토사자(兎絲子)가 난소적출(卵巢摘出)로 유도(誘導)된 흰쥐의 골다공증(骨多孔症)에 미치는 영향(影響) (Effects of Cuscutae Semen Extract on Prevention of Osteoporosis in Ovariectomized Rats)

  • Objectives: Osteoporosis, which occurs after menopause, is a kind of metabolic bone disorder. It develops when the bone mass begins to decrease radically, and its main symptoms are bone fracture and height-shortening. This thesis aims at what effects the Cuscutae Semen Extract(CSE) has on the prevention of osteoporosis in SD-rat that is caused by ovariectomy. Methods: The 24 female white rats, after removing their ovaries, were divided into the Normals, the Control group, and the CSE administrated group. For the next 8 weeks, distilled water to the normals and the control, and the CSE(45.9 mg/100 g) to the CSE administrated group were given in the mouths of them. After 8 weeks the rats were sacrificed. Weight, albumin, aspartate aminotransferase, alanine aminotransferase, total-cholesterol, triglyceride, phosphorus, calcium, tetraiodothyronine, estradiol, the weight of the femur, the amount of tibia ash, the area of trabecular bone, and the thickness of trabecular bone were measured. Results: The serum analysis shows that the calcium and phosphorous of the CSE administrated group significantly increased compared to that of the control group. Aspartate aminotransferase, alkalinephosphatase, total cholesterol, tetraiodothyronine of CSE group were decreased, but not so significant. Albumin, alanine aminotransferase, triglyceride and estradiol of the group showed the tendency to increase, but the significance wasn't admitted. Regarding the variation of bone, the femur weight and the ash content of tibia, CSE group was more increased than the control group but the significance wasn't admitted. The histological observation shows that the trabecular thickness was more increased than the control group, and trabecular area increased significantly compared to those of the control group. The number of osteoclast and osteoblast area of the CSE groups decreased significantly compared to those of the control group. Conclusions: From the result of the above study, CSE should be effective for the osteoporosis cure and precaution and deeper study through bedside and clinical demonstration is much needed from now on.

실험적 골 병소에 대한 콘빔형전산화단층영상과 초음파영상의 비교 (Comparison of cone-beam computed tomography and ultrasonography on experimental bone lesion)

  • Purpose : This study was performed to evaluate the diagnostic ability of ultrasonography in detection of bone defects and new bone formation. Materials and Methods : Experimental bony defects were prepared on the parietal bone samples acquired from 3.5 kg New Zealand male rabbits. The defects were evaluated using ultrasonography and CBCT, and examined histologically at interval of 1, 3, 6, and 8 weeks. Results : Ultrasonograph demonstrated hyperechogenicity in the defect area at 3 weeks and broadened hyperechogenicity from the margin of bone defect at 6 and 8 weeks due to new bone formation. On the CBCT images, new bone formation was first observed at 3 weeks around the margin of the defect, and showed gradually increase at 6 and 8 weeks. Histologic findings revealed existence of the fibroblasts and fibrous connective tissue with abundant capillary vessels only at 1 week, but osteoid tissue and newly formed trabecular bone at 3 weeks. Bone remodeling in the defect area was observed at 6 weeks and increased calcification and dense trabecular bone formation was observed at 8 weeks. Conclusions : Ultrasonograph proved to be a very useful diagnostic tool in detecting the bony defect and new bone formation. Additionally, ultrasonography provided valuable information regarding the blood supply around the defect area.

두충[Eucommia ulmoides Oliver] 조성물이 골다공증에 미치는 효과 (Effects of the Dietary Mixture of Eucommia ulmoides Oliver on Osteoporosis)

  • 두충의 골다공증 억제 효과를 연구하였다. 난소절제된 흰쥐에 기본사료, 칼슘, 비타민 D$_3$, 비타민 $K_1$를 함유하는 조성물과 두충추출물을 추가한 조성물을 7개월간 투여한 후 기본사료만 투여한 대조군과 비교하였다. 골 흡수 지표로 deoxypyridinoline을 분석한 결과 칼슘 비타민 조성물 투여군의 경우 대조군과 비교하여 9.0%, 두충 추가 조성물 투여군의 경우 26.9% 감소하였다. 경골의 trabecular 골면적을 측정한 결과 칼슘 비타민 조성물 투여군의 경우 대조군과 비교하여 9.9%, 두충 추가 조성물 투여군의 경우 42.0% 증가하여 골다공증 억제 효과를 확인할 수 있었다.

