본 연구는 미라 모발에 존재하는 멜라닌과립의 보존 상태와 모양을 규명하기 위해서 1250kV의 초고압전자현미경(high voltage electron microscope: HVEM)을 사용하여 3차 구조를 분석하였다. 미라 모발에서 피질에 분포하고 있는 멜라닌과립은 큐티클층에 인접한 바깥쪽 피질에 집중적으로 분포하고 있다. 이들 부위에는 다량의 멜라닌과립들이 무리를 지어 존재하고 있어서 현대인의 정상 모발에 있는 멜라닌과립의 분포양상과 뚜렷한 차이점을 나타내었다. 초고압전자현미경으로 ${\pm}60$도의 범위에서 1도 간격으로 촬영한 결과 멜라닌과립은 길쭉한 타원형의 형태로 크기가 다양하게 관찰되었다. 과립의 크기는 단축 직경이 $0.3-0.6{\mu}m$이고, 장축 직경은 $0.5-1{\mu}m$로 측정되었다. 결론적으로 초고압전자현미경은 일반 투과전자현미경보다 높은 해상도와 투과력을 가져서 두꺼운 절편의 생물 시료 구조분석에 용이하다.
The microstructural changes in Co nanoparticles and an Au-10at.%Pd thin film have been investigated using an in situ heating holder with a micro-electro-mechanical system (MEMS). In Co nanoparticles, two phases (face-centered cubic and hexagonal close-packed crystal structures) were found to coexist at room temperature and microstructures at temperatures, higher than $1,000^{\circ}C$, were observed with a quick response time and significant stability. The actual temperature of each specimen was directly estimated from the changes in the lattice spacing (Bragg-peak separation). For the Au-10at.%Pd thin film, at a set temperature of $680^{\circ}C$, the actual temperature of the sample was estimated to be $1,020^{\circ}C{\pm}123^{\circ}C$. Note that the specimen temperature should be carefully evaluated because of the undesired effects, i.e., the temperature non-uniformity due to the sample design of the MEMS chip, and distortion due to thermal expansion.
Objectives: To evaluate the biodurability of Korean Chrysotile(KC), the changes in fibers numbers and changes in the element composition of fibers from the lung of Sprague-Dawley rats instilled KC(average size $4.74{\mu}m$, $59,043{\times}10^6$ fibers/mg) was estimated. Methods: Rats were administered 1 mg KC(low group) or 2 mg KC(high group) by a single intratracheal instillation. At each time point(5 days, 5 weeks, 10 weeks), the numbers of KC fibers and the changes of element composition(atomic %) of KC fibers from the lung of the rats were analyzed with transmission electron microscope equipped with energy dispersive X-ray spectrometer. Results: Over time, the number of fibers within the lungs of animals were reduced. The average length of the low and high group is significantly reduced from 5 days after administration. Over time, the fiber ratio of at least $5{\mu}m$ remaining in the lung tissue of the low concentration group was up but the high group was reduced. From day 5 after administration, the composition ratio(Mg) was significantly decreased in all groups. Conclusions: Size and composition of Korean Chrysotile in the lung tissue of rats was changed from 5 days.
This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 1-year-old female catfish(Silurus asotus). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope. All data were collected from October in 1992 to May in 1993. The size of the nucleoli and number of the yolk granules increased as the oocyte grew. Yolk granules were deposited in the oocyte as fluid. Due to the presence of large early and late maturing oocytes, their ovaries were large, transparent, granular, and greenish in color. As the percentages of fish in LMO and RO stage increased from March to April, mean of GSI values(19.95%) increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Processes, microvilli, from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiate becomes more compact and devoid of pore canals during oocyte maturation. The electrophoretic pattern of major band in mature stage was much thicker(21k, 24k, 32k, 45k, 67∼110k, 170k dalton) than that in previtellogenic phase.
Electrical characteristics of soot particles in a LPG diffusion flame were studied for the control of soot particle coagulation. When a DC voltage was applied between two electrodes installed parallel to gas flow, ionic wind effect caused soot deposition on the cathode, implying that most of the soot particles were positively charged. Soot deposit on the cathode linearly increased and was saturated with respect to the strength of the applied voltage. The possibility of applying an AC voltage to enhance the particle coagulation was then investigated and the efficiency of the size control was checked with transmission electron microscope photographs. For the amplitude of 2 kV AC field, primary (spherical) soot particle size decreased from 30 ~ 40 nm to around 20 nm when the frequency of the applied AC voltage was 60 Hz and higher. Collisions between the soot particles in such a selected AC condition could lead to the formation of much bigger agglomerates of roughly 1-5 .mu.m in size.
Using mouse tissue, we studied electron opacity effect of tannic acid for transmission electron microscopic staining. Tannic acid-glutaraldehyde in 0.1M phosphate buffer was used as a fixative. To compare with this we have tested another method consisting of heavy metal staining after treatment of tannic acid in sodium tetraborate (borax) on glutaraldehyde-fixed sections. We have achieved equally consistent electron opacity in both methods. The elastin, collagen, basal lamina of skin and gap junctions of the epithelial cells gave excellent results, while it was good for glycogen, cilia, and plasma. Also fat cells and lipid droplets gave good preservation when tannic acid was added in the fixative. However, prolonged fixation in tannic acid-added fixative was hazardous for further processing, i.e., sectioning problem and deep electron opacity background.
$Al_2O_3$-copper alloy has been successfully made by gas atomization, mechanical alloying and hot pressing. In order to investigate microstructure and phase, it has been studied by using scanning electron microscope, transmission electron microscope and X-ray diffractometor. Mechanical properties have been examined using hardness tester and compressive tester according to annealing temperature. Although comparatively large Cu-Al powders are milled, the reaction between Cu-Al and $Cu_2O$ occurs and very fine $Al_2O_3$ particles in the matrix particles (5-10nm) are obtained. Compressive strength of this alloy is more than that of GlidCop Al60.
Ni nanowires were fabricated using anodic aluminum oxide (AAO) membrane as a template by electrochemical deposition. The nanowires were formed within the walls of AAO template with 200 nm in pore diameter. After researching proper voltage and temperature for electrochemical deposition, the length of Ni nanowires was controlled by deposition time and the supply of electrolyte. The morphology and microstructure of Ni nanowires were investigated by field emission scanning electron microscope (FE-SE), X-ray diffraction (XRD) and transmission electron microscope (TEM).
The effects of In and Be additions on the aging behaviors and mechanical properties in the Al-Cu-Li alloy were investigated using the transmission electron microscope, the scanning electron microscope, hardness and tensile strength measurement. The aging is promoted and the double hardness peak is appeared by the addition of In and Be in the Al-Cu-Li alloy. The strength is increased, but the elongation and toughness are decreased by the addition of In in the Al-Cu-Li alloy. And the Al-Cu-Li-In-Be alloy has good combination with the strength, the elogation and toughness than Al-Cu-Li-In alloy.
Multiwalled carbon nanotubes were massively produced by the catalytic reaction of C$_2$H$_2$ - Fe(CO)$\sub$5/ mixture at 750 - 950$^{\circ}C$ in a quartz tube reactor and over quartz substrates. Well-aligned MWNT array grows perpendicular to the quartz tube reactor and the quartz substrates at an average of 60 nm in diameter and up to several thousands of micrometers in length. This method does not require any pretreatment of substrates and CNTs are grown at atmospheric pressure. It could be suitable for mass production of multiwalled nanotubes. Scanning electron microscope and transmission electron microscope images of the nanotubes deposited on the substrates allowed us to monitor the quality of MWNTs grown under different operating conditions.
