Aspergillus Associated with Meju, a Fermented Soybean Starting Material for Traditional Soy Sauce and Soybean Paste in Korea

  • Aspergillus is an important fungal genus used for the fermentation of Asian foods; this genus is referred to as koji mold in Japan and China. A. oryzae, A. sojae, and A. tamari are used in the production of miso and shoyu in Japan, but a comprehensive taxonomic study of Aspergillus isolated from Meju, a fermented soybean starting material for traditional soy sauce and soybean paste in Korea, has not been conducted. In this study, various Aspergillus species were isolated during a study of the mycobiota of Meju, and the aspergilli were identified based on phenotypic characteristics and sequencing of the ${\beta}$-tubulin gene. Most strains of Aspergillus were found to belong to the following sections: Aspergillus (n = 220), Flavi (n = 213), and Nigri (n = 54). The most commonly identified species were A. oryzae (n = 183), A. pseudoglaucus (Eurotium repens) (n = 81), A. chevalieri (E. chevalieri) (n = 62), A. montevidensis (E. amstelodami) (n = 34), A. niger (n = 21), A. tamari (n = 15), A. ruber (E. rubrum) (n = 15), A. proliferans (n = 14), and A. luchuensis (n = 14); 25 species were identified from 533 Aspergillus strains. Aspergillus strains were mainly found during the high temperature fermentation period in the later steps of Meju fermentation.

콩 종류와 대두 가공식품에 함유된 isoflavones의 정량 (High Performance Liquid Chromatographic Analysis of Isoflavones in Soybean Foods)

  • 콩에 함유되어 있는 이소플라본은 골다공증 예방 및 치료제로써의 활발한 연구가 진행중이다. 콩은 우리의 식생활과 깊은 관련성이 있으므로 이소플라본의 함량을 HPLC로 정량분석 하였다. 한국산 황기와 콩제품인 콩나물, 두부, 콩기름, 메주, 된장, 간장, 대두, 약콩, 완두콩 및 강남콩 등에서 함유된 isoflavone을 5mM $NaH_2PO_4$ 완충용액(pH 4.6)과 메탄올을 혼합한 이동상 (4 : 6) UV 260 nm에서 분석하였다(Table 2). 콩식품에 함유된 isoflavone중 유리 daidzein 양은 콩나물에서 $43.49{\pm}3.41\;mg/kg$, 된장에서 $233.31{\pm}5.81\;mg/kg$, 약콩은 $35.66{\pm}4.70\;mg/kg$이 검출되어 대두 $(14.52{\pm}0.58\;mg/kg)$ 비해 높았다. (P<0.05)간장 및 콩 기름은 함량이 낮았고, 완두콩 및 강남콩의 함량은 측정치 이하였다. 유리 genistein은 콩나물이 $27.631{\pm}1.66\;mg/kg$,메주가 $18.75{\pm}1.33mg/kg$, 된장이 $291.52{\pm}6.81\;mg/kg$ 검출되었고, 대두는 $22.49{\pm}0.93mg/kg$으로 약콩$(61.10{\pm}7.73mg/kg)$ 보다 낮았다(P<0.01). 그러나 formononetin의 함량은 daidzein, genistein과는 다른 양상을 보였으며, 특히 황기는 daidzein과 genistein은 검출되지 않았으나 formononetin의 함량이 대두$(60.62{\pm}11.35\;{\mu}g/kg)$ 비해 $9629.73{\pm}0.57\;{\mu}g/kg$으로 약 160배 이상 많았고, 약콩은 대두와 유사하였으나, 콩기름, 완두콩, 강남콩에서는 formononetin이 검출되지 않았으며, 전통식품인 된장과 메주에도 상당량 함유되어 있었다.

Inhibition of Bacillus cereus Growth and Toxin Production by Bacillus amyloliquefaciens RD7-7 in Fermented Soybean Products

  • Bacillus cereus is a gram-positive, rod-shaped, spore-forming bacterium that has been isolated from contaminated fermented soybean food products and from the environment. B. cereus produces diarrheal and emetic toxins and has caused many outbreaks of foodborne diseases. In this study, we investigated whether B. amyloliquefaciens RD7-7, isolated from rice doenjang (Korean fermented soybean paste), a traditional Korean fermented soybean food, shows antimicrobial activity against B. cereus and regulates its toxin gene expression. B. amyloliquefaciens RD7-7 exhibited strong antibacterial activity against B. cereus and inhibited the expression of B. cereus toxin-related genes (groEL, nheA, nheC, and entFM). We also found that addition of water extracts of soybean and buckwheat soksungjang (Korean fermented soybean paste made in a short time) fermented with B. amyloliquefaciens RD7-7 significantly reduced the growth and toxin expression of B. cereus. These results indicate that B. amyloliquefaciens RD7-7 could be used to control B. cereus growth and toxin production in the fermented soybean food industry. Our findings also provide a basis for the development of candidate biological control agents against B. cereus to improve the safety of fermented soybean food products.

