This study was designed to investigate the collagen metabolism and tyrosinase activity of Meretrix extracts (ME). The effect of ME on type I procollagen production and collagenase activity in human normal fibroblasts HS68 after UVB (312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of ME was measured as well. Type I procollagen production was recovered by ME in UVB damaged HS68 cells. The increased collagenase activity after UVB damage was significantly recovered by ME. The tyrosinase activity and L-DOPA oxidation were significantly reduced as well. However, the effects on tyrosinase activity and L-DOPA oxidation were not powerful enough to be used as whitening agents. ME showed the anti-wrinkle effects and some whitening effects in vitro. These results suggest that ME may be a useful drug as an anti-wrinkle treatments.
UV exposure induces matrix metalloproteinases (MMPs) and extracellular matrix-degrading enzymes expression. We studied the protective effect of Rubus occidentalis seed against UVB-generated skin photoaging using human fibroblast cells (CCD-986sk). We used an ELISA kit to measure the supernatents of procollagen type I and MMP-1 in CCD-986sk cells after they were exposed to UVB irradiation. The CCD-986sk cells that were used with RC-E/E after the UVB irradiation caused higher levels of type I procollagen and lesser levels of MMP-1 compared with the control group. Furthermore, the RC-E/E treated group showed lesser MMP-1 levels and higher procollagen type I levels than the untreated counterpart. Therefore, it can be concluded that Rubus occidentalis seed can prevent from skin photoaging.

Although many studies have been performed to elucidate the molecular consequence of factors that regulate skin aging, little is known about the effect of green tea catechins except EGCG. The matrix metalloproteinase (MMP), can degrade matrix proteins and results in a collagen deficiency in photodamaged skin, are known to play an important role in photoaging. This study, investigated the effects of green tea catechins on the UVA-induced MMP-1 expression, activity of MMP-2 and synthesis of type I procollagen in human dermal fibroblasts. We examined eight catechins that naturally exist in green tea leaves and compared their efficacies among them. Most of catechins inhibited the expression of MMP-1 in dose dependent manner, and the levels were reduced, especially, 57.4 and 68.2% by treatment with $1{\mu}M$ of epigallocatechin-3-gallate (EGCG) and gallocatechin-3-gallate (GCG), respectively. Also, catechins significantly suppressed the activities of MMP-2. Catechins also induced the expression of type I procollagen, however, they acted only at the concentration below $1{\mu}M$ interestingly. Furthermore, when EGCG:GCG:ECG had the ratio of 0.5:1.5:.1.3, they presented the most effective on procollagen synthesis. Therefore, we concluded that catechins significantly inhibited MMPs and induced collagen synthesis. Taken together, all these results suggested that green tea catechins might be good natural materials act as an anti-photoaging and a skin-aging improving agent.

The tyrosinase activity after treatment of AGR was measured. Results : Type I procollagen production was recovered by AGR in UVB damaged HS68 cells. The increased collagenase activity after UVB damage was significantly recovered by AGR and the tyrosinase activity was significantly reduced. However, the L-DOPA oxidation was not changed. Conclusion : AGR showed the anti-wrinkle effects and whitening effects in vitro. These results suggest that AGR may have potential as an anti-aging ingredient in cosmetic herbs.

