연초(Nicotiana tabacum cv Xanthi) 세포배양에 의한 Ubiquinone 10의 생산 (Production of Ubiquinone 10 from the Callus Culture of Tabacco(Nicotiana tabacum cv Xanthi))

  • 식물세포의 기내배양에 의한 ubiquinone 10의 생산연구의 일환으로 Nicotiana tabacum cv Xanthi 품종으로부터 ubiquinone 10의 생성여부을 확인하고, 식물호르몬의 종류와 농도의 조건에 따른 ubiquinone 10의 생산증대를 조사하고자 본 실험을 수행하였다. Xanthi 캘러스 생장은 NAA 및 2,4-D와 혼합처리시 생장이 양호하였으며 NAA 단독처리구에서도 생장량이 증가되었고 암배양보다는 광배양상태에서 더 양호하였다. 또한 HPLC에 의해서 ubiquinone 10의 생성 여부를 조사한 결과 어느 조건에서도 모두 ubiquinone 10이 검출되었다. 식물호르몬에 의한 ubiquinone 10의 함량은 2,4-D와 NAA 혼합처리구에서보다 NAA 0.5 mg/L 단독처리구에서 ubiquinone 10의 함량이 증가되었으며, 특히 IBA 1mg/L와 NAA 0.5 mg/L 혼합처리구에서 더 양호한 경향을 보였고 단위 플라스크당 ubiquinone 10의 생산량도 더 높았다. 그러나 2,4-D 0.5 mg/L와 kinetin 0.5 mg/L 혼합한 처리구에서는 캘러스의 생장량은 비록 적었지만 ubiquinone 10의 함량이 가장 높아 단위 플라스크당 생산량은 제일 높은 경향을 보였다.

  • The effect of phytohormones, light and phosphate on in vitro production of ubiquinone 10 from the suspension cultures of Nicotiana tabacum cv. Xanthi callus was investigated. The inoculum size and cultured time in the suspension culture had to be at least over 2 % of medium volume at 15 days for the excellent growth of Xanthi callus. The growth of Xanthi callus in the suspension culture was improved by addition of NAA and 2,4-D, especially NAA 1.0mg/1 alone, at the light condition. The optimal concentration of phytohormone was 0.1 mg/l 2.4-D and 1.0 mg/l NAA for productivity of ubiquinone 10 in the suspenseion of Xanthi callus. Addition of 3mM KH$_2$PO$_4$ to the medium was more effective in promoting ubiquinone-l0 formation than other concentration in the light condition. Content and production of ubiquinone-l0 in the suspension cultures of Xanthi callus were the highest at the MS media containing 0.5mg/L kinetin, 0.5mg/L 2,4-D, 1.0mg/1 NAA, 3mM phosphate and 2 % inoculum in the light.

담자균 효모(酵母) Leucosporidium scottii와 관련 분류군균주(分類群菌株)의 ubiquinone 물질 (Ubiquinone compounds of the strains in Leucosporidium scottii and its related taxa)

  • 담자균효모 Leucosporidium scottii, Leucosporidium fellii, Leucosporidium lari-marini, Rhodosporidium fluviale의 ubiuqinone계를 고속액체크로마토그래피에 의해 결정하였다. Leucosporidium scottii 균주는 교배형 또는 self-sporulating 형에 관계없이 Q-9 또는 Q-10계를 가지고 있었다. 특히 동일 균주에서도 ubiquinone계의 변이가 관찰되었다. 이는 분류지표로서 중요시 되는 ubiquinone계의 재평가를 시사하는 새로운 사실로 간주된다. Leucosporidium fellii는 Q-9, Leucosporidium lari-marini는 Q-8, Rhodosporidium fuviale는 Q-10을 함유하고 있었다. L. lari-marini의 분류학적 위치는 다각적 연구에 기초하여 해명되어야 한다.

