Determination of Trace Amount of Germanium in Rocks and Sediments by Hydride Vapor Generation-ICP-AES (수소화합물 발생법-유도결합플라스마 원자방출 분광기를 이용한 암석및 퇴적물중 미량의 게르마늄 분석)

  • We examined the determination of trace amount of germanium in rocks and sediments by hydride vapor generation-ICP-AES. Germanium is formed volatile compounds with various types of acid reagents, but volatilizing of germanium was decreased in the presence of $H_3PO_4$. Sediments and rocks can be dissolved by mixed acids of $HF-HNO_3-H_3PO_4$ without volatilizing loss of germanium in open digestion system and it was possible to determine germanium by hydride generation-ICP-AES without further sample treatment. Detection limit of Ge is reached to 0.08 ppb under the condition of 5M $H_3PO_4$ and 1% $NaBH_4$ as a supporting acid and a reducing reagent, respectively. The measured values by hydride generation-ICP-AES agreed well with the reference values of SRMs as well as the values determined by solution nebulization-ICP-MS.

Structural Analysis of Spaceborne Two-axis Gimbal-type Antenna of Compact Advanced Satellite (차세대 중형위성용 2축 짐벌식 안테나의 구조해석)

  • A two-axis gimbal-type antenna for a Compact Advanced Satellite (CAS) is used to efficiently transmit high resolution image data to a ground station. In this study, we designed the structure of a two-axis gimbal-type antenna while applying a launch lock device to secure its structural safety under a launch environment. To validate the effectiveness of the structural design, a structural analysis of the antenna was performed. First, a modal analysis was performed to investigate the dynamic responses of the antenna with and without the mechanical constraints of the launch lock device. In addition, a quasi-static analysis was performed to confirm the structural safety of the antenna structure and bolt I/Fs between the antenna base and the satellite. The suitable range of constraint force on the launch lock device was also determined to ensure the structural safety and mechanical gapping of the ball & socket interfaces, which places multi-constraints on the azimuth and elevation stage of the antenna.

  • A direct-connected, continuous type combustion test facility was developed to test a supersonic combustor model used in scramjet engines. The facility requirements were determined by assuming the flight speed of Mach 5, yielding the combustor inlet flow speed of Mach 2. Also the cross-section of the supersonic combustor under test was assumed as $32mm{\times}70mm$. As a result, the facility was designed to have the flow total pressure of 548 kPaA, total temperature of 1,320 K, and flow rate of 0.776 kg/s. The facility consists of a turbo type air compressor, electric air heater, vitiation air heater and a two dimensional facility nozzle to accelerate the flow to Mach 2. Also, an oxygen supply system was added to compensate the vitiation. The exhaust de-pressurization system is not added. Designed pressure, temperature, and flow rate were verified through the test operation of the facility.

Photovoltaic Modified β-Parameter-based MPPT Method with Fast Tracking

  • Maximum power point tracking (MPPT) is necessary for photovoltaic (PV) power system application to extract the maximum possible power under changing irradiation and temperature conditions. The β-parameter-based method has many advantages over conventional MPPT methods; such advantages include fast tracking speed in the transient stage, small oscillations in the steady state, and moderate implementation complexity. However, a problem in the implementation of the conventional beta method is the choice of an appropriate scaling factor N, which greatly affects both the steady-state and transient performance. Therefore, this paper proposes a modified β-parameter-based method, and the determination of the N is discussed in detail. The study shows that the choice of the scaling factor N is determined by the changes of the value of β during changes in irradiation or temperature. The proposed method can respond accurately and quickly during changes in irradiation or temperature. To verify the proposed method, a photovoltaic power system with MPPT function was built in Matlab/Simulink, and an experimental prototype was constructed with a solar array emulator and dSPACE. Simulation and experimental results are illustrated to show the advantages of the improved β-parameter-based method with the optimized scaling factor.

