Preparation of Polyesteramides Based on Aliphatic Amine-Containing Phenol Derivatives via Interfacial Polymerization

  • A series of polyesteramides with randomly introduced ester/amide group ratio of 50/50 were newly synthesized by reacting terephthaloyl chloride, isophthaloyl chloride and sebacoyl chloride with tyramine and tyrosine. The polymerization was carried out by interfacial polymerization in two phase solvent systems, which gave various polyesteramides with moderate molecular weights in good yields. The chemical structures of the polymers were confirmed by $^1$H NMR, IR and elemental analysis. Tyrosine based polyesteramide was degraded thermally around 29$0^{\circ}C$ to give the polyesteramide, which was obtainable from tyramine. Thermal stability and degradation behaviors were examined by differential scanning calorimetry and thermogravimetric analyses.

  • An influence of bethanidine (B) onpressor effects of norepinephrine (NE) and tyramine (TR) was investigated in the whole rabbits. B, in a dose 1.0, 3.0 and 5.0 mg/kg, i.v., potentiated significantly the pressor effects of NE and TR. Reserpine and desipramine did not increase the NE effect that had been potentiated by B.B also made little modification of NE effect that had been potentiated by reserpine and desipramine. B increased the NE effect that had been potentiated by tranycypromine and guanethidine. The NE pressor effect potentiated by B was decreased by tranylcypromine, but not influenced by guanethidine. The TR pressor effect potentiated by B was not altered by reserpine and guanethidine, but decreased by desipramine and tranylcypromine. B increased the TR pressor effect that had been potentiated by guanethidine. B, when given after administration of reserpine, tranylcypromine or desipramine, exerted little influence on the TR effect. The mechanism of potentiation of NE and TR pressor effects by B seems to be similar to guanethidine, and the potency of B on the influence of NE and TR effects seems to be greater than guanethidine.

  • In order to investigate the effect of nifedipine, $Ca^{+2}$ channel antagoninst, on the action of some drugs participating in blood pressure, this experiment was peformed in rabbits. Nifedipine decreased the pressor actions of norepinephrine, angiotensin and carotid artery clamping, but did not affect the pressor actions of tyramine and depressor actions of acetylcholine and pilocarpine. Nifedipine inhibited the potentiated pressor action of norepinephrine and angiotensin, but did not influence the potentiated pressor action of tyramine in rabbits pretreated with chlorisondamine, ganglionic blocking agent. Nifedipine weakened the potentiated pressor action of norepinephrine, did not affect the pressor action of angiotensin and the potentiated pressor action of tyramine in rabbits pretreated with debrisoquine, sympathetic neuronal blocking agent.

Biosynthesis of Two Hydroxybenzoic Acid-Amine Conjugates in Engineered Escherichia coli

  • Two hydroxybenzoyl amines, 4-hydroxybenzoyl tyramine (4-HBT) and N-2-hydroxybenzoyl tryptamine (2-HBT), were synthesized using Escherichia coli. While 4-HBT was reported to demonstrate anti-atherosclerotic activity, 2-HBT showed anticonvulsant and antinociceptive activities. We introduced genes chorismate pyruvate-lyase (ubiC), tyrosine decarboxylase (TyDC), isochorismate synthase (entC), isochorismate pyruvate lyase (pchB), and tryptophan decarboxylase (TDC) for each substrate, 4-hydroxybenzoic acid (4-HBA), tyramine, 2-hydroxybenzoic acid (2-HBA), and tryptamine, respectively, in E. coli. Genes for CoA ligase (hbad) and amide formation (CaSHT and OsHCT) were also introduced to form hydroxybenzoic acid and amine conjugates. In addition, we engineered E. coli to provide increased substrates. These approaches led to the yield of 259.3 mg/l 4-HBT and 227.2 mg/l 2-HBT and could be applied to synthesize diverse bioactive hydroxybenzoyl amine conjugates.

  • 전신마취가토(全身麻醉家兎) 및 춘체가토(春體家兎)에 tetrabenazine(40mg/kg)을 투어(投與)한 후(後) norepinephrine 및 tyramine에 대(對)한 승압반응(昇壓反應) 및 심박증가(心搏增加) 반응(反應)을 검색하여 다음과 같은 결과를 얻었다. 1) 전신가토(全身家兎)에서 TBZ투여(投與) 2시간(時間) 이내(以內)에는 NE에 대(對)한 승압반응(昇壓反應)은 강화(强化)되지 않았다. 2) 전신가토(全身家兎)에서 TBZ투여(投與) 5시간후(時間後)에는 NE에 대(對)한 승압반응(昇壓反應)은 강화(强化)되었으며 10시간후(時間後)에는 강화(强化)는 최고(最高)에 달(達)하였고, 24시간(時間), 48시간후(時間後)에는 점차(漸次)이 강화(强化)는 소실(消失)되었다. 3) 춘체가토(春體家兎)에서 TBZ투여(投輿) 10시간후(時間後)에는 NE에 대(對)한 승압반응(昇壓反應)은 현저(顯著)히 강화(强化)되었고 심박증가반응(心搏增加反應)도 증강(增强)한 경향(傾向)을 보였다. 그러나 24시간 후(時間 後)에는 이러한 강화(强化)는 볼 수 없었다. 4) 전신가토(全身家兎)에서 TBZ투여(投與)로 NE승압반응(昇壓 反應)이 강화(强化)되어 있는 상태하(狀態下)에서는 desipramine으로써 더 이상 반응(以上 反應)이 증가(增加)되지 않았다. 5) 춘체가토(春體家兎)에서 TBZ투여(投與)로 NE승압반응(昇壓 反應)이 강화(强化) 되어 있는 상태하(狀態下)에서 desipramine으로써 그 반응(反應)은 더욱 증가(增加)되었다. 6) TBZ투여(投與) 2시간(時間) 및 6시간후(時間後)에 carbachol을 주사(注射)한 가토(家兎)에서는 NE에 대(對)한 승압반응(昇壓反應)의 강화(强化)는 현저(顯著)히 억제(抑制)되었다. 7) TBZ투여(投與) 10시간후(時間後), 전신가토(全身家兎)의 tyramine에 대(對)한 승압반응(昇壓反應)은 약화(弱化)되지 않았고 춘체가토(春體家兎)의 tyramine에 대(對)한 반응(反應)은 강화(强化)되었다. 8) TBZ로서 NE에 대(對)한 supersensitivity가 발생(發生)하고 tyramine에 대(對)한 반응(反應)은 약화(弱化)되지 않고 carbachol로서 NE에 대(對)한 supersensitivity가 방지(防止)되었음은, NE에 대(對)한 supersensitivity발생(發生)에 뇌내(腦內) catecholamine 감소(減少)에 따른 교감신경계(交感神經系) tone의 감소(減少)가 관계(關係)있음을 시사(示唆)하고 있다.

