흑색종에서의 I-131표지 혈관내피세포성장인자 수용체2항체를 이용한 방사면역치료 평가 (Evaluation of the Radioimmunotherapy Using I-131 labeled Vascular Endothelial Growth Factor Receptor2 Antibody in Melanoma Xenograft Murine Model)

  • 목적: 혈관내피성장인자(VEGF)와 그 수용체는 종양의 성장과 전이에 매우 중요한 역할을 한다 3개의 수용체가 알려져 있는데 그 중에서도 VEGFR2 (Flk-1/KDR)가 종양 angiogenesis에 매우 밀접하게 관련한 것으로 알려져 있다. 본 연구에서는 마우스 Flk-1 항체로 알려진 DC101에 I-131을 표지하여 매우 공격적인 종양으로 알려진 흑색 종의 치료 정도를 확인해 보고자 하였다. 방법: 하이브리도마 세포에서 Flk-1 항체인 DC101을 분리하여 western blot, ELISA, maldi-tof 방법을 이용하여 순도를 확인하고 항체 에 chloramin T를 이용하여 I-131을 표지하였다. 누드마우스에 B16F10세포를 주사하여 흑색종 모델을 만들고 평균 $200-250\;mm^3$으로 키워 $^{131}I$-DC101을 주사하여 영상과 시간별 장기섭취율(%ID/g)을 비교하고, 종양내 Flk-1 발현을 확인하기 위하여 면역염색 등을 시행하였다. 흑색종 동물모델을 5개 군으로 나누어 각각 치료를 시행하였다. 1군은 PBS만을, 2군은 $^{131}I$-DC101 $50\;{\mu}g(200\;{\mu}Ci)$을, 3군은 DC101 $50\;{\mu}g$을, 4군은 $^{131}I$-DC101 $30\;{\mu}g(200\;{\mu}Ci)$을, 5군은 $^{131}I$-DC101 $15\;{\mu}g(200\;{\mu}Ci)$을 각각 매 3일 ${\sim}$ 4일마다 주사하고 전체 5회를 주사하였고 종양볼륨을 측정하였다. 결과: $^{131}I$-DC101을 흑색종 모델에 정맥주사하고 78시간까지 영상을 얻은 결과 시간에 따라 종양섭취가 증가 하는 영상을 보였다. 시간대별 장기섭취를 정확히 확인하기 위하여 1시간, 6시간, 24시간, 48시간, 72시간 장기섭취율을 비교한 결과 시간에 따라 혈액내 방사능치가 서서히 감소하였고 다른 장기의 섭취도 시간에 따라 감소하였고 종양의 섭취는 48시간까지 증가하였다가 그 이후는 감소하였다. 흑색종 동물모델에 $^{131}I$-DC101 치료를 시행한 결과 3번째 주사까지는 각군간의 유의한 차이를 보이지 않다가 4번째 주사를 시행한 때부터 1군과 2군, 또는 1군과 4군간의 유의한 차이를 보이기 시작했다. 또한 5번째 주사 이후에는 5군에서도 유의한 차이를 보여 I-131 에 의한 효과가 뚜렷해짐을 확인하였다. 결론 마우스 Flk-1 항체로 알려진 DC101을 흑색종 모델에 정맥내 주사하였을 때, 종양성장억제 효과를 보이지 않는 항체양에서도 I-131을 표지하여 치료를 시행했을 경우에는 효율적인 종양성장억제 효과를 보였다.

Anti-Angiogenesis Effects Induced by Octaminomycins A and B against HUVECs

  • In the course of studies to discover natural products with anti-angiogenic properties, two cyclic octapeptides, octaminomycins A (1) and B (2), were isolated from the cultures of Streptomyces sp. RK85-270. Octaminomycins suppressed the vascular endothelial growth factor (VEGF)-induced proliferation, adhesion, tube formation, migration, and invasion of HUVECs. Anti-angiogenic activity was futher confirmed in vivo by the chicken chorioallantoic membrane assay. We also identified that 1 and 2 inhibited the phosphorylation of VEGF receptor 2, AKT, and ERK1/2 and the expression and activities of MMP-2 and MMP-9. These results suggest that 1 and 2 may serve as potential scaffolds for the development of therapeutic agents to angiogenesis-dependent diseases.

