery, region information notification, and data dissemination) to support mobile sink group. Thus, it causes high energy consumption and data delivery failure. Moreover, its per-sink mobility supporting scheme makes energy consumption and data delivery failure more serious. Thus, this paper proposes an Energy-efficient and Reliable Mobile Group communication protocol (ERMG) to support a mobile sink group, which exploits a virtual grid structure. For the group region discover and notification and the data dissemination, ERMG uses communications with only cell heads of grid cells that manages group sinks. ERMG also uses a per-grid based scheme to support sink mobility. Simulation results show that ERMG has 30% less energy consumption and 6% more desired delivery deadline success ratio than the existing protocols.

A Novel Pyruvate Kinase M2 Activator Compound that Suppresses Lung Cancer Cell Viability under Hypoxia

  • Pyruvate kinase M2 isoform (PKM2), a rate-limiting enzyme in the final step of glycolysis, is known to be associated with the metabolic rewiring of cancer cells, and considered an important cancer therapeutic target. Herein, we report a novel PKM2 activator, PA-12, which was identified via the molecular docking-based virtual screening. We demonstrate that PA-12 stimulates the pyruvate kinase activity of recombinant PKM2 in vitro, with a half-maximal activity concentration of $4.92{\mu}M$, and effectively suppresses both anchorage-dependent and -independent growth of lung cancer cells in non-essential amino acid-depleted medium. In addition, PA-12 blocked the nuclear translocalization of PKM2 in lung cancer cells, resulting in the inhibition of hypoxia response element (HRE)-mediated reporter activity as well as hypoxia-inducible factor 1 (HIF-1) target gene expression, eventually leading to the suppression of cell viability under hypoxia. We also verified that the effects of PA-12 were dependent on PKM2 expression in cancer cells, demonstrating the specificity of PA-12 for PKM2 protein. Taken together, our data suggest that PA-12 is a novel and potent PKM2 activator that has therapeutic implications for lung cancer.

Ligand Based Pharmacophore Identification and Molecular Docking Studies for Grb2 Inhibitors

  • Grb2 is an adapter protein involved in the signal transduction and cell communication. The Grb2 is responsible for initiation of kinase signaling by Ras activation which leads to the modification in transcription. Ligand based pharmacophore approach was applied to built the suitable pharmacophore model for Grb2. The best pharmacophore model was selected based on the statistical values and then validated by Fischer's randomization method and test set. Hypo1 was selected as a best pharmacophore model based on its statistical values like high cost difference (182.22), lowest RMSD (1.273), and total cost (80.68). It contains four chemical features, one hydrogen bond acceptor (HBA), two hydrophobic (HY), and one ring aromatic (RA). Fischer's randomization results also shows that Hypo1 have a 95% significant level. The correlation coefficient of test set was 0.97 which was close to the training set value (0.94). Thus Hypo1 was used for virtual screening to find the potent inhibitors from various chemical databases. The screened compounds were filtered by Lipinski's rule of five, ADMET and subjected to molecular docking studies. Totally, 11 compounds were selected as a best potent leads from docking studies based on the consensus scoring function and critical interactions with the amino acids in Grb2 active site.

Design of a Voltage Protection Circuit for DC-DC Converter of the Potable Device Application (소형 휴대기기용 DC-DC 변환기를 위한 전압 보호회로 설계)

  • In this paper, a potable device application for DC-DC converter was designed for voltage protection circuit. Voltage protection circuit to offer the under voltage lock out and over voltage protection consists of a comparator and bais circuits were implemented using. XFAB 1um CMOS process, SPICE simulations was confirmed through the characteristics. Simulation results, under voltage lock out input voltage is 4.8 V higher when the turn-on and, 4.2 V less when turn-off. When the input voltage is low voltage is applied can be used to prevent malfunction of the circuit. Over voltage protection is 3.8 V reference voltage when the output voltage caused by blocking circuit prevents device destruction can be used to improve the stability and reliability. The virtual control circuits of the DC-DC converter connected. According to the results of the abnormal voltage, voltage protection circuit behavior was confirmed. The proposed voltage protection circuit of the DC-DC converter cell is useful are considered.

