  • This study is expected to present instructions and analysis of Modern Dancheong pigments by its physical characteristics. Search pigments currently used and experiments proper mixing ratio of Jangdanyuksaek(Incarnadine). Samples are made by based on presented a mixture ratio which is less exfoliation, cracks and color change. And put into accelerated weathering test and ultraviolet ray degradation test. As a result in case of Jidang($TiO_2$) Rutile type is superior in discoloration and durability. Rutile type makes color difference remarkably because of oil absorption difference while mixing with Jangdan. Water paints which are used as a alternative present yellowness which means water paints lack in durability. whiting should be taken carefully as it has high brightness after degradation.

Feasibility of Red Tide Detection Around Korean Waters Using Satellite Remote Sensing

  • The monitoring activities at the National Fisheries Research and Development Institute (NFRDI) in Korea have been extended to include all the coastal waters of Korea after the outbreak of Cochlodinium polykrikoides blooms in 1995. We used several alternative methods including climatological analysis, spectral and optical methods which may offer potential detection of the major species of red tide in Korean waters. In the climatological analysis, NOAA, SeaWiFS, OCM satellite data was chosen using the known C. polykrikoides red tide bloom data and the area was mapped by helicopter reconnaissance and ground observation. The relationship between the distribution of sea surface temperature to C. polykrikoides bloom areas was studied. The anomalies of SeaWiFS chlorophyll a imageries against the imageries of non-occurring red tide for August, 2001 showed where the C. polykrikoides occurred. The anomalies of chlorophyll a concentrations from the satellite data during red tide outbreaks showed a similar distribution of C. polykrikoides in the red tide in August, 2001. The distribution between differences in sea surface temperatures during the day and at night also showed a possibility for red tide detection. We used a corrected vegetation index (CVI) to detect floating vegetation and submerged vegetation containing algal blooms. The results of from the optical absorption of C. polykrikoides in the ultraviolet band (340 nm) showed that if we use the optical characteristics from each red tide, we will be able to establish the feasibility of red tide detection.

Dyeability and functionality of Catechu(Part I) -Characteristics of Catechu and Dyeing properties of Cotton- (아선약의 염색성과 기능성(제1보) -아선약 색소의 특성과 면섬유에 대한 염색성-)

  • Catechu is widely used as in natural dyeing as well as in various medicines and tannage. This study investigated the dyeing properties and functionality of Catechu through a series of dyeing experiments using cotton fabric. It was shown that the absorbance of Catechu solution has two peaks at 220nm and 280nm. Thus, it can be concluded that the color constituent of Catechu is a catechol tannin and color tone is YR. The dyeability of Catechu increases as the concentration of the dye is increased. Its variation was shown to be similar to the isothermal absorption curve of the Freundlich. As dyeing time was increases, the dyeability did not change significantly after the 30 minute mark was passed. At high temperatures the dyeability was shown to increase. As the dyeing temperature was increased the color of dyeing fabrics changes from Y and YR to R. Experiments with pre-mordanting method showed that various colors can be obtained using Catechu. Cotton is effective in using K, Cu, Fe mordants, and the dyeability was shown to be improved with low pH. Both washing fastness and light fastness were shown to be low. However, the fabric color gradually changed to red was due to mailard reaction of catechol tannin causing by repeated washing and sunlight. The ultraviolet-cut ability was improved for cotton fabric dyed with Catechu. Also, dyed fabric with Catechu showed very good antimicrobial abilities at 99.9%.

Determiniation and Validation of Alibendol using High Pressure Liquid Chromatography in Human plasma (고속액체크로마토그라피법을 이용한 사람 혈장 중 알리벤돌(Alibendol)의 정량 및 검증)

  • The aim of this study was to develop and validate for determination of alibendol in human plasma by HPLC method. After precipitation of 500 ${\mu}l$ plasma samples by 50% methanol 50 ${\mu}l$ and 60% perchloric acid 30 ${\mu}l$ and the supernatant 50 ${\mu}l$ was injected into HPLC. The assay was performed isocratically using 10 mM potassium phosphate (pH 3.0) and acetonitrile (80 : 20, v/v) as mobile phase. The $C_{18}$ column (particle size $3.5{\mu}m$, $4.6{\times}50$ mm, Zorbax Eclipse) was used as a solid phase. The mobile phase was delivered at a flow-rate of 1.7 ml/min, detection was by ultraviolet absorption at 232 nm and concentrations were calculated on the basis of peak areas. In these conditions, alibendol can be separated from ethylparaben, the internal standard, and endogenous substances. The retention times of alibendol and ethylparaben were just about 2.6 and 3.5 minutes, respectively. This rapid HPLC method was validated by examining the precision and accuracy for inter- and intra-day analysis. The standard curve was linear ($R^2$=1.0000) over the concentration range of 0.05~20 ${\mu}g$/ml. The inter-day relative standard deviation (R.S.D.) and accuracy were 0.2~12.2% and 94.4~101.2% (82.7% at the lower limit of quatitation). The intra-day R.S.D. and accuracy were 0.1~11.8% and 98.8~102.5%, respectively. The method was successfully applied to the determination of alibendol in plasma for a pharmacokinetic study.

