Stable isotope approach for aquatic ecology and environmental sciences has been introduced as very useful technique since 1980s and also has been applied to investigate various issues in aquatic ecology and environmental study last 10 years in Korea. Especially carbon and nitrogen isotope ratios have been mainly used to understand food web energy flow and ecosystem structure. In addition, nitrogen isotope ratio has been applied for nitrogen cycle and source identification as well as biomagnification studies. However, large temporal or spatial variations of nitrogen isotope ratio of primary producer have been found in many aquatic environments, and it is regarded as the critical problems to determine trophic level of aquatic animals. Recently, the compound specific isotope analysis of nitrogen within individual amino acids has been developed as an alternative method for trophic ecology. This article introduces the progress history of stable isotope application in aquatic ecology and environmental sciences, and also suggests new direction based on future prospects in stable isotope ecology and environmental study.
Pneumocystis carinii causes serious pulmonary infection in immuno-suppressed patients. This study was undertaken to observe the cytoskeletal proteins of P. carinii by immune-electron microscopy. P. carinii infection was experimentally induced by immunosuppression of Sprague-Dawley rats for seven weeks, and their lungs were used for the observations of this study. The gold particles localized actin, tropomyosin, and tubulin. The actin was irregularly scattered in the cytoplasm of the trophic forms but was much more concentrated in the inner space of the cell wall of the cystic forms called the inner electron-lucent layer No significant amount of tropomyosin was observed in either trophic forms or cystic forms. The tubulin was distributed along the peripheral cytoplasm and filopodia of both the trophic and cystic forms rather than in the inner side of the cytoplasm. Particularly, in the cystic forms, the amount of tubulin was increased and located mainly in the inner electron-lucent layer of the cell wall where the actin was concentrated as well. The results of this study showed that the cell wall of P carinii cystic forms is a structure whose inner side is rich in actin and tubulin. The location of the actin and tubulin in P. carinii suggests that the main role of these proteins is an involvement in the protection of cystic forms from the outside environment by maintaining rigidity of the cystic forms.
This study compares the abundance and community structure of zooplankton organisms from pelagic regions, and considers particularly the trophic levels vs. zooplankton abundances and biomass. Zooplankton samples were collected three times from May to November 2022, at 30 temperate lakes and reservoirs, which belong to four different river basins. The total zooplankton abundance, biomass and species index were showed considerable spatial variation. The spatial pattern of rotifer abundance was similar to that of total zooplankton abundance, while there were not showed similar patterns of zooplankton biomass (㎍ L-1) in lentic ecosystems. The rotifer strongly dominated the zooplankton assemblage in smaller lentic system than that of larger. A total of 130 species of zooplankton were identified (83 rotifers, 34 cladocerans and 13 copepods). The total average of zooplankton abundance and biomass were 213.7±342.3 Ind. L-1 (n=129) and 1382.8±1850.4 ㎍ L-1, respectively. Total and average of zooplankton abundance were usually dominated by the rotifers (>56.9%), while those of zooplankton biomass were dominated by the cladocerans and copepods (>73.6%) in lentic ecosystems. Considering the Trophic State Index (TSI), the factors of zooplankton abundance and biomass were included in between meso- and eutrophic states(27 lakes, 90% of all). The mean abundance and biomass of zooplankton in eutrophic systems were higher than that of meso- and hypertrophic systems. From this result, we suggest that management strategy for the lentic ecosystem water environment has to be focused more on small-sized lakes and reservoirs, in terms of zooplankton assemblages.

