동백(冬柏)을 위시한 차나무과(科) 식물(植物) 종실(種實)에 함유(含有)된 항산화제(抗酸化劑)에 관한 연구(硏究) (Studies on the Antioxidative Substances in the Seeds of Some Theaceae Family)

  • Dried seeds of Camellia japonica and Thea sinensis were investigated to determine the nature of their antioxidative activity. Activity was measured by the induction period in the coupled oxidation of a substrate lard and extracts or isolates to be tested. 70% methanol and dichloromethane extracts were found to be antioxidative abilities. Their unsaponifiables revealed weak antioxidative activity, although hexane extracts did not show antioxidative effect on lard. Column chromatography for dlchloromethane extracts gave 4 fractions(only 2 fractions were potent). HPLC was used in isolating potent antioxidative components from the column fractions and the precolumn-passed methanol extracts. They were separated into 7 and 8 components, respectively. The column fractions obtained from both seeds comprised trans-p-coumaric acid. trans-p-ferulic acid and an unknown component with minor components such as chlorogenic acid and catechin. On the other hand, the most prominent components in the methanol extracts were an unidentified component. trans-pcoumaric acid, trans-p-ferulic acid, catechin and chlorogenic acid. The unknown compound isolated from the column fractions and methanol extracts was identified as epicatechin by $^1H-and\;^{13}C-NMR$. The antioxidative activities of these components were epicatechin > catechin > chlorogenic acid > trans-p-ferulic acid > trans-p-coumaric acid.

Effects of Concentrate to Roughage Ratio on the Formation of cis-9, trans-11 CLA and trans-11-Octadecenoic Acid in Rumen Fluid and Plasma of Sheep When Fed High Oleic or High Linoleic Acid Oils

  • A metabolism trial with four ruminally fistulated sheep was conducted in a $4{\times}4$ Latin square design to examine the effect of concentrate to roughage ratio (70:30 vs. 85:15) and oil source (soybean oil vs. rapeseed oil) on the ruminal fermentation pattern and $C_{18}$-fatty acids composition including trans11-$C_{18:1}$ (trans11-ODA) and cis9, trans11-18:2 (cis9, trans11-CLA) in the rumen fluid and plasma. Oil was added to the concentrate at 5% level of the total diet (DM basis) and chopped rye grass hay was fed as roughage. An increased level of concentrate (85%) within supplemented oil slightly lowered pH but increased ammonia concentration. Supplementation of rapeseed oil relatively increased pH and ammonia concentration. Higher concentrate level resulted in increased tendencies of total VFA concentration while oil source did not affect the total VFA concentration and VFA proportion. Whole tract digestibilities of DM, CP, EE, NDF and OM in diets slightly increased at higher concentrate level. Proportions of oleic acid ($C_{18:1}$) and linoleic acid ($C_{18:2}$) in the rumen fluid were influenced by the fatty acid composition of oil source but oil source did not affect the in vitro formations of trans11-ODA and cis9, trans11-CLA. Slightly increased trans11-ODA and cis9, trans11-CLA proportions, however, were observed from the sheep fed high roughage diet supplemented with both soybean oil and rapeseed oil. The $C_{18:1}$ and $C_{18:2}$ composition in supplemented oils responded to those in plasma of sheep. Effects of concentrate to roughage ratio and oil source on trans11-ODA and cis9, trans11-CLA proportions in plasma were found to be small. Proportion of cis9, trans11-CLA in plasma tended to be increased from the sheep fed high roughage diet and collection time at 9h post feeding.

Production of Trans-10, Cis-12 Conjugated Linoleic Acid by Megasphaera Elsdenii YJ-4: Physiological Roles in the Rumen

