해양생태독성시험 표준생물로서 미세조류의 특성 비교 연구(Skeletonema sp., Dunaliella tertiolecta) (Comparative Study on the Characteristics of Microalgae as Standard Species for Marine Ecotoxicity Tests (Skeletonema sp., Dunaliella tertiolecta))

  • 해양생태독성시험의 국제표준시험종 중 생산자에 속하는 대표적인 Skeletonema sp.와 Dunaliella tertiolecta의 생태독성학적 차이점을 알아보기 위해 각 표준시험법(규격)을 비교하였고 환경에 대한 종 적합성과 다양한 시험물질에 대한 민감도를 비교 분석하였다. 그 결과, 시험법의 경우 대부분 동일하였으나 시험 유효성의 기준에서 pH 변화 제한과 초기접종밀도에서 차이가 나타났으며 이는 D. tertiolecta의 낮은 성장률에 기인된 것으로 추정된다. 적합성에서는 두 종 모두 규격에서 요구하는 유효성의 기준을 연속 만족하여 시험수행의 일관성을 보였고 시험한계 염분범위는 Skeletonema sp.와 D. tertiolecta 각각 20 및 10 psu로 나타났다. 마지막으로 민감도의 경우, 시험규격에서 제시하는 참조물질, 실제 오염 배출수(선박평형수) 및 기타 다양한 화학물질에서 모두 Skeletonema sp.가 D. tertiolecta에 비해 독성 민감도가 상대적으로 높음을 확인하였으며 이는 해양생태독성시험 수행에 있어 생산자를 이용한 시험의 경우 최소 2종 이상의 다른 분류군의 미세조류를 이용하는 것이 시험결과의 신뢰성과 객관성을 높일 수 있는 방법임을 시사한다.

재원환자의 질환군에 따른 가정간호 요구도 (Needs of Home Care Nursing Services for Hospital Patients)

  • Purpose: The purpose of this study was to investigate the needs of home care nursing services in relation to the patients in hospital. Method: Subjects were 129 patients who admitted hospital and were selected through convenience sampling. Data were collected using the Home Health Care Need Assessment Questionnaire constructed by the Korea Health Industry Development Institute. Data were analyzed using SAS 8.12 program by applying percentage. Results: 1. According to the diagnosis of the subjects, the majority had cancer (25.0%), followed by musculoskeletal disease (15,6%), neuro/cerebral vascular disease (14.1%), digestive (10.9%) and respiratory disease (10.9%). 2. With regard to fundamental nursing service, subjects wanted to receive home care services for the following reasons: Problem identification and diagnosis (77.5%), vital sign check (49.6%); and intake and output measure (20.9%). 3. With regard to clinical laboratory tests, 62.8% wanted to receive blood tests, and followed by urine tests 26.4%, and wound drainage 26.4%. 4. With regard to medication and treatment service, 40.3% of the subjects wanted to receive intravenous fluid therapy, 26.4% intravenous antibiotics, and 26.4% the monitoring of fluid therapy. 5. With regard to therapeutic nursing service, 33.3% wanted to receive wound care, 26.4% ROM exercise, and 27.9% foley catheter change and care. 6. With regard to educational needs, 42.6% wanted education on infection monitoring, 41.4% on medication, and 34.9% on diet. 7. With regard to counseling needs, 65.9% wanted to receive telephone counseling about patient condition, 52.7% counseling about re-admission and 51.9% direct counseling about patient condition. In the group of injury and toxicity, and cardiovascular/circulatory diseases, 100% wanted telephone and direct counseling about the patient condition. Conclusion: Therefore, in order to improve the quality of hospital based home health care services, various factors that affect to the need of home health care should be analyzed and specified nursing care should be looked into.

국내의 지역 및 계절에 따른 콘크리트 배합별 환경영향평가 (Environmental Impact Assessment of Different Concrete Mixture Proportions according to Domestic Region and Season)

