The purpose of the study is to analyze the environmental characteristics of fish acute toxicity that is dependent on the harmfulness of ACQ (Alkaline Copper Quat)-Treated Wood and Oryzias latipes mortality in a comprehensive way, provide objective verification method on the eco-toxicity and environment-friendliness of landscaping materials and methods, and utilize it as a basic datum for evaluation criteria. The main results are summarized as follows : 1. As a result of analysis on the harmfulness characteristics, each experimental plot showed different values respectively. In particular, it has been found that in proportion to the volume of testing materials, COD and Cu increases at a constant rate, compared to the input water. In the plot C with three testing materials, COD increased 67 times more than that of the input water, and Cu increased up to 12.36mg/L. 2. In case of fish toxicity, plot C, B, A all showed a mortality rate of 100%, indicating that fish toxicity is strong. In particular, the mortality rate of each plot within the initial time of one and a half hour showed clearly, which suggests that the fish toxicity is influenced by the increased concentration of hazardous substances depending on the volume ratio of testing materials. 3. As a result of comparison and analysis on the harmfulness and fish toxicity, the harmfulness showed different values on each experimental plot but, we found that the changing rate of values of toxicity of COD and Cu is mutually similar to that of mortality in the initial hour according to the experiment of fish toxicity, which shows that COD and Cu are major factors to increase fish toxicity.
It has been known that Gentianae macrophyllae Radix is reputed to be effective in removal of 'Pungseub' and 'Heoyeol', and to be analgesic. Since ancient times, however, there have been many examples of substituting Lycoctoni Radix for Gentianae macrophyllae Radix. For that reason, this experiment was conducted to compare the diuretic, analgesic, anti-inflammatory, anti-pyretic, anti-bacterial effect, and toxicity of Lycoctoni Radix with those of Gentianae macrophyllae Radix. The results obtained in this work were summarized as follows; In the test of acute toxicity, Lycoctoni Radix was found to be toxic in comparison to Gentianae macrophyllae Radix which was found to be non-toxic. Both herbs were found to be diuretic and Gentianae macrophyllae Radix was more effective than the other and to be ineffective for the blood pressure and respiration. Lycoctoni Radix was found to be more analgesic than the other. Gentianae macrophyllae Radix was found to be more anti-inflammatory of the two and both demonstrated anti-pyretic effect. Anti-bacterial effect was detected only in Lycoctoni Radix. It is concluded that Gentianae macrophyllae Radix has mild diuretic, analgesic, anti-inflammatory and anti-pyretic effect, but Lycoctoni Radix, due to its toxicity, can not be substituted for Gentianae macrophyllae Radix.
The Japanese Good Laboratory Practice (GLP) Standard on Drugs was finalized as a guideline and implemented in April, 1983. This standard is intended to ensure the quality and integrity of the data from nonclinical toxicity studies submitted to the Ministry of Health and Welfare in support of applications for approval to manufacture or import new drugs or to be used in the reevaluation of previously approved drugs. The standard includes a guideline for organizational matters, personnel, facility, equipment, testing operation, documentation and conduct of studies. Principles and influences of implementation of Japanese GLP will be discussed briefly in comparison with foreign GLPs.
A growth inhibition assay using Chlorella sp. AG 10002 based on the OECD 201 standard test procedure was applied to the toxicity testing of endosulfan and its reported metabolites. Comparison of dry cell weight, optical density (OD) at 680 nm, and chlorophyll a concentration indicated that optical density at 680 nm of culture broth is convenient, rapid, and accurate method for cell growth. In this microalgae system, the $IC_{50}$ values of endosulfan, endosulfan sulfate, endosulfan lactone, and endosulfan ether were determined as 9.45, 18.8, 18.2 and 37.5 mg/L, respectively. In a while, endosulfan diol did not show a significant toxicity up to 50 mg/L. Since endosulfan is liable at acidic or alkaline conditions, treatment of endosulfan in pH 3, 4, and 11 for 3 days resulted in reduced toxicity, as expected. These results suggested that the microalgae system is useful to evaluate various toxic chemicals and provide a new notion for bioremediation of endosulfan in aqueous systems.
