The associations between serum leptin, adiponectin and intercellular adhesion molecule-1 in hypercholesterolemic patients

  • We examined the associations between adiponectin or leptin and serum ICAM-1 levels in seventy-six hypercholesterolemic patients (mean age 59 yrs, 25 males and 51 females, LDL-cholesterol>=130mg/dL at screening). Blood lipid profiles and HOMA-IR derived from fasting glucose and insulin concentrations were determined. Serum levels of adiponectin, leptin and ICAM-1 were analyzed using ELISA The results showed that serum levels of leptin were positively associated with serum levels of ICAM-1 independent of age, sex and BMI (r=0.392, p<0.001). Serum levels of adiponectin were negatively associated with serum levels of ICAM-1 independent of age, sex and BMI (r=-0.343, p<0.005). Stepwise multiple linear regression analysis showed that serum leptin was an independent factor to be associated with serum ICAM-1 levels after adjusting for age, sex, BMI, alcohol intake, smoking status, blood lipids such as total cholesterol, triglyceride, HDL cholesterol and LDL cholesterol and HOMA-IR (p<0.001). With respect to adiponectin, its association with serum ICAM-1 was attenuated but still significant when further adjustments were made for age, sex, BMI, alcohol intake, smoking status, blood lipids such as total cholesterol, triglyceride, HDL cholesterol and LDL cholesterol and HOMA-IR (p<0.005). In conclusion, this study suggests that adiponectin and leptin are associated with endothelial derived inflammation.

Effect of Fruiting Body of Cordyceps Militaris on Growth, Lipid and Protein Metabolism, and Enzyme Activities in Male Rats (동충하초 자실체가 흰쥐가 성장률, 지질과 단백질 대사 및 효소활성에 미치는 영향)

  • The effects of fruiting body of Cordyceps militaris on the growth, the food intake, the food efficiency ratio, the lipid metabolism, the serum protein level ad enzyme activity in male rats were studied. Sprague-Dawley rats were fed four types of diets for five weeks, respectiely: a control diet, a control diet supplemented with 2%, 3% or 4% fruiting body of Cordyceps militaris(CF) powder. In rats fed 2% or 3% CF diets the body weight gain, the food intake, the concentratons of hepatic triglyceride and serum LDL-cholesterol, the atherogenic index, and the total lipid, total cholesterol, triglyceride and phospholipid in serum were similar to those in rats fed the control diet. Whereas, in 4% CF diet these were significantly decrased. But the all CF diets feeding could not decrease the food efficiency ratio, the weights of liver, pancreas, spleen, kidney and heart, and the concentration of serum HDL-cholesterol. Also, it was shown that the concentrations of glucose, total protein, albumin, urea and creatinine, and the GOT, GPT, LDH, ${\gamma}$-GTP and ALP activities were the same levels in serum of rats of fed all the experimental diets.

Effect of Chitosan and N. O-Carboxymethyl Chitosan of Different Sources and Molecular Weights on Lipid Metabolism (급원과 분자량이 다른 Chitosan 과 N, O-Carboxymethyl Chitosan이 지방대사에 미치는 영향)

  • This study was performed to investigate the effect of chitosan and NOCC from different sources and of different molecular weights on lipid metabolism. Sprague-Dawley rats were blocked into 26 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400 ppm in diet. Various chitosan and NOCC sources were given at the level of 0%(w/w) of diet. Total serum cholesterol and serum triglyceride concentrations were little affected by chitosan and NOCC supplements. However, HDL-cholesterol concentration and HDL : total cholesterol ratio were increased , and liver lipid, cholesterol , and triglyceride concentration were decreased by chitosan and NOCC supplements. The cholesterol and lipid lowering activity depends on fiber sources(crab and shrimp); type (chitosan and NOCC); and molecular weight(low , medium , and high). Among cadmium-free groups, chitosan-fed groups showed greater activity than NOCC -fed groups in lowering cholesterol and lipid levels, and greater fecal excretion of lipids and bile acids. Crab chitosans were more effective in cholesterol and lipid lowering activity than shrimp chitosans. The group fed high molecular weight crab chitosan showed the highest fecal excretion of lipids and bile acids. This indicated that high molecular weight crab chitosan was most effective in interfering with cholesterol and lipid absorption.

