Cantharidin Overcomes Imatinib Resistance by Depleting BCR-ABL in Chronic Myeloid Leukemia

  • Cantharidin (CTD) is an active compound isolated from the traditional Chinese medicine blister beetle and displayed anticancer properties against various types of cancer cells. However, little is known about its effect on human chronic myeloid leukemia (CML) cells, including imatinib-resistant CML cells. The objective of this study was to investigate whether CTD could overcome imatinib resistance in imatinib-resistant CML cells and to explore the possible underlying mechanisms associated with the effect. Our results showed that CTD strongly inhibited the growth of both imatinib-sensitive and imatinib-resistant CML cells. CTD induced cell cycle arrest at mitotic phase and triggered DNA damage in CML cells. The ATM/ATR inhibitor CGK733 abrogated CTD-induced mitotic arrest but promoted the cytotoxic effects of CTD. In addition, we demonstrated that CTD downregulated the expression of the BCR-ABL protein and suppressed its downstream signal transduction. Real-time quantitative PCR revealed that CTD inhibited BCR-ABL at transcriptional level. Knockdown of BCR-ABL increased the cell-killing effects of CTD in K562 cells. These findings indicated that CTD overcomes imatinib resistance through depletion of BCR-ABL. Taken together, CTD is an important new candidate agent for CML therapy.

전통문양의 활용에 관한 기존연구 분석 - 의류학 분야를 중심으로 - (Analysis of Previous Study of Traditional Pattern - Focused on Textile & Clothing related Part -)

  • This study analyzed the "research trends and the design processes" of the traditional patterns by examining the previous researches on the application of the traditional patterns. For this purpose, 96 studies which include master's and doctoral theses and research papers published in 8 Korean academic journals on Clothing from 2000 to 2010 were chosen. The findings of this study are as follows: First, when the previous researches on the application of the traditional patterns were classified according to their subject matter, they were classified into: research on clothing design, research on textile design, research on the design of cultural commodities, and research on mixed design. Second, the researches that were classified according to their subject matter were re-classified according to their year of publication. It was shown that the most active research was on the clothing design. The research on textile design, when compared with clothing design, alternated an increase and an decrease in every other year. Also, research on the design of cultural commodities were fewer than the research on clothing design or textile design although it is on the increase from 2008. Third, when classified according to the types of design, it was found that the designs based on a flora were most common. Fourth, in most cases, in the process of the designs, the designswere dispensed with the original form or were simplified. The choice of the tone of color, in most cases, were based on the concept selected by the designer. The most common method for the expression of the designs was based on the printing. Also, when it came to the presentation of the designs, the original work greatly outnumbered an imaginary work. Based on this study, it is hoped that there will be more active research on the application of the traditional designs which, in turn, can popularize the aesthetic beauty of Korea.

서양음악전공 작곡가에 의해 작곡된 창작국악작품 연구 (A Study on Changjak Gukak Composed by the Composers who Majored in Western Music)

  • 이 연구는 서양음악 작곡가에 의한 창작국악작품을 분류 검토하여, 서양음악전공자들의 창작국악 작곡활동에 도움을 주고자 하는 목적으로 행해진 연구이다. 연구 결과, 기본적으로 서양음악 작곡가에 의한 창작국악은 서양의 전통적 작곡기법 또는 현대적 작곡기법의 바탕 위에 한국전통음악적인 요소가 활용되고 있음을 알 수 있다. 동기 주제 발전, 조성, 호모포니, 대위법적 전개, 푸가 형식, 소나타 형식, 바로크합주곡 형식, 서양예술작품의 선율 차용 등의 서양전통작곡기법이 사용되었고, 음색 선율, 음향 효과, 악기의 특수주법, 무조성, 변박과 폴리리듬, 불확정성 기법 등의 서양 현대음악 작곡기법이 사용되었다. 이와 함께 국악기 고유의 시김새, 국악장단, 국악의 음계와 선법, 국악의 형식, 국악 선율 등을 적용하여 한국적인 표현과 정체성을 반영하고 있다.