Thermodynamics of the Formation of Polymethylbenzene-Halogens Charge Transfer Complexes (IV) (폴리메틸벤젠과 할로겐 사이의 전하이동 착물생성에 관한 열역학적 연구 (제4보))

  • Ultraviolet spectrophotometric investigation have been carried out on the systems of pentamethylbenzene and hexamethylbenzene with iodine and iodine monochloride in carbon tetrachloride. The results reveal the formation of the one to one molecular complexes. The equilibrium constants were obtained in consideration of that absorption maxima due to the formation of the charge transfer complexes shift to blue with the increasing temperature. Thermodynamic parameters for the formation of the charge transfer complexes were calculated from these values. These results indicate that the complex formed between polymethylbenzene and iodine monochloride is more stable than that in the case of iodine. This may be a measure of their relative acidities toward polymethylbenzene, which is explained in terms of the relative electronegativities of halogen atoms. These results combined with previous studies of this series indicated that ${\lambda}_{max}$ shift to red with the increasing number of methyl groups on benezene ring and that the relative stabilities of these complexes increase in the order, Benzene < Toluene < Xylene < Durene < Mesitylene < Pentamethylbenzene < Hexamethylbenzene. The reason for the order found is thus additionally discussed.

High-Performance Liquid Chromatographic Quantification and Validation of Luteolin Glycosides from Sonchus brachyotus and Their Peroxynitrite-Scavenging Activity

  • In Korea, the leaves of Sonchus brachyotus (Compositae), an edible mountainous vegetable, are traditionally used to treat hepatitis and hemorrhage and are known to have diuretic action. The aqueous ethanolic extract of this plant was selected in our screening experiment using the peroxynitrite ($ONO_2^-$)-scavenging assay, and the present study was performed to qualitatively and quantitatively identify the active compounds from S. brachyotus and validate the present high-permormance liquid chromatography (HPLC) coupled with ultraviolet absorption detection method based on accuracy, precision and repeatability. Five phenolic substances including the main compound, luteolin $7-O-{\beta}-D$-glucuronopyranoside, as well as chlorogenic acid, luteolin 7-O-rutinoside, luteolin $7-O-{\beta}-D$-glucopyranoside, and luteolin, were found in the aqueous ethanolic extract of S. brachyotus. In the HPLC validation experiment, the linearity of the four compounds was established by $R^2$ values of more than 0.999 within the test ranges, and the recovery rate ranged from 98.2 - 105.3%. Luteolin 7-O-glucuronide was a predominant compound (143 mg/g of extract and 18.3 mg/g of the dry weight of plant material) with a potent peroxynitrite-scavenging effect ($IC_{50}$, $1.02{\pm}0.08{\mu}M$). Luteolin and its three glycosides together with chlorogenic acid were qualitatively and quantitatively determined using an HPLC method validated in the present study.

The Inactivation Effects of UV Light on Bacteriophage f2 (박테리오파지 f2에 대한 자외광선의 살균효과)

  • The effects of ultraviolet light on bacteriophage f2 were investigated to determine the inactivation kinetics and its mechanism. The 260nm light showed a little higher inactivation rate than the one of 300 nm. In this work, our main concern was whether structural and/or conformational changes in the protein capsid could occur by UV irradiation. The inactivation for the first 20 minutes irradiation was rapid with a loss of about 4 logs and followed by a slower rate during the next 40 minutes with no survival noted in the samples irradiated for 90 minutes or longer. The structural change of the protein capsid was examined by optical spectroscopic techniques and electron microscopy. The absorption spectra of the UV irradiated phages showed no detectable differences in terms of the spectral shape and intensity from the control phage. However, the fluorescence emission spectroscopic data, i.e. 1) fluorescence quenching of tryptophan residues upon irradiation of 300 nm light, 2) enhancement of fluorescence emission of ANS (8-aniline-1-naphthalene sulfonate) bound to the intact phages compared to the one in the UV-treated phages, and 3) decrease of energy transfer efficiency from tryptophan to ANS in the UV-treated samples, presented remarkable differences between the intact and UV-treated phages. Such a structural alteration was also observed by electron microscopy The UV-treated phages appeared to be broken and empty capsids. Therefore, the inactivation of the bacteriophage f2 by UV irradiation is thought to be attributed to the structural change in the protein capsid as well as damage in the viral RNA by UV irradiation.