  • 지역과 계절이 콘크리트의 전과정 환경영향에 미치는 효과를 정량적으로 평가하기위해 6331개의 레디믹스 콘크리트 배합을 분석하였다. 콘크리트의 환경영향은 국가 생애주기 데이터목록을 기반으로 산출한 환경부하 발생량을 분류화, 특성화, 정규화 및 가중치 단계를 거쳐 6가지 환경영향 범주(지구온난화, 자원고갈, 광화학산화물생성, 산성화, 부영향화, 인간독성)로 나타났다. 단위압축강도에서의 환경영향 지표를 평가하기 위해 콘크리트 압축강도로 무차원한 환경영향 지수로 정의하였다. 국내에서 가장 많이 사용되는 콘크리트의 압축강도($f_{ck}$)는 24 MPa와 27 MPa이다. $f_{ck}$이 24 MPa일때 환경영향 지표가 가장 낮은 지역은 울산이었으며 가장 높은 지역은 광주와 대구였다. 지역에 따른 환경영향의 차이는 지역에 따라 사용되는 혼화재의 종류와 치환율이 다르기 때문이다. 또한 압축강도 24 MPa일때, 콘크리트의 환경영향지수는 동절기가 하절기 및 표준기에 비해 약 5% 높았다. 반면에 콘크리트의 환경영향 지수는 콘크리트 압축강도가 35 MPa을 넘어서면서 계절의 영향은 미미하였다.

Establishment of the Korean Tolerable Daily Intake of Bisphenol A Based on Risk Assessments by an Expert Committee

  • Recently, reproductive and neurobehavioral effects of bisphenol A (BPA) have been documented, and thus a review was requested for BPA management direction by the government. Therefore, this study was performed to establish a Korean tolerable daily intake (TDI) for BPA. An expert committee, consisting of specialists in fields such as toxicology, medicine, pharmacology, and statistics, was asked to evaluate BPA health based guidance values (HbGVs). Although many toxicological studies were reviewed to select a point of departure (POD) for TDI, rat and mouse reproductive studies by Tyl et al. (2002, 2006), which were performed according to GLP standards and OECD guidelines, were selected. This POD was the lowest value determined from the most sensitive toxicological test. The POD, a NOAEL of 5 mg/kg bw/day, was selected based on its systemic toxicity as critical effects. An uncertainty factor of 100 including interspecies and intraspecies differences was applied to calculate the TDI. According to the evaluation results, a TDI of BPA for Korean was suggested at 0.05 mg/kg bw/day. In addition, the BPA exposure level based on food consumption by the Korean population was estimated as 1.509 ${\mu}g/kg$ bw/day, and the HI was evaluated at 0.03 when the TDI of 0.05 mg/kg bw/day was applied. This HI value of 0.03 indicated that hazardous effects would not be expected from BPA oral exposures. Although highly uncertain, further studies on low dose neurobehavioral effects of BPA should be performed. In addition, it is recommended that the 'as low as reasonably achievable' (ALARA) principle be applied for BPA exposure from food packaging materials in newborn infants and children.

Placenta Transfer and Toxicokinetics of Valproic Acid in Pregnant Cynomolgus Monkeys

  • Placenta transfer study in non-human primate (NHP) is one of the crucial components in the assessment of developmental toxicity because of the similarity between NHP and humans. To establish the method to determine placenta transfer in non-human primate, toxicokinetics of valproic acid (VPA), a drug used to treat epilepsy in pregnant women, were determined in pregnant cynomolgus monkeys. After mating, pregnancy-proven females were daily administered with VPA at dose levels of 0, 20, 60 and 180 mg/kg by oral route during the organogenesis period from gestation day (GD) 20 to 50. Concentrations of VPA and its metabolite, 4-ene-VPA, in maternal plasma on GDs 20 and 50, and concentrations of VPA and 4-ene-VPA in placenta, amniotic fluid and fetus on GD 50 were analyzed using LC/MS/MS. Following single oral administration of VPA to pregnant monkeys, concentrations of VPA and 4-ene-VPA were generally quantifiable in the plasma from all treatment groups up to 4-24 hours post-dose, demonstrating that VPA was absorbed and the monkeys were systemically exposed to VPA and 4-ene-VPA. After repeated administration of VPA to the monkeys, VPA was detected in amniotic fluid, placenta and fetus from all treatment groups, demonstrating that VPA was transferred via placenta and the fetus was exposed to VPA, and the exposures were increased with increasing dose. Concentrations of 4-ene-VPA in amniotic fluid and fetus were below the limit of quantification, but small amount of 4-ene-VPA was detected in placenta. In conclusion, pregnant monkeys were exposed to VPA and 4-ene-VPA after oral administration of VPA at dose levels of 20, 60 and 180 mg/kg during the organogenesis period. VPA was transferred via placenta and the fetus was exposed to VPA with dose-dependent exposure. The metabolite, 4-ene VPA, was not detected in both amniotic fluid and fetus, but small amount of 4-ene-VPA was detected in placenta. These results demonstrated that proper procedures to investigate placenta transfer in NHP, such as mating and diagnosis of pregnancy via examining gestational sac with ultrasonography, collection of amniotic fluid, placenta and fetus after Caesarean section followed by adequate bioanalysis and toxicokinetic analysis, were established in this study using cynomolugus monkeys.

