NMR Structure of Syndecan-4L reveals structural requirement for PKC signalling

  • Syndecans, transmembrane heparan sulfate proteoglycans, are coreceptors with integrin in cell adhesion process. It forms a ternary signaling complex with protein kinase C and phosphatidylinositol 4,5 bisphosphate (PIP2) for integrin signaling. NMR data indicates that cytoplasmic domain of syndecan-4 (4L) undergoes a conformational transition in the presence of PIP2, forming oligomeric conformation. The structure based on NMR data demonstrated that syndecan-4L itself forms a compact intertwined symmetric dimer with an unusual clamp shape for residues Leu$^{186}$ -Ala$^{195}$ . The molecular surface of the syndecan-4L dimer is highly positively charged. In addition, no inter-subunit NOEs in membrane proximal amino acid resides (Cl region) has been observed, demonstrating that the Cl region is mostly unstructured in syndecan-4L dimmer. However, the complex structure in the presence of PIP2 induced a high order multimeric conformation in solution. In addition, phosphorylation of cytoplasmic domain induces conformational change of syndecan-4, resulting inhibition of PKC signaling. The NMR structural data strongly suggest that PIP2 promotes oligomerization of syndecan-4 cytoplasmic domain for PKC activation and further induces structural reorganization of syndecan for mediating signaling network in cell adhesion procedure.

Combined resonant column and cyclic triaxial tests to estimate the dynamic behavior of undisturbed saturated clayey soils of Adapazarı, Turkey

  • Turkey is one of the most important earthquake regions in Europe. This region has been exposed to many earthquakes of different magnitudes from past to present. It is of great importance to determine the dynamic properties of the soils for structures to be built in earthquake zones. In order to minimize the damages that may occur, the behavior of the soils under repeated loads should be known and taken into consideration in the design. In this study, 4 different point borings were taken near active fault lines in the North Anatolian fault zone (NAFZ). In order to determine the dynamic parameters of soils, both dynamic triaxial (TRX) and resonant column (RC) tests were carried out on undisturbed samples at every 5 m. As a result of the experiments, Vs and Gmax values were obtained from the field and differences were determined. The dynamic behavior of the soil was examined at varying depths with the comparison of reference models in the literature and compatible results were obtained. Finally, the behavior at the transition region is highlighted. As a result, three shear modulus and dumping ratio models have been proposed for clay soils to be used in different soil conditions.

Research on Solar System Small Bodies using the Korean Small Telescopes Network

  • Small bodies in the solar system are pristine leftovers of planetesimals since the formation epoch (~4.6 Gyr ago). After the formation, icy planetesimals have been preserved in the distant cold place beyond 30 au (i.e., Trans-Neptunian region) until recently without any catastrophic processes but have just been injected into inner region (<~5 au from the Sun) to be observed as comets. On the contrary, asteroids are rocky primitive objects (although some of them contains icy volatiles) distributing in the mainbelt between Mars and Jupiter orbits. Because of frequent encounters in the mainbelt, asteroids have experienced a number of repeated impacts until the present day. Namely, it is important to investigate thermal alternation process of cometary volatiles and refractories in the solar radiation field, whereas collisional and subsequence phenomena of asteroidal bodies. Although recent spacecraft observations revealed the physical natures on the surfaces of comets and asteroids, their interiors still remain largely unexplored. It is likely that a sudden brightening of a comet is associated with rapid sublimation of internal CO and CO2 or phase transition of amorphous H2O. An episodic dust ejection from an asteroid is causally related to an impact among asteroids, sudden sublimation of remaining subsurficial volatiles, etc. Because these transient phenomena provide rare opportunities to investigate their interiors, immediate observations using any optical instruments are particular important. In my presentation, I will review some examples of such transient phenomena in the solar system and propose possible collaborative research using the Korean Small Telescope Network.

