Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats (감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. Effects of Nutrition Therapy and Drug Treatment on the Blood Lipid Levels in Patients with Hyperlipidemia according to Genetic Polymorphism of Apo CII (Apo CIII 유전자 다형성에 따라 영양치료 및 약물 병용치료가 고지혈증 환자의 혈중 지질 농도에 미치는 영향)

  • The purpose of this study was to investigate the effects of the nutrition therapy and drug(simvastatin) treatment on the levels of blood lipids for the hyperlipidemic patients according to genetic polymorphism of apo CIII. Subjects of the study consisted of 43(male: 9, female: 34 )hyperlipidemic patients registered to Kyung Hee Medical Center, Intakes of nutrients for the subjects were determined by 24-hr recall method through a personal interviews. The subjects were instructed to take the hypolipidemic and hypocholesterolemic diets. Compliance was monitored through food records, personal interview, and body-weight measurements. The patients changed their basal diet containing 40. 1g fat(182.7mg cholesterol./day) to a diet containing 30.7g fat(139.2mg cholesterol/day) after 12 weeks of nutrition therapy. The distribution of genotypes for apo CIII by Msp I was 32.5% of TT, 50.0% of TC, and 17.5% of CC. aT the beginning of the study, the level of blood triglyceride was the highest for patients with TT and TC. Following the nutrition therapy, patients with CC exhibited the largest drop of blood triglyceride. Following simvastatin treatment with nutrition therapy, blood LDL-cholesterol decreased in most of the patiens with TT. Meanwhile the distribution of genotypes for apo CII by the Fok I was 10.0% of TT, 47.5% of TC, and 42.5% of CC. The level of blood triglyceride was the highest for patients with CC. Following the nutrition therapy, patients with TC exhibited a significant drop for the blood triglyceride. Following simvastatin treatment with nutrition therapy the level of blood LDL-cholesterol decreased in most of the patients with CC. We concluded that dietary habits and food have been changed by nutrition therapy. And blood lipid levels have been decreased by the restriction of intakes of energy, fat and cholesterol. There was variation in the levels of blood lipids according to apo CII polymorphisms. The level of blood lipids responded to nutrition therapy and drug treatment in different was according to genetic polymorphisms. Accordingly, the choice of individualized therapy based on the patient\\`s genetic polymorphism is very important for effective therapy(Korean J Nutrition 33(8) : 813-823, 2000)

  • In order to evaluate the effects of SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG(Xinnao-Tong, Jiangzhiyanshou-Dan and Qingxintiaoqi-Tang) on hyperlipidemia, the experimental study was performed in rats without or with hyperlipidemia induced by 2% cholesterol fed-diet or Triton WR-1339. And the levels of serum total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol and phospholipid were measured. The results are summarized as follows : In the model of hyperlipidemia induced by 2% cholesterol Fed-diet 1. Both low & highly concentrated GANGJIYOUNSOODAN showed significantly decreasing effects on the level of serum total cholesterol while SIMNETONG & CHUNGSIMJOKlTANG didn't. 2. GANGJIYOUNSOODAN, SIMINETONG and CHUNGSIMJOKITANG showed decreasing effects on the level of serum triglyceride with no statistical significance. 3. Only highly concentrated GANGJIYOUNSOODAN showed increasing effects on the level of serum HDL-cholesterol while the others had no effects. 4. Only highly concentrated GANGJIYOUNSOODAN had decreasing effects on the level of serum LDL-cholesterol while the others had no effects. 5. Highly concentrated GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed significantly decreasing effects on the level of serum phospholipid while the other had no effects. In the model of hyperlipidemia induced by Triton WR-1339 6. SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed slightly decreasing effects on the levels of serum total cholesterol and triglyceride with no statistical significance. 7. None of SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed any effects on the level of serum HDL-cholesterol. 8. Highly concentrated GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed decreasing effects on the level of serum LDL-cholesterol but the other had no effects. 9. All of highly concentrated SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed decreasing effects on the level of serum phospholipid with statistical significance. These results show that GANGJIYOUNSOODAN and CHUNGSIMJOKITANG, in orders, have antihyperlipidemic effects. Further study on the mechanism and on the herb screening of these prescription should be followed.