Citron Essential Oils Alleviate the Mediators Related to Rosacea Pathophysiology in Epidermal Keratinocytes

  • Background: Citron is well known for an abundance of antioxidative and anti-inflammatory ingredients such as vitamin C, polyphenol compounds, flavonoids, and limonoids. Objective: In this study, we aimed to evaluate the effects of citron essential oils on rosacea mediators in activated keratinocytes in vitro. Methods: Normal human epidermal keratinocytes (NHEKs) were stimulated with $1{\alpha}$, 25-dihydroxyvitamin $D_3$ ($VD_3$) and interleukin 33 (IL-33) with LL-37 to induce rosacea mediators such as kallikrein 5 (KLK5), cathelicidin, vascular endothelial growth factor (VEGF), and transient receptor potential vanilloid 1 (TRPV1). These mediators were analyzed by performing reverse-transcription polymerase chain reaction (PCR), quantitative real-time PCR, immunocytofluorescence and enzyme-linked immunosorbent assay after NHEKs were treated with citron seed and unripe citron essential oils. Results: The messenger RNA (mRNA) and protein levels of KLK5 and LL-37 induced by $VD_3$ were suppressed by citron seed and unripe citron essential oils. Furthermore, the mRNA and protein levels of VEGF and TRPV1 induced by IL-33 with LL-37 were also suppressed by citron essential oils. Conclusion: These results show that citron essential oils have suppressive effects on rosacea mediators in activated epidermal keratinocytes, which indicates that the citron essential oils may be valuable adjuvant therapeutic agents for rosacea.

Tivozanib-induced activation of the mitochondrial apoptotic pathway and suppression of epithelial-to-mesenchymal transition in oral squamous cell carcinoma

  • The potential of tivozanib as a treatment for oral squamous cell carcinoma (OSCC) was explored in this study. We investigated the effects of tivozanib on OSCC using the Ca9-22 and CAL27 cell lines. OSCC is a highly prevalent cancer type with a significant risk of lymphatic metastasis and recurrence, which necessitates the development of innovative treatment approaches. Tivozanib, a vascular endothelial growth factor receptor inhibitor, has shown efficacy in inhibiting neovascularization in various cancer types but has not been thoroughly studied in OSCC. Our comprehensive assessment revealed that tivozanib effectively inhibited OSCC cells. This was accompanied by the suppression of Bcl-2, a reduction in matrix metalloproteinase levels, and the induction of intrinsic pathway-mediated apoptosis. Furthermore, tivozanib contributed to epithelial-to-mesenchymal transition (EMT) inhibition by increasing E-cadherin levels while decreasing N-cadherin levels. These findings highlight the substantial anticancer potential of tivozanib in OSCC and thus its promise as a therapeutic option. Beyond reducing cell viability and inducing apoptosis, the capacity of tivozanib to inhibit EMT and modulate key proteins presents the possibility of a paradigm shift in OSCC treatment.

Vascular Endothelial Growth Factor (VEGF) Gene Polymorphisms and Breast Cancer Risk in a Chinese Population

  • Vascular endothelial growth factor (VEGF) is a potent regulator of angiogenesis and thereby involved in the development and progression of solid tumours. Associations between three VEGF gene polymorphisms (-634 G/C, +936 C/T, and +1612 G/A) and breast cancer risk have been extensively studied, but the currently available results are inconclusive. Our aim was to investigate associations between three VEGF gene polymorphisms and breast cancer risk in Chinese Han patients. We performed a hospital-based case-control study including 680 female incident breast cancer patients and 680 female age-matched healthy control subjects. Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) analysis was performed to detect the three VEGF gene polymorphisms. We observed that women carriers of +936 TT genotypes [odds ratio (OR) =0.46, 95% confidence interval (CI) = 0.28, 0.76; P=0.002] or 936 T-allele (OR=0.81, 95% CI= 0.68, 0.98; P=0.03) had a protective effect concerning the disease. Our study suggested that the +1612G/A polymorphism was unlikely to be associated with breast cancer risk. The -634CC genotype was significantly associated with high tumor aggressiveness [large tumor size (OR=2.63, 95% CI=1.15, 6.02; P=0.02) and high histologic grade (OR=1.47, 95% CI= 1.06, 2.03; P=0.02)]. The genotypes were not related with other tumor characteristics such as regional or distant metastasis, stage at diagnosis, or estrogen or progesterone receptor status. Our study revealed that the VEGF -634 G/C and +936 C/T gene polymorphisms may be associated with breast cancer in Chinese Han patients.