Impedance Spectroscopy of (Pb0.92La0.08)(Zr0.95Ti0.05)O3 Ceramics above Room Temperatures

  • La modified lead zirconate titanate ceramics (Pb0.92La0.08)(Zr0.95Ti0.05)O3 = PLZT-8/95/5 were prepared using the conventional solid state reaction method in order to investigate the complex impedance characteristics of the PLZT-8/95/5 ceramic according to temperature. The complex impedance in the PLZT-8/95/5 ceramic was measured over a temperature range of 30~550 ℃ at several frequencies. The complex dielectric constant anomaly of the phase transition was observed near TU1 = 179 ℃ and TU2 = 230 ℃. A remarkable diffuse dielectric constant anomalous behaviour of the complex dielectric constant was found between 100 ℃ and 550 ℃. The complex impedance spectra below and above TU1 and TU2 were fitted by the superposition of two Cole-Cole types of impedance relaxations. The fast component in the higher frequency region may be due to ion migration in the bulk, and the slow component in the lower frequency region is interpreted to be the formation and migration of ions at the grain boundary or electrode/crystal interfacial polarization.

First Report of Pectobacterium brasiliense Causing Bitter Melon Soft Rot Disease in Korea

  • In the Goesan region, located in Chungcheongbuk-do, Korea, a significant outbreak of soft rot infections was documented in August 2021, affecting fruits of Momordica charantia, commonly known as bitter melon or bitter gourd. The symptoms included a noticeable transition to yellowing in the affected fruits, eventually leading to their collapse. The bacterial strain KNUB-09-21 was isolated from the diseased fruits. Molecular analysis, using the sequences of the 16S rRNA region and three housekeeping genes (dnaX, recA, and leuS), along with the results of compound utilization in the API ID 32 GN system, provide strong evidence for the identification of the isolate KNUB-09-21 as Pectobacterium brasiliense. The pathogenicity of strain KNUB-09-21 on M. charantia was confirmed through a controlled inoculation test. Within two days, inoculated fruits displayed soft rot symptoms closely resembling those observed in naturally affected fruits. This is the first report of soft rot on M. charantia in Korea.

  • Recently it is supposed to be a shift to regional geography. To understand the emergence of the new regional geography, It must be remarked that the progress of capitalist production and consumption change our conceptional apparatus such as representation of space. Region is one of the important mode of representations of space and time. In the Fordist capitalism of 20th century, development is regarded as diffusion of capitalist expansion of western worlds. State must support capitalist growth through regional policies which include constructing of infrastructure and regionalization of spatial division of labour. The regional development theories contributed as ideology and policy tools for state intervention. The region was simply one of the most logical classification tools of organizing geographical informations. In the theories of regional development, the concept of region was reduced to the formal unit of classification. As the transition from Fordism to flexible accumulation, the region is again acquiring its 'identity' and 'authenticity'. In this tendency of the revival of region, it is expected that good achievement could be made in the field of regional geography through relevant research methods. It is also believed one of the available means are historical approaches to the cultural and ecological regions. The historical approaches to cultural and ecological regions are not only correspondent with cultural development strategies of local governments, but also could convey regional identities through both narrativization of place and aestheticization of landscape.

DNA Sequence Analysis of 1-Nitropyrene-4,5-Oxide and 1-Nitropyrene-9,10-Oxide Induced Mutations in the hprt Gene of Chinese Hamster Ovary Cells

  • Nitropyrene, the predominant nitropolycyclic hydrocarbon found in diesel exhaust, is a mutagenic and tumorigenic environmental pollutant that requires metabolic activation via nitroreduction and ring oxidation. In order to determine the role of ring oxidation in the mutagenicity of 1-nitropyrene, its oxidative metabolites, 1-nitropyrene 4,5-oxide and 1-nitropyrene 9,10-oxide, were synthesized and their mutation spectra were determined in the coding region of hprt gene of CHO cells by a PCR amplification of reverse-transcribed hprt mRNA, followed by a DNA sequence analysis. A comparison of the two metabolites for mutation frequencies showed that 1-nitropyrene 9,10-oxide was 2-times higher than 1-nitropyrene 4,5-oxide. The mutation spectrum for 1-nitropyrene 4,5-oxide was base substitutions (33/49), one base deletions (11/49) and exon deletions (5/49). In the case of 1-nitropyrene 9,10-oxide, base substitutions (27/50), one base deletions (15/50), and exon deletions (8/50) were observed. Base substitutions were distributed randomly throughout the hprt gene. The majority of the base substitutions in mutant from 1-nitropyrene 4,5-oxide treated cells were $A{\rightarrow}G$ transition (15/33) and $G{\rightarrow}A$ transition (8/33). The predominant base substitution, $A{\rightarrow}G$ transition (11/27) and $G{\rightarrow}A$ transition (8/27), were also observed in mutant from 1-nitropyrene 9,10-oxide treated cells. The mutation at the site of adenine and guanine was consistent with the previous results, where the sites of DNA adduct formed by these compounds were predominant at the sites of purines. A comparison of the mutational patterns between 1-nitropyrene 4,5-oxide and 1-nitropyrene 9,10-oxide showed that there were no significant differences in the overall mutational spectrum. These results indicate that each oxidative metabolite exhibits an equal contribution to the mutagenicity of 1-nitropyrene, and ring oxidation of 1-nitropyrene is an important metabolic pathway to the formation of significant lethal DNA lesions.

