In vitro Propagation of Arbuscular Mycorrhizal Fungi using Ri t-DNA Transformed Carrot Roots (Ri t-DNA로 형질전환된 당근 뿌리를 이용한 Arbuscular 균근균의 기내증식)

  • This study was conducted to propagate the arbuscular mycorrhizal fungi in vitro using the hairy root of carrot transformed by Agrobacterium rhizogenes with Ri t-DNA. Mycorrhizal spores and roots in sudangrass plants were wet-sieved, surface-sterilized and inoculated onto the hairy root of carrot on the Modified Strullu & Romand (MSR) medium. The mycorrhizal spores of Glomus sp. propagated in vitro for 12 weeks was about $50{\mu}m$, and the shapes of spores were round or elliptic. Spores were formed mainly at the middle of the hyphae. Number of mycorrhizal spores propagated using dual culture of the transformed carrot roots and the mycorrhizal inoculum for 12 weeks were about 1,200 per plates.

  • Gram-positive bacteria, Bacillus subtilis BR151 and Staphylococcus aureus RN4220 were transformed with high efficiency by electroporation. The cells were incubated until late log phase, washed three times with 10% glycerol, 1mM HEPES, 12% sucrose and resuspended to $10^{10}{\sim}10^{11}cfu/ml$, then stored at -$70^{\circ}C$. Transformation efficiency of B. subtilis BR151 was $1.03{\times}10^7cfu/{\mu}g$ with cells washed with 10% glycerol and electroporated by 15KV/cm, 0.7msec pulse with pUB110. Transformation efficiency of S. aureus RN4220 was $4{\times}10^6cfu/{\mu}g$ with cells washed with 1mM HEPES + 10% glycerol and electroporated by 15KV/cm, 2.5msec pulse. The number of total transformants was 1000 when B. subtilis BRI51 was transformed with 100ng pUB110 DNA and the number of total transformants was 9000 when S. aureus RN4220 was transformed with 10ng pUB110 DNA

  • In this paper a hierarchical stereo matching algorithm to obtain the disparity in wavelet transformed domain by using locally adaptive window and weights is proposed. The pyramidal structure obtained by wavelet transform is used to solve the loss of information which the conventional Gaussian or Laplacian pyramid have. The wavelet transformed images are decomposed into the blurred image the horizontal edges the vertical edges and the diagonal edges. The similarity between each wavelet channel of left and right image determines the relative importance of each primitive and make the algorithm perform the area-based and feature-based matching adaptively. The wavelet transform can extract the features that have the dense resolution as well as can avoid the duplication or loss of information. Meanwhile the variable window that needs to obtain precise and stable estimation of correspondense is decided adaptively from the disparities estimated in coarse resolution and LL(low-low) channel of wavelet transformed stereo image. Also a new relaxation algorithm that can reduce the false match without the blurring of the disparity edge is proposed. The experimental results for various images show that the proposed algorithm has good perfpormance even if the images used in experiments have the unfavorable conditions.

In this paper, an Industrial HMI applied the transformed state splitting algorithm. this study suggests by applying a transformed the Successive state splitting algorithm, for the modeling in the questions of the expected data. So, you can save time and reliable and precise as high as 98.15 percent repregented recognition rate. HMI system applied to the voice of industrial equipment the man can not act directly in the industry environment was able to drive devices. Optimize the performance of the engine was the voice of HMI system.

  • Historians describe the early years of the 20th century as a period of "nationalism." During this period, Turkish nationalism transformed into a thought movement which emerged to defend Turkish national sovereignty during the collapse of the Ottoman Empire. Approaches towards nationalism in Turkey are based on the idea of national sovereignty and the ideas of national independence that developed subsequently. Nationalism in Turkey first transformed from Pan-Islamism into multinational Ottomanism, and finally developed into Turkish nationalism and patriotism. This process emerged as a movement of self-discovery in the multicultural structure of the Ottoman Empire and transformed into Turkism. The Balkan Wars (1912-1913) destroyed the foundation upon which Ottomanism was based, and led to the rise of Turkish nationalism, in other words, Turkism. The idea of nation in modern terms in the recent history of thought and nationalism subsequently developed based on this idea and emerged with the Turkism movement. Thus, Turkism became the movement of Turks in the empire, combined with political Turkism which was supported by the intellectuals who came to the Ottoman Empire from Russia. In this article, the formation of Turkist movements and the leading intellectuals of Turkish nationalism, who emerged at the end of the 19th century and at the beginning of 20th century in the Ottoman Empire, are investigated in order to examine the historical progress of nationalist approaches in a period in which a new national state was established and improved.

