Isolation of Ethanol-tolerant Strains of Yeast in Relation to Their Tolerant Mechanism (에탄올 내성 효모의 선별과 그의 에탄올 내성 기작)

  • The selection of ethanol-tolerant strains was applied to enrichment culture of YPD broth medium containing various concentrations of ethanol. Isolates were identified to be Saccharomyces cerevisiae, the others as S. dairensis, S. exiguus, S. telluris, Saccharomycodes ludwigii, Schwanniomyces occidentalis var. occidentalis and Zygosaccharomyces florentinus. Among isolates S. cerevisiae YO-1 was screened as having the highest ethanol tolerance and produced 18% (v/v) ethanol after 4 days fermentation. The change of fatty-acyl residues represents that a progressive decrease in fatty-acyl unsaturation and a proportional increase in saturation in phospholipids of yeast cells during fermentation affected the yeast viability. Supplementation ethanol to the cultures led to an increase of unsaturated fatty-acyl residues, especially $C_{16}$ or $C_{18}$ residues, along with a decrease in the proportion of saturated residues in cellular phospholipids. Increasing the amount of soy flour led to an increase in the maximum number of viable yeast cells and ethanol production. It was possible in 4 days to reach 21% (v/v) ethanol by adding 4% soy flour as source of unsaturated fatty-acyl residues to the fermentation medium. Soy flour not only increased yeast population but also enhanced the physiological properties of yeast cells to be ethanol tolerant in the anaerobic culture.

Perilla Leaf Fertilization Effect of Fertilizer by Chlorella and Seafood By-product Fermentation (클로렐라 및 수산부산물 발효 비료의 들깻잎 시비효과)

  • The effects of amino acid and/or urea liquid fertilizer application on the growth and phytochemicals of Perilla leaves were summarized as follows; The fresh weight of the perilla leaves was in the order of CF, CL, KLF, and control, and 39.7 g, 37.4 g, 36.5 g and 32.3 g were measured. The plant height increased by 71.6 cm in the CF treatment than in the control(54.6 cm). The number of nodes was 14.3 node in CF treatment and 19% more than control(12 node). The vitamin C content tends to be increased by fertilizing the amino acid solution in the perilla leaf. The components of polyvalent unsaturation of n-6 origin were measured in CF treatment, KLF treatment, and control in 10.19 mg, 10.18 mg, and 9.38 mg per 100 g, respectively. Glutamic acid, aspartic acid, leucine, arginine, alanine and lysin were contained in perilla leaf amino acid. Glutaminic acid content was found to be 455.1 mg, 495.6 mg, and 478.8 mg in the control, KLF and CF treatment per 100 g, respectively. Effective nutrition management using amino acid fertilizer optimizes crop yield and profitability, it is important to reduce the negative environmental risks of using fertilizer.

Effects of Dietary Fat with Various Ratios in n-6/n-3 and P/S on Immune Responses of Rats at Different Age (나이가 다른 흰쥐에서 n-6/n-3 비율과 P/S 비율이 다른 식이지방이 면역능력에 미치는 영향 연구)

  • Effect of age and dietary fatty acid composition on immune responses were investigated in Sprague-Dawley male rets. The animals weighing 88.6$\pm$2.2g were fed 10% dietary fat (W/W, 20% of calorie) with P/S ratio of 0.5, 1 and 2, and in each group, there were three different levels of n-6/n-3 fatty acid ratio; 2, 4 and 8(3$\times$3). The experimental periods were 1 month, 6 months and 12 months. The results of this study were; 1) Weights of thymus and spleen were significantly reduced with increasing age, moreover thymus weights were reduced with increasing the degree of unsaturation in dietarty fatty acid at 12 month. 2) The proportion of splenic lymphocyte in the total T-cell was increased with increased with age. The proportion of helper T-cell was not changed, while the proportion of suppressor T-cell was decreased. Thus at 12 month, the ratio of helpe $r^pressor T-cell was appeared to increase significantly, and showed the tendency to increase by consuming the low amount of dieraty n-3 fatty acid. 3) Proliferation stimulated by Con A or PWM reduced significantly at 12 month in which are high in dietary P/S ratio, representing the similar pattern with decrease in thymus weight. 4) Natural killer cell activities were shown significantly higher at 1 month than those at 6 month or 12 month.th.

