Production of Ubiquinone 10 from the Callus Culture of Tabacco(Nicotiana tabacum cv Xanthi) (연초(Nicotiana tabacum cv Xanthi) 세포배양에 의한 Ubiquinone 10의 생산)

  • The effect of phytohormones on in vitro production of ubiquinone 10 from the callus cultures of Nicotiana tabacum cv Xanthi was investigated. The growth of callus cultures of Xanthi was in proved by addition of NAA and 2,4-D, especially NAA 0.5 mg/L alone, at the light condition. Ubiquinone 10 was detected by HPLC, and confirmed from Xanthi callus cultured on the all of uppermedia. The ubiquinone 10 content in Xanthi tobacco callus cultured on the medium with NAA 0.5 mg/L only was higher than that of other mixed medium with NAA and 2,4-D. However addition of IBA 1 mg/L and NAA 0.5 mg/L to the medium was more effective in promoting ubiquinone 10 formation than that of NAA 0.5 mg/L only As the callus growth of Xanthi was considerabley restrained at concentration of kinetin, Content and production of ubiquinone low as the highest at kinetin 0.5mg/L and 2,4-D 0.5mg/L in the light.

  • Representative synthetic piericidin-like compounds, such as hydroxypyridine and hydroxyquinoline derivatives, which showed high inhibition activity against NADH-ubiquinone oxidoreductase on the respiratory chain revealed good fungicide activity. Especially, hydrolrypyridine ones showed high activity against rice blast (Pyricularia oryzae) and barley powdery mildew (Erysiphe graminis).

  • Isoprenoid quinone are essential compositions of the respiratory or photosynthetic electron transport system of microorganisms. Their chemotaxonomic significance as well as their physiological importance has been fully realized. We determined the ubiquinone types of the genus Trichoderma, Gliocladium, Verticillium, Aspergillus, and several mushroom such as Agaricus bisporus. Lentinus edodes, Pleurotus ostreatus, Flammulina velutips, Phellinus chrysoloma, Phellinus igniarius and Phellinus laevigatus. Most of Deuteromycotina had Q-10($H_2$), and all of mushroom had Q-9 as main ubiquinone type. Ubiquinone type in other fungal taxa.

  • Recently several studies have indicated that ubiquinone-l0 (CoQ-10) decrease with aging in various organs of the human body. The decrease of ubiquinone-10 in the aging process suggests the reduction of both energy production in mitochondria and anti-oxidation with aging. Based on these findings, Ubiquinone-10 is being focused at as an anti-aging agent in the cosmetic market.(omitted)

  • The effect of phytohormones, light and phosphate on in vitro production of ubiquinone 10 from the suspension cultures of Nicotiana tabacum cv. Xanthi callus was investigated. The inoculum size and cultured time in the suspension culture had to be at least over 2 % of medium volume at 15 days for the excellent growth of Xanthi callus. The growth of Xanthi callus in the suspension culture was improved by addition of NAA and 2,4-D, especially NAA 1.0mg/1 alone, at the light condition. The optimal concentration of phytohormone was 0.1 mg/l 2.4-D and 1.0 mg/l NAA for productivity of ubiquinone 10 in the suspenseion of Xanthi callus. Addition of 3mM KH$_2$PO$_4$ to the medium was more effective in promoting ubiquinone-l0 formation than other concentration in the light condition. Content and production of ubiquinone-l0 in the suspension cultures of Xanthi callus were the highest at the MS media containing 0.5mg/L kinetin, 0.5mg/L 2,4-D, 1.0mg/1 NAA, 3mM phosphate and 2 % inoculum in the light.

  • A strain of non-sulfur photosynthetic bacteria possessing large amount of ubiquinone-10 in the cell mass, was isolated from waste water collected from the Nakdong River. It was identified as Rhodobacter sp. N-2 by a series of studies on its morphological. cultural and physiological characteristics. The organism appeared to got higher cell mass under circumstances of anaerobic growth conditions than under photosynthetic aerobic cultures. Biotin and thiamin and niacin could be used as good growth factors.

  • Ubiquinone compounds of strains in Leucosporidium scottii, Leucosporidium fellii, Leucosporidium lari-marini, and Rhodosporidium fluviale were analyzed by the high performance liquid chromatography. The Q-9 or Q-l0 compounds, independent on mating or self-sporulating type, were determined in the analyses of Leucosporidium scottii strains. Particularly, there is heterogenous in the quinone compounds in the same strain of L. scottii. These results showed reassessment of the quinone compounds as a taxonomic criterion; L. fellii had the Q-9 compound, L. lari-marini the Q-8 compound, and R. fluviale the Q-10 compound, Taxonomic position of L. lari-marini should be elucidated on the basis of analysis of Q-compound with other aspects.

