Berberine suppresses in vitro migration of human aortic smooth muscle cells through the inhibitions of MMP-2/9, u-PA, AP-1, and NF-κB

  • Berberine, a type of isoquinoline alkaloid isolated from Chinese medicinal herbs, has been reported to have various pharmacological activities. Studies have demonstrated that berberine has beneficial effects on vascular remodeling and alleviates restenosis after vascular injury. However, its mechanism of action on vascular smooth muscle cell migration is not fully understood. We therefore investigated the effect of berberine on human aortic smooth muscle cell (HASMC) migration. Boyden chamber assay was performed to show that berberine inhibited HASMC migration dose-dependently. Real-time PCR and Western blotting analyses showed that levels of matrix metalloproteinase (MMP)-2, MMP-9, and urokinase-type plasminogen activator (u-PA) were reduced by berberine at both the mRNA and protein levels. Western blotting assay further confirmed that activities of c-Fos, c-Jun, and NF-${\kappa}B$ were significantly attenuated. These results suggest that berberine effectively inhibited HASMC migration, possibly by down-regulating MMP-2, MMP-9, and u-PA; and interrupting AP-1 and NF-${\kappa}B$ mediated signaling pathways.

Relationship between Plasminogen Activity and Plasminogen Inhibitor during the Culture of Porcine Oviduct Epithelial Cells

  • The present study was performed to identify changes of plasminogen activator (PA) and plasminogen activator inhibitor (PAI) in porcine oviduct epithelial cells (POECs) during the estrous cycle. POECs obtained from ovary in pre-ovulatory (Pre-Ov), early to mid-luteal stage (Early-mid L) and post-ovulatory stage (Post-Ov). For the examine of PA activity, $1{\times}10^5$ fresh cells of POECs were cultured in DMEM/Ham F-12 containing 10% FBS and 0.2% amphotericin under humidified atmosphere of 5% $CO_2$ in air and $38^{\circ}C$. The urokinase-type PA (uPA) was observed at 7 days of POECs culture. PA activity was measured with culture prolonged of 0, 3, 6, 12 and 24 h after culture of 7 days. The PA activity were high significantly (p<0.05) at 12 h of culture, but PA activity were decreased with culture periods increased. The PA activity in POECs of Post-Ov stage were higher significantly (p<0.05) than that of Early-mid L and Pre-Ov stage. When PAI-1 and PAI-2 were added during the POECs culture, the PA were observed significant low activity (p<0.05). The PA activity and protein expression were decreased by PA inhibitor. This results suggest that PAI-1 and PAI-2 have a suppressive action on change of PA activity during the estrous cycle of pigs. Specifically, this study using PA inhibitor was effect the PA activity and PAI expression in oviduct epithelial cells in pigs.

Effects of Meteorology and Air Pollution on Respiratory Disease affecting Vulnerable Populations (기상 및 대기질이 건강취약집단에 미치는 영향)

  • It is well known that atmospheric environments, including both meteorology and air quality, significantly affect public health, such as chronic lung disease and cancer, and respiratory infections. In this study, we have analyzed correlations between the number of daily respiratory outpatients and the atmospheric environments data for about ten years for the city of Busan, South Korea. The respiratory problem patients data have been categorized into two health-vulnerable groups by age over 65(DayPA_O65) and under 20(DayPA_U20), each of which shows relatively higher correlations with air quality and meteorology, respectively. However, time series analysis with factor separation results in that DayPA_O65 and DayPA_U20 show a higher relation with variance components and daily irregular factors of atmospheric concentrations, respectively.

A Study on Ventilation and Heat Transfer Coefficient of Passive Ventilation Skin (패시브환기외피의 통기성능 및 열관류율에 대한 연구)

  • This paper aims to evaluate performances of ventilation and insulation of 6types PVS(Passive ventilation skin) by numerical simulation. The results are as follows. 1) The result of Performance of ventilation by pressure difference, it was shown that the amount of ventilation changed bigger under 1Pa and amount of ventilation increased according to increase opening area (${\alpha}A$). Although same opening area of PVS, it can predict that pressure differences cause ventilation differences. 2) In case of same opening area of PVS, however, it was changed the amount of ventilation each types of PVS that is distinguished opening area by flow coefficient. 3) Dynamic U-value that represents performance of insulation PVS was similar change upper ${\alpha}A40\;cm^2/m^2$, great change in casse of 0.1 Pa pressure difference. In case of ${\alpha}A10\;cm^2/m^2$, it was changed bigger under 0.3 Pa pressure difference, ${\alpha}A20\;cm^2/m^2$ of PVS was changed under 0.2 Pa pressure difference.

