  • 3-Monochloro-1, 2-propanediol(3-MCPD) is a toxic compound, often present in different foods containing acid hydrolyzed(AH) protein, like seasonings and savory food products. In Korea, 3-MCPD is currently being a problem because of its toxicity in AH soybean sauce. The purpose of the present studies was to investigate the effects of 3-MCPD on male fertility, sperm and testosterone secretion. In vivo male fertility test was performed for observing the adverse effects of 3-MCPD on the function of male reproductive system and pregnancy outcome. (omitted)

  • TCDD (2,3,7,8-tetrachlorodibenzo-p-dioxin), one of the notorious toxic environmental pollutants, damages various organs including liver and is regarded as an endocrine disrupter. To investigate the effects of Houttuynia cordata Thunb (HCT) on the biochemical parameters of function, liver and serum of TCDD-treated rats were used. After 7 days from TCDD (1 $\mu\textrm{g}$/kg) injection, HCT (200 mg/kg) was administered into rats intraperitoneally for 4 weeks. The lipidperoxide content was examined by measuring the level of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum, and malondialdehyde (MDA) in liver tissue of rats. Prediction of PM10 concentration in Seoul, Korea using Bayesian network

  • Recent studies revealed that fine dust in ambient air may cause various health problems such as respiratory diseases and cancer. To prevent the toxic effects of fine dust, it is important to predict the concentration of fine dust in advance and to identify factors that are closely related to fine dust. In this study, we developed a Bayesian network model for predicting PM10 concentration in Seoul, Korea, and visualized the relationship between important factors. The network was trained by using air quality and meteorological data collected in Seoul between 2018 and 2021. The study results showed that current PM10 concentration, season, carbon monoxide (CO) were the top 3 effective factors in 24 hours ahead prediction of PM10 concentration in Seoul, and that there were interactive effects.

Exposure to Triclosan Induces Mortality through Oxidative Stress and DNA Damage in the Java Medaka Oryzias javanicus

  • To understand the detrimental effects of triclosan on Java medaka (Oryzias javanicus) embryos, fertilized embryos were exposed to different concentrations (1, 10, 50, 100, 200, 400, 600, 800, and 1,000 ㎍ l-1) of triclosan until hatching. Then, we examined the survival rate and developmental parameters as well as alterations in antioxidant constituents and DNA damage markers. The results showed dose-dependent mortality, hatching delays, and developmental abnormalities in the embryos. Additionally, there were significant increases in oxidative stress parameters and antioxidant responses, along with elevated DNA damage. These findings suggest that sublethal concentrations of triclosan induce toxic effects through oxidative stress on Java medaka embryos, as evidenced by changes in in vivo parameters and biochemical constituents.

Chronic Toxicities of Effluents from Dye Industry using Daphnia magna (물벼룩을 이용한 일부 염색폐수의 만성 수질독성 특성 연구)

  • Recent studies indicated the utility of whole effluent assessment as a measure to control discharge of toxic effluents to receiving water in Korea. However, most studies have been focussed on acute lethal effects of toxic wastewater with little consideration of chronic sublethal impacts which are of growing concern in protecting aquatic ecosystem. We conducted acute and chronic toxicity tests with effluents discharged from five different dyeing plants in Gyeong-gi province using a marine bacterium Vibrio fischeri and a freshwater macroinvertebrate Daphnia magna to demonstrate the importance of assessing chronic sublethal effects. Various levels of acute and chronic toxicities were observed in many samples tested in this study. In 21-d chronic toxicity tests using D. magna all samples showed effects on reproduction and growth. Notable mortalities were also noted in three out of five effluents. The result of the Microtox assay indicated that acute microbial toxicity existed in effluents from two out of five plants and acute daphnid toxicity was observed in only one effluent. The result of this study clearly suggests chronic toxicity tests are more suitable to assess biological effects of effluents because it was shown from this study that even an effluent with no acute toxicity could cause chronically lethal and/or sublethal adverse effects on aquatic biota which may affect the population dynamics in aquatic ecosystem.

