Recent Trends of Advanced Biosensors for Mycotoxin Analysis

  • A mycotoxin is a toxic secondary metabolite produced by organisms of the fungus kingdom, commonly known as molds and has been widely contaminated in agricultural products such as grains and cereals. Many methods including high performance liquid chromatography (HPLC) and gas chromatography (GC) have already been proposed and reviewed for mycotoxins. These methods are either expensive or time-consuming due to the complication of sample preparation and pre-concentration before determination. In addition, both methods are unsuitable for the routine screening of large sample numbers. A biosensor is a fictive analytical device that combines a biological component with a physicochemical detector for the detection of an analyte. Biosensors represent a rapidly expanding field, at the present time, with an estimated 60% annual growth rate; the major impetus coming from the health-care industry but with some pressure from other areas, such as food safety and environmental monitoring. Antibodies and aptamers are bioreceptors which have been used in the development of biosensors. There are many kinds of antibodies and aptamers specific to mycotoxin, and antibody (or aptamer)-based biosensors have been successfully developed for the detection of mycotoxin. The biosensors permit the rapid, sensitive, simple, and on-site detection of a range of mycotoxins and can be an alternative method to traditional methods such as HPLC and GC. This presentation provides the development trends of biosensors to mycotoxins and their application to food and agricultural products.

Headspace-SPME와 GC-ECD를 이용한 수중의 미량 Halonitromethane (HNM)류 분석 (Analysis of Trace Levels of Halonitromethanes (HNM) in Water using Headspace-SPME and GC-ECD)

  • HNMs는 독성이 강한 소독부산물들 중의 하나로 최근에 다양한 연구가 진행되고 있다. 본 연구에서는 headspace-solid phase microextraction (SPME) 전처리장치와 GC-ECD를 이용하여 9종의 HNMs를 동시분석할 수 있는 분석법을 개발하였다. 9종의 HNMs에 대한 검출한계(LOD)는 90~260 ng/L였으며, 정량한계(LOQ)는 270~840 ng/L였다. 수돗물과 해수를 이용하여 시료수의 matrix 영향을 살펴본 결과, 9종의 HNMs에 대해 80%~127%의 양호한 회수율을 나타내어 시료수의 matrix에 영향을 받지 않았다. 또한, 본 연구에서 개발된 headspace SPME GC-ECD 분석법은 용매류를 이용한 별도의 전처리 과정이 필요하지 않아서 친환경적이며 간편하고 빠른 자동화된 방법으로 HNMs 분석에 적합한 것으로 나타났다.

알코올을 주성분으로 하는 손소독제의 안전성 연구 (A Study on the Safety of Alcohol-based Hand Sanitizers)

  • Objectives: In this study, the safety of alcohol-based hand sanitizers (ABHSs) for quasi-drugs and cosmetics was investigated by analyzing the ethanol content, which is an active ingredient with a sterilizing effect, and methanol, which is toxic. Methods: Forty-one ABHSs were purchased at large supermarkets and online stores. Ethanol quantification was performed by gas chromatography-flame ionization detector, and methanol quantification was performed by headspace-gas chromatography-mass spectrometry. Results: The ethanol content of ABHS in quasi-drugs was 49.6-67.8%, which was suitable for standard manufacturing procedures for external disinfectants, and the ethanol content of ABHS in cosmetics was 9.1-61.3%. The methanol content of ABHS in quasi-drugs ranged from not detected(N.D.)-131.8 ppm, which was suitable for the methanol detection standard of ethanol raw materials in the Korean Pharmacopoeia. The methanol content of ABHS in cosmetics was 23.4-859.7 ppm, which was suitable for the detection limit of methanol in cosmetics. Conclusions: The ethanol and methanol content of ABHS was judged to be safe. When selecting an ABHS to be used for sterilization, it seems necessary to check the content of ethanol, an active ingredient, and use it according to its intended purpose.

