Histopathological Studies on the Effect of Persimmon Leaves on Cadmium Poisoning in Mice (감잎이 마우스의 카드뮴 중독에 미치는 병리조직학적 관찰)

  • This study was undertaken to find out the effect of persimmon leaves on histopathological changes of cadmium toxicity in mice. Seventy two BALB/c mice of male were divided into a control group(A) and five experimental groups (B, C, D, E, F) : group A received tap water and basal diet, group B received tap water and diet supplemented with 3% persimmon leaves alone, group C received basal diet and 300 ppm cadmium, group D, E and F received basal diet supplemented with 1, 3% and 7% persimmon leaves and 300 ppm cadmium respectively. Cadmium dissolved in tap water was used, and the persimmon leaves were mixed with feed. All mice were dissected on the 84th day. Pathological changes in liver, kidney, cortical osseous tissue of femoral shaft, bone trabecular of femur, and epiphyseal cartilage plate of femur were observed. Group B showed no significant changes as the control group. But group C showed the unclearness of specific cells in liver, the loss of architecture and necrosis of hepatocyte, degeneration and necrosis of renal convoluted tubules, desquamation and vacuolization of the greater part of the renal tubular epithelium, marked thinning of the cortical osseous tissue in femoral shaft, reduction of cancellous bone volume and decreaswe of trabecular number, and marked thinning of epiphyseal cartilage plate in femur. On the other hand, persimmon leaves-treated group showed a little convalescent changes an maintained their normal architectures in liver, kidney, cortical osseous tissue of femoral shaft, bone trabecular of femur, and epiphyseal cartilage plate of femur.

  • This paper was carried out to observe prostaglandin $F_{2{\alpha}}$-induced morphological changes in the placenta. White mice received intramuscular injections of $PGF_{2{\alpha}}$(containing dinoprost tromethamine 0.5mg/ml, Upjohn Co.) in once or twice with doses of 0.1ml on 10th to 18th day of their respective pregnancies, the histological changes of the placentae and ovaries were observed with light microscope. Abortion within 21 to 51 hours following $PGF_{2{\alpha}}$ administration occurred in the pregnant mice. Vacuolization of trophospongial cells, giant cells and myometrium under decidua basalis, spherical acidophilic inclusion bodies of various sixtes in trophospongial cells and giant cells. hydropic degeneration and necrosis of labyrinthine trophoblasts and yolk cells, and infltration of neutrophils in the placenta and the uterus were observed. In addition, there were decrease in glycogen of the placental labyrinth and the visceral yolk sac, but increase in glycogen deposit of mesometrial myometrium. Atrophy and increase in number of large lipid droplets of the luteal cells and proliferation of fibroblasts were also recognized in the corpora lutea.

Histological Responses of the Antarctic Bivalve Laternula elliptica to a Short-term Sublethal-level Cd Exposure

  • To develop fast and sensitive biomarkers for metal exposures in Antarctic marine organisms we examined histological alterations of an Antarctic sentinel bivalve species Laternula elliptica following a short-term exposure to a sublethal-level of Cd. Distinct histological alterations of tissues and cells of the gills, kidneys, and digestive glands were observed after 8-to 16-hours of exposure to Cd while an increase of Cd concentrations in tissues was not detectable. Most alterations were highly localized in the epithelium of the three tissues; epithelia were found to be detached from the remaining tissue parts. In addition ultra-structural changes such as cytosolic vacuolization, dilation of nucleus and rER membranes were detected in all three tissues, which suggested that the clams are subject to sublethal stresses. Thus, histological and ultrastructural changes on localized tissue parts were rapid and sensitive, suggesting that they may serve biomarkers for Cd exposures. Linkages between the shown ulrastructural changes and higher biological organization level responses are to be established by longer-term exposure experiments.