  • Recently several studies have indicated that ubiquinone-l0 (CoQ-10) decrease with aging in various organs of the human body. The decrease of ubiquinone-10 in the aging process suggests the reduction of both energy production in mitochondria and anti-oxidation with aging. Based on these findings, Ubiquinone-10 is being focused at as an anti-aging agent in the cosmetic market.(omitted)

Enzymatic and Energetic Properties of an Aerobic Respiratory Chain­Linked NADH Oxidase System in Marine Bacterium Vibrio natriegens

  • Membranes prepared from Vibrio natriegens oxidized both NADH and deamino-NADH as substrates. The maximum activity of the membrane-bound NADH oxidase was obtained at about pH 8.5 in the presence of 0.2 M NaCl, whereas that of the NADH:ubiquinone oxidoreductase was obtained at about pH 7.5 in the presence of 0.2 M NaCl. Electron transfer from NADH or deamino-NADH to ubiquinone-l or oxygen generated a considerable membrane potential (${\Delta}{\psi}$), which occurred even in the presence of $20{\mu}M$ carbonylcyanide m-chlorophenylhydrazone (CCCP). However, the ${\Delta}{\psi}$ was completely collapsed by the combined addition of $10{\mu}M$ CCCP and $20{\mu}M$ monensin. On the other hand, the activity of the NADH oxidase and the ${\Delta}{\psi}$ generated by the NADH oxidase system were inhibited by about $90\%$ with $10{\mu}M$ HQNO, whereas the activity of the NADH:ubiquinone oxidoreductase and the ${\Delta}{\psi}$ generated at the NADH:ubiquinone oxidoreductase segment were inhibited by about $60\%$. Interestingly, the activity of the NADH:ubiquinone oxidoreductase and the ${\Delta}{\psi}$ generated at the NADH:ubiquinone oxidoreductase segment were resistant to the respiratory chain inhibitors such as rotenone, capsaicin, and $AgNO_3$, and the activity of the NADH oxidase and the ${\Delta}{\psi}$ generated by the NADH oxidase system were very sensitive only to $AgNO_3$. It was concluded, therefore, that V. natriegens cells possess a $AgNO_3$-resistant respiratory $Na^+$ pump that is different from the $AgNO_3$-sensitive respiratory $Na^+$ pump of a marine bacterium, Vibrio alginolyticus.

유비퀴논, 이소플라본, γ-aminobutyric acid가 강화된 기능성 요구르트 개발 (Development of a functional yogurt fortified with ubiquinone, isoflavone, and γ-aminobutyric acid)

  • 탈지유에 홍국발효 콩 분말을 1:1의 비율로 첨가한 기능성 요구르트(SMP+MFSP)를 제조하여 대조군의 탈지유 요구르트(SMP)와 미량의 생리활성 물질 및 항산화 활성을 비교한 결과는 다음과 같다. 젖산발효 24시간 이후의 기능성 요구르트의 평균 pH는 3.96, 적정산도 1.03%, 생균수 9.68 log CFU/g으로 나타나 일반적인 호상 요구르트의 품질기준에 적합한 것으로 나타났다. 또한 대조군에 함유되지 않은 비배당체형의 이소플라본인 daidzein과 genistein의 총량은 젖산발효 48시간에 $201.21{\pm}6.29{\mu}g/g$으로 측정되었으며, GABA의 농도는 대조군에 비해 11.8배 높은 $107.22{\pm}3.06{\mu}g/g$으로 비교되었다. 대조군에 비해 기능성 요구르트의 유비퀴논의 총 농도는 평균 32.3배 높게 측정되었으나 젖산발효 시간에 따른 유의적인 농도변화는 나타나지 않았다(p>0.05). DPPH와 ABTS의 라디컬 소거활성으로 측정된 항산화 활성은 기능성 요구르트의 trolox 당량 값(${\mu}g$ TE/g)이 대조군에 비해 평균 7.9배 높게 비교되었으며, 시료에 함유된 유리 이소플라본의 농도와 밀접한 양의 상관성을 보여 주었다. 따라서 본 연구의 기능성 요구르트는 고부가가치의 probiotics 건강식품으로서의 활용가치가 높을 것으로 평가되었다.

Chemosystematics and Molecular Phylogeny of a New Bioflocculant-Producing Aspergillus Strain Isolated from Korean Soil

  • The ubiquinone and G+C contents of the bioflocculant-producing fungus, a new Aspergillus strain, were detennined using high-perfonnance liquid chromatography. The internal transcribed spacers 1 and 2 (ITS1 and ITS2), and the 5.8S ribosomal DNA (rDNA) of the strain were amplified and sequenced. The strain contained ubiquinone-l0($H_2$)as a major quinone and the G+C content was 49 mol%. A phylogenetic analysis of the ITS regions indicated that the strain belonged to the genus Aspergillus according to its previously classified morphological characteristics. Based on a sequence homology search, the strain was most closely related to Petromyces muricatus (anamorph, A. muricatus; accession number, AJ005674). The sequence of a new Aspergillus strain in ITS1 and ITS2, and 5.8S rDNA showed 97% homology to P. muricatus. Therefore, the strain is believed to be a new bioflocculant-producing Aspergillus strain.