Characteristics of Transesterification Reaction of Soy Bean Oil by Acid Catalysts (산촉매에 의한 대두유의 전이에스테르화 반응 특성)

  • Characteristics of the transesterification reaction between triglycerides in soy bean oil and methanol were investigated in the presence of acid catalysts. such as sulfuric acid and PTS (p-toluene sulfonic acid). Concentrations of diglyceride and monoglyceride which were intermediates in the reaction mixtures, were far below 10% of triglyceride under any reaction conditions. Thus, conversion of the reaction could be determined from the concentration of triglyceride. Dried PTS had more superior catalytic power than sulfuric acid for transesterification reaction between soy bean oil and methanol. When transesterification reaction of soy bean oil was catalyzed by 1 wt% of PTS at methanol stoichiometric mole ratio of 2 and $65^{\circ}C$, final conversion reached 95% within 48 hours. If FAME (fatty acid methyl ester) was added into reaction mixture of soy bean oil, methanol and PTS catalyst, it converted reaction mixture into homogeneous phase, and substantially increased reaction rate. When reaction mixture was freely boiling which had equal volumetric amount of FAME to soy bean oil, methanol stoichiometric mole ratio of 2 and 1 wt% of PTS, final conversion achieved value of 94% and temperature approached to $110^{\circ}C$ within 2 hours.

Ramie Leaf Extracts Suppresses Adipogenic Differentiation in 3T3-L1 Cells and Pig Preadipocytes

  • The present study was carried out to evaluate the anti-obesity effect of different concentrations of extracts of hot air-dried ramie leaf (HR) and freeze-dried ramie leaf (FR) in 3T3-L1 cells and pig preadipocytes. To analyze the effect on cell proliferation, cells were treated with $25{\mu}g/mL$ or $100{\mu}g/mL$ HR or FR extract for 2 days. Cell differentiation was evaluated by measuring glycerol-3-phosphate dehydrogenase and lipoprotein lipase (LPL) activities and intracellular triglyceride content. Treatment with either HR or FR extracts inhibited the proliferation of 3T3-L1 cells and pig preadipocytes in a dose-dependent manner. HR extract treatment inhibited the differentiation of both cell types more effectively than FR treatment. The extent of triglyceride accumulation decreased significantly in both cells following either HR or FR treatment. Furthermore, LPL activity significantly decreased after treatment with HR or FR extract. These results indicated that HR and FR extracts may inhibit proliferation and differentiation of 3T3-L1 cells and pig preadipocytes. Further studies are needed to explore the anti-obesity effect of HR and FR extracts.

Analysis of Serum Lipids and Lipoprotein Electrophoresis Fractions in Hypertensive Patients

  • We analyzed the concentration of serum total cholesterol (TC), high density lipoprotein cholesterol (HDLC), low density lipoprotein cholesterol (LDLC), triglyceride (TG) and the serum lipoprotein electrophoresis fractions in hypertensive patients (hypertension group, n=182). The average concentration of lipids and lipoprotein fractions in the hypertension group was compared to that of the normal group. The average concentrations of serum TC, LDLC and TG in the hypertension group were significantly higher than those of the normal group (3.3%, 11% and 70%, respectively) (P<0.05). But HDLC was nonsignificantly lower (2%) (P<0.05). In the hypertension group, the percentages of patients who had an abnormally high level of total cholesterol, HDLC, LDLC and TG were 12.1%, 8.2%, 24.2% and 44.5%, respectively. The average of ${\beta}$-lipoprotein (27.5%) and pre ${\beta}$-lipoprotein (12.6%) were significantly higher, whereas ${\alpha}$-lipoprotein (15.2%) was significantly lower in the hypertension group than in the normal group (P<0.05). The percentages of patients who had an abnormally high level of very low density lipoprotein (pre ${\beta}$-LP), LDLC (${\beta}$-LP) and chylomicron were 31.3%, 17.0% and 12.6%, respectively. This study suggests that an increasing of total cholesterol, LDLC and TG, pre ${\beta}$-LP, ${\beta}$-LP and chylomicron are associated with a risk factor for hypertension.

Effects of taeeumjoweetang Herbal-Acupuncture on plasma and liver lipid composition and antioxidative capacity in rat fed high fat diet (태음조위탕(太陰調胃湯) 약침(藥鍼)이 고지방 급여 흰쥐의 혈장 및 간장의 지질구성과 항산화능에 미치는 영향)

  • Objectives : In order to measure the Effects of taeeumjoweetang Herbal- Acupuncture at joksamri(ST36) and kwanweun(CV4) on liver and plasma lipid composition and antioxidative capacity in rat fed high fat diet. Results : Concentrations of plasma triglyceride, total cholesterol and LDL-cholesterol showed a tendency to increase in the high fat diet group. However Herbal-Acupuncture groups showed a lower values than control groups. HDL-cholesterol showed a tendensy to decrease in high 131 fat diet groups and in high fat diet groups, these values showed no significantly different. Liver total cholesterol values showed no significantly different in all treatment groups. Triglyceride concentration showed a high value in control group and other treatment groups showed no significantly different. plasma GOT and GPT values showed a tendency to increase in high fat diet group. however these values decreased in Herbal-Acupuncture group. The concentration of TBARS in liver and plasma showed a high values in high fat diet group, however these values showed a tendency to decrease in aqua- acupuncture group. Glutathione peroxidase, superoxide dismutase and catalase activity values showed a low values in high fat diet group, however these values showed a tendency to increase in Herbal-Acupuncture group.

