Phospholipase C-${\gamma}l$ (PLC-${\gamma}l$) expression is associated with cellular transformation. Notably, PLC-${\gamma}$ is up-regulated in colorectal cancer tissue and breast carcinoma. Because exotoxins released by Clostridium botulinum have been shown to induce apoptosis and promote growth arrest in various cancer cell lines, we examined here the potential of Clostridium difficile toxin A to selectively induce apoptosis in cells transformed by PLC-${\gamma}l$ overexpression. We found that PLC-${\gamma}l$-transformed cells, but not vector-transformed (control) cells, were highly sensitive to C. difficile toxin A-induced apoptosis and mitotic inhibition. Moreover, expression of the proapoptotic Bcl2 family member, Bim, and activation of caspase-3 were significantly up-regulated by toxin A in PLC-${\gamma}l$-transformed cells. Toxin A-induced cell rounding and paxillin dephosphorylation were also significantly higher in PLC-${\gamma}l$-transformed cells than in control cells. These findings suggest that C. difficile toxin A may have potential as an anticancer agent against colorectal cancers and breast carcinomas in which PLC-${\gamma}l$ is highly up-regulated.
Al-Si coated boron steel and Zn coated DP steel plates were laser-welded to manufacture a Tailor Welded Blank (TWB) for a car body frame. Hot-stamping heat treatment ($900^{\circ}C$, 5 min) was applied to the TWB weld, and the microstructural change and transformation mechanism were investigated in the Al-rich area near the bond line of the Al-Si coated steel side. There was Al-rich area with a single phase, $Fe_3(Al,Si)$, which was transformed to ${\alpha}-Fe$ (Ferrite) after the heat treatment. It could be explained that the $Fe_3(Al,Si)$ phase was transformed to ${\alpha}-Fe$ during heat treatment at $900^{\circ}C$ for 5 min and the resultant ${\alpha}-Fe$ phase was not transformed by rapid cooling. Before the heat treatment, the microstructures around the $Fe_3(Al,Si)$ phase consisted of martensite, bainite and ${\alpha}-Fe$ while they were transformed to martensite and ${\delta}-Fe$ after the heat treatment. Due to the heat treatment, Al was diffused to the $Fe_3(Al,Si)$ and this resulted in an increase of Al content to 0.7 wt% around the Al-rich area. If the weld was held at $900^{\circ}C$ for 5 min it was transformed to a mixture of austenite (${\gamma}$) and ${\delta}-Fe$, and only ${\gamma}$ was transformed to the martensite by water cooling while the ${\delta}-Fe$ was remained unchanged.
Background: Ginsenosides, which have strong biological activities, can be divided into polar or less-polar ginsenosides. Methods: This study evaluated the phytochemical diversity of the saponins in Panax ginseng (PG) root, American ginseng (AG) root, and Panax notoginseng (NG) root; the stem-leaves from Panax ginseng (SPG) root, American ginseng (SAG) root, and Panax notoginseng (SNG) root as well as the saponins obtained following heating and acidification [transformed Panax ginseng (TPG), transformed American ginseng (TAG), transformed Panax notoginseng (TNG), transformed stem-leaves from Panax ginseng (TSPG), transformed stem-leaves from American ginseng (TSAG), and transformed stem-leaves from Panax notoginseng (TSNG)]. The diversity was determined through the simultaneous quantification of the 16 major ginsenosides. Results: The content of ginsenosides in NG was found to be higher than those in AG and PG, and the content in SPG was greater than those in SNG and SAG. After transformation, the contents of polar ginsenosides in the raw saponins decreased, and contents of less-polar compounds increased. TNG had the highest levels of ginsenosides, which is consistent with the transformation of ginseng root. The contents of saponins in the stem-leaves were higher than those in the roots. The transformation rate of SNG was higher than those of the other samples, and the loss ratios of total ginsenosides from NG (6%) and SNG (4%) were the lowest among the tested materials. In addition to the conversion temperature, time, and pH, the crude protein content also affects the conversion to rare saponins. The proteins in Panax notoginseng allowed the highest conversion rate. Conclusion: Thus, the industrial preparation of less-polar ginsenosides from SNG is more efficient and cheaper.