실험적 골조직 병소부에 관한 방사선학적 연구 (A RADIOGRAPHIC STUDY ON EXPERIMENTAL BONY LESIONS)

  • The author studied the effect of periapical bone loss on radiographic interpretation by using a dry adult human mandible. Artificial bony lesions were created at the apices of two mandibular second molars and four mandibular bicuspids. The jaw was stabilized and various artificial lesions were radiographed under ideal circumstances. Radiographic pictures were recorded and compared with the size of the actual lesions. The obtained results were as follows: 1. The size of actual lesions were always larger than their radiographic pictures. 2. The size of actual lesion had a greater discrepancy in the molar area where the cortical plates were havier, but the lesions in the bicuspid areas were only slightely larger than their radiographic pictures, and in these areas the cortical plates were quite thin. 3 Periapical lesions located in cancellous bone did not appear on a radiograph, regardless of the size of the created lesion, but when the junctional bone was involved or the cortical plate was perforated, the definite bone destructive change was observed on the roentenogram. 4. Removal of the entire buccal or lingual plate did not affect the trabecular pattern of bone on the roentgenogram. 5. When the cancellous bone and junctional bone were removed simultaneously, altered trabecular patterns were observed on the roentgenogram.

Antiosteoporotic activity of ‘Dae-Bo-Won-Chun’ in the ovariectomized rats

  • The preventive effect of herbal formulation, ‘Dae-Bo-Won-Chun’(DBWC), on the progress of bone loss induced by ovariectomy (OVX) was studied in rats. From light microseopic analyses, a porous or erosive appearances were observed on the surface of trabecular bone of tibia in ovariectomized rats. Whereas those of the same bone in sham rats were composed of fine particles. The trabecular bone area and trabecular thickness in ovariectomized rats decreased by 50% from those in sham-operated rats, these decreases were completely inhibited by administration of DBWC at concentration of 10mg/kg per day for 7 weeks. The mechanical strength in femur neck was decreased by ovariectomy, and this was significantly suppressed by the administration of DBWC. Serum phosphorus, alkaline phosphatase, and thyroxine levels in ovariectomized rats were increased compared to those in sham-operated rats, and increases were completely inhibited by the administration of DBWC. These results strongly suggest that DBWC is effective in preventing the development of bone loss induced by ovariectomy in rats.

속단(續斷)이 난소적출 흰쥐의 골조직 대사에 미치는 영향 (Effects of Phlomis Umbrosa on Prevention of Osteoporosis in Ovariectomized Rats)

This thesis aims at what effects the Phlomis umbrosa will have for the prevention of the osteoporosis in the SD-rat that is caused by the ovariectomy. The 24 female white rats, after their ovaries are removed, were divided into the Normals, the Control group, Phlomis umbrosa Administrated group. After then, for 8 weeks, D.W to the normals and the control, and the Phlomis umbrosa(46 mg/100 g) to the Phlomis umbrosa group were given in the mouths of them. After 8 weeks the rats were sacrificed. And the weight, albumin, AST, ALT, total-cholesterol, triglyceride, P, Ca, T4, estradiol, the weight of the femur, the amount of tibia ash, the area of trabecular bone, the thickness of trabecular bone were measured. The serum analysis shows that the total cholesterol of the Phlomis umbrosa Administrated group reduced remarkably, compared to that of the control group. AST, ALT, ALP, T4 of Phlomis umbrosa group were decreased, but not in distinct measure. Estradiol of Phlomis umbrosa group increased substantially compared to that of the control group, but albumin, triglyceride, Phosphorous, calcium of the group showed the tendency to increase, but not so remarkably. The histological observation shows that the area and the thickness of (trabecular bone), the number of osteoclast of the Phlomis umbrosa groups increased significantly compared to those of the control group. Conclusions: The above experiment shows that Phlomis umbrosa has the prevention effect by supplying estradiol.

홍화자·녹용·자하거 약침이 골다공증에 미치는 영향 (A Study on the effects of the Carthami semen·Cervi pomtotrichum cornu·Hominis placenta aquacupuncture on the osteoporosis in the rats)

  • Objectives : This study was designed to evaluate on the effects of the Carthami semen Cervi pomtotrichum cornu Hominis placenta aquacupuncture on the osteoporosis in the rats Materials and Methods : In order to induce estrogen deficient osteoporosis, ovariectomy was done on rats. Then aquacupunctures were injected into the pai shu points corresponding to urinary bladder meridian of human body in experimental groups. And such indexes were measured as the change of body weight, bone mineral density, histological change of tibia, and levels of osteocalcin, bone alkaline phosphatase, calcium, phosphorus in serum. Results : 1. Significant changes of trabecular area % in epiphysis of tibia were shown between control group and experimental group II and between experimental group I and group II. 2. Significant changes of trabecular area % in diaphysis of tibia were shown between control and experimental groups. 3. Significant changes of serum osteocalcin were shown between control group and experimenta group I . 4. Significant changes of levels of serum bone alkaline phosphatase were shown between control group and experimental group I and II. Conclusions : The Carthami semen Cervi pomototrichum cornu Hominis placenta aquacupuncture is effective to estrogen deficient osteoporosis, ovariectomy was done on rats.