Proficiency testing by interlaboratory comparisons is used to determine the performance of individual laboratories. In order to verify the quality of acute toxicity testing with Daphnia magna, National Institute of Environmental Research in South Korea is regularly organizing interlaboratory comparisons to estimate the analytical accuracy of different laboratories. Total 58 laboratories located in South Korea took part in interlaboratory proficiency testing scheme with three proficiency testing samples. TU(Toxic Unit) values of each laboratory were determined and robust z-score was calculated in order to evaluate the proficiency levels. Based on the robust z-score classification, 74% of the participant laboratories showed a satisfactory performance (43 laboratories). The main reason of 'unsatisfactory' performance seemed to be considered that the unsuitable management of test organism incubation system and the lack of experience on the identification of the test organism condition by effect of toxicity.
Honey bees (Apis mellifera) forage in agricultural areas, and are exposed to diverse pesticide poisoning. Toxic effects on Apis mellifera of different groups of pesticides were tested in the laboratory; fungicide (Metconazole), herbicide (Glyphosate), acaricide (Amitraz), organophosphate insecticide(Fenitrothion) and neonicotinoid insecticides(Thiacloprid, Thiamethoxam, Imidacloprid, Acetamiprid, Dinotefuran and Clothianidin). Commercial formulations were serially diluted from the recommended concentration (RC) to 10-6 times to carry out feeding and contact tests. Toxicity was transformed into lethal dose (LD50) and hazard question (HQ). The acute toxicity of pesticides showed similar patterns between feeding and contact tests. But feeding tests showed greater toxic to honey bee than contact test. The organophosphate and nitro-neonicotinoid insecticides were highly toxic with HQ values ranging greater than 1. However, cyano-neonicotinoids of Thiacloprid and Acetamiprid showed low toxicity. Even at the RC, 24 hr mortalities were 18 and 30%. The acaricide (Amitraz) showed intermediate level of toxicity at RC but negligible at the concentration lower than 10-1 times. A fungicide(Metconazole) and herbicide(Glyphosate) showed minimal impacts. The results imply that the selective use of pesticides could help conservation of pollinators in agricultural production systems.
In this study, the potential toxicity of isoprocarb was demonstrated using zebrafish embryos. We treated isoprocarb (0, 29, and 58 mg/L) to the zebrafish embryos for 72 h then, we estimated morphological changes and apoptotic cell numbers. The increasing extent of apoptosis from the anterior to posterior region of developing zebrafish larvae was correlated with toxicity in the overall development process, including growth and normal organ formation. The appearance of abnormalities in the isoprocarb-treated groups in comparison to normal developing zebrafish larvae was verified using quantitative image analysis based on ImageJ software program. The vascular system comprising a complex interconnection of blood vessels was visualized in vessel-fluorescent transgenic zebrafish (fli1:eGFP). The main vasculature was malformed on isoprocarb treatment, and this was also related to cardiac defects. Taken together, normal embryonic development in zebrafish was interrupted owing to the acute toxicity of isoprocarb.
A method for rapid acute toxicity test based on temperature control of Ceriodaphnia dubia has been developed and evaluated. A new toxicity test based on temperature control (TTBTC) which are based on temperature control, was developed and compared for the adsorption of the better methodology for short-term toxicity screening. Initially, daphnid larval are exposed to toxicants and at the same time the temperature of the water bath containing media is increased to high temperature $(35.5^{\circ}C).$ After 1.25 hrs of contact time, the number of the daphnids, either living (no toxic effect) or dead (toxic effect), is counted by the naked eyes. Effect of exposure time on test sensitivity was not significantly different between 1 to 1.5 hr. Comparison of the rapid 1.25 hr acute toxicity test developed in this study and the standard 48 hr acute toxicity test using heavy metals, cyanide and pentachlorophenol indicated that the 1.25 hour test provides an acceptable level of sensitivity in toxicity test for C. dubia.
Fiftythree species(35 families and 52 genera)of Korean medicinal plants which have been frequently used in oriental herb prescriptions or have been used as folklorics were evaluated on their short term acute toxicity and potential antitumor activities against P-388 murine lymphocytic leukemia model in vivo. Among these plants Acorus gramineus (Araceae), Agrimonia pilosa (Rosaceae), Aralia elata (Araliaceae), Dryopteris crassirhizoma (Aspidiaceae), Syringa reticulata (Oleaceae) and Calystegia japonica (Convolvulaceae) exhibited potent acute toxicity resulting from severe weight loss and death. Agrimonia pilosa (Rosaceae) showed about 33% of increased life span in comparison with that of control group mouse, but others exhibited no significant antitumor activities.
