High-precision THz Dielectric Spectroscopy of Tris-HCl Buffer

  • Tris-HCl buffer solution is extensively used in biochemistry and molecular biology to maintain a stable pH for biomolecules such as nucleic acids and proteins. Here we report on the high-precision THz dielectric spectroscopy of a 10 mM Tris-HCl buffer. Using a double Debye model, including conductivity of ionic species, we measured the complex dielectric functions of Tris-HCl buffer. The fast relaxation time of water molecules in Tris-HCl buffer is ~20% longer than that in pure water while the slow relaxation time changes little. This means that the reorientation dynamics of Tris-HCl buffer with such a low Tris concentration is quite different from that of pure water.

유즙내에서 메티실린 내성을 지닌 젖소 유방염 주요 원인균에 대한 항생제와 EDTA-Tris의 병합의 효과 (The Effect of Antibiotics in Combination with EDTA-Tris on the Methicillin-Resistant Major Pathogens of Bovine Mastitis in Milk)

  • The combined effects of EDTA-Tris and eighteen antimicrobial agents have been evaluated in eight clinically isolated methicillin-resistant bacteria (Staphylococcus aureus, Escherichia coli, Streptococcus uberis and Streptococcus agalactiae) from bovine mastitis. The antimicrobial activity was evaluated by measuring the minimal bactericidal concentration (MBC) for the antibiotics alone or in combination with EDTA-Tris in Mueller-Hilton broth and milk. Combined use of EDTA-Tris and antibiotics potentiated or antagonized activity of antibiotics against mastitic pathogens. Milk increased the antibiotic potency of erythromycin and spiramycin on S. aureus. Culture in milk changed patterns of EDTA-Tris combinational effects compared with that in standard Mueller-Hilton broth. Combined with EDTA-Tris in milk, synergic effects were observed in colistin, dihydrostreptomycin, kanamycin, erythromycin, gentamycin, oxytetracycline, streptomycin to E. coli, Str. uberis, and Str. agalactiae. However, significant antagonistic effects of milk on antibiotic susceptibility in combination with EDTA-Tris were noted in neomycin, streptomycin, penicillin, roxithromycin, and amoxicillin. This study indicates that combination therapy of EDTA-Tris with antibiotics in bovine mastitis should be used with caution because of the possible antagonistic effects of antibiotic combination with EDTA-Tris on mastitic pathogens. In addition, antibiotic susceptibility test in combination with EDTA-Tris in milk culture condition can be benefit in search of effective treatment regimen for some antibiotic-resistant bacteria of mastitis.

Preservation of Simmental bull sperm at 0℃ in Tris dilution: effect of dilution ratio and long-distance transport

  • Objective: This study aimed to assess the impact of the dilution ratio of Tris diluent, storage at 0℃, and long-distance transportation on the spermatozoa of Simmental cattle. It also validated the feasibility of the regional distribution of fresh semen. Methods: In experiment 1, semen was diluted at four dilution ratios (1:6, 1:9, 1:12, and 1:15) to determine the optimal dilution ratio of Tris diluent. In experiment 2, we assessed sperm viability, progressive motility (objectively assessed by computer-assisted sperm analyzer), and acrosome intactness in Tris dilutions kept at constant 0℃ for 1, 3, 6, 9, and 12 days. We compared them to Tianshan livestock dilutions (Commercial diluent). In experiment 3, semen was diluted using Tris diluent, and sperm quality was measured before and after long-distance transport. Artificial insemination of 177 Simmental heifers compared to 156 using Tianshan Livestock dilution. Results: The outcomes demonstrated that 1:9 was the ideal Tris diluent dilution ratio. The sperm viability, Progressive Motility, and acrosome integrity of both Tris and Tianshan dilutions preserved at 0℃ gradually decreased over time. sperm viability was above 50% for both dilutions on d 9, with a flat rate of decline. The decrease in acrosome integrity rate was faster for Tianshan livestock dilutions than for Tris dilutions when stored at 0℃ for 1 to 6 days. There was no significant difference (p>0.05) in sperm viability between semen preserved in Tris diluent after long-distance transportation and semen preserved in resting condition. The conception rates for Tris dilution and Tianshan livestock dilution were 49.15% and 46.15% respectively, with no significant difference (p>0.05). Conclusion: This shows that Tris diluent is a good long-term protectant. It has been observed that fresh semen can be successfully preserved for long-distance transport when stored under 0℃ conditions. Additionally, it is feasible to distribute semen regionally.