Study on Changes in Endogenous Stem Cells in the Salivary Gland of Streptozotocin-induced Diabetic Rats

  • Type1 diabetes mellitus (DM) is generally known to be caused by destruction of insulin-producing pancreatic ${\beta}$ cells or an immune-related problem. Polydipsia is a representative symptom of DM, and it has been reported that this condition is closely related to xerostomia and is considered that hyposalivation from the salivary gland results in this phenomenon. Although various studies have reported that induction of diabetes reduces endogenous stem cells in other organs (heart, brain etc.), diabetes-related changes in endogenous stem cells in the salivary gland have not yet been well established. Therefore, in this study, to verify the change in salivary gland stem cells after diabetes, salivary gland tissues in the control and diabetes-induced groups were processed by histochemistry (Masson's trichrome staining) for morphological analysis, TUNEL assay for cell death, and immunohistochemistry (Ki-67 and c-Kit) for cell proliferation and maturation. Diabetes induced by STZ leads to vacuolization, apoptosis, and reduction in proliferating cells/salivary gland stem cells in salivary glands of rats. This result suggests that diabetes may be associated with reduction in salivary gland function such as degeneration and inhibition of regeneration in the salivary gland.

The Effect of Rutin on Antioxidant and Anti-inflammation in Streptozotocin-induced Diabetic Rats

  • The study examined the antioxidant activities and anti-inflammatory effects of rutin from the streptozotocin (STZ)-induced diabetic rats. Results revealed that the levels of plasma glucose and serum glucose were remarkably higher in the STZ-treated group compared to other groups and were significantly reduced in the STZ+rutin treated group compared to the STZ-treated group. In terms of weight, it significantly increased in all experimental groups during the experiment period except for STZ-induced diabetic group. The weight of the STZ-treated group was remarkably reduced compared to other groups. Regarding the weight of each body organ, the STZ-treated group showed higher organ weight compared to the other groups while STZ+rutin-treated group showed significantly reduced kidney and liver weights compared to those of STZ-treated group. In the pancreas tissue of the STZ-treated group, ${\beta}$-cell destruction and vacuolization were observed. Inflammation in the heart, liver, kidney, and retina tissues were also vividly recorded. In the STZ+rutin administered group, the heart and retina tissues were shown to be preserved normally while the liver and kidney tissues showed reduced histopathology in general compared to the STZ-treated group. Conclusionally, the rutin has the effect on the antioxidant activities and anti-inflammation in the STZ-induced diabetic rats.

A HISTOLOGIC STUDY ON THE RESPONSES OF PULP IN EXPERIMENTAL TOOTH MOVEMENT OF WHITE RATS (흰쥐의 실험적 치아이동시 치수의 반응에 관한 조직학적 연구)

  • The author investigated on the responses of pulp and dentine following tooth movements. The material consisted of fifty-four intact teeth from twenty-seven adult white rats. The half of the teeth were employed as controls and the other half served as experimental group. These teeth were moved with forces ranging from 30 grams to 120 grams for from 3 to 7 days. All these were extracted immediately after the force was relieved. The main pulp changes in the experiment were vacuolization of the pulp tissue and circulatory disturbances. The magnitude of the force had an important role. In addition to these changes, the resorption in dentine and cementum was observed, which was related to the magnitude of the force and the duration of experiment.

Apoptosis of MCF7 Cells Treated with PKC Inhibitors and Daunorubicin

  • The present study was performed to observe the role of protein kinase C (PKC) inhibitors (H-7, staurosporine) and daunorubicin in the cell death process of MCF7 cells; and examined whether or not the type of induced cell death was apoptosis. The usefulness of the combined therapy of PKC inhibitors and daunorubicin to improve the adverse effect of daunorubicin was also investigated. Cell death was induced by treatment with PKC inhibitors or daunorubicin. Characteristic morphologic features of cell shrinkage, chromatic condensation, and cytoplasmic vacuolization were observed. These treatments also stimulated the cleavage of poly-(ADP-ribose) polymerase (PARP), an early event in apoptosis. With slight differences in the percentage of apoptosis-induced cells, staurosporine, H-7 and daunorubicin effectively induced apoptosis in MCF7 cells. Furthermore, combined treatment of PKC inhibitors and daunorubicin significantly drove the cells into an apoptotic state. Hence, our results revealed the possible therapeutic value of combined therapy for the prevention of drug resistance and adverse side effects.

