본 연구는 생체 내 세포 및 조직배양기로서의 면역결핍동물을 개발할 목적으로 proximal Ick promoter와 DT-A유전자를 이용하여 형질전환생쥐를 생산하였고 형질전환생쥐의 면역기관에서 Di-phteria toxin이 발현되어 T-cell이 결핍되는지를 분석하였다. 총 암수 2마리의 형질전환생쥐를 PCR과 South-ern blotting으로 분석하여 얻었으며 이식유전자의 copy수는 약 2∼3 copy가 정착된 것으로 확인되었다. 형질전환생쥐의 thymus, spleen, liver 조직을 분리한 후 total RNA를 추출하여 poly(dT) primer 와 DT 특이적 primer를 이용하여 RT-PCR수행 결과 형질전환생쥐의 thymus, spleen, liver에서 DT gene이 발현되고 있는 것을 확인할 수 있었다. 형질전환생쥐의 이들 조직간에 DT 발현량에는 큰차이는 없는 것으로 확인되었다. 형질전환생쥐의 혈액에서 적혈구, 백혈구 ,혈소판, 헤모글로빈 등이 정상생쥐보다 감소되었고 특히 백혈구수와 혈소판의 수가 크게 감소되어 있는 것으로 나타났다. 또한 형질전환생쥐의 혈액을 CD3 antibody를 이용하여 FACS 분석을 실시하여 형질전환생쥐의 혈액 중 T-cell이 수가 비정상적으로 줄어든 것을 확인할 수 있었다. 본 연구에서는 Ick-DT 형질전환생쥐에서 DT유전자의 발현에 의한 T-cell 결핍을 유도할 수 있는 것으로 나타나 이를 바탕으로 돼지를 이용한 사람의 이종장기 배양용 형질전환동물을 생산하여 응용될 수 있을 것으로 사료된다.
동일 종내 미꾸라지(Misgurnus mizolepis) 웅성발생성 인공처녀생식(intraspecific androgenesis)기술을 개발하기 위해 미꾸라지 난 유전물질의 불활성화와 이에 따른 웅성발생성 반수체 유도 조건을 최적화하였다. 다양한 자외선(UV)농도$(0\sim25,650ergs/mm^2)$를 이용하여 미꾸라지 난의 유전학적 불활성화를 시도하였으며 수정률, 부화율 및 반수체 출현율을 평가한 결과 10,800 ergs의 농도에서 가장 우수한 반수체 유도 효율을 보였으며 최초 처리 난 중 50%이상의 웅성발생성 반수체 수율을 확보할 수 있었다. 웅성발생성 개체는 전형적인 haploid syndrome의 특징을 나타내었고 flow cytometry분석 결과 정확히 미꾸라지 반수체(1.4 pg/cell)의 DNA함량을 보유하는 것으로 나타났다. 인위적으로 삽입된 형질전환 유전자 표지를 이용하여 부계 특이적인 인공처녀생식 여부를 확인하였으나 일부 반수체자어들에서$(8\sim11%)$ 모계 유전물질의 잔류가 검출되었다.
The use of transgenic fish has so far been chiefly limited by the lack of predictable, strong, tissue specific, and position-independent expression of transgenes. For genetic analysis, expression of a marker transgene, easily screenable in the living fish, could facilitate studies of gene targeting, insertional mutagenesis, lineage, and mutational analysis. The expression of a chimeric transgene (nos-npt II) has been examined in 9 years old transgenic poplars (Populus koreana x P. nigra) growing in a nursery. The expression of the gene in twenty six independentely transformed plants were examined by 1) enzyme (NPT II) assay, 2) RT-PCR, and 3) resistance to kanamycin. High NPT II activities in young leaves of all the transformed plants were found even without a selection pressure for antibiotics for 9 years. However, the activity varied with the positions of leaves in the stem in that young leaves showed higher activity than did mature tissues. When leaf segments were cultured in the presence of 150 mg/l kanamycin, only those from young leaves produced vigorously growing callus. However, as in the case of NPTII assay, the leaf segments from mature leaves did not form callus well on the media. RT-PCR with nptII specific primers also showed that amplification products were observed only when RNAs from young tissues were used. The total RNA gel showed that while RNA in young leaves are relatively stable and in a large quantity, those in old leaves were mostly degraded. All the above results suggest that the gene is transcriptionally active only in young tissue even though it is attached to a constituitive promoter. Therefore, the expression of foreign gene in poplar plants seemed to be affected by the metabolic state of the cells and thus vary greatly with the developmental stages and the age of tissue.
Gene-manipulated mice were discovered for the first time about a quarter century ago. Since then, numerous sophisticated technologies have been developed and applied to answer key questions about the fundamental roles of the genes of interest. Functional genomics can be characterized into gain-of-function and loss-of-function, which are called transgenic and knock-out studies, respectively. To make transgenic mice, the most widely used technique is the microinjection of transgene-containing vectors into the embryonic pronucleus. However, there are critical drawbacks: namely position effects, integration of unknown copies of a foreign gene, and instability of the foreign DNA within the host genome. To overcome these problems, the ROSA26 locus was used for the knock-in site of a transgene. Usage of this locus is discussed for the gain of function study as well as for several brilliant approaches such as conditional/inducible transgenic system, reproducible/inducible knockdown system, specific cell ablation by Cre-mediated expression of DTA, Cre-ERTM mice as a useful tool for temporal gene regulation, MORE mice as a germ line delete and site specific recombinase system. Techniques to make null mutant mice include complicated steps: vector design and construction, colony selection of embryonic stem (ES) cells, production of chimera mice, confirmation of germ line transmission, and so forth. It is tedious and labor intensive work and difficult to approach. Thus, it is not readily accessible by most researchers. In order to overcome such limitations, technical breakthroughs such as reporter knock-in and gene knock-out system, production of homozygous mutant ES cells from a single targeting vector, and production of mutant mice from tetraploid embryos are developed. With these upcoming progresses, it is important to consider how we could develop these systems further and expand to other animal models such as pigs and monkeys that have more physiological similarities to humans.