Hypoglycemic Effect of Exo- and Endo-biopolymers Produced by Submerged Mycelial Culture of Ganoderma lucidum in Streptozotocin-Induced Diabetic Rats

  • The hypoglycemic effect of an exo-biopolymer (EXO) and endo-biopolymer (ENDO) produced from submerged mycelial culture of Ganoderma lucidum was investigated in streptozotocin (STZ)-induced diabetic rats. Both the EXO and ENDO showed hypoglycemic potential, however, the former proved to be more potent than the latter. The administration of the EXO at the dose of 100 mg/kg body weight (BW) significantly reduced the plasma glucose level (23.5%) and increased the plasma insulin level (2.2 fold) in the diabetic animals. The EXO also lowered the plasma total cholesterol, triglyceride, low-density lipoprotein (LDL) cholesterol, and athrogenic index by 14.7, 31.4, 24.1, and 45.4%, respectively, and reduced the liver total cholesterol and triglyceride levels by 6.7 and 25.8%, respectively. It increased the plasma high-density lipoprotein (HDL) cholesterol (37.7%), compared to the control group. Furthermore, the alanine transaminase (ALT) and aspartate transaminase (AST) showed lower activities in the EXO administered groups than the other experimental groups. Taken together, these results suggest that the exo-biopolymer may alleviate the blood glucose level by increased insulin secretion.

Isolation of Hyperlipidemic Substances from Methanol Extract of Paeoniae Radix (작약의 메탄올 추출물로부터 항고지혈 활성성분의 분리)

  • We previously showed that a methanol extract of Paeoniae radix decreased total cholesterol level in rats with hyperlipidemia. In order to isolate the active ingredient(s), the methanol extract of Paeoniae radix was fractionated with chloroform/methanol(4:1) solution and isolate into soluble part and insoluble part of the the methanol extract. Above two parts were tested on the experimentally induced hypercholesterolemia in rats for the lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high choleserol diet for 7 days. After oral administration of above samples for 4 weeks, serum lipid profile was verified on these rats by measuring total cholesterol, triglyceride, high density lipoprotein cholesterol and low density lipoprotein cholesterol. The chloroform/methanol(4:1) soluble part and insolule part showed lowering activity of total cholesterol level and triglyceride level at 4 week point significantly(p<0.01 and p<0.05) compare with the control group and the soluble part was more effective.

Effects of Foot Reflexology on Essential Hypertension Patients (본태성 고혈압 환자에게 미치는 발반사마사지 효과)

  • Purpose: This study was to evaluate the effects of foot reflexology on blood pressure, serum lipids level and life satisfaction in essential hypertension patients. Method: The research design used was a nonequivalent control group pretest-posttest design. Foot Reflexology was used as the experimental treatment from June 23rd, 2003 until August 31st, 2003. Thirty-four subjects were assigned to an experimental group(18) and control group(16). Foot Reflexology was administered twice a week for 6 weeks and self foot Reflexology was administered twice a week for 4 weeks on the experimental group. Result: There was a significant decrease in systolic blood pressure but no significant decrease indiastolic pressure in the experimental group compared to the control group. The total cholesterol level in the experimental group compared to the control group was not significantly decreased after foot reflexology. However, the triglyceride level in theexperimental group compared to the control group was significantly decreased after foot reflexology. On the other hand, high density lipoprotein and low density lipoprotein levels in the experimental group compared to the control group was not significantly decreased after foot reflexology. Life satisfaction in the experimental group compared to the control group was significantly improved after foot reflexology. Conclusion: The results proved that foot reflexology was an effective nursing intervention to decrease systolic pressure, and triglyceride but not for the blood cholesterol and to improve life satisfaction. Therefore, blood cholesterol should be further evaluated in a larger group of subjects and for a longer period. Further research is regarded as necessary to evaluate and to compareeffects of self-foot reflexology and foot reflexology.

Effects of Chungganhaeju-tang(Qingganjiejiu-tang) on Alcohol Metabolism and Alcoholic Liver Damages (청간해주탕이 알코올대사 및 손상간에 미치는 영향)