Supplementing punicalagin reduces oxidative stress markers and restores angiogenic balance in a rat model of pregnancy-induced hypertension

  • Pre-eclampsia (PE) is a pregnancy disorder that is characterised by severe hypertension and increased risks of foetal and maternal mortality. The aetiology of PE not completely understood; however, maternal nutrition and oxidative stress play important roles in the development of hypertension. The treatment options for PE are currently limited to anti-hypertensive drugs. Punicalagin, a polyphenol present in pomegranate juice, has a range of bioactive properties. The effects of supplementation with punicalagin on angiogenesis and oxidative stress in pregnant rats with induced hypertension were investigated. The pregnant rats were randomly divided into five experimental groups (n=12 per group). Hypertension was induced using an oral dose of NG-nitro-L-arginine methyl ester (L-NAME, 50 mg/kg/day) on days 14-19 of pregnancy. Punicalagin (25, 50 or 100 mg/kg) was given orally on days 14-21 of pregnancy. Punicalagin treatment at the tested doses significantly reduced diastolic, systolic, and mean arterial blood pressure in L-NAME treated rats from day 14. Punicalagin also restored angiogenic balance by increasing the expression of vascular endothelial growth factor and downregulating vascular endothelial growth factor receptor-1/fms-like tyrosine kinase-1. Punicalagin, significantly increased the placental nitric oxide levels as compared to PE group. The increased levels of oxidative stress in rats with PE were markedly decreased by treatment with punicalagin. Punicalagin at the tested doses markedly (p<0.05) enhanced the placental antioxidant capacity in L-NAME-treated rats. The raised catalase activity observed following L-NAME induction was significantly (p<0.05) and restored to normal activity levels in punicalagin treatment. Further, 100 mg dose of punicalagin exhibited higher protective effects as compared to lower doses of 25 and 50 mg. This study shows that supplementation with punicalagin decreased blood pressure and oxidative stress and restored angiogenic balance in pregnant rats with induced PE.

Differential antiangiogenic and anticancer activities of the active metabolites of ginsenoside Rg3

  • Background: Epimers of ginsenoside Rg3 (Rg3) have a low bioavailability and are prone to deglycosylation, which produces epimers of ginsenoside Rh2 (S-Rh2 and R-Rh2) and protopanaxadiol (S-PPD and R-PPD). The aim of this study was to compare the efficacy and potency of these molecules as anti-cancer agents. Methods: Crystal violet staining was used to study the anti-proliferatory action of the molecules on a human epithelial breast cancer cell line, MDA-MB-231, and human umbilical vein endothelial cells (HUVEC) and compare their potency. Cell death and cell cycle were studied using flow cytometry and mode of cell death was studied using live cell imaging. Anti-angiogenic effects of the drug were studied using loop formation assay. Molecular docking showed the interaction of these molecules with vascular endothelial growth factor receptor-2 (VEGFR2) and aquaporin (AQP) water channels. VEGF bioassay was used to study the interaction of Rh2 with VEGFR2, in vitro. Results: HUVEC was the more sensitive cell line to the anti-proliferative effects of S-Rh2, S-PPD and R-PPD. The molecules induced necroptosis/necrosis in MDA-MB-231 and apoptosis in HUVEC. S-Rh2 was the most potent inhibitor of loop formation. In silico molecular docking predicted a good binding score between Rh2 or PPD and the ATP-binding pocket of VEGFR2. VEGF bioassay showed that Rh2 was an allosteric modulator of VEGFR2. In addition, SRh2 and PPD had good binding scores with AQP1 and AQP5, both of which play roles in cell migration and proliferation. Conclusion: The combination of these molecules might be responsible for the anti-cancer effects observed by Rg3.

  • 식생활의 서구화와 대장암 검진의 증가로 인하여 국내의 대장암 유병률은 지속적으로 증가하고 있으며, 최근 보고에 의하면 남자에서는 종양 발생률 2위, 여자에서는 종양 발생률의 3위를 차지하고 있다. 비록 대장내시경이 대장암의 진단과 선별검사에 아주 효과적인 방법이지만, 여전히 대장암의 20-25%는 이미 진단 당시에 전이를 동반하고 있는 것으로 되어 있다. 최근 10년 동안 이러한 전이성대장암의 고식적인 치료로 irinotecan과 oxaliplatin 등의 약제들의 개발과 이들의 다양한 조합에 관련된 연구들이 보고되어 왔으며, 분자생물학적인 발전에 힘입은 표적 치료제의 개발과 이에 대한 다양한 연구들은 향후에도 진행성대장암 환자들의 종양 반응률과 생존기간을 증가시킬 것으로 기대된다.