Characteristics and Dyeability of Chelidonium majus var. asiaticum Extracts with Different Solvents (추출 용매에 따른 애기똥풀 색소의 특성 및 염색성)

  • The purpose of this study is to examine the influence of the pigment characteristic and changes in dying conditions on the dying properties by extracting Chelidonium majus var. asiaticum using distilled water and ethanol as solvents. Changes in dying conditions include variations in dye concentrations, dyeing temperatures and time on dye uptake, and K/S Value was compared according to these changes. Additionally, color changes were observed through mordant. Ultraviolet-visible spectrum was used to investigate the pigment characteristic, and as a result, tannin was identified in distilled-water-extract, whereas berberine and chlorophyll were identified in ethanol-extract. In FT-IR analysis, tannin in distilled-water-extract was verified as hydrolyzable tannin. For ethanol extract, chlorophyll was verified through absorption band of C-H, which is aliphatic spectrum around $2920cm^{-1}$ and $2850cm^{-1}$. From GC/MS analysis, oil components as well as terpine compounds were detected in ethanol-extract, and this, in turn, brings expectation regarding functionality. When dying in silk, dye uptake increased as concentration of the extract increased, and the optimum dyeing temperature and time were $40^{\circ}C$ and 60 minutes respectively. Dyed fabrics' colors were all basically Y-series colors, and adjustment in brightness and revelation of khaki color were also available depending on the type of the mordant. Color fastness, except for washing fastness, was good in silk dyed with distilled-water-extract. Thus, it can be concluded that by selecting the right extracting method and by doing proper dyeing and mordant according to the needs, these dyed fabrics can be used as eco-friendly, functional clothing material.

Structural and Optical Properties of SnS Thin Films Deposited by RF Magnetron Sputtering (RF 마그네트론 스퍼터링법으로 제조한 SnS 박막의 구조적 및 광학적 특성)

  • SnS thin films with different substrate temperatures ($150 {\sim}300^{\circ}C$) as process parameters were grown on soda-lime glass substrates by RF magnetron sputtering. The effects of substrate temperature on the structural and optical properties of SnS thin films were investigated by X-ray diffraction (XRD), Raman spectroscopy (Raman), field-emission scanning electron microscopy (FESEM), energy dispersive X-ray spectroscopy (EDS), and Ultraviolet-visible-near infrared spectrophotometer (UV-Vis-NIR). All of the SnS thin films prepared at various substrate temperatures were polycrystalline orthorhombic structures with (111) planes preferentially oriented. The diffraction intensity of the (111) plane and the crystallite size were improved with increasing substrate temperature. The three major peaks (189, 222, $289cm^{-1}$) identified in Raman were exactly the same as the Raman spectra of monocrystalline SnS. From the XRD and Raman results, it was confirmed that all of the SnS thin films were formed into a single SnS phase without impurity phases such as $SnS_2$ and $Sn_2S_3$. In the optical transmittance spectrum, the critical wavelength of the absorption edge shifted to the long wavelength region as the substrate temperature increased. The optical bandgap was 1.67 eV at the substrate temperature of $150^{\circ}C$, 1.57 eV at $200^{\circ}C$, 1.50 eV at $250^{\circ}C$, and 1.44 eV at $300^{\circ}C$.

Effects of Succinylation and Partial Proteolysis of Soybean Protein Isolates on Functional Properties and Protein-Protein Interaction (숙시닐화 및 부분가수분해가 대두단백질 분리물의 기능적 특성과 단백질-단백질 상호작용에 미치는 영향)

  • Soybean protein isolates were acylated with succinic anhydride and partially hydrolyzed with trypsin. Chemical modification decreased protein contents of samples and, in amino acid composition, tyrosine was increased comparatively. And lysine was increased remarkably by partial proteolysis. Succinylation and trypsin treatment increased the aqueous solubility and shifted the isoelectric potint that showed high pH-dependence of protein solubility. Protein solubility was influenced by salt concentration such as $NaCl,\;CaCl_2,\;NaNO_3$ and $NaH_2PO_4$. Chemical modification increased the absorption of oil and water, emulsification properties and foam capacity, but decreased foam stability, ultraviolet absorbance and bulk density. Protein-protein Interaction between soybean protein isolates and beef protein increased the emulsifying activity, emulsifying activity index and foaming properties, but it didn't have any influence on emulsion stability.
