Hemp is cultivated to produce fiber and seeds for extracting vegetable oil. This study was carried out to obtain reliable data on fatty acid profile including ${\gamma}-linolenic$ acid (GLA) in seeds of hemp accessions with regard to suitability for both human nutrition and pharmaceutical purposes. The ratio of unsaturated fatty acid including ${\alpha}-linolenic$ acid were high but saturated fatty acid including palmitic acid were low in the seeds of hemp accessions, perilla and evening primrose. The ratio of GLA in fatty acid profile of evening primrose and hemp seeds were 12.5% and 2.1 %, respectively but not detected in perilla seeds. In the fatty acid profile of hemp seeds accessions, the range and coefficient of variation of GLA were $0.8%{\sim}3.8%$ and 42.9%, respectively which are highest among the fatty acid. Among the major characteristics and fatty acid ratios, flowering date and ${\alpha}-linolenic$ acid showed highly positive correlation but not showed any significant correlation between THC (Tetrahydrocannabinol) and any fatty acids. Among the fatty acid ratios, negative correlation were showed between palmitic acid and ${\gamma}-linolenic$ acid, and showed highly negative correlation between oleic acid and ${\alpha}-linolenic$ acid, linoleic acid and a-linolenic acid. The accessions of high GLA ratio were originated from Europe including Central Russia, and were short in flowering date and stem height.
Carthamus tinctorius L. (known as safflower) is a valuable oil plant whose importance is increasing rapidly in the world due to its high adaptation to arid regions. The seeds of this unique plant are especially used in edible oil, soap, paint, varnish and lacquer production. Its flowers are used in vegetable dye production and medicinal purposes beside its features as a coloring and flavoring in food. After the oil is removed, the remaining pulp and plant parts are used as animal feed, and dry straw residues are used as fuel. Beside all these features, its usage as a herbal medicinal plants for various diseases has gained importance on recent years. In this study, it was designed a plant metabolomic approach which transfers all the recent data processing strategies of untargeted metabolomics in clinical applications to the present study. Q-TOF LC/MS-based analysis of the extracts (70% ethanol, hexane, and chloroform) for both seed and flowers was performed using a C18 column (Agilent Zorbax 1.8 µM, 100 × 2.1 mm). Differences were observed in seed and fruit extracts and these differences were visualized using principal component analysis (PCA) plots. The total number and intersections of the peaks in the extracts were visualized using peak count comparison graph. Based on the experimental results, the number of the detected peaks for seeds was higher than the ones for the flowers for all solvent systems to extract the samples.
Seed viability is affected by storage conditions and rapid loss of viability in storage is the major cause for low germination. This study was carried out to examine the effect of packaging materials and storage temperature on seed germination rate over 10 years in two species (Capsicum annuum L. and Brassica rapa L. ssp. pekinensis) and determine effective storage conditions for maintaining seed viability. Seeds were packaged in aluminum poly pouches under vacuum, polyethylene bottles, and paper bags containing silica gel and stored under one of two controlled conditions ($15^{\circ}C$, RH 40% or $5^{\circ}C$, RH 30%) or at ambient condition. Seed germination was recorded at 6-month intervals for 10 years. The seeds of both species showed no decline in viability until 6.5 years at 15 or $5^{\circ}C$, irrespective of packaging materials. However, under ambient conditions, the seeds of chili pepper and Chinese cabbage in paper bags lost viability after 4 and 5 years, respectively. By contrast, seeds of both species in vacuum-aluminum poly pouches exhibited a 99% germination rate after 6 years under ambient conditions. Pepper seeds in the vacuum-aluminum poly pouches maintained a 93% germination rate after 10 years in ambient conditions. These results indicated that a special seed storage facility for maintaining viability of chili pepper and Chinese cabbage seed might not be essential and seed testing would not be necessary for 10 years, if chili pepper and Chinese cabbage seeds were packed in ambient/vacuum-aluminum poly pouches or $5^{\circ}C$/vacuum-aluminum poly pouches.