  • The tris(hydroxymethyl)aminomethane (Tris)-catalyzed hydrolysis of p-nitrotrifluoroacetanilide was kinetically studied. On the contrary to the previously reported results (R. M. Pollack and T. C.Dumsha, J. Am. Chem. Soc., 1973, 95, 4463), the dependence of the rate on [Tris] consists of an initial curve portion and a subsequent linear increase. This indicates that erroneous conclusions were made in the reported work due to the insufficient amount of data. The initial portions of the rate[Tris] profiles are attributed to the catalyzed breakdown of the tetrahedral intermediate, and the linear portions to the general base-catalyzed water attack at the substrate.

  • In Tris-iso-butanol (TIB; Tris buffer pH 8.8 and 1% iso-butanol)-treated chloroplasts, oxygen evolving activity was more inhibited than Tris-treated chloroplasts, but restored highly by 2,6-dichlorophenol-indophenol (DCPIP) and photoreactivation. To understand the mechanism of this results of TIB in photosynthetic electron transport, system, oxygen consumption and evolution of PS I and PS II were measured and protein of the chloroplasts was analysed. In Tris- and TIB-treated chloroplasts, oxygen evolving activity was increased according to the light intensity. Under 48 W·m-2 light intensity, the oxygen evolving activity in both chloroplasts were similar but as the light intensity was increased, TIB-treated chloroplasts showed higher activity. Under 240 W·m-2 light intensity, TIB-treated chloroplasts showed about 25% higher oxygen evolving activity than Tris-treated chloroplasts. Oxygen evolving activity was increased after photoreactivation in both Tris-treated and TIB-treated chloroplasts. Addition of NH4Cl increased the activity in both chloroplasts but in TIB-treated chloroplasts the increase was 30% higher than that in Tris-treated chloroplasts. In PS I, oxygen evolving activity was not inhibited by both treatments whereas in PS II, significant difference was observed between two treatments. Addition of Mn2+ and Ca2+ enhanced oxygen evolution in both Tris- and TIB-treated chloroplasts. Though enhancement was higher in TIB-treated chloroplasts. No difference was observed n protein analysis of the two thylakoid membrane.

In-situ Thermally Curable Hyper-branched 10H-butylphenothiazine

  • A hyper branched 10-butylphenothiazine with in-situ thermally curable methacrylate (1,3,5-tris-[$\{$10-Butyl-3-(4-(2-methyl-acryloyloxy)-phenyl)-7-yl-10H-phenothiazine$\}$]-benzene, (tris-PTMA)) was synthesized successfully. From the TGA thermogram of tris-PTMA was thermally stable up to $336^{\circ}C$. In the first heating scan of DSC thermogram, tris-PTMA showed glass transition temperature (Tg) at $140^{\circ}C$ and broad endothermic process in the region of $144-179^{\circ}C$, which is thermally curing temperature. In the second heating process, $T_g$ exhibited at $158.7^{\circ}C$ and endothermic process was not observed. Thermally cured tris-PTMA showed no big change in the UV-visible spectrum after washing with organic solvent such as methylene chloride, chloroform, toluene, indicating that thermally cured film was very good solvent resistance. Thermally cured tris-PTMA was electrochemically stable and the HOMO energy level of tris-PTMA was -5.54 eV. The maximum luminance efficiency of double layer structured polymer light-emitting diode based on in-situ thermally cured tris-PTMA was 0.685 cd/A at 16.0 V, which was higher than that of the device without thermally cured tris-PTMA (0.348 cd/A at 15.0 V).