Circ-SNX27 sponging miR-375/RPN1 axis contributes to hepatocellular carcinoma progression

  • Hepatocellular carcinoma (HCC) is a prevalent malignant tumor with high fatality. It has yet to be reported whether circ-SNX27 can affect the progression of HCC. This study attempted to analyze circ-SNX27's precise role and underlying mechanisms in HCC. HCC cell lines and tumor specimens from HCC patients were analyzed using quantitative real-time PCR and Western blotting to quantify the expressions of circ-SNX27, miR-375, and ribophorin I (RPN1). Cell invasion and cell counting kit 8 experiments were conducted for the evaluation of HCC cell invasion and proliferation. Caspase-3 Activity Assay Kit was utilized to gauge the caspase-3 activity. Luciferase reporter and RNA immunoprecipitation assays were executed to ascertain the relationships among miR-375, circ-SNX27, and RPN1. To determine how circ-SNX27 knockdown affects the growth of HCC xenografts in vivo, tumor-bearing mouse models were constructed. Elevated expressions of circ-SNX27 and RPN1 as well as a reduced miR-375 expression were observed among HCC cells and HCC patient tumor specimens. Knocking-down circ-SNX27 in HCC cells abated their proliferative and invasive abilities but raised their caspase-3 activity. Moreover, the poor levels of circ-SNX27 inhibited HCC tumor growth among the mice. Circ-SNX27 enhanced RPN1 by competitively binding with miR-375. Silencing miR-375 in HCC cells promoted their malignant phenotypes. Nonetheless, the promotive effect of miR375 silencing was reversible via the knockdown of circ-SNX27 or RPN1. This research demonstrated that circ-SNX27 accelerated the progression of HCC by modulating the miR-375/RPN1 axis. This is indicative of circ-SNX27's potential as a target for the treatment of HCC.

  • In this study, the fashion trend after the 1990s and the characteristics of fashion in the postmodernistic viewpoint that mainly effected fashion were observed, and the characteristics of fashion illustration expression was observed in the viewpoint of its link. It can be seen that mutually close characteristics are mutually linked. As the result of this study, the characteristics of fashion illustration can be categorized into three areas. and the sub-concepts are as following. The first category is the pursuit of feminine beauty of individual multi-dimension. This includes- 1) retro feminine beauty; 2) fetishistic and ecstatic feminine beauty; 3) mixed composition genderless (mixed composition style. ageless style and raceless style): and 4) beauty of body deconstruction and reconstruction (deformation due to partial transformation, change into animalization, mechanization and spirituelization. and beautification of crudeness). The second is the intentional deviation of expression and production technique. This includes-1) deviation from traditional perspective; 2) deviation from composition concept: and 3) breaking away from the existing material and expression technique of material. The third is the pursuit of authority-rejection form of expression. This is an area that should be noticed in the fashion illustration expression of the 1990s, as it shows the most diverse phenomenon. The sub-concepts of this category include- 1) separation of art 2) minimalism (minimalism in structure and production method); 3) flatness; 4) completion of incompletion; 5) accidentalism; 6) sportive;7) absence of figure; 8) intentional childishness 9) story development; 10) techno mechanism and cyber character; and 11) animation style.

건폐탕(健肺陽)이 호흡기 뮤신의 생성 및 유전자 발현에 미치는 영향 (Effect of Geonpye-tang(GPT) on Production and Gene Expression of Respiratory Mucin)