Ethanol extracts from 46 plants were tested for their fungicidal activity against six plant diseases consisting of Maynaporthe grisea, Rhizoctonia solani, Botrytis cinerea, Phytophthora infestans, Puccinia recondita, and Erysiphe graminis in the greenhouse studies. Strong activity at 5 and 10 mg/pot was produced from the extracts of Helianthus annuus flowers and Zea mays leaves against P. grisea. In a test with B. cineara, extracts of H. annuus leaves, H. annuus flowers, Chrysanthmum coronarium var. spatiosum, Cucurbita moschata seeds, Lycopersicon esculentum, Z. mays, and Z. mays leaves had strong activities at 5 mg/pot. In a test with P. recondita, strong activity was obtained from the extracts of Capsicum frutescens, C. moschata seeds, H. annuus seeds, L. esculentum, and Malva veticillata at 5 mg/pot. Against E. graminis, extracts of Cucumis sativus, H. annuus seeds, Salanum tuberosum, Z. mays, and Z. mays leaves produced strong activities at 10 mg/pot. All the extracts were ineffective against P. infestans and R. solani. Among seven extracts tested, the extracts of H. annuus leaves and flowers were highly effective against all the strains of B. cinerea resistant to carbendazim, procymidone, and diethofencarb. Furthermore, potent fungicidal activity was produced from the extracts of C. coronarium var. spatiosum and C. moschata seeds against the SSR, SRR, and RSR strains of B. cinerea, and Z. mays and Z. mays leaves against SSR and RSR. Extract of L. esculentum showed very strong activity only against RRS of B. cinerea.
'Sangwon', a new cultivar for vegetable soybean, was developed from the cross between 'Keunolkong' and 'Oshimamidori', and was released at the National Institute of Crop Science (NICS) in 2007. The goal to develop a vegetable soybean cultivar with green pod, early maturity, large seed size, high yield, lodging tolerance, and resistance to disease such as soybean mosaic virus (SMV). 'Sangwon' has light green pod, early maturity, large seed, short plant height, and lodging tolerance. 'Sangwon' has determinate growth habit, white flower, gray pubescence, and oval leaf shape. The matured seeds have a yellow seed coat with light brown hilum, and a yellow cotyledon. 'Sangwon' has 5.8 cm fresh pod length, 13.2mm fresh pod width, 69.5 g seed weight per 100 green seeds, 44.0% green seed protein content, and 14.8% green seed oil content. At the regional yield trials (RYT) for vegetable soybean from 2005 to 2007, 'Sangwon' shows strong resistance to soybean mosaic virus (SMV) and tolerance to lodging in fields. Fresh pods of 'Sangwon' were harvested at the beginning of August. In the same tests, fresh pod of 'Sangwon' (10.39ton/ha) yielded 5% higher than 'Hwaeomputkong' (9.90ton/ha).
Asia has 60% of the world population and only about 34.5% of the world arable land. However, Asians traditionally consume very large amount of horticultural produce and depend heavily on vegetables for the sources of horticultural food crop. The vegetable production in Asia is highly intensive and multiple and successive cropping are routinely practiced. Concomitant with the rapid economic growth in many east Asian countries, the demands for high-quality horticultural crop have been drastically increased among many Asians and their interest in high-quality products, outstanding cultivars, machinery and equipment, production technology has been sharply increased in recent years. It is therefore believed that establishing and maintaining closer and regular relations among scientists and societies could provide an excellent momentum for promotion of horticulture on a global basis and both the ISHS and the ASHS could play decisive roles for this kind of promotion.
An efficient transient expression system has been developed and characterized for the production of foreign genes in seedlings. The seedlings can be easily produced from commercial seeds used for vegetable sprouts. In principal, a chemical abrasive was employed to generate wounds in seedlings prior to vacuum-infiltration with Agrobacterium tumefaciens bearing the target gene. This optimized chemical wounding-assisted agro-infiltration process resulted in up to 15-fold increase in $\beta$-glucuronidase (GUS) enzyme activity. This procedure has been used efficiently to express hepatitis B surface antigen (HBsAg) protein in a transient mode. Therefore, seedlings with proper wounds can be suggested as a convenient tool for the production of useful recombinant proteins.