Characterization of a New Acidophilic Acetobacter sp. Strain HA Isolated from Korean Traditional Fermented Vinegar

  • A new strain of acidophilic, acetogenic bacterium, Acetobacter sp. strain HA was isolated by selective enrichment from the traditionally fermented rice wine vinegar in Korea. It was a gram-negative, non-motile short rod and oxidized acetate and lactate. The optimal temperature and pH for growth were $28^{\circ}C$ and 4.0, respectively. The strain HA differed from other Acetobacter species by growing well on methanol, xylitol, inositol, dulcitol, D-xylose, L-arabinose, and D-mannose as sole sources of carbon and energy. The isolated strain HA did not produce $\gamma$-pyrones from glucose and did not produce ketone bodies from glycerol. The quinone system used in this study was an ubiquinone-9 isoprene unit. The guanine-plus-cytosine content of the DNA was 50.7 mol%, and the major cellular fatty acids were $C_{18:1} and C_{16:0}$.

Acinetobacter marinus sp. novo and Acinetobacter seohaensis sp. nov., Isolated from Sea Water of the Yellow Sea in Korea

  • Two Gram-negative, nonmotile, coccobacilli, SW-$3^T$ and SW-$100^T$, were isolated from sea water of the Yellow Sea in Korea. Strains SW-$3^T$ and SW-$100^T$ contained ubiquinone-9 (Q-9) as the predominant respiratory lipoquinone and $C_{18:1}\;{\omega}9c$ and $C_{16:0}$ as the major fatty acids. The DNA G+C contents of strains SW-$3^T$ and SW- $100^T$ were 44.1 mol% and 41.9 mol%, respectively. A neighbor-joining tree based on l6S rRNA gene sequences showed that the two isolates fell within the evolutionary radiation enclosed by the genus Acinetobacter. Strains SW-$3^T$ and SW-$100^T$ exhibited a l6S rRNA gene similarity value of 95.7% and a mean DNA-DNA relatedness level of 9.2%. Strain SW-$3^T$ exhibited l6S rRNA gene sequence similarity levels of 93.5-96.9% to the validly described Acinetobacter species and fifteen Acinetobacter genomic species. Strain SW-$100^T$ exhibited l6S rRNA gene sequence similarity levels of less than 97.0% to the other Acinetobacter species except Acinetobacter towneri DSM $14962^T$ (98.0% similarity). Strains SW-$3^T$ and SW-$100^T$ exhibited mean levels of DNA-DNA relatedness of 7.3-l6.7% to the type strains of some phylogenetically related Acinetobacter species. On the basis of phenotypic, phylogenetic, and genetic data, strains SW-$3^T$ and SW-$100^T$ were classified in the genus Acinetobacter as two distinct novel species, for which the names Acinetobacter marinus sp. novo (type strain SW-$3^T$=KCTC $12259^T$=DSM $16312^T$) and Acinetobacter seohaensis sp. novo (type strain SW-$100^T$=KCTC $12260^T$=DSM $16313^T$) are proposed, respectively.

광합성(光合成) 세균(細菌) T-20주(株)의 동정(同定) (Identification of Phototrophic Bacterium T-20)

  • 태국의 토양으로부터 분리한 젤라틴 분해능을 갖는 광합성세균 T-20주(株)는 직경이 $0.6{\sim}0.7{\mu}m$의 적색 간균으로써, Bacteriochlorophyll a의 흡수극대는 870nm이었고 또한, 퀴논종(種)은 UQ-8을 나타내었다. 유기 화합물로써 citrate, aspirate, glutamate, fractose 등을 이용하였다. 이상의 결과로부터 T-20주는 Bergey's Manual of Systematic Bacteriology(1989)에 기재된 Rhodocyclus gelatinosus와 일치한다.