대기 중 휘발성유기오염물질의 환경, 개인 및 인체 노출의 상관성 연구 (A Correlation Study between the Environmental, Personal Exposures and Biomarkers for Volatile Organic Compounds)

  • Volatile organic compounds (VOCs) are an important public health problem throughout the world. Many important questions remain to be addressed in assessing exposure to these compounds. Because they are ubiquitous and highly volatile, special techniques must be applied in the analytical determination of VOCs. Personal exposure measurements are needed to evaluate the relationship between microenvironmental concentrations and actual exposures. It is also important to investigate exposure frequency, duration, and intensity, as well as personal exposure characteristics. In addition to air monitoring, biological monitoring may contribute significantly to risk assessment by allowing estimation of absorbed doses, rather than just the external exposure concentrations, which are evaluated by environmental and personal monitoring. This study was conducted to establish the analytic procedure of VOCs in air, blood, urine and exhaled breath and to evaluate the relationships among these environmental media. The subjects of this study were selected because they are occupationally exposed to high levels of VOCs. Environmental, personal, blood, urine and exhalation samples were collected. Purge & trap, thermal desorber, gas chromatography and mass selective detector were used to analyze the collected samples. Analytical procedures were validated with the“break through test”, 'quot;recovery test for storage and transportation”,“method detection limit test”and“inter-laboratory QA/QC study”. Assessment of halogenated compounds indicted that they were significantly correlated to each other (p value < 0.01). In a similar manner, aromatic compounds were also correlated, except in urine sample. Linear regression was used to evaluate the relationships between personal exposures and environmental concentrations. These relationships for aromatic and halogenated are as follows: Halogen $s_{personal}$ = 3.875+0.068Halogen $s_{environmet}$, ($R^2$= .930) Aromatic $s_{personal}$ = 34217.757-31.266Aromatic $s_{environmet}$, ($R^2$= .821) Multiple regression was used to evaluate the relationship between exposures and various exposure deter-minants including, gender, duration of employment, and smoking history. The results of the regression model-ins for halogens in blood and aromatics in urine are as follows: Halogen $s_{blood}$ = 8.181+0.246Halogen $s_{personal}$+3.975Gender ($R^2$= .925), Aromatic $s_{urine}$ = 249.565+0.135Aromatic $s_{personal}$ -5.651 D.S ($R^2$ = .735), In conclusion, we have established analytic procedures for VOC measurement in biological and environmental samples and have presented data demonstrating relationships between VOCs levels in biological media and environmental samples. 소방 실화재 훈련에서 사용하는 압축목재 가연물에서 발생하는 유해물질 특성 (Characteristics of Hazardous Substances Generated from Combustible Compressed Wood Used during Live Fire Training for Firefighters)

  • Objectives: To identify and investigate through qualitative and quantitative analysis the hazardous substances generated when compressed wood was burned at a live fire-training center. Methods: Four types of compressed wood that are actually used in live fire training were burned in a chamber according to KS F2271. The gaseous material was sampled with a gas detector tube and conventional personal samplers. Results: 1,3-butadiene, benzene, toluene, xylene, formaldehyde, hydrogen chloride, hydrogen cyanide, ammonia, carbon monoxide, and nitric acid were detected. In particular, 1,3-butadiene (497.04-680.44 ppm), benzene (97.79-125.02 ppm), formaldehyde (1.72-13.03 ppm), hydrogen chloride (4.71-15.66 ppm), hydrogen cyanide (3.64-8.57 ppm), and sulfuric acid (3.85-5.01 ppm) exceeded the Korean Occupational Exposure Limit as measured by sampling pump according to the type of compressed wood. Conclusions: We found through the chamber testing that firefighters could be exposed to toxic substances during live fire training. Therefore, firefighter protection is needed and more research is required in the field.