  • Objectives: This experiment was conducted to verify the effects of Chungganhaeju-tang(Qingganjiejiu-tang) on the alcohol metabolism and liver functions, by measuring the activity levels of ADH and ALDH, as well as glucose, triglyceride, and BUN. Damage of the liver cells caused by alcohol was determined through the examination of serum levels of AST, ALT, ALP, LDH, and uric acid. Methods: Sprague-Dawley rats were used in this experiment and the rats were divided into control and experiment groups. Chungganhaeju-tang(Qingganjiejiu-tang) extract was orally administered in the experiment group for three weeks. Each group was further classified into two sub-groups, and control group's blood was taken without oral ingestion of alcohol, while the experiment group' s blood was withdrawn after ingestion of alcohol. Evaluation of damage level was done considering the presence of extract and alcohol. Results: In this experiment, Chungganhaeju-tang(Qingganjiejiu-tang) significantly suppressed the activity of ADH which is a precursor enzyme of acetaldehyde, but didn't cause significant changes in the activity of ALDH which is a catabolic enzyme. Decreased glucose level due to alcohol consumption was recovered back to the normal level and increased levels of triglyceride, BUN, AST, ALT, ALP, LDH, and uric acid were significantly reduced. Conclusions: These experiment results suggest that Chungganhaeju-tang(Qingganjiejiu-tang) inhibits the formation of acetaldehyde in the metabolism of alcohol, and affects the recovery of weakened liver functions due to alcohol.

The Effect of Green Tea on the Lipid Composition of Serum and Liver and the Activities of Antioxidative Enzymes in Rats (녹차가 흰쥐의 혈청 및 간의 지질성분과 항산화계 효소 활성도에 미치는 영향)

  • This research was performed to Investigate the effects of green tea on the lipid composition of serum and liver and the specific activities of antioxidative enzymes. Male Sprague Dawley rats were fed 10% fat diet with lard and fish oil. Powdered green tea was added to the lard and fish oil diet at the level of 0.1% and 1%. After 6 weeks of feeding, serum and liver were obtained from experimental rats. Then we measured the concentration of total cholesterol, HDL-cholesterol and triglyceride. From liver cytosolic fraction, we analized the specific activities of superoxide dismutase, glutathione peroxidase and glutathione S-transferase. The level of total cholesterol and triglyceride were decreased and the ratio of HDL-cholesterol to total cholesterol was increased by the fish oil in the serum. But in the liver, the level of total cholesterol was increased by the fish oil and green tea than the lard. The specific activities of glutathione S-transferase were more increased in the fish oil than the lard. There was not effect of the green tea of daily dose on the lipid composition of serum and liver and the specific activities of antioxidative enzymes in rats.

A Study on Health Behaviors by a Risk Level of Metabolic Syndrome among Petty Merchants in Traditional Markets (재래시장 영세상인의 대사증후군 위험도별 건강행위에 대한 연구)

  • Purpose: The purpose of this study was to identify the health behaviors by a risk level of metabolic syndrome (MS) among petty merchants in traditional markets. Methods: The data were collected through questionnaires and physical examination on abdominal circumstance, triglyceride, HDL, blood pressure, and FBS of 177 subjects in S city, Korea. The data were analyzed by descriptive statistics, ${\chi}^2$-test (or fisher's exact test) and ANOVA using SPSS/WIN 18.0 program. Results: The prevalence of MS was 15.8%, and risk group was 72.9%. The abdominal circumstance and triglyceride levels were higher in MS group than those of the risk group and normal group. The HDL and systolic blood pressure levels were also higher in MS group than those of the normal group. Conclusion: The prevalence of MS and risk groups was high among petty merchants in traditional markets. However, these merchants did not practice health behaviors nor take institutional health management benefit. Therefore, governmental level community based health management services are requested for them.

Hypoglycemic and Hypolipidemic Effects of Crude Saponin Fractions from Panax ginseng and Gynostemma pentaphyllum (인삼 및 틀의 초 saponin분획의 항당뇨 및 항고지혈 효과)

  • Crude saponin fractions were isolated using non-ionic resin chromatography from Ginseng Radix Alba (PG) and Gynostemmae Herba+ (GP). These saponin fractions were orally administered to strep- tozotocin (STZ)-induced diabetic rats for 2 weeks and to high-fat diet-induced obese rats for 4 weeks. Treatment with either PG saponin or GP saponin at a dose of 1 mg/kg significantly decreased the plasma glucose level to that of glibenclamide treated or normal groups. The increased plasma triglyceride (TG) level in diabetic rats was decreased by 50% with PG or GP saponin treatment. Combined administration of PG and GP saponins with different ratios (total dose of 1 mg/kg) also had the similar effects on the blood glucose and TG levels with that of PG or GP alone. Treatments with GP (1 mg/kg) or GP (0.5 mg/kg) and PG (0.5 mg/kg) together significantly suppressed the rise in TG levels induced by high-fat diet whereas slightly suppressed the rise in the total cholesterol level. The body weight gain was also decreased both in the two saponin treated groups. These results demonstrate that either alone or mixture of PG and GP have similar degree of effects on hyperglycemia and hyperlipidemia.