  • Objectives : In this study, the author tried to investigate whether Geonpye-tang(GPT) significantly affects PMA-, EGF- or TNF-alpha-induced MUC5AC mucin production and gene expression from human airway epithelial cells. Materials and Methods : Effects of the agent on PMA-, EGF- or TNF-alpha-induced MUC5AC mucin production and gene expression from human airway epithelial cells (NCI-H292) were investigated. Confluent NCI-H292 cells were pretreated for 30 min in the presence of GPT and treated with PMA (10ng/ml) or EGF (25ng/ml) or TNF-alpha (0.2nM), to assess both effect of the agent on PMA- or EGF- or TNF-alpha-induced MUC5AC mucin production by enzyme-linked immunosorbent assay (ELISA) and gene expression by reverse transcription-polymerase chain reaction (RT-PCR). Possible cytotoxicity of the agent was assessed by examining the rate of survival and proliferation of NCI-H292 cells after treatment with the agent over 72 hrs (SRB assay). Results : (1) GPT significantly inhibited PMA-induced and EGF-induced MUC5AC mucin production from NCI-H292 cells. However, GPT did not affect TNF-alpha-induced MUC5AC mucin production. (2) GPT significantly inhibited the expression levels of PMA-, EGF- or TNF-alpha-induced MUC5AC genes in NCI-H292 cells (3) GPT did not show significant cytotoxicity to NCI-H292 cells. Conclusion : This result suggests that GPT can affect the production and gene expression of respiratory mucin observed in diverse respiratory diseases accompanied by mucus hypersecretion. This can explain the traditional use of GPT in oriental medicine. Effects of GPT with their components should be further investigated using animal experimental models that reflect pathophysiology of airway diseases through future studies.

  • 현대사회는 산업의 다양화로 디자인 영역은 다각화로 확장되어 가고 있으며 여러 학문들과 통합과 통섭 등 을 통하여 여러 가지 새로운 기능성을 제시하고 있다. 사람들의 사고방식, 감성의 변화, 그리고 행동양식에도 큰 변화가 일어나 새로운 표현방식과 미적 가치를 추구하다. 그러함 속에서 전통에 대한 새로운 인식 혹은 새로운 소재로서 수묵화 표현이 늘어나고 있다. 본 고 에서는 수묵이 나타나게 된 역사적, 사회적 배경을 기초하여 그 표현방식과 분석을 중심으로 고찰하였고 동양적 전통미와 정서적 감성에 부합되는 미적 가치로서 수묵화적 표현을 제시하였다. TV-CM디자인에 나타난 수묵표현의 활용에 대해 연구하는 것으로 수묵의 개념, 특징, 분류, 표현방식을 분석하고 새로운 수묵기법 혹은 동양적 표현을 통하여 디자인 영역을 확대하고자 한다.

가미진해탕(加味鎭咳湯)과 소청룡탕가석고(小靑龍湯加石膏)가 PMA로 유발된 기도뮤신의 생성 및 MUC5AC gene 발현에 미치는 영향 (Effects of Gamijinhae-tang and Socheongryong-tang-ga-seokgo on PMA- induced Production of Airway Mucin and Expression of Airway MUC5AC Gene)

Materials and Methods : Confluent NCI-H292 cells were pretreated for 30 min in the presence of JHT and STS and treated with PMA (10ng/ml), to assess the effects of the agents on PMA-induced mucin production by enzyme-linked immunosorbent assay (ELISA). Also, the effects of the agents on PMA-induced MUC5AC gene expression from the same cells were investigated. Possible cytotoxicities of the agent were assessed by examining the rate of survival and proliferation of NCI-H292 cells after treatment of agents during 48 hrs. Results : (1) JHT and STS did not show significant cytotoxicity to NCI-H292 cells. (2) JHT significantly decreased PMA-induced mucin production from NCI-H292 cells. However. STS did not affect mucin production. (3) JHT significantly inhibit the expression levels of PMA-induced MUC5AC gene in NCI-H292 cells. STS slightly decreased the expression levels of PMA-induced MUC5AC gene. Conclusion : These results suggest that JHT can not only affect the production of mucin but also affect the expression of the mucin gene, and this explains the traditional use of JHT in oriental medicine. The effects of JHT and STS with their components should be further investigated using animal experimental models that simulate pathophysiology of airway diseases through future studies.