  • The spermatogenesis of Korean slug, Incilaria fruhstorferi are observed by electron microscope. The results are as follows: The spermatogenesis of Korean slug, Incilaria fruhstorferi, is processed through the five stages; Spermatogonia, primary spermatocyte, secondary spematocyte, spermatid and spermatozoon. The spermiogenesis, the differentiation of the spermatid, is also processed through the five stages. In stage 1, the numerous and round mitochomdria in the cytoplasm are moved around the nucleus of spermatid. In stage 2, the nucleus of spermatid transformed into the oval shape, and the oval nucleus is surrounded by many rough endoplasmic reticulum. In stage 3, the oval nucleus of spermatid is changed to be curved as an arrow, and then two centrioles appeared behind nucleus. The centriole is sucked into the cytoplasm. and almost all the chromatins are changed into heterochromatins. In stage 4, the nucleus of spermatid are transformed into the oval shape, when the lamella plate chromatin of spermatid form in the nucleoplasm. In stage 5, the oval nucleus is then transformed into the stream-line shape when the lamella plate chromatin of spematid gradually concentrated in the nucleus, and long axoneme ($65{\mu}m$ in length) form from the distal centriole. Two long mitochondria in the middle piece and the main piece of spermatozoon array spirally along a long axoneme, and the mitochondria matrix is gradually filled with electron-dense glycogen particles ($0.1{\mu}m$ in size). The axoneme of spermatozoon consists of typical 9+2 microtubular pattern.

  • Various types of high strength steel sheets were usually used for improving the automobile safety and fuel efficiency by reducing the vehicle weight. The present study aimed to develop the TRIP (transformation induced plasticity) aided high-strength low carbon steel sheets by using a reverse transformation process. The 0.1C-4~8Mn steels were reverse-transformed by slow heating to intercritical temperature region and then furnace cooled to the room temperature. Granular type retained austenite was observed in 4Mn steel and lath type retained austenite was also observed in 6~8Mn steel. The results show that the 6Mn steel under reverse transformed at $625^{\circ}C$ for 6 hrs has maximum elongation up to 39%. The optimum strength-elongation combination was 3,888 ($kg/mm^2{\times}%$) when the 8Mn steel was reverse transformed at $625^{\circ}C$ for 12 h.

  • Image identification is the process of checking whether the query image is the transformed version of the specific original image or not. In this paper, image identification method based on feature composition is proposed. Used features include color distance, texture information and average pixel intensity. We extract color characteristics using color distance and texture information by Modified Generalized Symmetry Transform as well as average intensity of each pixel as features. Individual feature is quantized adaptively to be used as bins of histogram. The histogram is normalized according to data type and it is used as the signature in comparing the query image with database images. In matching part, Manhattan distance is used for measuring distance between two signatures. To evaluate the performance of the proposed method, independent test and accuracy test are achieved. In independent test, 60,433 images are used to evaluate the ability of discrimination between different images. And 4,002 original images and its 29 transformed versions are used in accuracy test, which evaluate the ability that the proposed algorithm can find the original image correctly when some transforms was applied in original image. Experiment results show that the proposed identification method has good performance in accuracy test. And the proposed method is very useful in real environment because of its high accuracy and fast matching capacity.

An alternative approach to extreme value analysis for design purposes

  • The asymptotic extreme value distributions of maxima are a natural choice when designing against future extreme events like flood peaks or wave heights, given a stationary time series. The generalized extreme value distribution (GEV) is often utilised in this context because it is seen as a convenient single expression for extreme event analysis. However, the GEV has a drawback because the location of the distribution bound relative to the data is a discontinuous function of the GEV shape parameter. That is, for annual maxima approximated by the Gumbel distribution, the data is also consistent with a GEV distribution with an upper bound (no lower bound) or a GEV distribution with a lower bound (no upper bound). A more consistent single extreme value expression for design purposes is proposed as the Weibull distribution of smallest extremes, as applied to transformed annual maxima. The Weibull distribution limit holds here for sufficiently large sample sizes, irrespective of the extreme value domain of attraction applicable to the untransformed maxima. The Gumbel, Type 2, and Type 3 extreme value distributions thus become redundant, together with the GEV, because in reality there is only a single asymptotic extreme value distribution required for design purposes - the Weibull distribution of minima as applied to transformed maxima. An illustrative synthetic example is given showing transformed maxima from the normal distribution approaching the Weibull limit much faster than the untransformed sample maxima approach the normal distribution Gumbel limit. Some New Zealand examples are given with the Weibull distribution being applied to reciprocal transformations of annual flood maxima, where the untransformed maxima follow apparently different extreme value distributions.

Isolation and Characterization of a Salt Inducible Promoter from Chlorella vulgaris PKVL7422

  • Chlorella is a eukaryotic organism that can be used as an industrial host to produce recombinant proteins. In this study, a salt-inducible promoter (SIP) was isolated from the freshwater species Chlorella vulgaris PKVL7422 from the screening of genes that were upregulated after salt treatment. Several cis-acting elements, including stress response elements, were identified in the isolated SIP. Moreover, the Gaussia luciferase gene was cloned after the SIP and transformed into C. vulgaris to test the inducibility of this promoter. Reexamination of transcriptome of C. vulgaris revealed that genes involved in the synthesis of methyl jasmonic acid (MeJA), gibberellin (GA), and abscisic acid (ABA) were upregulated when C. vulgaris was treated with salt. Furthermore, the expression level of recombinant luciferase increased when the transformed C. vulgaris was treated with salt and MeJA, GA, and ABA. This study represents the first report of the C. vulgaris SIP and highlights how transformed microalgae could be used for robust expression of recombinant proteins.
