Effect of Combined Supplementation Catechin and Vitamin C on Growth Performance, Meat Quality, Blood Composition and Stress Responses of Broilers under High Temperature (고온 환경에서 카테킨 및 비타민 C 첨가가 육계의 생산성, 계육품질, 혈액성분 및 스트레스 지표에 미치는 영향)

  • The study was carried out to investigate the effects of dietary combined supplementation of antioxidants as catechin and vitamin C on growth performance, meat quality, blood profiles and stress responses of broilers exposed to high temperature. For this experiment, a total of 360 21-day-old male Ross 308 broilers were used. Treatments were assigned with 6 replicates per treatment and 10 birds per replicate in a 2 × 3 factorial design with vitamin C (0, 250 mg/kg) and catechin (0, 600, 1,200 mg/kg). The heat stress environment was maintained at temperature 32±1℃ and relative humidity 60±5% for 24 hours until the end of the experiment. The supplemented antioxidants had no significant difference in weight gain, feed intake and feed conversion ratio (P>0.05). The content of total cholesterol in blood had no interaction, but decrease (P<0.01) in the supplemented catechin group. Also, the supplementation with catechin showed increase in the SOD activity of blood, and lower corticosterone and IgM levels of broilers. The contents of HSP70 and MDA in liver decrease (P<0.05) with the supplementation of antioxidants, and HSP70 showed an interaction between groups. DPPH radical scavenging ability in breast meat increased (P<0.01) in catechin, but meat quality did not show difference according to treatments. Respiratory rate decreased (P<0.05) in catechin, but no interaction with vitamin C. In conclusion, the combination of vitamin C and catechin can alleviate stress under high temperature, such as HSP70 and MDA, but further study on the optimal supplemental level is needed.

Study on High Sensitivity Metal Oxide Nanoparticle Sensors for HNS Monitoring of Emissions from Marine Industrial Facilities (해양산업시설 배출 HNS 모니터링을 위한 고감도 금속산화물 나노입자 센서에 대한 연구)

  • A sensor is needed to continuously and automatically measure the change in HNS concentration in industrial facilities that directly discharge to the sea after water treatment. The basic function of the sensor is to be able to detect ppb levels even at room temperature. Therefore, a method for increasing the sensitivity of the existing sensor is proposed. First, a method for increasing the conductivity of a film using a conductive carbon-based additive in a nanoparticle thin film and a method for increasing ion adsorption on the surface using a catalyst metal were studied.. To improve conductivity, carbon black was selected as an additive in the film using ITO nanoparticles, and the performance change of the sensor according to the content of the additive was observed. As a result, the change in resistance and response time due to the increase in conductivity at a CB content of 5 wt% could be observed, and notably, the lower limit of detection was lowered to about 250 ppb in an experiment with organic solvents. In addition, to increase the degree of ion adsorption in the liquid, an experiment was conducted using a sample in which a surface catalyst layer was formed by sputtering Au. Notably, the response of the sensor increased by more than 20% and the average lower limit of detection was lowered to 61 ppm. This result confirmed that the chemical resistance sensor using metal oxide nanoparticles could detect HNS of several tens of ppb even at room temperature.

Alteration Yield Traits of Sweetpotato (Ipomoea batatas L.) Cultivars Suitable for Paddy Cultivation (논 재배에 적합한 가공용 고구마 품종 선발을 위한 수량성 평가)

  • Several domestically consumed sweetpotato derivatives, such as sweetpotato starch for processing raw materials, frozen and refrigerated paste, and powder are dependent on imports. This study was conducted to examine the suitability of sweetpotato cultivars of twelve varieties (powdery-type and viscous type) cultivated in paddy fields, for use in starch, chips, dried products, and beverages. The two-year average yield results of the four cultivars suitable for starch (in order of highest to lowest yield) was as follows: Gogeonmi (4,018 kg/10a); Daeyumi (3,615 kg/10a); Jinhongmi (3,426 kg/10a); Singeonmi (2,837 kg/10a). The starch content was 20.2%, 18.2%, 21.2%, and 20.6% in Daeyumi, Gogeonmi, Singeonmi, and Jinghongmi, respectively. The total amount of starch was higher in Daeyumi (730 kg/10a) and Gogeonmi (731 kg/10a) than that in Singeonmi and Jinghongmi. The yield of Pungwonmi and Shinjami were 4,443 and 3,602 kg/10a, respectively. Powdery-type sweetpotatoes (Daeyumi and Gogeonmi) showed the low decay rates of all cultivars (0.8 and 0%, respectively). The yield of the storage root formation and storage root swelling stages by water-logging treatment decreased by 16.5% and 15.4% for Pungwonmi, and by 17.2% and 10.0% for Shinjami. Drainage management of paddy fields is necessary to reduce the damage caused by water-logging. Our results suggest that cultivation of sweetpotato varieties suitable for processing raw materials in paddy fields will enable stable yields of sweetpotato with a high starch content.