저장 감귤의 부패에 관여하는 Penicillium spp.에 대한 Iminoctadine tris(albesilate)와 Kresoxym-methyl의 방제 효과 (Protective Effect of Iminoctadine tris(albesilate) and Kresoxim-methyl Fungicides to Citrus Postharvest Diseases caused by Penicillium spp.)

  • 본 연구는 감귤을 부패시키는 Penicillium spp.에 대한 kresoxim-methyl 과 iminoctadine tris(albesilate)의 방제효과를 검정하기 위하여 실시하였다. P. italicum과 P. digitatum의 균사생장에 대한 iminoctadine tris(albesilate)의 $EC_{50}$값은 각각 0.01-0.02와 $0.01{\mu}g$ a.i./mL이었지만, 아직 미동정인 새로운 유형의 Penicillium spp.균은 iminoctadine tris(albesilate)가 $0.64{\mu}g$ a.i./mL들어 있는 PDA배지에서도 거의 균사생장이 억제되지 않았다. 또한 P. italicum과 P. digitatum의 발아와 발아관의 생장에 대한 iminoctadine tris(albesilate)의 $EC_{50}$값은 $0.0013-0.0025{\mu}g$ a.i./mL이었지만 새로운 유형치 Penicillium spp.는 $0.2{\mu}g$ a.i./mL 농도에서도 포자 발아가 거의 억제되지 않았다. P. italicum과 P. digitatum 그리고 미동정 Penicillium spp.의 균사생장에 대한 kresoxim-methyl의 $EC_{50}$은 각각 0.08-0.16, 0.04 그리고 $0.16{\mu}g$ a.i./mL이었으며 P. italicum과 새로운 유형의 Penicillium spp.의 포자 발아 및 발아관 생장에 대한 $EC_{50}$$0.04{\sim}0.08{\mu}g$ a.i./mL이며 P. digitatum에 대해서는 $0.01{\sim}0.02{\mu}g$ a.i./mL였다. Kresoxim-methyl과 iminoctadine tris(albesilate)의 저장병에 대한 방제효과를 알아보기 위하여 이들 약제를 각각 $155{\mu}g$ a.i./mL과 $200{\mu}g$ a.i./mL의 농노로 수상살포하고 7일 후 수확하여 90일간 저장한 결과 무처리구와 thiophanate-methyl $700{\mu}g$ a.i./mL을 처리한 대조구에서의 부패율은 각각 $20.3{\pm}10.0%\;and\;19.5{\pm}9.6%$으로 높은데 반하여 tresoxim-methyl과 imonoctadine tris(albesilate) 처리구에서의 부패율은 각각 $3.6{\pm}1.8%$$5.9{\pm}1.8%$로써 방제효과가 높았다. 이들 처리로부터 저장 후 0, 30, 60, 90일에 각각 iminoctadine tris(albesilate)와 kresoxim-methyl의 잔류량을 조사해본 결과 각 조사 기간 중 iminoctadine tris(albesilate)의 최대잔류량은 0.10 ppm이었고 kresoxlm-methyl의 최대 잔류량은 0.45 ppm이었다.

