Tissue Factor Inhibitor from Aster scaber

  • Tissue factor (TF) is a cell surface receptor of coagulation factor Ⅶ and is the principal initiator of the vertebrate coagulation cascade. TF is found in high levels in some organs such as brain, lung and placenta, whereas blood monocytes, endothelial cells contain only trivial amount of TF when quiescent, and is stimulated to synthesize TF by infections or vascular lesions. TF is reported to be found in high levels in atherosclerotic plaques, cancer cells. TF activation in various cells in many infectious or immunologic diseases tells us the physiologic importance of TF. We screened many edible vegetables for TF inhibitor, by measuring the prothrombin time to detect the TF activity, and we picked Aster scaber to isolate the TF inhibitory substance. Aster scaber showed two kinds of anti thrombotic activity, one is TF inhibition and the other is elongation of plasma recalcification time. The anti thrombotic substances were found to be saponins which has echinocystic acid as aglycone.

  • Several forms of gangliosides have been separated from various types of biological matrices using cyclodextrin-modified capillary zone electrophoresis (CZE). Quantitative analysis of phospholipids from biological fluids was achieved by micellar electrokinetic capillary chromatography (MECC) using 35mM sodium dodecyl sulfate. Phosphorylation, one of the most important post-translational modifications of proteins at serine, threonine and tyrosine residues in small peptides were identified and quantitative analyses of phosphopeptides were performed. Seven different neuropeptides which are relative the pain reachanism in the vertebrate central nervons system were also separated by CZE.

  • Sturgeons and paddlefishes are frequently referred to as 'living fossils' among the actionpterygian lineage. They are increasingly facing threats to their existence because of various anthropogenic pressures. In this study, we present the complete mitogenome sequence of the Russian sturgeon Acipenser gueldenstaedtii (Acipenseriformes; Acipenseridae). The mitogenome showed highly homogeneous molecular features compared to previously known vertebrate mitogenomes. Phylogenetic tree inferred from concatenated protein-coding and tRNA genes unambiguously revealed the monophyly of A. gueldenstaedtii, Acipenser stellatus, and Huso huso. Genetic information of the endangered A. gueldenstaedtii will provide baseline data needed to develop molecular markers for stock identification and assessment of population diversity and also to develop future conservation strategies.

PromoterWizard: An Integrated Promoter Prediction Program Using Hybrid Methods

  • Promoter prediction is a very important problem and is closely related to the main problems of bioinformatics such as the construction of gene regulatory networks and gene function annotation. In this context, we developed an integrated promoter prediction program using hybrid methods, PromoterWizard, which can be employed to detect the core promoter region and the transcription start site (TSS) in vertebrate genomic DNA sequences, an issue of obvious importance for genome annotation efforts. PromoterWizard consists of three main modules and two auxiliary modules. The three main modules include CDRM (Composite Dependency Reflecting Model) module, SVM (Support Vector Machine) module, and ICM (Interpolated Context Model) module. The two auxiliary modules are CpG Island Detector and GCPlot that may contribute to improving the predictive accuracy of the three main modules and facilitating human curator to decide on the final annotation.

Identification and Expression Patterns of kif3bz during the Zebrafish Embryonic Development

  • We are reporting the identification, expression patterns, and possible biological functions of zebrafish kif3b (kif3bz) encoding 475 amino acids. Kif3Bz contains the kinesin motor domain, catalytic domain, KISc domain, and one single coiled coil domain. Phylogenetic analysis indicates that kif3bz is a highly conserved gene among the tested vertebrates. First of all, both maternal and zygotic messages of kif3bz were evenly distributed in the blastomeres at 2-cell stage. Its ubiquitous expression throughout the blastomeres continued till 40% epiboly. However, kif3bz transcripts became restricted in Kupffer's vesicle at tailbud and 6-somite stages. At 13-somite stage, kif3bz expression pattern became specific to the telencephalon, diencephalon, trigeminal placode, and somites. Such expression patterns were further intensified in the telencephalon, diencephalons, hind brain, pronephric ducts, optic vesicles, and spinal cord neurons in the 23-somite stage embryos, and last till 24 hpf. We discussed possible functions of Kif3Bz related to the vertebrate embryonic development.