패션 인플루언서의 체형이 자기표현 및 자기제시의도, 인플루언서 추천의도에 미치는 영향 - 친근감의 매개 역할을 중심으로 - (The Effects of Fashion Influencers' Body Types on Self-Expression, Self-Representation Intentions, and Recommendation Intentions - Focusing on the Mediating Effect of Familiarity -)

  • This study examines the effects of fashion influencers' body types (realistic versus ideal body types) on self-expression, self-representation, and recommendation intentions, as mediated by familiarity toward influencers. Although fashion influencers lead to a positive consumer response compared to traditional advertisements, previous research on the effects of fashion influencers on consumers is limited. Thus, this study tests the role of consumers' socio-psychological aspects in understanding how and why fashion influencers affect consumers' behavioral intentions associated with self-expression, self-representation, and influencer recommendation. A total of 180 women in their 20s and 30s participated in the survey. The responses were collected after showing them stimuli featuring fashion influencers with either ideal or realistic body shapes. The data were analyzed using SPSS18.0 for descriptive statistics, and AMOS 18.0 for confirmatory factor analysis and structural equation modeling. The results showed that participants who were shown realistic body types perceived familiarity, which generated positive effects on self-expression, self-representation, and recommendation intentions. Hence, the effects of influencers' body types on recommendation intention are mediated by familiarity. Self-expression and self-representation intentions also increase influencer recommendation intention. Comparatively, participants who were shown ideal body types only induced higher self-representation intention, which increased their recommendation intention. The current findings can help fashion marketers select the appropriate influencers who fit their target customers as promotional models, as well as to induce changes in consumers' behavioral intention.

In vitro Study of the Antagonistic Effect of Low-dose Liquiritigenin on Gemcitabine-induced Capillary Leak Syndrome in Pancreatic Adenocarcinoma via Inhibiting ROS-Mediated Signalling Pathways

  • Background: To investigate in-vitro antagonistic effect of low-dose liquiritigenin on gemcitabine-induced capillary leak syndrome (CLS) in pancreatic adenocarcinoma via inhibiting reactive oxygen species (ROS)-mediated signalling pathways. Materials and Methods: Human pancreatic adenocarcinoma Panc-1 cells and human umbilical vein endothelial cells (HUVECs) were pre-treated using low-dose liquiritigenin for 24 h, then added into gemcitabine and incubated for 48 h. Cell viability, apoptosis rate and ROS levels of Panc-1 cells and HUVECs were respectively detected through methylthiazolyldiphenyl-tetrazoliumbromide (MTT) and flow cytometry. For HUVECs, transendothelial electrical resistance (TEER) and transcellular and paracellular leak were measured using transwell assays, then poly (ADP-ribose) polymerase 1 (PARP-1) and metal matrix proteinase-9 (MMP9) activity were assayed via kits, mRNA expressions of p53 and Rac-1 were determined through quantitative polymerase chain reaction (qPCR); The expressions of intercellular adhesion molecule 1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1) and PARP-1 were measured via western blotting. Results: Low-dose liquiritigenin exerted no effect on gemcitabine-induced changes of cell viability, apoptosis rate and ROS levels in Panc-1 cells, but for HUVECs, liquiritigenin ($3{\mu}M$) could remarkably elevate gemcitabine-induced decrease of cell viability, transepithelial electrical resistance (TEER), pro-MMP9 level and expression of ICAM-1 and VCAM-1 (p<0.01). Meanwhile, it could also significantly decrease gemcitabine-induced increase of transcellular and paracellular leak, ROS level, PARP-1 activity, Act-MMP9 level, mRNA expressions of p53 and Rac-1, expression of PARP-1 and apoptosis rate (p<0.01). Conclusions: Low-dose liquiritigenin exerts an antagonistic effect on gemcitabine-induced leak across HUVECs via inhibiting ROS-mediated signalling pathways, but without affecting gemcitabine-induced Panc-1 cell apoptosis. Therefore, low-dose liquiritigenin might be beneficial to prevent the occurrence of gemcitabine-induced CLS in pancreatic adenocarcinoma.