전통 장류에서 분리한 Biogenic Amines 저감 유산균 Pediococcus pentosaceus의 분리 및 특성 (Characterization of Biogenic Amine-reducing Pediococcus pentosaceus Isolated from Traditionally Fermented Soybean Products)

  • 장류 식품에서 biogenic amines 저감 기능과 유해균 저해 능력을 동시에 지닌 유산균을 선발하기 위해, 전통방식으로 제조한 장류 시료로부터 2종의 균주를 분리하였다. 생화학적 동정 및 16S rRNA 유전자 염기 서열을 분석 결과 이들 균은 유산균인 Pediococcus pentosaceus로 동정되었다. 질소원으로 0.1% (w/v) histamine과 0.1% tyramine이 첨가된 최소 합성 배지에서 $30^{\circ}C$, 48시간 배양 후 잔류 amine을 분석한 결과, LE22 균주의 경우 histamine은 23.7%, tyramine은 15.7%가 감소한 반면, LE17의 경우 histamine은 13.7%, tyramine은 25.9%가 감소하였다. 장류에서 발견되는 주요 유해균에 대한 항균 효과를 조사한 결과, 두 균주 모두 유해균들에 대해 항균 작용을 보였다. 두 균주의 발효 특성을 고려했을 때 이들은 유산균 종균으로써 산업적 장류 생산에 적용할 수 있을 것으로 보인다.

  • In the course of continuous work on the leaves of Allium tuberosum (Liliaceae), compounds 1 and 2 were isolated from the ethylacetate and butanol-soluble fraction, and identified as N-p-coumaroyl tyramine and bis(p-hydroxyphenyl) ether, respectively, on the basis of spectral data and physicochemical results.

Biogenic Amine Formation in "Bez Sucuk," a Type of Turkish Traditional Fermented Sausage Produced with Different Meat: Fat Ratios

  • This study aims to evaluate biogenic amine levels of bez sucuks (BS) produced with different meat:fat ratios. For this, three BS groups were manufactured with meat:fat ratios of 90:10 (BS10), 80:20 (BS20), and 70:30 (BS30). The pH and water activity values and biogenic amine amounts of sucuk samples were determined during processing and storage periods and the pH values of the initial mixtures of BS samples were in the range 5.51-5.74, decreasing to 4.72-4.94 by the $14^{th}$ day. The water activity values of BS samples showed significant decreases as a result of the drying stage and reached to range 0.913-0.935 on the $14^{th}$ day of processing (p<0.05). Although BS10 had the highest tyramine (434.12 mg/kg), histamine (5.69 mg/kg), cadaverine (12.48 mg/kg), putrescine (17.83 mg/kg), 2-phenylethylamine (15.43 mg/kg), and tryptamine (122.41 mg/kg) levels at the end of processing stage (p<0.05), spermine and spermidine levels did not differ between the BS samples due to their utilization of different meat:fat ratios (p>0.05). Similarly, the tryptamine (205.11 mg/kg), putrescine (43.57 mg/kg), and tyramine (766.23 mg/kg) levels of BS10 were higher than BS20 and BS30 samples at the end of storage (p<0.05). The results showed that BS10 with the highest meat ratio had the highest tryptamine, putrescine, and tyramine levels at the end of the processing and storage period.

Biogenic Amine Degradation by Bacillus Species Isolated from Traditional Fermented Soybean Food and Detection of Decarboxylase-Related Genes

  • Biogenic amines in some food products present considerable toxicological risks as potential human carcinogens when consumed in excess concentrations. In this study, we investigated the degradation of the biogenic amines histamine and tyramine and the presence of genes encoding histidine and tyrosine decarboxylases and amine oxidase in Bacillus species isolated from fermented soybean food. No expression of histidine and tyrosine decarboxylase genes (hdc and tydc) were detected in the Bacillus species isolated (B. subtilis HJ0-6, B. subtilis D'J53-4, and B. idriensis RD13-10), although substantial levels of amine oxidase gene (yobN) expression were observed. We also found that the three selected strains, as non-biogenic amineproducing bacteria, were significantly able to degrade the biogenic amines histamine and tyramine. These results indicated that the selected Bacillus species could be used as a starter culture for the control of biogenic amine accumulation and degradation in food. Our study findings also provided the basis for the development of potential biological control agents against these biogenic amines for use in the food preservation and food safety sectors.