Diversification of Rice Quality for Processing. Physicochemical Characteristics and Inheritance of Floury Endosperm Mutants (특수 가공용 미질개발 : 분상질배유 돌연변이 계통의 이화학적특성과 유전)

  • This study was carried out to assess the agronomic characters and physicochemical properties of floury and chalky-endosperm mutant lines induced by chemical mutagen treatment to rice varieties, Hwacheongbyeo and IR24. Linkage analysis of a floury-endosperm gene was carried out using linkage testers. The grain size of brown rice of the mutants was smaller than that of the original varieties. The l, 000-grain and 1$\ell$ weight were lighter in the mutants compared with those in the original varieties. The compound starch granules in the endosperm cell of the mutants showed a loosely-packed crystalline structure. Amylose contents in mutants ranged from 16.9 to 28.5%. Crude protein contents of the mutants were not significantly different from the original rice variety, Hwacheongbyeo, but white core mutant(line 47106) derived from IR24 showed higher protein(l1.32%) compared with IR24(8.30%). The mutants showed slightly harder gel characteristics, and much lower viscosity in Amylograph than original varieties. Steamed rice-cakes from mutant lines showed greater volume than those from original varieties. During the process of alcohol fermentation, Brix in the mutants(especially floury mutants) decreased faster and the alcohol production after 10-day fermentation was much greater in the mutants than in the original varieties. Three different gene loci for floury endosperm characteristics were identified from the allelism test among mutant lines, and the genes were tentatively symbolized as flo-a, flo-b and flo-c, respectively. A floury gene, flo-a, was linked with lg(liguleless) gene in the linkage group N, with R.V. 5.76$\pm$1.72%.

Effect of soil stabilizer on the bioavailability of arsenic in paddy soil (안정화제가 논토양 내 비소의 생물유효도에 미치는 영향)

  • In this study, we sought to identify a soil stabilizer that can be applied to paddy fields vulnerable to arsenic (As) pollution. To this end, we conducted a pot experiment in which we evaluated the effects of different stabilizers on the bioavailability of As in paddy soil. As candidate stabilizers, we assessed calcium superphosphate (CSP), sulfur, and steel slag, which were applied at the rates of 0.7 and 1.4, 0.1 and 0.2, and 7.0 and 14.0 Ma ha-1, respectively. On day 67 after rice transplantation, we detected significantly lower concentrations of As in the solutions of paddy soil treated with 1.4 Ma ha-1 CSP (96.9 ㎍ L-1) and 0.2 Ma ha-1 sulfur (207.2 ㎍ L-1) compared with the As concentrations in control (314.5 ㎍ L-1) and steel slag-treated (268.6-342.4 ㎍ L-1) soil. Compared with the As concentrations in control brown rice (0.16 mg kg-1), concentrations in brown rice were lowest in the pots treated with 1.4 Ma ha-1 CSP (0.09 mg kg-1). Furthermore, in response to CSP treatments, we detected increases in the weight of rice grains (50.0-50.4 g/pot) compared with the control (40.4 g/pot) and other treatments (26.9-48.1 g/pot), which we speculate could be attributed to the reduction in As toxicity to rice owing to a decline in soil solution As contents and the fertilization effect of the CSP treatment. Collectively, our findings indicate a high-level application of CSP (1.4 Ma ha-1) to paddy soil has a comparatively beneficial effect in mitigating the bioavailability of As.

Changes in chemical composition and physiological activity of Jeju-Tatary buckwheat tea according to leaching temperature (제주 타타리메밀의 침출 조건에 따른 제주 타타리메밀침출차의 이화학적 특성 및 생리활성)