Hypoxia can indeed impact the survival of protists, which play a crucial role in marine ecosystems. To better understand the protistan community structure and species that can thrive in hypoxic waters, we collected samples from both the surface and bottom waters during the hypoxic period in Jinhae and Masan Bays and the non-hypoxic period in Jinhae Bay. Subsequently, we utilized metabarcoding techniques to identify the protistan species. During hypoxia, with dissolved oxygen concentrations of 0.8 mg L-1 in Jinhae Bay and 1.8 mg L-1 in Masan Bay within the bottom waters, the phylum Dinoflagellata exhibited the highest amplicon sequence variants richness among the identified protist phyla. Following the Dinoflagellata, Ochrophyta and Ciliophora also displayed notable presence. In hypoxic waters of Jinhae and Masan Bays, we identified a total of 36 dinoflagellate species that exhibited various trophic modes. These included one autotrophic species, 14 mixotrophic species, 9 phototrophic species with undetermined trophic modes (either autotrophic or mixotrophic), 2 kleptoplastidic species, and 10 heterotrophic species. Furthermore, the hypoxic bottom water exhibited a greater number of heterotrophic dinoflagellate species compared to the non-hypoxic surface water within the same water column or the non-hypoxic bottom water. Therefore, feeding by mixotrophic and heterotrophic dinoflagellates may be partially responsible for their dominance in terms of the number of species surviving in hypoxic waters. This study not only introduces the initial documentation of 26 dinoflagellate species surviving in hypoxic conditions but also establishes a foundation for a more comprehensive understanding of the ecophysiology of dinoflagellates in hypoxic marine environments.
Recently, quantitative analyses of food web structure based on carbon and nitrogen stable isotopes are widely applied to environmental assessments as well as ecological researches of various ecosystems, particularly rivers and streams. In the present study, we analyzed carbon and nitrogen stable isotope ratios of POM (both planktonic and attached forms), zooplankton, benthic macroinvertebrates and fish collected from 6 sites located at Nakdong River. Samples were collected from upstream areas of 5 weirs (Sangju, Gangjeong-Goryeong, Dalseong, Hapcheon-Changnyeong, and Changnyeong-Haman Weirs) and one downstream area of Hapcheon-Changnyeong Weir in dry season (June) and after rainy season (September). We suggested ranges of their carbon and nitrogen stable isotope ratios and calculated their trophic levels in the food web to compare their temporal and spatial variations. Trophic levels of organisms were relatively higher in Sangju Weir located at upper part of Nakdong River, and decreased thereafter. However, the trophic levels were recovered at the Changnyeong-Haman Weir, the lowest weir in the river. The trophic level calculated by nitrogen stable isotope ratios showed more reliable ranges when they were calculated based on zooplankton than POM used as baseline. The suggested quantitative ecological information of the majority of biological communities in Nakdong River would be helpful to understand the response of river food web to environmental disturbances and can be applied to various further researches regarding the quantitative approaches for the understanding food web structure and function of river ecosystems as well as restoration.
Compound specific isotope analysis of amino acids (CSIA-AAs) is being highlighted as an alternative approach for overcoming some restrictions in application of stable isotope analysis of bulk tissue (SIA) for trophic position (TP) estimation. However, this approach has rarely been applied in Korea. The present study determines TP of two Polychaeta (Nephtyidae and Glyceridae) and two fish species(Platycephalus indicus and Lophius litulon) collected from the Geum River estuary using nitrogen isotope ratio of amino acid and compared with the TP values estimated by SIA. The Polychaeta species, sampled in two sites, showed similar TP between SIA(2.7 and 3.1) and CSIA-AAs (2.6 and 3.1). However, for both fish species, TP values displayed a large difference between SIA (3.1 and 2.3) and CSIA-AAs (3.8 and 3.7). In this study TP values estimated by CSIA-AAs showed more similar to the previously reported gut content analysis of both fishes compared with the results of SIA. Current study suggests the applicability of nitrogen isotope ratio of amino acid to understand coastal ecosystem structure and trophic ecology.

Goals, research direction, and operational structure of the Global Ocean Ecosystem Dynamics (GLOBEC), which is co-sponsored by the Scientific Committee on Oceanic Research (SCOR) and the Intergovemmental Oceanographic Commission (IOC), are described. Following the Joint Global Ocean Flux Study (JGOFS) activities on the lower trophic level in ecosystem during 1980s-90s, scientists have considered the responses of the mid and higher trophic levels to the environmental or climate change. The scientific steering committee of the GLOBEC published the implementation plan in 1999, and the IGBP will support the GLOBEC as one of its core projects for 10 years. The GLOBEC programme has four major components: the research foci, framework activities, regional programmes, and integrating activity. The core of GLOBEC research is the four research foci: retrospective analyses, process studies, predictive and modelling capabilities, and feedbacks. Currently, four regional programmes have been undergoing in the world ocean: Southern Ocean GLOBEC (SO-GLOBEC), Small Pelagic Fishes and Climate Changes (SPACC), Cod and Climate Change (CCC), and Climate Change and Carrying Capacity (CCCC). Also, national GLOBEC programmes were already established in 9 nations.