Pathological findings on encephalomyocarditis virus infections of swine in Korea (국내에서 발생한 돼지 뇌심근염 바이러스 감염증의 병리학적 관찰)

  • Pathological studies by light and electron microscope were carried out on the twenty piglets naturally affected by encephalomyocarditis virus infection. Gross findings included pale or yellow, small necrotic foci on myocardium, together with pulmonary edema and liver congestion in some cases. On light microscopy, nonsuppurative interstitial endocarcitis, epicarditis and myocarditis, myocardial infarction, and dystrophic calcification or fibtosis were observed in heart. Perivascular cuffings, gliosis and nonsuppurative meningitis were appeared in brain. Focal or diffuse necrosis with mononuclear cell infiltration in lacrimal gland and multifocal necrosis in liver were observed in some cases. Congestion and edema of lung, hyperemia, hemorrhage and deletion of lymphocytes of lymph nodes and spleen were recognized. On electron microscopy, severe swelling and vacuolization of mitochondria and sarcoplasmic reticulum, large intracellular vacuolation and edema, separation and fragmentation of myofibrils were observed. Virus particles were seen in the sarcoplasm of degenerated cardiac muscle cell.

A Study on the Fine Structural Changes of Porcine Ovarian Follicles during Atresia (돼지 난소내 여포의 폐쇄에 따른 미세구조의 변화에 관한 연구)

  • This experiment has been done in order to study the correlation between the ultrastructure changes and the atresia phenomenon of the follicles in porcine ovary. The ovaries were assorted according to the presence or absence of corpus luteum. Thereafter, the follicles were classified into normal, pyknotic, necrotic and cystic groups by atretic characteristics on the histological observation, and then their ultrastructures were examined with an electron microscope. The results were as followings. 1. In normal group, granulosa cells represented the ultrastructural characteristics of protein-synthesizing cells. Since the initiation of atresia, the line structure of granulosa cells showed many of the characteristic features of steroid-secreting cells, followed by gradual pyknosis. 2. In necrotic group of the ovary without corpus luteum, the theca interns became hypertrophic and displayed the ultrastructural features of active steroidsecreting cells. But this phenomenon was not seen in the follicles of the ovary with corpus luteum. 3. Degenerative changes of cumulus cells were similar to those of granulosa cells, and the degenerating oocytes showed the degeneration of cellular organelles, cytoplasmic vacuolization and disappearance of microvilli on the surface. The degeneration of granulosa cells tended to procede that of oocytecumulus complex in the follicles of the ovary having no corpus luteum, but this tendency was reversed in the case of presence of corpus luteum.   • Structures of the adventitious root meristem induced from callus culture of tobaco (Nicotiana tabacum cv. NC 82) leaves were investigated by light and transmission electron microscopy. Structural differences between in vitro root and callus cells were also examined by the microscopy. The submicroscopic features of the in vitro root cells were as follows. Intercellular spaces were not developed and nuclei with two nucleoli were observed occasionally. Plasmodesmate were found in groups or sing1y on transverse and longitudinal walls. Amyloplast solely filled with starch grains, with one to five electron - dense bands, was surrounded by single membrane. in the callus cells, vacuolization of central part in the cytoplasm having mitochondria with swollen cristae and starch grains like those of in vitro root cells was a distinct feature. Vesicles which were found between cell wall and plasma membrane may be arisen by a process of protoplasmic invagination. By comparing of ultrastructures between the cells of callus and in vitro roots we found that the distinct differences lied on thickened cell walls and hypertrophed vacuoles in the former, and less thickened cell walls and several small vacuoles in the later.