Characterization of Forest Fire Emissions and Their Possible Toxicological Impacts on Human Health

  • In flight particulate matter particularly emissions generated by incomplete combustion processes has become a subject of global concern due to the health problems and environmental impacts associated with them. This has compelled most countries to set standards for coarse and fine particles due to their conspicuous impacts on environment and public health. This contribution therefore explores forest fire emissions and how its particulates affects air quality, damage to vegetation, water bodies and biological functions as architects for lung diseases and other degenerative illnesses such as oxidative stress and aging. Soot was collected from simulated forest fire using a clean glass surface and carefully transferred into amber vials for analysis. Volatile components of soot were collected over 10 mL dichloromethane and analyzed using a QTOF Premier-Water Corp Liquid Chromatography hyphenated to a mass selective detector (MSD), and Gas Chromatograph coupled to a mass spectrometer (GC-MS). To characterize the size and surface morphology of soot, a scanning electron microscope (SEM) was used. The characterization of molecular volatiles from simulated forest fire emissions revealed long chain compounds including octadec-9-enoic acid, octadec-6-enoic acid, cyclotetracosane, cyclotetradecane, and a few aromatic hydrocarbons (benzene and naphthalene). Special classes of organics (dibenzo-p-dioxin and 2H-benzopyran) were also detected as minor products. Dibenzo-p-dioxin for instance in chlorinated form is one of the deadliest environmental organic toxins. The average particulate size of emissions using SEM was found to be $11.51{\pm}4.91{\mu}m$. This study has shown that most of the emissions from simulated forest fire fall within $PM_{10}$ particulate size. The molecular by-products of forest fire and particulate emissions may be toxic to both human and natural ecosystems, and are possible precursors for various respiratory ailments and cancers. The burning of a forest by natural disasters or man-made fires results in the destruction of natural habitats and serious air pollution.

  • 소독부산물의 일종인 트리할로메탄류(THMs)는 정수처리 공정에서 소독제로 사용되는 염소와 수중에 잔존하는 유기물질이 반응하여 생성된다. 일반적으로 염소계 및 브롬계 THMs는 일반적인 수돗물에서 검출되기 때문에 잘 알려져 있으나, 요오드계 THMs (I-THMs)의 경우는 수중에 요오드 이온이 존재할 때 생성된다. I-THMs는 약품취를 유발하며, 또한 염소계나 브롬계 THMs에 비해 인체독성이 더 강한 것으로 알려져 있다. 현재 I-THMs 분석을 위한 공인된 분석법은 없는 실정이다. 10종의 THMs 분석을 위해 headspace 전처리장치와 GC/ECD를 이용하여 최적화된 분석법을 개발하였으며, 개발된 분석법에 의한 검출한계(LOD)와 정량한계(LOQ)는 각각 12 ng/L~56 ng/L 및 38 ng/L~178 ng/L로 나타났다. 강물, 해수 및 하수처리장 최종방류수의 matrix 영향을 평가하였으며, 본 연구에서 개발된 분석법은 별도의 전처리 과정이 필요치 않아 간편하고 빠르며 자동화된 방법으로 수중에 미량으로 함유된 I-THMs 분석에 적합한 것으로 나타났다.

지하공동구내 가연성케이블의 열화접촉으로 인한 화재위험성 예측평가 (Study on the Fire Risk Prediction Assessment due to Deterioration contact of combustible cables in Underground Common Utility Tunnels)