Effect of Dietary n6 Gamma-linolenic Acid and n3 Alpha-linolenic Acid on Plasma Lipid Composition and Platelet Aggregation in Human Subjects (사람에서 식이의 n6 Gamma-Linolenic Acid와 n3 Alpha-Linolenic Acid가 혈장 지질조성과 혈소판 응집반응에 미치는 영향)

  • To observe the effect of dietary n6 linoleic acid, n6 gamma-linolenic acid and n3 alphalinolenic acid aon plasma lipid composition and platelet aggregation, twenty college women were divided into 4 groups and treated for 2 weeks with experimental diets supplying fat at 23% cal which were different only in fatty acid composition. Dietary fat was corn oil(CO) as a source of n6 linoleic acid(LA), perilla oil(PO) for n3 alpha-linolenic acid(ALA) and evenign primrose oil(EPO) for n6 gamma-linolenic acid(GLA). Plasma cholesterol level was slightly decreased by PL(13.5g) but significantly increased by equal amount of CO. However, there was similar hypocholeaterolemic effect when double amount of CO(27.0g), was supplemented. Therefore, total fat unsaturation may be more important factor for plasma cholesterol-lowering effect than the structure of fatty acid itself. Plasma cholesterol level was not lowered by supplement of GLA in CO diet. There was similar trend in hypotriglyceridemic effect by PO and CO as in plasma cholesterol. Plasma TG level was rather increased but not significantly by GLA supplement to CO diet. Overall, plasma lipid-lowering effect was greater by ALA than LA and GLA effect was not greater than by LA. GLA supplement did not significantly improve lipid compositions to prevent against CHD. There was no significant change both in fatty acid composition in platelet and ADP-induced platelet aggregation by GLA supplement to corn oil diet and by ALA in PO diet in young women.

Effect of Individual Fatty Acids on Synthesis and Secretion of Apolipoprotein and Lipoprotein in hep-G2 Cells

  • The effects of individual fatty acids, differing in their degree of unsaturation(18:0, 18:1, 18:2 and 18:3) on the biosynthesis and secretion and lipids were investigated in Hep-G2 cells. Synthesis of apolipoprotein was measured by the incorporation of 3H-leucine into apolipoprotein(d<1.21g/ml) and synthesis of lipids was measured by the incorporation of 3H-glycerol and 14C-acetate into various lipid classes. Inclusion of 1.0mM of each fatty acids into the culture medium significantly increased the synthesis of total apolipoprotein and Apo B(p<0.05). However, addition of fatty acid did not affect the synthesis of cellular and medium protein. Among different fatty acids tested, oleic acid had the greatest effect on Apo B synthesis. While stearic, linoleic and linolenic acid, all had similar effects. The secretion of triglyceride into the medium markedly increased in all fatty acid groups being 5-6 times over the albumin control. The triglyceride secretion was the highest int he oleic acid group. The secretion of phospholipid and cholesterol also increased with triglyceride output. A positive relationship existed between the output of lipoprotein-triglyceride and Apo B. Since the synthesis of Apo B was significantly increased when various fatty acids were included into the culture medium, part of the apparently stimulated synthesis of the apolipoprotein may be in response to the increased formation and secretion of lipoprotein lipids.

  • Oxidation/reduction titrations are important quantitative procedures for many chemicals. Several widely used analytical methods for cosmetic ingredients are based on the redox reactions. In this article, we summarized basic theories of redox titration and applications. Determination of unsaturation properties based on iodine or bromine number, quantitation of hydrogen peroxide or peroxide materials in several cosmetic ingredients and measurement of titanium dioxide, widely used sunscreen agent, in cosmetics are discussed here.