In previous reports, pVPSV.IGR2.1 transgenic mouse were described that brain tumor and lymphoma by reason of Vasopressin-SV40 T antigen. In this study, we produced pVPSV.IGR3.6 transgenic mouse that used pVPSV.IGR3.6 vector. Expression of transgene was vary different in transgenic mouse. We obtained 6 transgenic mouse line, moreover they had died at the age of 2-6 weeks without transmitting the transgene to their offspring, and had tumorigenesis on same location with pVPSV.IGR2.1 transgenic mouse. Only a founder mouse was investigated for expression of fusion gene. Here we extended this transgenic approach to the study of tumor progression. From the mouse, we confirmed brain tumor cell, after then cultured for investigate characterization. In this report, we demonstrate that reduction of survival rate in transgenic mouse fused vasopressin gene length, acquisition of brain tumor cell, composition with astrocyte cells and neuronal cells. Finally, cells had no change with increase of passage.
High production of milk and its components are necessary to allow maximal growth of developing piglets. In this study, transgenic pigs were produced containing the $\alpha$-lactalbumin gene, whose product is a potential limiting component in the production of milk. Two lines of transgenic pigs were produced to analyze the effects that overproduction of the milk protein $\alpha$-lactalbumin may have on milk production and piglet growth. Transgenic pigs were produced through microinjection of the bovine $\alpha$-lactalbumin gene. The gene construct contained 2.0 kb of 5 flanking region, the 2.0 kb coding region and 329 bp of 3 flanking region. Sows hemizygous for the transgene produced as much as 0.9 g of bovine $\alpha$-lactalbumin per liter of pig milk. The production of the bovine protein caused approximately a 50 % increase in the total $\alpha$-lactalbumin concentration in pig milk throughout lactation. The concentration of bovine $\alpha$-lactalbumin was highest on day 0 and 5 of lactation and decreased as lactation progressed. The ratio of bovine to porcine $\alpha$-lactalbumin changed during the sow's lactation. This ratio was 4.3 to 1 on day 0 of lactation, but by day 20 of lactation the ratio was 0.43 to 1. This suggested that the bovine transgene and the endogenous porcine gene were under slightly different control mechanisms. The higher level of total $\alpha$-lactalbumin present on day 0 of lactation was correlated with higher lactose percentage on day 0 in transgenic sows (3.8 %) as compared to controls (2.6 %) (P < 0.01). Although there was also a trend for higher lactose percentage in transgenic sows on day 5 and 10 of lactation, no significant differences were observed. These data suggest that $\alpha$-lactalbumin is limiting early in lactation of swine. Furthermore, higher concentrations of $\alpha$-lactalbumin early in lactation may boost milk output.

Nonstructural protein 3 (NS3) encoded by RNA3 of Rice stripe virus (RSV), known to be a suppressor of gene silencing, was cloned and sequenced. The cloned NS3 gene is composed of 636 nucleotides encoding 211 deduced amino acids, and showed a high degree of similarity with the equivalent genes isolated from Korea, Japan and China. The NS3 gene promoted the enhancement of transient gene expression and suppressed transgene co-silencing. In the transient GFP expression via agroinfiltration, GFP expression was dramatically enhanced in terms of both protein yield and expression period in the presence of NS3. The highest accumulation of GFP protein reached to 6.8% of total soluble proteins, which corresponded to a two-fold increase compared to that obtained in the absence of NS3. In addition, NS3 significantly suppressed the initiation of GFP co-silencing induced by the additive GFP infiltration in GFP-transgenic Nicotiana benthamiana. The NS3 gene was also found to be a stronger suppressor than Cucumber mosaic virus 2b. These observations are believed to be derived from the strong suppressive effect of NS3 on gene silencing, and indicate that NS3 could be used as an effective enhancer for the rapid production of foreign proteins in plants.
The pronuclear injection of metallothionein-human growth hormone (MT-hGH) gene into rabbit zygotes was performed to establish in vitro developmental system and to detect the presence of the injected gene by nested PCR. Mature female New Zealand White rabbits were superovulated by eGG and hCG treatments. The rabbits were mated and the zygotes were collected from the oviducts 18-22 h after hCG injection by flushing with D-PBS. Two to three picoliters of MT-hGH gene was microinjected into male pronuclei. The foreign gene-injected zygotes were cultured in TCM-199 or RD mediurn containing 10% FCS with a monolayer of rabbit oviductal epithelial cefls in a 5% $CO_2$ incubator. The presence of injected DNA in rabbit embryos or blastomeres at different developmental stages .vas detected by a nested PCR analysis. The results are summarized as follows ; 1.The developmental rate of the MT-hGH gene-injected zygotes to blastocyst was significantly higher in TCM-199 medium (68.1%) than in RD medium (42.9%). 2.The gene injection into pronuclei at 18 or 22 hours post hCG treatment during pronuclear stage did not much affect on the in vitro development of the rabbit embryos. 3.The rate of gene-positive embryos detected by the nested PCR analysis was significantly decreased when they developed to blastocysts. The results indicate that the screening of transgene in rabbit embryos by nested PCR analysis could be a prornisible method for the preselection of transgenic embryos. Furthermore, the preselection of transgenic embryos would greatly reduce hoth the cost and effort of production of transgenic animals.