Bacillus licheniformis 와 Saccharomyces rouxii 첨가에 의한 된장의 풍미향상 (Flavour Improvement of Soybean Pastes by the Addition of Bacillus licheniformis and Saccharomyces rouxii)

  • 한국인의 기호에 맞는 상업적 된장의 풍미향상을 위하여 된장제조시 Bacillus licheniformis와 Saccharomyces rouxii를 첨가하여 $27^{\circ}C{\pm}\;1$에서 40일간 발효시킨 결과 B. licheniformis 와 S. rouxii의 첨가로 환원당의 증량이 억제되었고, S. rouxii는 에틸 알콜의 생산을 증가시키고 암모니아태 질소의 양을 감소시켰다. B. licheniformis는 아미노태 질소와 적정산도를 증가시켰다. A. oryzae의 생육은 숙성중 감소되었고 B. licheniformis와 S. rouxii의 첨가로 더욱 촉진되었다. B. licheniformis와 S. rouxii를 혼용한 시료에 유리아미노산 함량이 가장 많았으며 각시료 모두 휘발성 성분으로 아세트알데히드, 아세톤, 에틸 알콜, 에틸 아세테이트, 메틸 알콜, 프로필 알콜, 이소부틸 알콜, 이소아밀 알콜 등이 검출되었으며 재래식 된장에서는 이소부틸 알콜, 이소아밀 알콜 등이 검출되지 않았다. 시료의 전체적인 풍미는 B. licheniformis 와 S. rouxii를 혼용한 시료가 가장 우수하였다.

제주 전통된장의 품질 특성 (Quality Characteristics of Jeju Traditional Doenjang)

  • 제주도내 된장 가공업체에서 제조된 전통된장의 수분, 조단백질, 조지방, 조회분의 함량은 각각 37.33~47.43%, 11.06~14.28%, 0.08~0.53%, 12.75~15.97%로 나타났다. 된장제품의 pH는 4.82~5.12, 산가는 17.84~27.55 mg/g, 염도는 10.66~13.91%, 아미노산성 질소함량은 301.06~573.89 mg%을 보였다. 제주 전통된장은 유기산 중에서 lactic acid의 함량이 가장 높았다. 총당 함량은 4.21~24.19 mg/g으로 전화당인 fructose와 glucose 함량은 0.68~6.21 mg/g와 0.81~10.44 mg/g의 범위를 나타내었다. 총 지방산중에 필수지방산인 linoleic acid의 비율이 61.54~66.79%로 높았다. 된장의 구성아미노산은 모두 17종이 검출되었으며, 구성 아미노산의 총 함량은 시료간에 13.89~16.46%의 범위를 보였다. 제주 전통된장의 isoflavone의 함량은 274.87~451.49 mg/kg으로 시료간에 다양한 범위를 나타내었다. 이와 같은 결과는 된장의 제조에 사용된 콩의 품종, 발효기간, 발효미생물, 제조방법 등에 차이에 기인한 것으로 생각된다.

재래 및 개량메주와 납두의 배합이 된장 발효에 미치는 영향 (Effects of mixture of improved Meju, Korean traditional Meju and Natto on soybean paste fermentation)

  • 개량식과 재래식 된장의 기호와 향기의 단점을 개선하고 장점을 유지하기 위하여 재래식, 개량식 그리고 납두를 혼합 제조한 된장의 이화학적 변화와 관능검사에 의한 적정배합 비율과 숙성정도를 검토하였다. pH는 개량식 메주에 납두의 배합량이 높을수록 증가하였으며 재래식 메주의 배합에서는 감소하였다. 재래식 메주에서의 질소화합물 함량은 개량식에서 보다 3배 높았으며 숙성도 빨랐다. 갈색색소의 변화는 초기의 값에 따르며 재래식이 높았다. 개량식과 재래식 그리고 납두를 혼합한 구가 관능적 평가가 높았으며 그 비율은 개량식, 재량식 그리고 남두의 비가 48 : 48 : 5의 비율이었다.