Analytical Analysis of PT Ferroresonance in the Transient-State (과도상태에서 PT 철공진의 해석적 분석)

  • When a circuit breaker is opened, a large capacitance around the buses, the circuit breaker and the potential transformer (PT) might cause PT ferroresonance. During PT ferroresonance, the iron core repeats saturation and unsaturation even though the supplied voltage is a rated voltage. This paper describes an analytical analysis of PT ferroresonance in the transient-state. To analyze ferroresonance analytically, the iron core is modelled by a simplified two-segment core model in this paper. Thus, a nonlinear ordinary differential equation (ODE) for the flux linkage is changed into a linear ODE with constant coefficients, which enables an analytical analysis. In this simplified model, each state, which is either saturated or unsaturated state, corresponds to one of the three modes, i.e. overdamping, critical damping and underdamping. The flux linkage and the voltage in each state are obtained analytically by solving the linear ODE with constant coefficients. The proposed transient analysis is effective in the more understanding of ferroresonance and thus can be used to design a ferroresonance prevention or suppression circuit of a PT.

Age Related Changes of Microsomal Fatty Acid Composition and Lipid Peroxidation in 2-acetylaminofluorene Treated rats. (2-Acetylaminofluorene의 투여와 나이에 따른 쥐의 세포막 지방산 조성 및 지질과산화물 생성의 변화)

  • For studying the effect of different dietary fats on carcinogenesis, fatty acid composition of membrane and lipid peroxidation were measured. Male Sprague-Dawley rats were fed on diet containing 15%(w/w) beef tallow or soybean oil. A single dose of 50 mg 2-AAF/kg B.W. was injected i.p. in each diet group 10 times. Rats were sacrifled after 1, 5, 10, and 15 weeks from the first injection. By 2-AAF injection, !ipid peroxidation increased slightly compared to control group. The rats fed on different fats had similar MDA production and those fed on soybean oil had slightly higher free radical concentration measured by ESR. In young rats, iipid peroxidation level was high and hydroxy radical production was higher in soybean oil group than in beef tallow group. With age, the lipid peroxidation values were decreased initially then increased. The fatty acid composition in microsomal membrane was reflected by dietary fatty acid composition. In soybean oil group, monoenoic acid was lower and polyunsaturated fatty acid was higher than beef tallow group. Linoleic acid contents showed the most discrepancy among groups. By 2-AAF treatment, iinoleic acid content and unsaturation index increased in soybean oil group. But in beef tallow group, there was no difference in fatty acid contents.

A study for design method minimizing wetland's influence by tunnel excavation

  • In recent, as Republic of Korea has been interested in environmental problem increasingly and became a member of many organizations or institutions related to environmental preservation such as a Ramsar convention, fundamental and completed methods to prevent ground water's drying up and leakage in tunnel excavation are requested. In this paper, we have studied the anticipated problems by tunnel excavation under the wetland and described the effective designed method to maintain the wetland's ecosystem environment. To accomplish this purpose, firstly, we investigated the wetland's ecosystem, ground's hydraulic properties and analysed the foreign similar case for tunnel excavation near the wetland. And by numerical analysis, we analyzed the runoff and infiltration quantity of water and hydraulic behaviour properties by saturation and unsaturation concept in rock mass and wetland. Finally, we established the effective countermeasure to minimize the ecosystem's bad influence by tunnel excavation.

Modeling and Performance Analysis of MAC Protocol for WBAN with Finite Buffer

  • The IEEE 802.15.6 standard is introduced to satisfy all the requirements for monitoring systems operating in, on, or around the human body. In this paper, analytical models are developed for evaluating the performance of the IEEE 802.15.6 CSMA/CA-based medium access control protocol for wireless body area networks (WBAN) under unsaturation condition. We employ a three-dimensional Markov chain to model the backoff procedure, and an M/G/1/K queuing system to describe the packet queues in the buffer. The throughput and delay performances of WBAN operating in the beacon mode are analyzed in heterogeneous network comprised of different user priorities. Simulation results are included to demonstrate the accuracy of the proposed analytical model.