The tyrosinase activity after treatment of SP was measured as well. Results : Type I procollagen production was recovered by SP in UVB damaged HS68 cells. The increased collagenase activity after UVB damage was significantly recovered by SP. The tyrosinase activity was significantly reduced as well. However, the L-DOPA oxidation was not changed. Conclusion : SP showed the anti-wrinkle effects and whitening effects in vitro. These results suggest that SP may be a potential pharmacopuncture as an anti-aging pharmacopuncture treatments.
Biosynthetic processing of fibrillar procollagens is essential for producing mature collagen monomers that polymerize into fibrils by a self-assembly process. The metalloproteinase ADAMTS-2 is the major enzyme that processes the N-propeptide of type I procollagen in the skin and also of type II and type III procollagens. Mutations in the ADAMTS-2 gene cause dermatospraxis in animals and Ehlers-Danlos syndrome VIIC in humans, both of which are characterized by the accumulation of type I pN-collagen and the formation of abnormal collagen fibrils in the skin. Despite its importance in procollagen processing, little is known about the regulation of ADAMTS-2 expression. Here, we demonstrate that ADAMTS-2 can be regulated by 1,25-dihydroxyvitamin D3, an inducer of type I procollagen synthesis. This steroid hormone induced ADAMTS-2 mRNA ${\sim}3-fold$ in MG-63 human osteosarcoma cells and MC3T3-E1 murine osteoblastic cells. This induction was dose- and time-dependent in MG-63 cells. In contrast, secreted ADAMTS-2 protein was increased only 1.4-fold with 1,25-dihydroxyvitamin D3. Finally, 1,25-dihydroxyvitamin D3 in the presence of ascorbate increased levels of secreted ADAMTS-2 1.9-fold over ascorbate treatment alone, which did not appreciably change ADAMTS-2 expression. These data indicate that the regulation of ADAMTS-2 is coupled with the synthesis of type I procollagen through 1,25-dihydroxyvitamin D3 signaling and may involve translational or posttranslational control.
The natural plants were screened for new cosmetic materials. Many parts of Nelumbo nucifera has been investigated for anti-wrinkling and anti-furrowing effects. The leaves. flowers and stamens of Nelumbo nucifera were extracted with ethanol or distilled water. The effects of cell viability and $H_2O_2-induced$ cytotoxicity were studied on CCD-986sk human fibroblast cell line by MTT assay. We measured various effects related to skin such as the synthesis of MMP-1 and type I procollagen. The ethanol extract of Nelumbo nucifera leaves (ENL) at concentrations of 30 ${\mu}g/ml$ showed maximum cell viability on CCD-986sk cells by 129.70%. and $H_2O_2-induced$ cytotoxicity was blocked by ENL (5. 10. 20. 30. and 40 ${\mu}g/ml$) concentration-dependently. The synthesis of MMP-1 and type I procollagen were positive in ENL. These results suggest that ENL has anti-oxidant and anti-furrowing effects.
Purpose: Solar ultraviolet (UV) radiation causes inflammation and matrix metalloproteinase (MMP) overexpression and extracellular matrix depletion, leading to skin photoaging such as wrinkle formation, dryness, and sagging. Activation of MMP is influenced by various molecules such as reactive oxygen species (ROS), proinflammatory cytokines, and transient receptor potential vanilloid type (TRPV)-1, which are increased in UV-irradiated skin cells. Aralia elata (AE) ethanolic extract was reported to inhibit ROS generation caused by UVB-irradiation in keratinocytes. In this study, we investigated the photoprotective effect of AE ethanolic extract on UVB-irradiated human keratinocytes (HaCaT) and human dermal fibroblasts (HDF). Methods: AE was freeze-dried, extracted in 70% ethanol, and concentrated. Skin cells were treated with AE extract for 24 h and then exposed to UVB ($55mJ/cm^2$). After 48 h of incubation, proinflammatory cytokines, MMP-1, type-1 procollagen, and TRPV-1 levels were measured by ELISA or Western blotting. Results: Treatment with AE extract ($100{\mu}g/mL$) significantly inhibited UVB-induced IL-6, IL-8, and $PGE_2$ production in HaCaT by 25.6%, 5.3%, and 70.2%, respectively, and also inhibited elevation of MMP-1 and TRPV-1 caused by UVB irradiation by 20.0% and 41.9%, respectively (p < 0.05). In HDF, AE extract treatment significantly inhibited both elevation of MMP-1 and reduction of type-1 procollagen caused by UVB irradiation (p < 0.05). In addition, type-1 procollagen was elevated by AE extract treatment in normal HDFs (p < 0.05). Conclusion: AE 70% ethanol extract has photoprotective ability via reduction of proinflammatory mediators, TRPV-1 and MMP-1 production, and elevation of collagen synthesis. Our findings suggest that AE extract might be a good natural material to protect against UVB-induced premature skin aging.
목적 : 본 연구는 녹용약침액의 주름 개선 효과에 대하여 여러 가지 in vitro 실험을 통해 확인해 보고자 계획되었다. 방법 : 녹용약침액의 항산화효과를 DPPH(1,1-diphenyl-2-picrylhydrazyl) 자유라디칼 소거법을 이용하여 측정하였고, 또, elastase 억제 효과를 측정하였다. 그리고 녹용약침액이 사람 정상 섬유아세포 HS 68에서 UVB 조사 후 type I procollagen의 생산량 회복에 미치는 효과를 ELISA법을 이용하여 측정하였다. 결과 : 녹용약침액에서 우수한 DPPH 자유라디칼 소거 효과가 관찰되었다. 또한 통계적으로 유의한 elastase 활성 억제 효과를 관찰할 수 있었다. 사람 섬유아세포 HS 68을 이용한 실험에서는 녹용약침액 처치군에서 UVB 조사로 감소된 type I procollagen이 유의하게 회복되는 것이 관찰되었다. 결론 : 본 연구 결과에 의하면, 녹용약침액에는 유효한 주름개선 효과가 있어, 미용약침 소재로 개발할 수 있을 것으로 사료된다.
Objectives : Draconis Resina (DR), the resin of Daemonorops draco Bl., is used to circulate the blood and to stop bleeding. It also has been used to generate flesh including ulceration. The present study investigated the effects of DR extract on collagen metabolism in human fibroblasts and tyrosinase activity in mushroom tyrosinase.Methods : The effect of DR extract on type I procollagen production (collagen type I synthesis) and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts cell line. Hs68 cells after ultraviolet B (UVB, 312 nm) irradiation was measured using the enzyme - linked immunosorbent assay (ELISA). The tyrosinase activity was also measured to find out the whitening effects in mushroom tyrosinase by ELISA method.Results : There was no cytotoxicity at DR extract at concentrations of 10 μg/ml, 30 μg/ml, and 100 μg/ml. DR extract significantly inhibited the increase of collagenase activity, whereas it did not show on the reduction of type I procollagen in UVB damaged Hs68 cells. DR extract did not reduce the L - DOPA oxidation. However, it significantly reduced the tyrosinase activity by DR extract at concentraions of 0.1 mg/ml, 1 mg/ml and 10 mg/ml.Conclusions : In conclusion, DR showed the anti-wrinkle and whitening effects via the inhibition of collagenase production and the tyrosinase activity. These results suggest that DR may have potential as an anti-aging ingredient in cosmetic herb markets.