육미지황환(六味地黃丸) 에탄올 추출물이 난소제거 흰쥐의 경골 소주골에 미치는 영향 (Effect of Ethanol Extract of Yukmijiwhang-Whan on Trabecular Bone Area in OVX Rats)

  • Bone is continuously remodeled during adult life with the resorption of old bone by osteoclasts and its subsequent replacement by osteoblast. Bone homeostasis is maintained by a balance between activities of osteoblasts and osteoclasts, but an imbalance between resorption and formation results in bone diseases including osteoporosis. Osteoblasts line up on the bone surface, especially regions of new bone formation, lay down bone matrix (osteoid) in orderly lamellae and induce its mineralization. Thus, the increased activity of osteoblasts is helpful to treat and prevent osteoporosis. In this study, we examined whether 80% EtOH extract of yukmijiwhang-whan is capable of affecting osteoblast proliferation using human osteoblast-like cell line, MG-63 and Saos-2. In an in vivo experiment, extract of yukmijiwhang-whan was administered for 9 weeks to ovariectomized (OVX) rats. At necropsy, uterus weights were measured, and trabecular bone areas (TBAS) of tibia and the sixth lumbar vertebra were measured by bone histomorphology. The maximum cell proliferation of MG-63 caused by extract of yukmijiwhang-whan at $5\;{\times}\;10^{-6}\;mg/ml$ was approximately 115% compared with control. In Saos-2, cell proliferation was approximately 145% of control at $5\;{\times}\;10^{-4}\;mg/ml$ and maximum alkaline phosphatase (ALP) activity was approximately 143% of control at $5\;{\times}\;10^{-5}\;mg/ml$. In animal study, however, the tibia and lumbar TBAS of the yukmijiwhang-whan group did not increased than the OVX control group. In conclusion, the 80% EtOH extract of yukmijiwhang-whan increased proliferation of osteoblasts but did not prevent bone loss in OVX rats.

디지털방사선영상에서 추출한 해면질골의 강도와 미세구조의 형태계측학적 분석에 대한 연구 (A study of trabecular bone strength and morphometric analysis of bone microstructure from digital radiographic image)

  • Purpose : To evaluate the relationship between morphometric analysis of bone microstructure from digital radiographic image and trabecular bone strength. Materials and Methods : One hundred eleven bone specimens with 5 mm thickness were obtained from the mandibles of 5 pigs. Digital images of specimens were taken using a direct digital intraoral radiographic system. After selection of ROI (100 × 100 pixel) within the trabecular bone, mean gray level and standard deviation were obtained. Fractal dimension and the variants of morphometric analysis (trabecular area, periphery, length of skeletonized trabeculae, number of terminal point, number of branch point) were obtained from ROI. Punch sheer strength analysis was performed using Instron (model 4465, Instron Corp., USA). The loading force (loading speed 1 mm/min) was applied to ROI of bone specimen by a 2 mm diameter punch. Stress-deformation curve was obtained from the punch sheer strength analysis and maximum stress, yield stress, Young's modulus were measured. Results: Maximum stress had a negative linear correlation with mean gray level and fractal dimension significantly (p<0.05). Yield stress had a negative linear correlation with mean gray level, periphery, fractal dimension and the length of skeletonized trabeculae significantly (p < 0.05). Young's modulus had a negative linear correlation with mean gray level and fractal dimension significantly (p < 0.05). Conclusions : The strength of cancellous bone exhibited a significantly linear relationship between mean gray level, fractal dimension and morphometric analysis. The methods described above can be easily used to evaluate bone quality clinically.

Chronic Alcohol Consumption Induced Tibial Bone Loss and Resulted in Osteopenia in Growing Young Male Rats

  • To determine the deleterious effects of chronic alcohol consumption on bone especially in adolescents or young adults, 8 week-old Sprague Dawley male rats were fed with Lieber-Decarli ethanol liquid diet, containing $36\%$ of energy as ethanol, ad libitum (ethanol group) or isocaloric normal liquid diet (control group) for 7 weeks. Body weight was significantly lower in ethanol group than that in control group after 1 week of feeding to the end. liver weight and the ratio of liver or kidney weight to body weight in ethanol group were significantly increased when compared to those in control group. Ethanol group showed significantly lower serum protein and albumin levels (p<0.05), higher total cholesterol and HDL-cholesterol levels (p<0.05), and AST, ALT and BUN activities than control group, but serum triglyceride, Ca and phosphate levels were not different. Ethanol group had significantly lower tibial trabecular bone area and serum osteocalcin level than control group (p<0.05), but urinary Ca and NTx (cross-linked N-telopeptide of type I collagen) concentrations and serum testosterone and parathyroid hormone levels were not different. In conclusion, chronic alcohol consumption in growing young male rats may result in osteopenia through the reduction of bone formation as well as liver malfunction.