  • The antimutagenicities of methanol extracts from traditional and commercial kochujang (Korean Red Pepper Soybean Paste) and their ingredients were evaluated in Salmonella/mammalian microsome assay system. The traditional kochujang showed higher antimutagenic effects than the commercial one against Ν-methyl-Ν'-nitro-Ν-nitrosoguanidine (MNNG) in the Salmonella typhimurium TA100, a base-pair substituted mutant strain.(omitted)

순창 재래식 된장의 암세포 성장억제 효과 (Growth Suppression Effect of Traditional Fermented Soybean Paste(Doenjang) on the Various Tumor Cells)

  • Methanol extract and its fraction of traditional fermented soybean paste(doenjang) from Soonchang area were studied for growth suppression on the various tumor cells and suppression components, by using HPLC and GC were analysed. Hexane fraction of methanol extract was indicated 79%, 76%, 67%, 66%, 78% of growth suppression on L1210, P338D1, HepG2, WiDr and SNU 1 tumor cells, respectively. Ethylacetate fraction of methanol extract also showed 81%, 75%, 75%, 76% and 82% of growth suppression on the same tumor cells, respectively. Peak 8 obtained from HPLC of ethylacetate fraction indicated 81%, 77%, 77%, 75% and 79% of growth suppression on the same tumor cells and identified as a genistein, by comparing with standard one by HPLC analysis. Hexane fraction of methanol extract contained oleic acid, linoleic acid and palmitic acid.

한국 전통 된장 및 콩 추출물의 KB 세포에 대한 증식 억제효과 (Inhibitory Effects of Doen-jang(Korean Fermented Soybean Paste) and Soybean Extracts on the Growth of KB Cells)

  • The present study was designed to investigate whether traditional Korean fermented soybean paste(doen-jang) and soybean extracts have inhibitory effects on the growth of KB cell, an oral epithelioid cancer cell. When KB cell ATCC CCL-17 was cultivated for 48 flours with the addition of 0.5% of the five types or doen-jang extract, the growth of KB cell was inhibited by all types of extract, and ethyl acetate extract showed the highest inhibitory effect. In case of soybean extract, all types of extract also showed KB cell inhibitory effects, however, generally less than those of doen-jang extract. When ethyl acetate extract of doen-jang was added in different concentrations and KB cell was cultivated for 24 hours and 45 hours, strong inhibitory effect began to appear from the concentration of 1.25 mg/ml. Although soybean extract showed such a tendency, its effect was lower than that of doen-jang extract. These results indicate that doen-jang extract has inhibitory effect against KB cell, and particularly ethyl acetate extract has the highest effect. The effect of doen-jang extract might be possibly enhanced by the fermentation of soybeans. It is assumed that doen-jang extract may be used to develop nontoxic medicines for preventing and treating oral diseases.

"증보산림경제"의 장류(醬類) 조리 가공에 관한 연구 (A Study on Manufacturing of Korean Sauce Described in "Jeungbosallimgyeongje")

  • The “Jeungbosallimgyeongje” was literature reviewed about the manufacture of Korean sauces. Furthermore, in order to investigate the changes made by time period, other literatures, “Eumsigdimibang(1670s)”, “Sallimgyeongje(1715)”, “Gyuhapchongseo(1815)” and “Choson­mussangyorijebeop(1930)”, were compared. The ingredients mentioned included soy beans, flour, barley, elm trees, red beans and blue beans, etc. In addition, the shapes and sizes of dried soybean paste brick were varied. “Manchojang”, which designated the kind of hot pepper paste, appeared in this book for the first time. During its manufacturing process, it was characteristic to add dried bean paste, sea kelp and fish to produce a novel and higher quality product. From the above mentioned books, we found out that Koreans used only the soybeans and Chinese a mixture of buckwheat, flour and barley in addition to soybeans to make their traditional sauces. According to the“ Gyuhapchongseo” , there was a slight difference in ingredients to add for the manufacture of fish sauce, but the manufacturing methods and the one year period needed for maturing the ingredients were the same.. However, in the “Chosonmussangsinsikyrijebop”, fish sauce and meat sauce were classified separately and their manufacturing methods were different as well. In conclusion, the ingredients of used for the sauces recorded in “Jeungbosallimgyeongje” were various and at first hot pepper sauce made from “Manchojang” appeared and additionally red peppers were added to five kinds of Korean paste and red pepper powder were added to two kinds of Korean paste. The manufacturing method of the sauces changed according to time period, for example, only soybean has been used in Korean traditional sauces and other ingredients used as for Chinese ones eventually disappeared.