Gene Expression Profiling of Acetaminophen Induced Hepatotoxicity in Mice

  • Microarray analysis of gene expression has become a powerful approach for exploring the biological effects of drugs, particularly at the stage of toxicology and safety assessment. Acetaminophen (APAP) has been known to induce necrosis in liver, but the molecular mechanism involved has not been fully understood. In this study, we investigated gene expression changes of APAP using microarray technology. APAP was orally administered with a single dose of 50 mg/kg or 500 mg/kg into ICR mice and the animals were sacrificed at 6, 24 and 72 h of APAP administration. Serum biochemical markers for liver toxicity were measured to estimate the maximal toxic time and hepatic gene expression was assessed using high-density oligonucleotide microarrays capable of determining the expression profile of >30,000 well-substantiated mouse genes. Significant alterations in gene expression were noted in the liver of APAP-administered mice. The most notable changes in APAP-administered mice were the expression of genes involved in apoptosis, cell cycle, and calcium signaling pathway, cystein metabolism, glutatione metabolism, and MAPK pathway. The majority of the genes upregulated included insulin-like growth factor binding protein 1, heme oxygenase 1, metallothionein 1, S100 calcium binding protein, caspase 4, and P21. The upregulation of apoptosis and cell cycle-related genes were paralleled to response to APAP. Most of the affected gene expressions were returned to control levels after 72 hr. In conclusion, we identified potential hepatotoxicity makers, and these expressions profiling lead to a better understanding of the molecular basis of APAP-induced hapatotoxicity.

케라틴 펩타이드에 의한 모발 및 두피 특성 변화 연구 (Study on Changes of Hair and Scalp Characteristics by Keratin Peptides)

  • 본 연구에서는 Fervidobacterium islandicum AW-1를 이용하여 케라틴 펩타이드를 생산하고, 두피 및 모발에 관련된 인자를 확인하여, 화장품 원료로서 케라틴 펩타이드의 가능성을 알아보았다. 모유두 세포주에 케라틴 펩타이드를 농도에 따라 세포 독성 및 증식을 살펴본 결과, 세포 독성은 나타나지 않았고, 세포증식에도 효과가 없었다. 케라틴 펩타이드를 함유한 헤어 샴푸, 헤어 에센스를 제조하여 인체 첩포 시험을 거쳐 무자극을 확인하였다. 30 명의 건강한 성인 피험자를 대상으로 4 주 동안 샴푸 및 에센스를 2 그룹으로 나누어 사용한 결과, 육안평가에서 윤기, 탈모, 두피 트러블, 푸석거림에서 통계적으로 유의한 긍정적 결과를 나타내었다. 두피 수분량은 샴푸와 에센스 사용 전 대비 2, 4 주 후에 통계적으로 유의한 증가를 나타내었다. 두피에 대한 경피수분손실양은 에센스를 사용한 그룹에서 사용 전 대비 4주 후에 통계적으로 유의한 감소를 나타내었다. 두피 피지량 역시, 에센스를 사용한 그룹에서 사용 전 대비 4 주 후에 통계적으로 유의한 감소를 나타내었다. 정상 모발, 손상 모발을 이용한 헤어 샴푸와 에센스의 사용 전후의 빗질에 의한 마찰력을 측정한 결과, 사용 전 대비 모두 통계적으로 유의한 변화를 나타내었다. 정상 모발의 경우, 사용 전보다 빗질의 마찰력이 증가하였고, 손상 모발의 경우 사용 전보다 빗질의 마찰력이 감소하였다. 이를 통해 케라틴 펩타이드는 모발 및 두피 관련한 제품에 사용할 수 있는 원료로 확인하였다.

해양적조생물제어를 위한 살조물질 Thiazolidinedione 유도체(TD49) 평가 (Assessment of New Algicide Thiazolidinedione (TD49) for the Control of Marine Red Tide Organisms)