Effect of Leaf Mustard (Brassica juncea) on Lipid Metabolism of Rat with Chroethanol Administration (갓(Brassica juncea) 첨가식이가 만성적인 알콜 투여시 흰쥐의 지방대사에 미치는 영향)

  • This study was undertaken to investigate the effects of mustard leaf diet on the lipid metabolism under chronic alcohol administration. Sprague-Dawley male rats were fed either AIN-76 diet(control), control with ethanol, AIN-76 diet plus 5% mustard leaf(mustard leaf), or mustard leaf with ethanol lot 30 days. On the 21st day, all of the rats were given an oral dose of ethanol and blood-ethanol concentration were monitored for the next 5 hours. Lipid and enzyme determinations in serum and liver were carried out after 30 days. The results obtained were summarized as following: 1) Supplementing 5% of mustard leaf did not recover the body weight loss due to chronic alcohol administration. 2) There were no significant differences in blood ethanol concentrations among the experimental groups. 3) Mustard leaf diet decreased the plasma total cholesterol, triglyceride levels increased due to the chronic alcohol administration, but not HDL-, LDL-cholesterol, and liver lipids. 4) Mustard leaf diet decreased ${\gamma}$ -GTP level increased by chronic alcohol administration. Overall, these data suggest that mustard lear can have a recovery function, which was not via ethanol metabolism on the symptoms of alcohol related diseases.(Korean J Human Ecology 1(1) : 94~102, 1998)

Effect of Mixed Onion Pills on Plasma Triglyceride and Total Cholesterol Levels in Overweight or Obese Men (혼합 양파환이 과체중 및 비만 남성의 혈중 지질 농도에 미치는 영향)

  • The effect of mixed onion pills on body composition, blood pressure, and concentrations of blood glucose and lipids were studied in 10 men ($38.11{\pm}2.82$ years old). The mixed onion pills was produced by mixing quercetin, onion, mulberry leaves and buckwheat. Ten volunteers (male, BMI 23.0~35.4 kg/$m^2$) were chosen for this study. The volunteers consumed 1.2 g of mixed onion pills (containing about 30 mg of quercetin) per day for 2 months. The administration of the mixed onion pills did not produce any weight and BMI changes in the subjects. The systolic blood pressure and the level of blood glucose were slightly lowered by the supplementation of the mixed onion pills. After 2 months both the concentration of plasma triglycerides and total cholesterol of the subjects were lowered by 12.4% and 5.0%, respectively (triglyceride; pre $251.67{\pm}45.63$ mg/dL vs. post $220.44{\pm}44.67$ mg/dL and total cholesterol; pre $207.25{\pm}10.39$ mg/dL vs. post $196.63{\pm}10.47$ mg/dL, p<0.05). These results suggest that the mixed onion pills may possibly lower the level of plasma lipids in overweight or obese men (BMI greater than 23 kg/$m^2$).

Effect of Lactobacillus acidophilus based probiotic product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs

  • The aim of this study was to evaluate the effect of Lactobacillus acidophilus probiotic (LAP) product supplementation on the blood profile, fecal noxious gas emission, and fecal shedding of lactic acid bacteria and coliform bacteria in healthy adult Beagle dogs. In total, 14 Beagle dogs with an average initial body weight of 10.19 ± 0.61 kg were randomly assigned into two dietary treatments,with and without LAP supplementation, for a 28-day feeding trial. At the end of the experiment, there was no significant (p > 0.05) difference in the concentration of serum total cholesterol, high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), white blood cell (WBC), red blood cell (RBC), blood lymphocyte percentage, fecal hydrogen sulfide (H2S) and total mercaptans (R.SH) emission, and fecal coliforms counts. However, the serum concentrations of the triglyceride and fecal ammonia (NH3) emission of the LAP treatment were significantly (p < 0.05) decreased in the group compared with the CON dogs. Fecal total lactic acid bacteria counts were significantly (p < 0.05) increased in the LAP treatment. In conclusion, the supplementation of LAP in Beagle dog diets could decrease the blood triglyceride level and enhance the gut Lactobacillus count which may have positive effects on dogs.