In order to improve the availability of phytase and probiotics together, a phytase gene from Aspergillus ficuum has been expressed in Lactobacillus. In this study, the transformed Lactobacillus with phytase gene was fed to pigs to determine its effect on pig production, feed conversion and gut microbes. Forty eight, 60-day-old, castrated pigs (Duroc${\times}$Landrace${\times}$Pietrain) were assigned to 6 groups, 8 pigs for each group. Group 1 was the control, group 2 was added with chlortetracycline (500 mg/kg), group 3 was added with the transformed Lactobacillus (500 mg/kg) with 20% (w/w) of calcium monohydrogen phosphate (CMP, $CaHPO_{4}$) removed, group 4 was added with the natural Lactobacillus (500 mg/kg) with 20% (w/w) of CMP removed, group 5 was added with the transformed Lactobacillus (500 mg/kg) with 40% (w/w) of CMP removed, group 6 was added with phytase (500 mg/kg) with 40% (w/w) of CMP removed. The results showed: i) the average daily gain (ADG) was improved in groups 2, 3 and 4 (p<0.05); ii) the diarrhea rates in the groups added with Lactobacillus were lower than in the other groups (p<0.05), in which the transformed Lactobacillus had more effect on reducing digestive disease; iii) the transformed Lactobacillus was most effective in improving the digestibilities of crude protein (CP), calcium (Ca), phosphorus (P), compared with the other groups (p<0.05); iv) Lactobacillus could increase lactic acid bacterium number and ammonia concentrations, and decrease pH values and E. coli number in pig feces (p<0.05); v) the phytase activity in the feces of pigs fed with the transformed Lactobacillus was 133.32 U/g, which was higher than in group 4 (9.58 U/g, p<0.05), and was almost the same as group 6 (135.94 U/g); vi) the transformed Lactobacillus could increase serum concentrations of IgA, triglyceride, and glutamic oxaloacetic transaminase activity (p<0.05), and had no significant effect on other serum indexes (p>0.05).
Objectives : The authors performed this study to further understand Saam acupuncture treatment in an aspect of the use of two patterns i.e. fixed- and transformed-pattern. Methods : The authors did documentary survey based on Do Hae Kyo Kam Sa Am Do In Chim Beop. 1. We investigated the frequency of the use of fixed- and transformed-pattern as well as the examples of use. 2. We surveyed related literatures and classified transformed-pattern. We did study on the principles of prescriptions of fixed- and transformed-pattern and researched the utilization. Results and Conclusions : 1. Fixed-pattern occupys 35.3% and transformed-pattern takes much more part. 2. Fixed-pattern is based on Nangyoung 69nan, and has the prescription of two enhancement and two inhibition by adaptation of Sangsaengsanggeuk(相生相剋) into self meridian and other meridian, and suggests the importance of the order of needling. 3. There are main points and assistant points in tonifying and suppressing of Fixed-pattern, accordingly, it has a system like Gunshinjwasa(君臣佐使). 4. Transformed-pattern is classified into following 3 types: first, the use of Nangyoung 75nan; second, the substitution of Junghuyl(井穴) with Hyunghyul(滎穴) or Haphyul(合穴) according to Nangyoung 73nan; third, the substitution of the same Osuhyul(五輸穴) in other meridians.

Sample size calculations plays an important role in a bioequivalence trials and is determined by considering power under the alternative hypothesis. The regulatory guideline recommends that $2{\times}2$ crossover design is conducted and raw data is log-transformed for statistical analysis. In this paper, we discuss the sample size calculation in $2{\times}2$ crossover design with the log-transformed data.
