Agrobacterium-Mediated Co-transformation of Multiple Genes in Metarhizium robertsii

  • Fungi of the Metarhizium genus are a very versatile model for understanding pathogenicity in insects and their symbiotic relationship with plants. To establish a co-transformation system for the transformation of multiple M. robertsii genes using Agrobacterium tumefaciens, we evaluated whether the antibiotic nourseothricin has the same marker selection efficiency as phosphinothricin using separate vectors. Subsequently, in the two vectors containing the nourseothricin and phosphinothricin resistance cassettes were inserted eGFP and mCherry expression cassettes, respectively. These new vectors were then introduced independently into A. tumefaciens and used to transform M. robertsii either in independent events or in one single co-transformation event using an equimolar mixture of A. tumefaciens cultures. The number of transformants obtained by co-transformation was similar to that obtained by the individual transformation events. This method provides an additional strategy for the simultaneous insertion of multiple genes into M. robertsii.

  • Gi(氣) transformation theory is the foundation of Traditional Chinese Medicine[TCM]. This theory matured during the period of "Hwangjenaegyeong(黃帝內經)", and later in the history of TCM was a source of significant impact on the development process and direction of general medical theory. The concept of "gi transformation[氣化]" mainly originates from the definition of the three levels which are nature, the relationship between nature and human, and the body's own metabolism, and is used to describe the usage and the resulting changes of 'gi(氣)'. Gi transformation possesses six characteristics which are permanence, universality, representationalism, directional, variability, and orderliness. The research of the concept and characteristics of gi transformation[氣化] in "Hwangjenaegyeong" will help identify the important academic value and practical significance of the formation of the basic theory of TCM as the result of the gi transformation theory, and highlight the unique characteristics of TCM.

  • The object of the research is to estimate for pearlite structure of quenched carbon steels. The effects of temperature on physical properties metallic structures and the latent heat by phase transformation were considered. In this study a set of constitutive equations relevant to the analysis of thermo-elasto plastic materials with pearlite phase transformation during quenching process way presented on the basis of continuum thermo-dynamics. The iso-thermal transformation curve of the SM50C was formlated by cubic spline curve. The formulated equations of evolution in pearlite transformation was used for structure analysis. The volume fraction of pearlite was obtained from the results of calculated metallic structure by Finite element equation.

  • Adenoviruses are icosahedral virions containing double-stranded linea DNA. They are 70 nm to 90 nm in diameter and capsid is composed of 252 capsomeres. Several members of this group, including types commonly associated with respiratory disease in man, are capable of producing malignant tumors in young hamsters and a few types have been shown to be oncogenic in young rat. Previous report involving effect of caffein on transformation induced by Adenovirus type 12 [9] has been carried out. The present report represents a continuation of previous study. To obtain evidence concerning the effect of MNNG (N-methyl-N'-nitro-N-nitroguanidine) on transformation, investigation of adenovirus type 12 of this group was undertaken. For practical consideration it was desirable to investigate the effect of MNNG on the adenovirus type 12 induced transformation in L cell. Results were as following 1. Adeno virus type 12 induced transformation was enhanced in the presence of MNNG. 2. Yields of adeno type 12 virus in L cell were slightly inhibited by treatment of MNNG.

Degradation in Steels: Transformation Plasticity

  • Super-plastic deformation that originates from transformation plasticity has favorable aspects for steels with improved strength and ductility. However, it also causes undesirable deformation of products or specimens, leading to their degradation. This article reviews recent investigations of transformation plasticity. A combination of newly suggested models, numerical analyses, and novel experiments has attempted to reveal the mechanism. Since the nature of the transformation plasticity is still unclear, there are significant challenges still to be solved. Fundamental understanding of transformation plasticity will be essential for the development of advanced steels.

Digital Filter Design using the Symbol Pulse Invariant Transformation

  • In general, when IIR digital filter are designed from analog filters, the bilinera transformation and the impluse invariant tramsformation are commonly used. It is known, however, that high frequency response of digital filters designed by these transformations can not be well approximated to the sampled analog signals. In this paper, the symbol pulse invariant transformation is analyzed theoretically so that the symbol pulse invariant transformation which was originally application to a rectangular pulse is newly applied to double rate pulse signals and generic shape pulse signals. Also, the relation of spectra between a transfer function of digital filter and one of analog filter is considered. Further, we apply to design the digital high pass filters using the symbol pulse invariant transformation method.

  • The phase transformation behaviors of Ni-Ti shape memory alloys were investigated through a DSC(Differential Scanning Calorimetry). The annealing temperatures were applied from $600^{\circ}C,\;700^{\circ}C,\;800^{\circ}C,\;and\;900^{\circ}C$ for their effects on the phase transformation behaviors. Based on the results of DSC, phase transformation behaviors of shape memory alloys can be predicted by Liang's phase transformation model or Tanaka's phase transformation model.

Augmented Rotation-Based Transformation for Privacy-Preserving Data Clustering

  • Multiple rotation-based transformation (MRBT) was introduced recently for mitigating the apriori-knowledge independent component analysis (AK-ICA) attack on rotation-based transformation (RBT), which is used for privacy-preserving data clustering. MRBT is shown to mitigate the AK-ICA attack but at the expense of data utility by not enabling conventional clustering. In this paper, we extend the MRBT scheme and introduce an augmented rotation-based transformation (ARBT) scheme that utilizes linearity of transformation and that both mitigates the AK-ICA attack and enables conventional clustering on data subsets transformed using the MRBT. In order to demonstrate the computational feasibility aspect of ARBT along with RBT and MRBT, we develop a toolkit and use it to empirically compare the different schemes of privacy-preserving data clustering based on data transformation in terms of their overhead and privacy.

  • Agrobacterium-mediated genetic transformation has been widely used for the production of genetically modified transgenic plants to obtain specific desired traits. Most of the molecular mechanisms that underlie the transformation steps have been well elucidated over the years. However, a few steps, such as nuclear targeting, T-DNA integration, and Agrobacterium-plant proteins involved remain largely obscure and are still under extensive studies. This review describes the major steps involved in the molecular mechanism of Agrobacterium-mediated transformation and provides insight in the recent developments in studies on the Agrobacterium-mediated genetic transformation system. Some factors affecting the transformation efficiency are also briefly discussed.

Development of Non-protoplast transformation System in Aspergillus oryzae

  • Aspergillus oryzae is a filamentous fungus classified in the group Aspergillaceae Ascomycetes. It is an important microorganism for industrial production of enzymes and fermented food productions. It secrets large quantities of proteins or enzymes into the culture medium which makes this organism appealing for the production of heterologous proteins. Recently Electric field-mediated transformation method, electroporation, has been applied to fungal transformation. In this study, fungal transformation was carried out by bypassing the protoplast isolation step, decreasing the culturing time and non-protoplast transformation for the increment of transformation efficiency. Transformants were obtained with electroporation in optimal condition 2,500 voltage, 1,540 ohm and 0.50 capacitance. More than 1,000 transform ants were obtained with 6-10 hrs cultured mycelia without enzyme treatment, called non-protoplast transformation.