Galactooligosaccharide Synthesis by Active ${\beta}$-Galactosidase Inclusion Bodies-Containing Escherichia coli Cells

  • In this study, a galactooligosaccharide (GOS) was synthesized using active ${\beta}$-galactosidase (${\beta}$-gal) inclusion bodies (IBs)-containing Escherichia coli (E. coli) cells. Analysis by MALDI-TOF (matrix-assisted laser desorption/ionization-time of flight) mass spectrometry revealed that a trisaccharide was the major constituent of the synthesized GOS mixture. Additionally, the optimal pH, lactose concentration, amounts of E. coli ${\beta}$-gal IBs, and temperature for GOS synthesis were 7.5, 500 g/l, 3.2 U/ml, and $37^{\circ}C$, respectively. The total GOS yield from 500 g/l of lactose under these optimal conditions was about 32%, which corresponded to 160.4 g/l of GOS. Western blot analyses revealed that ${\beta}$-gal IBs were gradually destroyed during the reaction. In addition, when both the reaction mixture and E. coli ${\beta}$-gal hydrolysate were analyzed by high-performance thin-layer chromatography (HP-TLC), the trisaccharide was determined to be galactosyl lactose, indicating that a galactose moiety was most likely transferred to a lactose molecule during GOS synthesis. This GOS synthesis system might be useful for the synthesis of galactosylated drugs, which have recently received significant attention owing to the ability of the galactose molecules to improve the drugs solubility while decreasing their toxicity. ${\beta}$-Gal IB utilization is potentially a more convenient and economic approach to enzymatic GOS synthesis, since no enzyme purification steps after the transgalactosylation reaction would be required.

The Effect of social Support on Chronic Stress and Immune System in Male Manufacturing Workers (사회적 지지가 만성적 스트레스와 면역체계에 미치는 영향)

  • Objectives : To examine whether cumulative chronic stress influences the immune status, and to verify the effect of social support on the relationship between these two dimensions in male manufacturing workers. Methods : A total of 39 workers were recruited for this study. A structured-questionnaire was used to assess general characteristics, job characteristics (work demand and decision latitude), psychosocial distress, and social support. The serum levels of CD4 and CD8 were measured as immune markers, and were collected between 8:00 and 10:00am in order to standardize the markers. Nonparametric statistics were used to estimate the differences between job characteristics and the immune markers. Results : General characteristics, and health-related behaviors, were not associated with CD4, CD8 or CD4/CD8. No relationships were found between job characteristics and the mean levels of immune reactivity. These results were consistent, even after controlling for social support. Social support failed to modify the relationship toward work demand, decision latitude or psychosocial distress to CD4, CD8, and CD4/CD8. Conclusion : Cumulative chronic life stress might not influence the immune status, and the effects of social support on the immune function under chronic stress, may not play a crucial role in modifying the relationships. This implication supports that the effect of stress on the immune function may be determined by the characteristics of that stress. further research should effectively considers the type, magnitude and timing of a stress event, and modifiable factors, such as personality traits, coping style, and hormone excretion levels, on the alteration of immune status.

Isotopic Fissile Assay of Spent Fuel in a Lead Slowing-Down Spectrometer System

  • A lead slowing-down spectrometer (LSDS) system is under development to analyze isotopic fissile content that is applicable to spent fuel and recycled material. The source neutron mechanism for efficient and effective generation was also determined. The source neutron interacts with a lead medium and produces continuous neutron energy, and this energy generates dominant fission at each fissile, below the unresolved resonance region. From the relationship between the induced fissile fission and the fast fission neutron detection, a mathematical assay model for an isotopic fissile material was set up. The assay model can be expanded for all fissile materials. The correction factor for self-shielding was defined in the fuel assay area. The corrected fission signature provides well-defined fission properties with an increase in the fissile content. The assay procedure was also established. The assay energy range is very important to take into account the prominent fission structure of each fissile material. Fission detection occurred according to the change of the Pu239 weight percent (wt%), but the content of U235 and Pu241 was fixed at 1 wt%. The assay result was obtained with 2~3% uncertainty for Pu239, depending on the amount of Pu239 in the fuel. The results show that LSDS is a very powerful technique to assay the isotopic fissile content in spent fuel and recycled materials for the reuse of fissile materials. Additionally, a LSDS is applicable during the optimum design of spent fuel storage facilities and their management. The isotopic fissile content assay will increase the transparency and credibility of spent fuel storage.