  • Megaspahera elsdenii YJ-4, which was previously isolated as a producer of trans-10, cis-12 CLA, was studied for its carbon source on the CLA production. M. elsdenii YJ-4, was incubated with glucose and lactose, and cultured in batch and continuous culture systems with linoleic acid at various pHs to investigate CLA production. Batch cultures of the ruminal bacterium, M. elsdenii YJ-4, were resistant to stearic acid and linoleic acid, and little growth inhibition was observed even when the fatty acid concentration in the culture was as much as 4 mg $ml^{-1}$. Stationary phase batch cultures (0.25 mg bacterial protein $ml^{-1}$) that had been grown on lactate and incubated with linoleic acid (0.20 mg $ml^{-1}$) produced approximately 12 ${\mu}g$ trans-10, cis-12 CLA mg $protein^{-1}$ and little cis-9, trans-11 CLA was detected. Some linoleic acid was converted to hydrogenated products (chiefly stearic acid), but these fatty acids were less than 5 ${\mu}g$ mg bacterial $protein^{-1}$. Stationary phase batch cultures that had been grown on glucose produced at least 3-fold less trans-10, cis-12 CLA than ones grown on lactate. Cells from lactate-limited continuous cultures produced less trans-10, cis-12 CLA than those from batch culture, but only if the pH was greater than 6.4. When the pH of the lactate-limited continuous cultures was lower than 6.4, trans-10, cis-12 CLA and hydrogenated products declined. Cells from glucose-limited continuous cultures produced less trans-10, cis-12 CLA and hydrogenated products than the cells that had been limited by lactate, but pH had little impact on this production. These results support the idea that M. elsdenii YJ-4 could be one of the major producers of trans-10, cis-12 CLA which causes cows to produce milk with a low fat content.

일반 고체지와 저트랜스 고체지를 이용한 베이커리 제품의 특성 비교 (Comparison of the Bakery Products Made Commercial Fat or Low Trans Fat)

  • Breads and cookies made with conventional or low-trans fat(margarine and shortening) were analyzed by determining their trans fat content, textural properties and color values after production. The trans fatty acids content in conventional margarine was three times higher than that of low-trans margarine. Moreover, the content of trans fatty acids in the conventional shortening was 49 times higher than that of low-trans shortening. The trans fatty acid contents of cookies and breads made with low-trans fat, can be reduced more than 2 to 75 times compared to cookies and breads made with conventional fat. In the produced cookies and breads, the color values and textural properties(hardness, springiness, cohesiveness, gumminess and chewiness) did not differ significantly when the conventional fat or low-trans fat was used.

  • [$^{2}$H$_{2}$] Tranylcypromine hydrochloride (trans-3, 3-dideuterio-2-phenylcyclopropylamine HCL) was synthesized for application to the metabolic studies. Mass fragmentation processes for the tranylcypromine and its two synthetic intermediates .gamma-phenyl-.gamma.-butyrolactone and trans-2-phenylcyclopropanecarboxylic acid were described based upon comparisons between labeled and unlabeled compounds.

  • A new UPLC-DAD method for simultaneous determination of p-coumaric acid and trans-cinnamic acid in propolis was developed using Halo-C18 column and step gradient elution of MeCN and $0.1%H_3PO_4$ in 8 min. The method was validated by specificity, linearity, limit of detection, limit of quantification, precision(intra- and inter-day variability) and recovery tests. The validated method was sufficiently applicable for quantitative analysis of propolis products as well as propolis collected from different regions in Korea. The established method is simple, appropriate and efficient, and can be used for quality assessment of Korean propolis.

Structure-Guided Identification of Novel Phenolic and Phenolic Amide Allosides from the Rhizomes of Cimicifuga heracleifolia

  • Two phenolic allopyranosides and two phenolic amide allopyranosides, along with eight known phenolic compounds, including cimicifugic acids, shomaside B, fukiic acid, isoferulic acid, and piscidic acid, were isolated from the n-butanolic extract of rhizomes of Cimicifuga heracleifolia. On-line spectroscopic data for UV, NMR, and MS from a combination of LC-NMR and LC-MS techniques directly and rapidly provided sufficient structural information to identify and confirm all the structures of major phenolic compounds in the extract, in addition to their HPLC profiles. This combined analytic information was then used as a dereplication tool for structure-guided screening in order to isolate unknown phenolic compounds in the extract. Successive fractionation and purification using semi-preparative HPLC acquired four unknown allopyranosides, and their structures were identified as cis-ferulic acid 4-O-${\beta}$-D-allopyranoside, trans-ferulic acid 4-O-${\beta}$-D-allopyranoside, trans-feruloyltyramine 4-O-${\beta}$-D-allopyranoside, and trans-feruloyl-(3-O-methyl)dopamine 4-O-${\beta}$-D-allopyranoside, based on a subsequent spectroscopic interpretation.