  • 전세계적으로 연안해역에서 확산되고 있는 유해유독성 식물플랑크톤의 대발생은 수산자원생물자원에 심각한 피해를 입힌다. 본 연구에서는 유해성 미세조류 대발생을 제어하기 위해 개발된 신물질 Thiazolidinedione 유도체(TD49)의 살조능을 유해성 미세조류 성장단계(초기성장기, 대수증식기, 안정기)에 따라 조사하였다. TD49는 $Heterosigma$ $akashiwo$, $Chattonella$ $marina$ 그리고 $Chattonella$ sp.의 세포를 사멸시켰으며, 특히 낮은 농도($0.02{\mu}M$)의 TD49는 대수증식기와 안정기보다 초기 성장기에서 우수한 살조효과를 보였다. 또한 모든 성장단계에서 유해생물을 제어 할 수 있는 TD49의 농도는 $2{\mu}M$로 측정되었다. 무각 편모조류인 $Heterosigma$ $akashiwo$, $Chattonella$ $marina$ 그리고 $Chattonella$ sp.은 세포벽이 약하여 TD49물질에 의해 세포가 쉽게 파괴되어 우수한 살조효과를 보였다. 결과적으로 본 연구에서 개발된 Thiazolidinedione 유도체(TD49)는 유해적조생물 $H.$ $akashiwo$, $C.$ $marina$ 그리고 $Chattonella$ sp.를 제어할 수 있는 우수한 물질로 판단되었으나, 추후 현장 실용을 위해 메소코즘과 같은 인공생태계를 이용한 해양생태계 위해성 평가가 체계적으로 추진되어야 한다.

실내사육조건에서 한국산 물벼룩 종간 life cycle 비교 (Comparison of Life Cycle of Several Korean Native Freshwater Cladocerans in Laboratory Culture Conditions)

  • 국내서식 물벼룩종을 이용한 표준생태독성시험법 개발을 위한 기초연구로서 국내환경에서의 대표성, 실내사육가능성 및 독성시험에 적합한 크기 등을 고려하여 선발된 4종의 한국산 물벼룩 Daphnia sp., Daphnia obtusa, Moina macrocopa, S.imocephalus vetulus과 Daphnia magna에 대한 실내사육조건에서의 life cycle을 조사하였다. 국내산 물벼룩 4종과 D. magna에 대한 실내사육조건에서의 전 생애에 걸친 생육특성을 조사한 결과 D. magna가 어미 1마리당 평균 358마리의 새끼를 출산하였고 국내산 물벼룩 중에는 Daphnia sp.가 297마리의 새끼를 출산하여 가장 출산새끼수가 많았으며 그 다음으로는 M. macrocopa, S. vetulus, D. obtusa 순이었다. 평균수명은 D. magna가 50일로 가장 길었으며 국내산 물벼룩 중에는 Daphnia sp.가 39일로 조사되었으며 그 다음으로는 S. vetulus, D. obtusa M. macrocopa, 순이었다. 특히 M. macrocopa의 경우 번식력이 우수하면서도 세대기간이 짧아 기존의 번식독성시험법에서 제시하는 21일보다 짧은 10일 정도의 단기간에 번식영향을 평가할 수 있을 것으로 판단된다.

Associations of Low Environmental Exposure to Multiple Metals with Renal Tubular Impairment in Korean Adults

  • Recently several studies reported that the renal toxicity of lead (Pb) and cadmium (Cd) may exist in even a low level exposure. In terms of the deterioration of tubular function, it affects the loss of divalent metals and leads to other complications, so renal tubular effect of heavy metals should be well managed. Considering the exposure to heavy metals in reality, it is hard to find the case that human is exposed to only one heavy metal. We designed a cross-sectional study using Korean Research Project on the Integrated Exposure Assessment (KRIEFS) data to investigate the renal effects of multiple metal exposure in general population. We used blood Pb and urinary Cd as exposure measures, and urinary N-acetyl-${\beta}$-D-glucosaminidase (NAG) and ${\beta}_2$-microglobulin (${\beta}_2$-MG) as renal tubular impairment outcome. We conducted linear regression to identify the association between each heavy metal and urinary NAG and ${\beta}_2$-MG. And then, we conducted linear regression including the interaction term. Of 1953 adults in KRIEFS (2010~2011), the geometric mean of blood Pb and urinary Cd concentration was $2.21{\mu}g/dL$ (geometric $SD=1.49{\mu}g/dL$) and $1.08{\mu}g/g\;cr$ (geometric $SD=1.98{\mu}g/g\;cr$), respectively. In urinary Cd, the strength of the association was also high after adjusting (urinary NAG: ${\beta}=0.44$, p < 0.001; urinary ${\beta}_2$-MG: ${\beta}=0.13$, p = 0.002). Finally, we identified the positive interactions for the two renal biomarkers. The interaction effect of the two heavy metals of ${\beta}_2$-MG was greater than that of NAG. It is very important in public health perspective if the low level exposure to multiple heavy metals has an interaction effect on kidney. More epidemiological studies for the interaction and toxicological studies on the mechanism are needed.