Seungchon Weir was constructed in 2010 on the downstream region of Yeongsan River watershed for four major river projects. The purpose of our study was to determine whether fish compositions and distributions differ between the upper ($U_w$) and lower regions ($L_w$) of Seungchon Weir during May - December 2011 along with hydroacoustic fish surveys using Eco-Sounder monitoring ($E_SM$) technique. Total number of fish species sampled by conventional fish sampling at all sites was 29, and 21 and 24 species occurred in the $U_w$ and $L_w$, respectively. Most dominant species was Hemiculter eigenmanni (43%) in the $U_w$ and Opsaiichthys uncirostris amurensis (21%) in the $L_w$, indicating a distinct differences of the species composition. According to hydroacoustic approach of ESM, fish density was significantly higher (p < 0.05) in the $L_w$ section of 250 m than any other sections, This outcome indicates that fishes moving toward the upstream were blocked by weir construction. According to analysis of tolerance guilds, tolerant species (TS) were made of 88% and 79% in the $U_w$ and $L_w$ region, respectively, while the proportions of sensitive species were < 0.1% of the total in the both regions. Trophic guild analysis showed that the proportion of omnivore species were 58% and 40% in the $U_w$ and $L_w$ region, respectively, while insectivores were made of 16% and 21% in the $U_w$ and $L_w$ region, respectively. Both analyses of tolerance guilds and trophic guilds indicate the biological degradation of the waterbody. Fish community structure analysis, based on species diversity, dominance, and evenness indices, showed higher community stability in the $L_w$ region than $U_w$ region. Exotic species of largemouth bass were largely increased in this survey (14%), compared to the past (< 1%), indicating a modification of fish trophic structure.

Korea is one of the most advanced countries in kelp aquaculture. The brown algae, Undaria pinnatifida and Saccharina japonica are major aquaculture species and have been principally utilized for human food and abalone feed in Korea. This review discusses the diversity, population structure and genomics of kelps. In addition, we have introduced new cultivar development efforts considering climate change, and potential carbon sequestration of kelp aquaculture in Korea. U. pinnatifida showed high diversity within the natural populations but reduced genetic diversity in cultivars. However, very few studies of S. japonica have been conducted in terms of population structure. Since studies on cultivar development began in early 2000s, five U. pinnatifida and one S. japonica varieties have been registered to the International Union for the Protection of New Varieties of Plants (UPOV). To meet the demands for seaweed biomass in various industries, more cultivars should be developed with specific traits to meet application demands. Additionally, cultivation technologies should be diversified, such as integrated multi-trophic aquaculture (IMTA) and offshore aquaculture, to achieve environmental and economic sustainability. These kelps are anticipated to be important sources of blue carbon in Korea.
The object of this study was to evaluate lotic ecosystem health using multiple eco-metric approaches such as water chemistry diagnosis, physical habitat health evaluations, and biological integrity modeling at 100 streams of four major watersheds. For the study, eight chemical water quality parameters such as nutrients (N, P) and organic material were measured and 11-metric models of Qualitative Habitat Evaluation Index (QHEI) and multiple eco-metric health assessment model (MEHA) were applied to the four major watershed. Nutrient analysis of nitrogen (N) and phosphorus (P) in all watersheds indicated a eutrophic state depending on the locations of sampling streams. Physical habitat health, based on the QHEI model, averaged 114 (range: 56 - 194), judging as a "good condition" by the criteria of Plafkin et al. (1989). In addition, primary (H1 - H4), secondary (H5 - H7), and tertiary habitat metric variables (H8 - H11) were analyzed in relation to the physical habitat degradations. The plots of tolerant species ($P_{TS}$) and sensitive species ($P_{SS}$) to water quality showed that the proportions of $P_{TS}$ had positive linear functions with nutrients, and that the $P_{SS}$ had inverse linear relations with the chemical variables. The model of eco-metric health assessment showed that mean MEHA was 20.4, indicating a fair condition. Overall, our data suggest that water chemistry, based on nutrients and organic matter, directly modified the trophic structures in relation to food chain in the aquatic ecosystems, and then these directly influenced the compositions of tolerance/sensitive species, resulting in degradations of overall ecological health.