  • 동물에서 맛을 느끼는 것과 의사소통이 라는 행위는 생명을 유지하는데 있어 매우 중요한 수단이다. 섭취 할 수 있는 것과 먹어서는 안 되는 물질을 가려내고, 위험으로부터 벗어 날 수 있는 정보를 가장 빠르게 전달 할 수 있는 수단이다. 이러한 기능을 가지고 있는 몸 안의 기관으로는 혀가 있는데, 다양한 근육과 신경의 분포를 이용해서 위에서 언급한 기능을 수행하게 된다. 혀의 다양한 기능과 특징적인 구조에도 불구하고 현재까지는 혀의 감각기관으로써의 기능과 구조에 대한 연구가 대부분인 실정이다. 물론 발생학적 측면에서 다양한 접근이 이루어지고 있으나, 이 또한 신경의 분화와 유도에 대해서 초점을 맞추고 있는 것 이 사실이다. 생물학에 있어서 가장 중요한 초점은 항상 그 구조와 기능이지만 이러한 구조와 기능을 보다 정확하게 이해하기 위해서는 발생학적인 측면에서의 접근이 반드시 필요하고, 특히 Epithelium과 Mesenchyme의 interaction으로 생기는 외배엽성 기원 기관의 하나로써 혀의 papillae에 대한 고찰을 하고자 한다 (Jung et al 1998). 또한 현재까지 진행되어진 신경분포와 감각기관으로의 혀의 역할과 발생 과정에 대해서도 알아보고 이러한 현시점의 발생학과 형태학을 이해하는데 필요한 하나의 Model system으로써의 혀에 대해 이야기하고자 한다.

A Phylogenetic Analysis for Hox Linked Gene Families of Vertebrates

  • The human chromosomes 2, 7, 12 and 17 show genomic homology around Hox gene clusters, is taken as evidence that these paralogous gene families might have arisen from a ancestral chromosomal segment through genome duplication events. We have examined protein data from vertebrate and invertebrate genomes to analyze the phylogenetic history of multi-gene families with three or more of their representatives linked to human Hox clusters. Topology comparison based upon statistical significance and information of chromosome location for these genes examined have revealed many of linked genes coduplicated with Hox gene clusters. Most linked genes to Hox clusters share the same evolutionary history and are duplicated in concert with each other. We conclude that gene families linked to Hox clusters may be suggestion of ancient genome duplications.

Cultivation of Cells on Cytodex 1 Microcarrier Culture (Cytodex 1을 이용한 Microcarrier 배양법에서의 세포의 증식성 조사)

  • Microcarrier culture technics are widely used for the massive production of the vertebrate animal cells. In this study, attempt was made to establish the microcarrier cell culture system using Cytodex. Various factors affecting the growth of cells on microcarrier culture were also discussed. In conclusion, the yield of cells in microcarrier culture was several times greater than those in roller bottle and flask culture methods, based on the volume of culture media.

  • In this research, a comprehensive study is performed upon the design of a quadruped walking robot. In advance, the walking posture and skeletal configuration of the vertebrate are analyzed to understand quadrupedal locomotion, and the roles of limbs during walking are investigated. From these, it is known that the forelimbs just play the role of supporting their body and help vault forward, while most of the propulsive force is generated by hind limbs. In addition, with the study of the stances on walking and energy efficiency, design criteria and control method for a quadruped walking robot are derived. The proposed controller, though it is simple, provides a useful framework for controlling a quadruped walking robot. In particular, introduciton of a new rhythmic pattern generator relieves the heavy computational burden because it does not need any computation on kinematics. Finally, the proposed method is validated via dynamic simulations and implementing in a quadruped walking robot, called AiDIN(Artificial Digitigrade for Natural Environment).

A Comparison of the Responses of Lower Vertebrate Intestines to Prostaglandin $E_1\;and\;E_2$

  • 1. The isolated strips of guinea-pig, fowl and reptiles (snake and tortoise) showed consistenly excitatory responses to $PGE_1\;and\;E_2$, which were dose-dependent. 2. Frog intestine revealed inhibitory responses to both $PGE_1\;and\;PGE_2$ except a small of $PGE_2$ (1-10 ng/ml) caused slight contraction. 3. The intestines of pieces showed inconsistent responses to $PGE_1\;and\;E_2$. In fresh-water fish(carp), $PGE_1$ produced relaxation under the dose of 50 ng/ml, and contraction by the large doses, but $PGE_2$ consistently caused contraction in dose-dependent manner. However, the strips of sea-water fish revealed the different responses to PGE compound: $PGE_1$ caused relaxation and $PGE_2$ conversly contraction even though in small degree. 4. These results that there are genera differences in the responses of the longitudinal strips of intestine to $PGE_1\;and\;PGE_2$ was assumed to be possibly correlated with evolutionally primitive function of gut.