A novel BTK gene mutation, c.82delC (p.Arg28 Alafs*5), in a Korean family with X-linked agammaglobulinemia

  • X-linked agammaglobulinemia (XLA) is a hereditary humoral immunodeficiency that results from Bruton's tyrosine kinase (BTK ) gene mutations. These mutations cause defects in B-cell development, resulting in the virtual absence of these lymphocytes from the peripheral circulation. Consequently, this absence leads to a profound deficiency of lg all isotypes, and an increased susceptibility to encapsulated bacterial infections. A 15-month-old Korean boy presented with recurrent sinusitis and otitis media after 6 months of age, and had a family history of 2 maternal uncles with XLA. Laboratory tests revealed a profound deficiency of Ig isotypes, and a decreased count of $CD19^+$ B cells in the peripheral circulation. Based on his family history and our laboratory test results, he was diagnosed with XLA. We performed BTK gene analysis of peripheral blood samples obtained from family members to confirm the diagnosis. Mutational analysis revealed a novel hemizygous frameshift mutation (c.82delC, p.Arg28Alafs*5), in the BTK gene. His mother and maternal grandmother were heterozygous carriers of this mutation and his two maternal uncles were hemizygous at the same position. After XLA diagnosis, intravenous immunoglobulin (400 mg/kg, monthly) treatment was initiated; recurrent sinusitis and otitis media were subsequently brought under control. To our knowledge, this is the first reported case of a Korean pedigree with a novel mutation in the BTK gene.

A Spatial Interpolation Model for Daily Minimum Temperature over Mountainous Regions (산악지대의 일 최저기온 공간내삽모형)

  • Spatial interpolation of daily temperature forecasts and observations issued by public weather services is frequently required to make them applicable to agricultural activities and modeling tasks. In contrast to the long term averages like monthly normals, terrain effects are not considered in most spatial interpolations for short term temperatures. This may cause erroneous results in mountainous regions where the observation network hardly covers full features of the complicated terrain. We developed a spatial interpolation model for daily minimum temperature which combines inverse distance squared weighting and elevation difference correction. This model uses a time dependent function for 'mountain slope lapse rate', which can be derived from regression analyses of the station observations with respect to the geographical and topographical features of the surroundings including the station elevation. We applied this model to interpolation of daily minimum temperature over the mountainous Korean Peninsula using 63 standard weather station data. For the first step, a primitive temperature surface was interpolated by inverse distance squared weighting of the 63 point data. Next, a virtual elevation surface was reconstructed by spatially interpolating the 63 station elevation data and subtracted from the elevation surface of a digital elevation model with 1 km grid spacing to obtain the elevation difference at each grid cell. Final estimates of daily minimum temperature at all the grid cells were obtained by applying the calculated daily lapse rate to the elevation difference and adjusting the inverse distance weighted estimates. Independent, measured data sets from 267 automated weather station locations were used to calculate the estimation errors on 12 dates, randomly selected one for each month in 1999. Analysis of 3 terms of estimation errors (mean error, mean absolute error, and root mean squared error) indicates a substantial improvement over the inverse distance squared weighting.

  • Lithium-ion battery (LiB) with high energy density and efficiency has been utilized for the electric vehicle (EV) and energy storage system (ESS) as well as portable devices. However, as explosion accidents have frequently happened till lately, all-solid-state lithium secondary battery (ALSB) began to get in a spotlight because it can secure a very high safety and energy density by substituting flammable organic liquid electrolyte to nonflammable inorganic solid electrolyte. In spite of ALSB's certain merits, it has shown much poorer performance of cells than one of LiB due to some challenges, which have been small or never dealt with in the LiB system. Hence, although plenty of studies made progress to solve them, an approach about design of all-solid-state electrode (ASSE) has been limited on account of difficulty of ALSB's experiments. That is why the virtual 3D structure of an all-solid-state electrode has to be built and used for the prediction of cell performance. In this study, we elucidate how to form the 3D ASSE structure and what to be needed for the simulation of characteristics on ALSB. Furthermore, the ultimate orientation of 3D modeling and simulation for the study of ALSB are briefly suggested.

Obstacle Avoidance of Unmanned Surface Vehicle based on 3D Lidar for VFH Algorithm (무인수상정의 장애물 회피를 위한 3차원 라이다 기반 VFH 알고리즘 연구)

  • In this paper, we use 3-D LIDAR for obstacle detection and avoidance maneuver for autonomous unmanned operation. It is aimed to avoid obstacle avoidance in unmanned water under marine condition using only single sensor. 3D lidar uses Quanergy's M8 sensor to collect surrounding obstacle data and includes layer information and intensity information in obstacle information. The collected data is converted into a three-dimensional Cartesian coordinate system, which is then mapped to a two-dimensional coordinate system. The data including the obstacle information converted into the two-dimensional coordinate system includes noise data on the water surface. So, basically, the noise data generated regularly is defined by defining a hypothetical region of interest based on the assumption of unmanned water. The noise data generated thereafter are set to a threshold value in the histogram data calculated by the Vector Field Histogram, And the noise data is removed in proportion to the amount of noise. Using the removed data, the relative object was searched according to the unmanned averaging motion, and the density map of the data was made while keeping one cell on the virtual grid map. A polar histogram was generated for the generated obstacle map, and the avoidance direction was selected using the boundary value.