  • 최근 지하공동구(Underground Common Utility Tunnels)는 도시민이 일상생활을 영위하는데 필요한 전기, 통신, 상수도, 도시 가스, 하수도뿐만 아니라 냉난방시설, 진공 집진관, 정보처리케이블 등의 시설물을 2종 이상 공동으로 수용하기 위한 지하시설물로서 국가의 중추기능을 담당하는 시설이지만, 화재 사고시 신속한 대처가 힘들고 각종 케이블 연소시 발생하는 유독가스 및 연기에 의해 공동구 내에 진입하여 진압하기가 힘들다. 따라서 화재발생시 막대한 재산피해 및 통신두절 등 국가의 중추신경이 마비됨은 물론 시민불편사항을 초래하게 된다. 따라서 본 논문은 지금까지 발생 되어온 국내,외 공동구 화재사례에서 화재발생의 주요원인으로 전기공사로 인한 합선 및 가연성케이블에 의한 열화접촉으로 화재가 발생하는 것이 대다수를 차지하고 있음에 착안하여 실제 공동구 모형을 제작하고 화재를 재현함으로서 과학적으로 화재의 성상을 분석하는데 그 목적이 있다. 화재실험은 지하공동구 내에 정온식 감지선형 감지기(Line type fixed temperature detector), 방화문(Fire door), 연결살수설비(Connection deluge set), 및 환기설비를 설치하고, 송 배전케이블은 일정구간 내화(Fireproof)도료로 도장하며, 난방관은 내화 피복된 상태에서 실험하였다. 그 결과 Type II의 경우 최고온도가 $932^{\circ}C$로 측정되었고, 일정한 온도에서 정온식감지선형감지기가 화재위치를 정확하게 수신반에 표시되었다. 그리고 Type III의 경우인 송 배전케이블은 일정구간 내화도료로 도장한 것은 내화성능이 없는 것으로 나타났고, 내화피복(Fireproof covered)된 난방관은 약 30분 정도의 내화성능이 있는 것으로 나타났다. 또한 화재뮬레이션 결과는 실물화재시험시의 화재하중(Fire load)을 입력하여 실시한 결과로서 최고온도가 $943^{\circ}C$로 실물화재실험시의 $932^{\circ}C$와 거의 일치 하였다. 따라서 공동구 화재하중만으로 화재시뮬레이션을 실시하여 화재성상에 대한 예측이 가능한 것으로 판단되며 시뮬레이션으로 얻은 열방출률(Heat release rate), 연기층의 높이, 산소(O2), 일산화탄소(CO), 이산화탄소(CO2)의 농도 등의 결과 값들은 실제 화재실험시의 값으로 적용시킬 수 있는 것으로 판단된다. 향후 본 연구에서 구축한 국내 지하공동구 화재사고에 대한 실험자료 및 매년 지속적으로 화재사례들을 분석하여 축적하고 법 규정 및 관리 메뉴얼 등을 보완함으로써 국내 지하공동구 화재사고에 대한 보다 신뢰성 있는 정보를 제공해 줄 수 있을 것이며, 효과적이고 체계적으로 지하공동구의 신설 및 유지 관리 보수에 기여할 것으로 기대된다.

일부 대체세정제 제조업체의 물질안전보건자료의 실태와 그 화학물질의 유해성 평가에 관한 연구 (Survey of Actual Conditions of Material Safety Data Sheet and Quantitative Risk Assessment of Toxic Substances : Substitutes for Degreasing Agents)

  • Since the regulation of MSDS (Material Safety Data Sheets) had started from July 1996, employers were required to furnish MSDS for the chemicals in use in their workplace. However, many MSDS did not contain upright information for the chemicals, and they were not updated regularly, and were not written in the standard format required by the Industrial Safety and Health Act (ISHA). The purposes of this study were 1) to examine the reliability of MSDS for mixed solvents, 2) to provide reliable MSDS to employers or employees, 3) to find out any difficulties in implementing MSDS after the initiation, and 4) to promote regular MSDS updating and to ensure the reliability of MSDS for chemical manufacturers. To check the reliability of MSDS of mixed chemicals, 21 samples of mostly degreasing solvents were collected along with their MSDS from the work place. The samples were analyzed by gas chromatography-mass selective detector(GC-MSD). Their components were classified as saturated hydrocarbon, cyclic hydrocarbon, aromatics, and halogen containing hydrocarbon, and the amount of each class were measured. Manufacture's MSDS were compared with the actual composition of the collected samples, and further examined the reliability by checking whether the chemicals analyzed were included in the MSDS correctly. Finally, each item of MSDS was evaluated whether the MSDS correspond to the regulation required by ISHA. The results were following: 1) most of the degreasing solvents in MSDS were incorrect in their composition and contents, 2) the information in the MSDS including hazard classification, exposure level, toxicity, regulatory information were incorrectly provided, and 3) some MSDS did not disclose carcinogens in their MSDS. Continuous monitoring of MSDS was required to ensure reliability of MSDS. The Chemicals containing hydrocarbons from C10-C15 need to be tested to provide toxicity data. In addition, governmental support for providing correct MSDS was recommended to ensure reliability of MSDS. The MSDS regulation relating to the confidential business information may need to be revised to ensure reliability of MSDS.