Genetic Studies of Redcell Types for Individual Identification and Parentage Verification in Horse Breeds. (개체식별 및 친자판정을 위한 말의 적혈구항원형에 관한 연구)

  • The present study was carried out to investigate the redcell types of horse breeds. A total of 210 horses (73 Korean native horses, 118 crossbreed horses, and 19 Mongolian horses) were tested a redcell types by serological procedure, and their phenotypes and gene frequencies were estimated. The blood groups phenotypes observed with highest frequency were Aa (27.4%, 63.6%, 63.2%), Ca (97.3%, 94.9%, 89.5%), K- (97.3%, 99.2%, 84.2%), Pa (39.7%, 44.9%, 42.1%), and Ua (71.2%, 70.3%, 63.2%) in the Korean native horse, crossbreed horse, and Mongolian horse, respectively. In the D system and Q system, phenotypes observed with highest frequency were Dbcm/dghm (12.3%), Dbcm/cgm (14.4%), Dcgm/dghm (15.8%), and Qc (56.2%), Qabc (36.4%), Qc (31.6%) in the Korean native horse, crossbreed horse, and Mongolian horse, respectively Alleles observed with highest frequency were A- (0.287), Ca(0.827), Ddghm (0.226), K- (0.985), Pa (0.358), Qc (0.494), U-(0.529) in the Korean native horse, Aa (0.529), Ca (0.776), Dbcm (0.306), K- (0.995), P- (0.531), Q- (0.504), U- (0.548) in crossbreed horse, and Aa (0.421), Ca (0.895), Ddghm (0.421), K- (0.842), Pa (0.447), Qc (0.448), Ua (0.632) in Mongolian horse. Dcfgk and D- alleles were not detected in these horses. These results present basic information for estimating the genetic relationships between the Korean native horse, and developing a system for parentage verification and individual identification in these horses.

Synthesis of Polyester Polyols by Using Double Metal Cyanide Catalyst and Physical Properties of Polyurethanes Produced by the Polyols (이중금속시안염 촉매에 의한 폴리에스테르 폴리올 제조 및 이를 이용한 폴리우레탄의 물성)

  • Copolymerizations of propylene oxide (PO) and phthalic anhydride (PA) have been performed in the presence of double metal cyanide (DMC) catalyst as a means of incorporating ester groups in the polyol backbone. DMC catalyst was effective for the copolymerization and the reactivity ratios measured by modified Kelen-$T{\ddot{u}}d{\ddot{o}}s$ equation were $r_1(PA)\;=\;0$, and $r^2(PO)\;=\;0.248$. Four different Polyol samples containing 1.0, 2.1, 7.52, and 11.42 mol% of PA unit were utilized for the synthesis of thermoplastic polyurethanes of their hard segments of 19 wt%. As the incorporated amount of PA increases, the elongation of the resulting polyurethane decreases and the tensile strength and the tensile modulus increase. The modulation of the incorporated amount of PA into polyol backbone was proven to be a feasible way to tune the physical properties of polyurethanes.