Effects of Dietary Fat Sources on Occurrences of Conjugated Linoleic Acid and trans Fatty Acids in Rumen Contents

  • The effects of dietary sources of C18:2 n-6 or C18:3 n-3 fatty acids on the occurrence of conjugated linoleic acid (CLA) and time-dependent changes of free fatty acid fractions in rumen contents were investigated. Sheep (n=4) fitted with rumen fistula were used in a 44 Latin square design wxith each 14 d period. Sheep were fed one of four diets consisting of grass hay and concentrates in a ratio of 70:30. Dietary treatments were 100% concentrates (served as the control), and concentrates were replaced by safflower seed at 30% (SFS), safflower meal at 18% - safflower oil at 12% (SFO), and safflower meal at 18%-linseed oil at 12% (LNO). At the end of each experimental period, rumen contents from each sheep were collected before feeding and at 1, 3, 6 and 12 h after feeding. The levels of cis-9, trans-11 CLA in free fatty acid fraction were considerably increased in all treated groups relative to the control, but not significantly. The increase in cis-9, trans-11 CLA was slightly higher in SFS and SFO groups than group fed diet containing linseed oil. The level of cis-9, trans-11 CLA in free fatty acid fraction was reached to the maximum value at 1hr after feeding and, thereafter gradually decreased to near the value before feeding. The generation of trans-11 C18:1 was significantly higher in all treated groups than that of control. The level of trans-11 C18:1 was linearly increased after feeding of experimental diets, reaching the maximum value at 3 h. Feeding of diets containing polyunsaturated fats to sheep resulted in a marked increase in the levels of trans-11 C18:1 and a slight increase of CLA in free fatty acid fraction of rumen contents. Our results support that endogenous synthesis of CLA from trans-11 C18:1 may be involved the primary source of CLA in dairy product. (Asian-Aust.

  • The conversion of trans-cinnamic acid to phenylalanine using phenylalanine ammonia lyase (PAL) was examined. The optimum concentration of trans-cinnamic acid for the reaction was observed at 100 mM in cells and at 20 mM in cell free extracts, respectively. The production of L-phenylalanine was increased in both experiments as the concentration of ammonia was increased up to 10 M. The optimal pHs for the maximal conversion of trans-cinnamic acid to L-phenylalanine were 9.5 and 9.0 in experiments carried out with cells and with cell free extracts, respectively.

트랜스형 불포화 지방산이 mice 혈중 지질농도에 미치는 영향 (Effect of Trans-unsaturated Fatty Acid on Serum Lipid Levels in Mice)

  • 본 연구에서는 각기 다른 형태의 지방산이 ICR mice의 혈중 지질 농도에 미치는 영향을 평가하고자 하였다. 이를 위하여 8주령 수컷 ICR mice를 일반 식이섭취군(C), 10%의 트랜스 불포화 지방산 섭취군(TFA-1), 30% 트랜스 불포화 지방산 섭취군(TFA-2), 50% 트랜스 불포화 지방산 섭취군(TFA-3), 50% 포화 지방산 섭취군(SFA), 불포화 지방산 섭취군(USFA) 으로 나누어 식이 하였다. 혈중 총 콜레스테롤의 수치는 TFA-3 군과 SFA 군의 지질함량이 다른 군에 비하여 높았으며, 중성지방지수 역시 높게 나타났다. 총 콜레스테롤과 중성지방수치를 이용하여 나타낸 LDL 콜레스테롤의 경우에도 50%의 트랜스 지방산을 섭취한 군에서 포화 지방산을 섭취한 군과 유사한 결과가 나타났다. 이에 반해, 10%, 30%의 트랜스 지방산을 섭취한 군에서는 일반식이 섭취군과 비슷한 결과가 나타났다. 이는 많은 양의 트랜스 불포화지방산을 짧은 기간 동안 섭취하더라도 포화지방산을 섭취한 것과 유사한 효과를 나타낸다는 것을 의미한다.