  • At the aim of finding a Fehased amorphous alloy with a wide supercooled liquid region (${\Delta}T_{x}=T_{x}-T_{g}$) before crystallization, the changes in glass transition temperatudfI$T_{g}$ and crystallization temperature ($T_{x}$) by the dissolution of additional M elements were examined for the $Fe_{80}P_{10}C_{6}B_{4}$(x~6at%. M= transition metals) amorphous alloys. The ${\Delta}T_{x}$ value is 27K for the Fe,,,P,,,C,,R, alloy and increases to 40K for the addition of M=4at%Hf, 4at%Ta or 4at%Mo. The increase in ${\Delta}T_{x}$ is due to the increase of $T_{x}$ exceeding the degree in the increase in $T_{g}$. The $T_{g}$ and $T_{x}$ increase with decreasing electron concentration (e/a) from about 7 38 to 7.05. The decrease of e/a also implies the increase in the attractive bonding state between the M elements and other constitutent elements. It is therefore said that $T_{g}$ and $T_{x}$ increase kith increasing attractive bonding force.

  • The regional innovation policies so far have been separated from the social problems facing the local communities. The regional innovation policies, regarding the region as the location of the business, have focused on the invigoration of business innovation activities. However, as the recent emergence of the new paradigm of innovation policy aiming the sustainability, 'transformative innovation policy,' has led to a search for regional innovation policies that begin with solving the local social problems. This research paper deals with regional innovation theory that starts from searching for solutions and system transformation for social problems such as climate crisis and energy problems. The objective is to present a new framework called 'transformative regional innovation policy' and to improve its content through case studies by combining the results of the transformative innovation policy and the regional innovation policy studies. In particular, the contribution of this paper is to analyze and discuss the concept of the transition platform, which aims to solve the local social problems, through the case studies of Gussing, Austria and Esbjerg, Denmark. Lastly, it discusses the derived implications of the cases applied in Korean society.

A Study on the Transition of Salmi in Dapo Type Kong-Po(拱包) - Focused on Inner and Outer Same Chulmok(出目) - (다포계 공포의 살미 변천에 관한 연구 - 내외 2출목·내외 3출목 공포를 중심으로 -)

  • Integrated-Salmi can be suggested as a character of Da-po type Kong-po in Korea's traditional architecture. However, only few studies are made on this subject yet. This study is to investigate the integration process of Salmi through cases of certain Kong-po which have the same number of inner and outer Chul-mok and are also known to precede in time. The results of the study are like below. First, Integrated-Salmi is more dominant in inner and outer 3 Chul-mok Kong-po than in inner and outer 2 Chul-mok Kong-po. Second, While inner and outer 2 Chul-mok prevail in Yeong-nam region, inner and outer 3 Chul-mok prevail in Ho-nam region. Third, integrated Salmi of Inner and outer 3 Chul-mok, first appeared in internal Jusangpo to be carried over to internal Juganpo then eventually to outer Salmi. Fourth, the reason why integrated form precede in internal salmi of inner and outer 3 Chul-mok Kong-po is due to 2 main factors. First of which is the impact of integrated Boaji that developed in Da-po type since the early 16th century. The second factor is downward expansion of Cho-gak's Boaji which was adopted to cope with disorders made in structure of Chum-Cha, due to inner and outer 3 Chul-mok type buddhist temples' wide reconstruction throughout Jeolla region following the Japanese invasion of Korea in 1592. This study is to disclose the regularity of development process of Salmi's Cho-gak(草刻) which is a characteristic of Korea's Da-po type Kong-po.