Expression, Purification, and Biological Characterization of The Amino-Terminal Fragment of Urokinase in Pichia pastoris

  • Urokinase (uPA) and its receptor (uPAR) play an important role in tumor growth and metastasis. Targeting the excessive activation of this system as well as the proliferation of the tumor vascular endothelial cell would be expected to prevent tumor neovasculature and halt the tumor development. In this regard, the amino-terminal fragment (ATF) of urokinase has been confirmed as effective to inhibit the proliferation, migration, and invasiveness of cancer cells via interrupting the interaction of uPA and uPAR. Previous studies indicated that ATF expressed in Escherichia coli was mainly contained in inclusion bodies and also lacked posttranslational modifications. In this study, the biologically active and soluble ATF was cloned and expressed in Pichia pastoris. The recombinant protein was purified to be homogenous and confirmed to be biologically active. The yield of the active ATF was about 30 mg/l of the P. pastoris culture medium. The recombinant ATF (rATF) could efficiently inhibit angiogenesis, endothelial cell migration, and tumor cell invasion in vitro. Furthermore, it could inhibit in vivo xenograft tumor growth and prolong the survival of tumor-bearing mice significantly by competing with uPA for binding to cell surfaces. Therefore, P. pastoris is a highly efficient and cost-effective expression system for large-scale production of biologically active rATFs for potential therapeutic application.

The Candidate Tumor Suppressor Gene SLC8A2 Inhibits Invasion, Angiogenesis and Growth of Glioblastoma

  • Glioblastoma is the most frequent and most aggressive brain tumor in adults. Solute carrier family 8 member 2 (SLC8A2) is only expressed in normal brain, but not present in other human normal tissues or in gliomas. Therefore, we hypothesized that SLC8A2 might be a glioma tumor suppressor gene and detected the role of SLC8A2 in glioblastoma and explored the underlying molecular mechanism. The glioblastoma U87MG cells stably transfected with the lentivirus plasmid containg SLC8A2 (U87MG-SLC8A2) and negative control (U87MG-NC) were constructed. In the present study, we found that the tumorigenicity of U87MG in nude mice was totally inhibited by SLC8A2. Overexpression of SLC8A2 had no effect on cell proliferation or cell cycle, but impaired the invasion and migration of U87MG cells, most likely through inactivating the extracellular signal-related kinases (ERK)1/2 signaling pathway, inhibiting the nuclear translocation and DNA binding activity of nuclear factor kappa B ($NF-{\kappa}B$), reducing the level of matrix metalloproteinases (MMPs) and urokinase-type plasminogen activator (uPA)-its receptor (uPAR) system (ERK1/2-$NF-{\kappa}B$-MMPs/uPA-uPAR), and altering the protein levels of epithelial to mesenchymal transitions (EMT)-associated proteins E-cardherin, vimentin and Snail. In addition, SLC8A2 inhibited the angiogenesis of U87MG cells, probably through combined inhibition of endothelium-dependent and endothelium-nondependent angiogenesis (vascular mimicry pattern). Totally, SLC8A2 serves as a tumor suppressor gene and inhibits invasion, angiogenesis and growth of glioblastoma.

Nucleophilicity와 Basicity의 연관성에 관한 연구

  • 본 연구에서는 DFT를 이용하여 Nucleophilicity와 Basicity의 연관성에 대한 계산화학적 분석을 수행하였다. Basicity는 선정된 모델 분자의 protonation 반응에서 생성물과 반응물의 enthalpy 변화량인 양성자 친화도(Proton affinity, PA) 값을 구하여 분석하였다. 계산한 결과는 실험을 통해 얻은 PA 결과와 경향성이 거의 일치함을 확인하였다. Nucleophilicity는 모델 분자들과 $CH_3Br$ (electrophile)의 $SN_2$반응에서 gibbs free energy of activation(${\Delta}G^{\ddag}$) 값으로 그 경향성을 분석하였다. 또한 용매의 종류를 다르게 하여 용매에 따른 ${\Delta}G^{\ddag}$ 값의 경향성도 확인하였다. 각 용매에 따라 구한 ${\Delta}G^{\ddag}$와 PA의 상관관계를 비교하였으나, 큰 연관성은 보이지 않았다. 이에 ${\Delta}G^{\ddag}$와 PA의 상관관계를 보여줄 수 있는 parameter를 찾기 위하여 각 모델 분자의 Electronegativity와 Polarizability를 계산하여 연관성을 비교해보았다. Polarizability를 적용했을 때 Nucleophilicity와 Basicity사이의 연관성을 나타낼 수 없었던 반면, Electronegativity를 적용하여 Basicity와 Nucleophilicity의 연관성 보일 수 있음을 이론적으로 규명하였다.