칸나비디올(CBD)의 항산화 활성 및 인간 모유두 세포 증식에 미치는 영향 (Antioxidant Activity of Cannabidiol (CBD) and Effect on Its Proliferation in Human Dermal Papilla Cells)

  • 최근 세계 여러 나라에서 대마초 및 대마제품을 합법화하고 대마를 이용한 다양한 치료법에 대한 연구가 활발히 진행되고 있다. 그러나 대마에는 생물학적 효과가 아직 확립되지 않은 여러 화합물들이 포함되어 있다. 본 연구에서는 인간 모유두 세포(HDPC)의 모발 성장에 대한 칸나비디올(CBD)의 효과를 조사하였다. 2,2'-Azino-bis (3-ethylbenzothiazolin-6-sulfonic acid) (ABTS) 및 2,2-diphenyl-1-picrylhydrazyl (DPPH) 라디칼 소거 분석법을 활용하여 CBD의 항산화 활성을 측정하였다. 모유두 세포에서 CBD의 세포생존률은 WST-1 분석법으로 측정하였다. CBD 처리에 의한 모유두 세포에서 모발 성장과 관련된 인자의 발현은 real-time PCR 및 western blot으로 측정하였다. CBD의 항산화 활성 측정결과, DPPH 및 ABTS 자유 라디칼 소거 활성의 IC50 값은 각각 15.46±0.24 μM 및 13.90±0.06 μM으로 뛰어난 활성 산소 제거능을 나타냈다. CBD 처리군은 대조군에 비해 세포 증식이 증가하는 경향을 나타냈다. 또한 모유두 세포에서 Real-Time PCR과 Western blotting을 통해 모발 성장 관련 인자를 측정한 결과, CBD 처리로 인하여 성장 관련 인자들이 증가하는 것으로 나타났다. 종합적으로, 항산화 활성이 높은 CBD는 모유두 세포에서 세포 증식을 증가시키고 모발 성장 관련 인자들을 긍정적으로 조절합니다. 이러한 결과는 CBD가 탈모증에 대한 잠재적으로 활용될 수 있음을 시사한다.

Insulin Induces Transcription of VEGF in Arnt-dependent but HIF-l$\alpha$-Independent Pathway

  • Hypoxia is a pathophysiological condition that occurs during injury, ischemia, and stroke. Hypoxic stress induces the expression of genes associated with increased energy flux, including the glucose transporters Glutl and Glut3, several glycolytic enzymes, nitric oxide synthase, erythropoietin and vascular endothelial growth factor. Induction of these genes is mediated by a common basic helix-loop-helix PAS transcription complex, the hypoxia-inducible factor-l${\alpha}$ (HIF-1${\alpha}$)/ aryl hydrocarbon receptor nuclear translocator (ARNT). Insulin plays a central role in regulating metabolic pathways associated with energy storage and utilization. It triggers the conversion of glucose into glycogen and triglycerides and inhibits gluconeogenesis. Insulin also induced hypoxia-induced genes. However the underlying mechanism is unestablished. Here, we study the possibility that transcription factor HIF-1${\alpha}$ is involved in insulin-induced gene expression. We investigate the mechanism that regulates hypoxia-inducible gene expression In response to insulin We demonstrate that insulin increases the transcription of hypoxia- inducible gene. Insulin-induced transcription is not detected in Arnt defective cell lines. Under hypoxic condition, HIF- l${\alpha}$ stabilizes but does not under insulin treatment. Insulin-induced gene expression is inhibited by presence of PI-3 kinase inhibitor and Akt dominant negative mutant, whereas hypoxia-induced gene expression is not. ROS inhibitor differently affects insulin-induced gene expressions and hypoxia-induced gene expressions. Our results demonstrate that insulin also regulates hypoxia-inducible gene expression and this process is dependent on Arnt. However we suggest HIF-l${\alpha}$ is not involved insulin-induced gene expression and insulin- and hypoxia- induces same target genes via different signaling pathway.