  • In this study, Jeju Tatary buckwheat tea's chemical composition and physiological activities were compared according to the leaching temperature (60, 80, 100 ℃). As the leaching temperature is increased, the degree of browning is induced. However, there was no significant change in pH. The total polyphenol content was higher at 80 ℃ than at 60 ℃ leaching temperature, but significantly decreased at 100 ℃ leaching temperature (60 ℃: 17.06 mg GA/g, 80 ℃: 20.09 mg GA/g, 100 ℃ :18.45 mg GA/g). There were high content of flavonoid and rutin as the leaching temperature increased. Consistently, 2,2-diphenyl1-picrylhydrazyl (DPPH) radical scavenging activity and tyrosinase inhibitory activity were significantly higher with increasing temperature (DPPH % inhibition: 60 ℃: 41.88%, 80 ℃: 46.01%, 100 ℃: 46.80%/tyrosinase inhibitory activity: 60 ℃: 9.38%, 80 ℃: 22.94%, 100 ℃: 28.17%). However, there was no significant difference in DPPH radical scavenging activity between 80 and 100 ℃. A cytotoxicity test was performed by treating with Jeju Tatary buckwheat extract into mouse macrophage cells (Raw264.7). 100 and 200 ㎍/mL treatment (100 ℃ extract) were significantly upregulated the survival rate, but there was no significant difference in other concentrations. Collectively, most of the bioactive components, antioxidant activity, and tyrosinase inhibitory activity were induced as the leaching temperature increased. However, the content of polyphenols which are known to have antioxidant activity, was significantly reduced at 100 ℃ leaching temperature. Several reports have demonstrated that leaching at too high temperature lowered the overall acceptability, so the optimal leaching condition of Tatary Buckwheat is 80 ℃, 5 min in this study.

  • Objectives: The purpose of this study is to explore the effectiveness and safety of frequently used clinical herbal medicines (Yijung-tang [Lizhong-tang, LJT], Pyeongwi-san [Pingwei-san, PWS], and Shihosogan-tang [Chaihu Shugan-tang, SST]) in patients with functional dyspepsia (FD) when administered according to herbal medicine and Korean medicine pattern identification. The results of this study will be used to standardize the diagnostic instrument used in Korean medicine and to investigate biomarkers of Korean medicine pattern identification. Methods: This study will be a randomized, assessor-blind, 3-arm, parallel, open-label, multi-center clinical trial. A total of 300 FD participants will be recruited from 3 Korean medical hospitals and assigned to the LJT (n=100), PWS (n=100), and SST (n=100) groups according to FD pattern identification. The patients will take the medication for 8 weeks, 3 times a day, before or between meals. The primary outcome will be total dyspepsia symptom (TDS) and the secondary outcomes will be adequate relief (AR) for dyspepsia, overall treatment effect (OTE), visual analogue scale (VAS), functional dyspepsia-related quality of life (FD-QoL), gastrointestinal symptom score (GIS), and pattern identification questionnaires. For the exploratory outcomes, we will analyze blood and fecal metabolome profiles, microbiota from fecal and saliva samples, single nucleotide polymorphism (SNP), and results of Korean medicine diagnosis device measurements (heart rate variability, and tongue, pulse, and abdominal diagnosis). Conclusions: The results of this study will prove objectivity for Korean medicine pattern identifications, and the effectiveness and safety of herbal medicines for the population with FD.

Verification of Anti-Inflammatory Efficacy of Apple Mango (Mangifera indica L.) Peel in LPS-Activated Macrophage (LPS에 의해 활성화된 대식세포에서 애플망고 껍질(Mangifera indica L. Peel)의 항염증 효능 검증)

  • The purpose of this study was to investigate the antioxidant and anti-inflammatory activities of hot water (AMPW) and 70% ethanol (AMPE) extracts of apple mango (Mangifera indica L.) peel. The antioxidant activities were measured using a total polyphenol, electron-donating, 2,2'-azinobis [3-ethylbenzothiazoline6-sulfonic acid] (ABTS) radical scavenging assay. The total polyphenol content of AMPW and AMPE was 66.08 ± 0.62 mg TAE/100 g and 100.13 ± 0.23 mg TAE/100 g, respectively. As a result of measuring the electrondonating ability, at a concentration of 1,000 ㎍/ml, AMPW and AMPE showed an effectiveness of 86% and 94%, respectively. The ABTS assay showed 80% and 98% respective radical scavenging activity for AMPW and AMPE, at a concentration of 1,000 ㎍/ml. The cell viability on macrophage cells was performed using a 3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyl-tetrazoliumbromide (MTT) assay, and the results showed more than 90% cell viability at a 100 ㎍/ml concentration. Anti-inflammatory activity was verified by confirming nitric oxide (NO) production inhibitory activity, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) protein and mRNA expression inhibitory activity from lipopolysaccharide (LPS)-treated RAW 264.7 cells. The NO production inhibitory effects were measured using the Griess assay, which confirmed 45% and 40% inhibition after treatment with AMPW and AMPE, respectively. Moreover, the protein and mRNA expression of inflammatory-related factors iNOS and COX-2, decreased in a concentrationdependent manner. In conclusion, this study showed antioxidant and anti-inflammatory effects of Mangifera indica L. peel and revealed its promising potential for application as an antioxidant and anti-inflammatory agent.