Tris(2-cyclohexylaminoethyl)amine-Zn(II) 착물의 안정성 (Stability of Tris(2-cyclohexylaminoethyl)amine-Zn(II) Complex)

  • Cyclohexanone,과 tris(2-aminoethyl)amine을 이용하여 Schiff 연기 축합반응으로 tren의 유도체인 tris(2-cyclohexylaminoethyl)amine (L)을 합성하였다. 또한 합성한 tren의 유도체인 tris(2-cyclohexylaminoethyl)amine (L)과 Zn(II) 착물의 열역학적 특성과 안정도 상수, Zn(II)와의 착물 조성비 등을 순환전압전류법과 열량계법으로 측정하였다. Zn(II)과 [Zn(II)-L] 순환전압전류 곡선을 0${sim}$-1.5 V vs. Ag/AgCl의 가전압 범위에서 측정하였다. 금속인 경우, -1.02V와 -0.48V vs. Ag/AgCl에서 각각 환원피이크와 산화피이크가 나타났으며, 금속착물인 경우에는 -1.19V와 -0.45V vs. Ag/AgCl에서 각각 환원피이크와 산화피이크가 나타났다. 또한 피이크 전류(IP)는 주사속도의 평방근 $(v^{1/2})$에 비례하였으며 이것은 전류의 유형이 확산 지배적인 전류임을 나타낸다. 그리고 [Zn-L] 착물에 대해서 전압전류법적으로 구한 안정도상수는 logK$_f$ = 5.8, 결합비는 1:1을 나타내었다. 또한 열량계법적으로 [Zn-L] 착물의 열역학적 파라메타를 조사한 결과, 리간드 L과 Zn(II)는 1:1의 4 배위수를 가지는 착물을 이룬다는 것을 알 수 있었고, 이때 25 ${\circ}$C에서 logK=5.4, ${\Delta}H$= -53.0 kJ/mol, ${\Delta}$G의 값은 -31.1 kJ/mol이었으며 T${\Delta}$S는 -21.9 J/K${\cdot}$mole이었다.

Glycerol-free TRIS 배지내 glucose를 이용한 개 정자 동결: 포도당 농도, 노출시간 (Dog Sperm Cryopreservation Using Glucose in Glycerol-free TRIS: Glucose Concentration, Exposure Time)

  • 개 정액 동결을 위한 glucose가 첨가된 glycerol-free TRIS 희석액을 개발하기 위해 glycerol-free TRIS내 알맞은 glucose의 양과 0.3 M glucose가 첨가된 glycerol-free TRIS내 정자의 적정 노출시간을 조사하였다. 여섯 마리의 수캐의 사출액을 0.04 M glucose가 첨가된 glycerol-free TRIS내에서 $4^{\circ}C$까지 100분 동안 냉각한 후 서로 다른 glucose농도 (0 M, 0,04 M, 0.1 M, 0.2 M, 0.3 M)의 glycerol-free TRIS에서 30분 동안 냉각하여 동결하였다. $37^{\circ}C$에서 25 초 동안 융해한 후 정자의 막 고유성과 첨단체 고유성을 검사하였다. 부가적으로 0.3 M glucose가 첨가된 glycerol-free TRIS내 정자의 적정 노출시간에 따른 정자의 동결 후 운동성, 생존성, DNA 고유성을 확인하였다. 막 고유성과 첨단체 고유성은 각각 6-carboxyfluoresceindiacetate(6-CFDA)/propidium iodide(PI) fluorescent staining와 Pisum sativum agglutinin conjugated-fluorescein isothiocyanate 방법에 의해 검사하였다. DNA 고유성은 terminal deoxynucleotidyl transferase dUTP nick end labeling로 염색하여 flow cytometry로 검사하였다. 0.2 M 또는 0.3 M glucose가 첨가된 glycerol-free TRIS에서 동결된 정자가 낮은 농도의 glucose가 첨가된 희석액에서 동결된 정자보다 막 고유성이 높게 나타났으며(p<0.05), 첨단체 고유성은 0.3 M 군에서 높게 나타났다(p<0.05). 운동성은 50 분 군에서 높게 나타났으나(p<0.05), DNA fragmentation index는 노출시간에 따라 차이가 없었다. 본 연구 결과 개정자가 0.3 M glucose가 첨가된 glycerol-free TRIS에서 $4^{\circ}C$, 50 분간 냉각 후 동결과 융해 후 더 높은 생존성을 나타냈다.