Behavior of a steel bridge with large caisson foundations under earthquake and tsunami actions

  • The main focus of this study is to numerically investigate the influence of strong earthquake and tsunami-induced wave impact on the response and behavior of a cable-stayed steel bridge with large caisson foundations, by assuming that the earthquake and the tsunami come from the same fault motion. For this purpose, a series of numerical simulations were carried out. First of all, the tsunami-induced flow speed, direction and tsunami height were determined by conducting a two-dimensional (2D) tsunami propagation analysis in a large area, and then these parameters obtained from tsunami propagation analysis were employed in a detailed three-dimensional (3D) fluid analysis to obtain tsunami-induced wave impact force. Furthermore, a fiber model, which is commonly used in the seismic analysis of steel bridge structures, was adopted considering material and geometric nonlinearity. The residual stresses induced by the earthquake were applied into the numerical model during the following finite element analysis as the initial stress state, in which the acquired tsunami forces were input to a whole bridge system. Based on the analytical results, it can be seen that the foundation sliding was not observed although the caisson foundation came floating slightly, and the damage arising during the earthquake did not expand when the tsunami-induced wave impact is applied to the steel bridge. It is concluded that the influence of tsunami-induced wave force is relatively small for such steel bridge with large caisson foundations. Besides, a numerical procedure is proposed for quantitatively estimating the accumulative damage induced by the earthquake and the tsunami in the whole bridge system with large caisson foundations.

Expression of Codon Optimized β2-Adrenergic Receptor in Sf9 Insect Cells for Multianalyte Detection of β-Agonist Residues in Pork

  • ${\beta}_2$-adrenergic receptor (${\beta}_2-AR$) was expressed efficiently using Bac-to-Bac Baculovirus Expression System in Sf9 cells as a bio-recognition element for multianalyte screening of ${\beta}$-agonist residues in pork. Sf9 cells were selected as the expression system, and codon optimization of wild-type nucleic acid sequence and time-dependent screening of expression conditions were then carried out for enhancing expression level and biological activity. Under optimum conditions of multiplicity of infection (MOI) = 5 and 48 h post transfection, the protein yield was up to 1.23 mg/ml. After purification by chromatographic techniques, the purified recombinant protein was applied to develop a direct competitive enzyme-linked receptor assay (ELRA) and the efficiency and reliability of the assay was determined. The IC50 values of clenbuterol, salbutamol, and ractopamine were 28.36, 50.70, and $59.57{\mu}g/l$, and clenbuterol showed 47.61% and 55.94% cross-reactivities with ractopamine and salbutamol, respectively. The limit of detection (LOD) was $3.2{\mu}g/l$ and the relevant recoveries in pork samples were in the range of 73.0-91.2%, 69.4-84.6%, and 63.7-80.2%, respectively. The results showed that it had better performance compared with other present nonradioactive receptorbased assays, indicating that the genetically modified ${\beta}_2-AR$ would have great application potential in detection of ${\beta}$-agonist residues.

Encapsulation of Lactobacillus rhamnosus GG Using Milk Protein-Based Delivery Systems: Effects of Reaction Temperature and Holding Time on Their Physicochemical and Functional Properties

  • Microencapsulation is a protective process for materials that are sensitive to harsh conditions encounted during food manufacture and storage. The objectives of this research were to manufacture a milk protein-based delivery system (MPDS) containing Lactobacillus rhamnosus GG (LGG) using skim milk powder and to investigate the effects of manufacturing variables, such as reaction temerpature and holding time, on the physiccohemical properties of MPDS and viability of LGG under dairy food processing and storage conditions. MPDS was prepared using chymosin at varing reaction temperatures from 25℃ to 40℃ for 10 min and holding times from 5 to 30 min at 25℃. The morphological and physicochemical properties of MPDS were evaluated using a confocal laser scanning microscope and a particle size analyzer, respectively. The number of viable cells were determined using the standard plate method. Spherical-shaped MPDS particles were successfully manufactured. The particle size of MPDS was increased with a decrease in reaction temperature and an increase in holding time. As reaction temperature and holding time were increased, the encapsulation efficiency of LGG in MPDS was increased. During pasteurization, the use of MPDS resulted in an increase in the LGG viability. The encapsulation of LGG in MPDS led to an increase in the viability of LGG in simulated gastric fluid. In addition, the LGG viability was enhanced with an increase in reaction temperature and holding time. In conclusions, the encapsulation of LGG in MPDS could be an effective way of improving the viability of LGG during pasturization process in various foods.