Differential Effects of Transforming Growth Factor-β on NKG2D Ligands Expression and NK Cell-mediated Immune Responses in Primary and Metastatic Colon Cancer (원발성 및 전이성 대장암에서 TGF-beta가 NKG2D 리간드 발현과 NK 세포 매개 면역반응에 미치는 영향)

  • Transforming growth factor-β (TGF-β) is a multifunctional cytokine that affects not only the survival and growth of cancer cells but also the activity of immune cells. Although it has been generally accepted that cancer cell-derived TGF-β could promote the survival and growth of early cancer cells and have immunosuppressive roles, it has been known that TGF-β has differential effects according to the type or stage of cancer cells. Therefore, it is hard to clearly define its role in cancer progression and immune responses. This study investigated the effects of TGF-β signaling on the expression of five NKG2D ligands and the NK cell-mediated anticancer immune response in the primary colon cancer cell line KM12C and its two metastatic cell lines, KM12SM and KM12L4A. At the surface protein level, exogenous TGF-β decreased the expression of MICA, MICB, ULBP1, and ULBP2, and galunisertib increased the expression of MICA, MIAB, ULBP1, ULBP2, and ULBP3 in KM12C. However, KM12SM and KM12L4A showed no significant changes in the expression of NKG2DLs after treatment with TGF-β or galunisertib. TGF-β signaling inhibition via galunisertib improved the NK cell-mediated anticancer immune response against KM12C but did not show a significant response to KM12SM and KM12L4A. Therefore, the suppression of TGF-β signaling could improve the NK cell-mediated anticancer immune response against KM12C. However, an increase in NKG2DLs expression and an enhanced NK cell-mediated cancer immune response is hard to expect due to the alteration of TGF-β signaling in KM12SM and KM12L4A.

Predicting the Effects of Agriculture Non-point Sources Best Management Practices (BMPs) on the Stream Water Quality using HSPF (HSPF를 이용한 농업비점오염원 최적관리방안에 따른 수질개선효과 예측)

  • Non-point source (NP) pollutants in an agricultural landuse are discharged from a large area compared to those in other land uses, and thus effective source control measures are needed. To develop appropriate control measures, it is necessary to quantify discharge load of each source and evaluate the degree of water quality improvement by implementing different options of the control measures. This study used Hydrological Simulation Program-FORTRAN (HSPF) to quantify pollutant discharge loads from different sources and effects of different control measures on water quality improvements, thereby supporting decision making in developing appropirate pollutant control strategies. The study area is the Gyeseong river watershed in Changnyeong county, Gyeongsangnam-do, with agricultural areas occupying the largest proportion (26.13%) of the total area except for the forest area. The main pollutant sources include chemical and liquid fertilizers for agricultural activities, and manure produced from small scale livestock facilities and applied to agriculture lands or stacked near the facilities. Source loads of chemical fertilizers, liquid fertilizers and livestock manure of small scale livestock facilities, and point sources such as municipal wastewater treatment plants (WWTPs), community WWTPs, private sewage treament plants were considered in the HSPF model setup. Especially, NITR and PHOS modules were used to simulate detailed fate and transport processes including vegitation uptake, nutrient deposition, adsorption/desorption, and loss by deep percolation. The HSPF model was calibrated and validated based on the observed data from 2015 to 2020 at the outlet of the watershed. The calibrated model showed reasonably good performance in simulating the flow and water quality. Five Pollutants control scenarios were established from three sectors: agriculture pollution management (drainge outlet control, and replacement of controlled release fertilizers), livestock pollution management (liquid fertilizer reduction, and 'manure management of small scale livestock facilities) and private STP management. Each pollutant control measure was further divided into short-term, mid-term, and long-term scenarios based on the potential achievement period. The simulation results showed that the most effective control measure is the replacement of controlled release fertilizers followed by the drainge outlet control and the manure management of small scale livestock facilities. Furthermore, the simulation showed that application of all the control measures in the entire watershed can decrease the annual TN and TP loads at the outlet by 40.6% and 41.1%, respectively, and the annual average concentrations of TN and TP at the outlet by 35.1% and 29.2%, respectively. This study supports decision makers in priotizing different pollutant control measures based on their predicted performance on the water quality improvements in an agriculturally dominated watershed.