This experiment was conducted to determine the effect of priming duration and temperature on germinability of vegetable seeds. Priming chemicals were -0.50 MPa PEG 8000 for carrot, 50 mM $K_3PO_4$ for lettuce, 200 mM $KH_2PO_4$ for onion, and 100 mM $Ca(NO_3)_2$ for Welsh onion seeds. Priming temperature varied from $10^{\circ}C$ to $25^{\circ}C$, and priming duration varied from 12 hours to 5 days depending on crops. Percent germination of carrot seeds was significantly affected by the seed treatment and priming temperature. Priming at $20^{\circ}C$ for 3 days was most effective in reducing number of days to attain 50% of the final germination percentage (T50) and mean number of days to germination (MDG). Radicle protrusion in the priming solution was as high as 14% when primed at $20^{\circ}C$ for 4 days. Percent germination of lettuce seeds was affected significantly by the seed treatment and priming duration. Priming lettuce seeds at $20^{\circ}C$ for 2 days increased percent germination and shortened T50 and MDG without radicle protrusion. Percent germination of onion seeds was affected significantly by the seed treatment, priming duration, and temperature. Priming at either $10^{\circ}C$ or $15^{\circ}C$ for 3, 4, or 5 days did not affect percent germination of onion seeds, but was effective in reducing T50 and MDG. Decline in percent germination of onion seeds was precipitous as the length of priming duration increased from 3 to 5 days. Percent germination of welsh onion seeds was not affected by the priming duration and temperature. Priming at $15^{\circ}C$ for 4 days was most effective in reducing T50 and MDG, increased in duration of priming at $15^{\circ}C$ or $20^{\circ}C$ increased radicle protrusion.

Luffa cylindrica Roem (sponge gourd) belongs to the Cucurbitaceae family and has been cultivated as an ornamental plant in Korea. Recently, its cultivation area has been increased with an increase in demand for it as an ingredient in cosmetics, herbal medicines, and health supplements. We analyzed inorganic components of sap collected from land race sponge gourds. We also measured antioxidant enzyme activities and antimicrobial activities of the plant, seed, and sap to examine functional properties of sponge gourd. The sap of the sponge gourd contained high levels of K, Ca, P, and Mg, with the most abundant mineral in the sap being K ($470mg{\cdot}L^{-1}$). The amounts of Ca and Mg were 2 and 1.7 times more than those found in cucumber (Cucumis sativus), respectively. Ascorbate peroxidase was more active than catalase and superoxide dismutase in various plant parts of sponge gourd. Antioxidant activities were much lower in stems than in other plant parts such as leaves, roots, flowers, fruits, seeds, and sap. In addition, sap showed a very low level of antimicrobial activity against two food-borne pathogens, Vibrio parahaemdyticus and Propionibacterium acne, and none against the other eight tested food-borne pathogens. Antimicrobial activities against Candida albicans and Malassezia furfur, which causes dermatitis, appeared to be higher in sap than in other parts of sponge gourd plants. Overall, the antimicrobial activity against Malassezia furfur appeared to be higher than against Candida albicans.
Soybean is very useful crop to supply vegetable protein for human. However, cultivation arear of this economically important crop is gradually diminished in upland field. Hence, cultivation area of soybean is increased in paddy field. During the growth duration of soybean, excessive moisture injury is serious problem for sustainable production and supply. We investigated protein expression according to different period of seed soaking and germination after seed soaking. For comparison on expression of protein according to different condition, we performed two-dimensional electrophoresis. After electrophoresis analysis, we selected differentially expressed protein spots according to different condition such as soaking period and germination after soaking to identify protein function by using MALDI-TOF. Results revealed that pattern of expression of protein according to soaking period and germination after soaking were generally not different in major spots. However, degree of expression of protein in some protein spots was increased in accordance with decrease of soaking period. Especially, in Hwangkeum-Kong, Danyeop-Kon, and Pecking, the degree of expression of protein was remarkably increased for 4 days after soaking. But, according to germination after soaking, degree of expression of protein in germinated seeds of all cultivars was higher than un-germinated seeds. In results of MALDI-TOF analysis, specific proteins were identified by different soaking period such as Allergen Gly m Bd 28K, P24 oleosin isoform B. Also, in accordance with germination, degree of protein expression of the related protein, Gibberellin was increased in un-germinated seeds of Iksan-Kong. In ungerminated seeds of Sinpaldal-kong, proteins were identified as down-regulated by soaking such as ATP binding and Inhibitor II', proteinase.