  • New quinoline compounds were designed, synthesized, and examined with submitochondria. Most compounds showed high activity against NADH-ubiquinone oxidoreductase. Inhibition activity was mainly affected by the length of the lipophilic part, regardless of bulkiness or location of a phenyl group in the side chain. The $\beta-methyl$ group was demons)rated to be the optimal functionality on the nuclei of the quinoline derivatives so 4hat either deletion or insertion of a methylene on the group eliminated its activity.

HQNO-sensitive NADH:Quinone Oxidoreductase of Bacillus cereus KCTC 3674

  • The enzymatic properties of NADH:quinone oxidoreductase were examined in Triton X-100 extracts of Bacillus cereus membranes by using the artificial electron acceptors ubiquinone-1 and menadione. Membranes were prepared from B. cereus KCTC 3674 grown aerobically on a complex medium and oxidized with NADH exclusively, whereas deamino-NADH was determined to be poorly oxidized. The NADH oxidase activity was lost completely by solubilization of the membranes with Triton X-100. However, by using the artificial electron acceptors ubiquinone-1 and menadione, NADH oxidation could be observed. The activities of NADH:ubiquinone-1 and NADH:menadione oxidoreductase were enhanced approximately 8-fold and 4-fold, respectively, from the Triton X-100 extracted membranes. The maximum activity of FAD-dependent NADH:ubiquinone-1 oxidoreductase was obtained at about pH 6.0 in the presence of 0.1M NaCl, while the maximum activity of FAD-dependent NADH:menadione oxidoreductase was obtained at about pH 8.0 in the presence of 0.1M NaCl. The activities of the NADH:ubiquinone-1 and NADH:menadione oxidoreductase were very resistant to such respiratory chain inhibitors as rotenone, capsaicin, and $AgNO_3$, whereas these activities were sensitive to 2-heptyl-4-hydroxyquinoline-N-oxide (HQNO). Based on these results, we suggest that the aerobic respiratory chain-linked NADH oxidase system of B. cereus KCTC 3674 possesses an HQNO-sensitive NADH:quinone oxidoreductase that lacks an energy coupling site containing FAD as a cofactor.

The Membrane-Bound NADH:Ubiquinone Oxidoreductase in the Aerobic Respiratory Chain of Marine Bacterium Pseudomonas nautica

  • Each oxidoreductase activity of the aerobic respiratory chain-linked NADH oxidase system in the marine bacterium Pseudomonas nautica was stimulated by monovalent cations including $Na^+,\;Li^+,\;and\;K^+$. In the presence of NADH or deamino-NADH as electron donors, $GH_2$ formation was approximately 1.3-fold higher in the presense of 0.08 M of $Na^+\;than\;K^+$, Whereas the other reductase activities were not significantly higher in $Na^+\;than\;K^+$. The optimal pH of NADH (or deamino-NADH):ubiquinone-1 oxidoreductase was 9.0 in the presence of 0.08 M NaCl. The activity of NADH (or deamino-NADH):ubiquinone-1 oxidoreductase was inhibited by about 33% with $60{\mu}M$ 2-heptyl-4-hydroxyquinoline-N-oxide (HQNO). The activity of NADH (deamino-NADH): ubiquinone-1 oxidoreductase was inhibited by about 32 to 38% with $80{\mu}M$ rotenone, whereas the activity was highly resistant to capsaicin. On the other hand, electron transfer from NADH or deamino-NADH to ubiquinone-1 generated a membrane potential (${\Delta}{\psi}$) which was larger in the presence of $Na^+$ than that observed in the absence of $Na^+$. The ${\Delta}{\psi}$ was almost completely collapsed by $5{\mu}M$ carbonylcyanide m-chlorophenylhydrazone(CCCP), and approximately 50% inhibited by $100{\mu}M$ rotenone, or $60{\mu}M$ 2-heptyl-4-hydroxyquinoline (HQNO). Also, HQNO made the ${\Delta}{\psi}$ very unstable. The results suggest that the enzymatic and energetic properties of the NADH:ubiquinone oxidoreductase of P. nautica are quite different, compared with those of other marine halophilic bacteria.