  • The purpose of this study is to find out effects of prescriptions according to the formula, manipulation of Rhei Rhizoma, configuration. The following results were reached through investigations on the prescriptions using Rhei Rhizoma as a main component. Objectives : Analysis of prescriptions According to the formula : Liquid Extract Prescriptions were used widely on three parts to treat fever and damp heat in the interior organs. Powder Prescriptions were taken with hot water, thin porridge, tea etc. to treat damp heat, congestion of phlegm, acute episodes. Liquid Mixed Pill treat congestion of QI, damp heat, phlegm. Honey Mixed Pill treat accumulated fever, distension, acute excessive fever. Paste Pill treat blood stagnation, excessive toxic-fever, epidemic diseases. External Application treat inflammation by injury, swelling due to severe fever by internal damage. Methods : Analysis of prescriptions According to the manipulation of Rhei Rhizoma : Prescriptions including Liquor processed Rhei Rhizoma treat excessive toxic-fever, congestion of phlegm, blocking orifices on the upper side. Steamed Rhei Rhizoma strengthen effects of making evacuate and cooling of heat. Processed Rhei Rhizoma with vinegar strengthen effects of removing blood stagnation by activating blood movement, releasing gathering. Results : Analysis of prescriptions According to the Composition of Rhei Rhizoma : 41% of the total prescriptions were on the area of less than 20%. In case of lower groups show increased frequency of combination with Pharbitidis Semen, Persicae Semen, Scutellariae Radix and manipulation of baking, steaming, roasting. In case of higher groups show increased frequency of treating excess syndrome, critical illness, acute severe illness, and using proccesed Rhei Rhizoma with vinegar. Treatment of damp heat on the liver and gallbladder, disorder of the spleen and stomach is done mostly by prescriptions on the area of less than 30%. Conclusions : Rhei Rhizoma-Coptidis Rhizoma pair treat damp heat, heat toxins in blood, and Constipation caused by excessive heat. Rhei Rhizoma-Glycyrrhizae Radix pair relieve effects of Rhei Rhizoma passing blocked feces, removing the poison, activating blood movement, releasing gathering with the effects of Glycyrrhizae Radix relaxing tension by harmonizing Middle. Rhei Rhizoma-Magnoliae Cortex pair are used to treat damp heat in middle area, excessive heat in the stomach and intestine. Rhei Rhizoma-Pharbitidis Semen pair act on both blood system and QI system treating edema, damp, stagnation, heat toxins, feces. Rhei Rhizoma-Persicae Semen pair treat blood stagnation with fever on blood system.

Effects of Reactive Oxygen Species on Sperm Function, Lipid Peroxidation and DNA Fragmentation in Bovine Spermatozoa (소 정자에 있어서 활성산소계가 정자 기능과 지방산화 및 DNA 절편화에 미치는 영향)

  • Objective : To evaluate the effects of the reactive oxygen species (ROS) generated with a xanthine (X) and xanthine oxidase (XO) system on sperm function, the change of sperm characteristics, lipid peroxidation, and DNA fragmentation in bovine spermatozoa. Materials and Methods: ROS were produced using a combination of 1000 uM X and 50 mU/ml XO. The ROS scavengers: superoxide dismu tase (SOD) (200 U/ml) and catalase (500 U/ml) were also tested. Spermatozoa were incubated for 2 hours in BWW medium with a combination of X-XO supplemented with or without ROS scavengers at $37^{circ}C$ under 5% $CO_2$ incubator. Sperm movement characteristics by CASA (computer-aided sperm analysis), HOST (hypoosmotic swelling test), Caionophore induced acrosome reaction, malondialdehyde formation for the analysis of lipid peroxidation, the percentage of DNA fragmentation using the method of TdT-mediated nick end labelling (TUNEL) by flow cytometry were determined after 2 hours incubation. Results: The action of ROS on bovine spermatozoa resulted in a decreased in capacity for sperm motility, Ca-ionophore induced acrosome reaction and membrane integrity, an increased in malondialdehyde formation and the percentage of sperm with DNA fragmentation. In the effects of antioxidant, catalase completely alleviated the toxic effects induced by the ROS in terms of sperm function and characteristics, however SOD exhibited no capacity to reduce the toxic effects. Conclusion: The ROS can induce significant damages to sperm functions and characteristics. The useful ROS scavengers can minimized the defects of sperm function and various damages of spermatozoa.

  • cis-Dichlorodiammineplatinum (II) (cis-Platin) inhibits the proliferation and growth of the tumor cells by way of inhibiting DNA and protein synthesis of the cancer cells. Although cis-Platin is very effective antitumor drug, it also produces many other side effects. Thus the author has studied the effects of cis-Platin on the proximal epiphyseal plate in the tibia of the rat. The results were as follows: In the chondrocyte of the proliferative zone, sacculated, and fragmented cisternae of rough endoplasmic reticulum, some mitochondria with disorganized mitochondrial cristae and distorted procollagens were observed, and in the matrix some large matrix granules and dispersed collagen fibrils were revealed on the 1st, 3rd day and 1st week group of cis-Platin treated rats. In the chondrocyte of the proliferative zone of cis-Platin treated rats on the 2nd and 3rd week group, parallely arranged rough endoplasmic reticulum and many procollagens were shown, and in the matrix a number of large matrix granules and many small matrical granules as well as collagen fibrils were revealed. Consequently it is suggested that though cis-Platin induces the degenerative changes of the chondrocyte resulting in components of the cartilagenous matrix, these toxic effects are regressed with time.

Effects of stranded oils and dispersant clean-up on intertidal gastropods (유류오염과 유처리제를 이용한 전화작업이 조간대에 서식하는 고둥류에 미치는 영향)

  • An experimental approach was applied to test the effects of stranded oils and dispersant cleanup on marine gastropods, Nodilittorina exigua, littorina brevicula and Purpura clavigera. They were exposed to Labuan crude, Dubai crude and Bunker C fuel oil. Direct oil contact caused death of gastropods within 96 hours. N. exigua and L. brevicula were more sensitive than P. clavigera at the exposure of Bunker C fuel oil. Toxic effects of Bunker C oil was slower than crude oils. direct contact to concentrated dispersant killed gastropods, while clean-up with diluted dispersant still gave severe damage. P. clavigera could escape from dispersed crude oil below 250 ppm. Oiling and dispersant clean-up may have severe effects on marine gastropods by rendering them washed out to sea.

  • Zearalenone (ZEA) is a phenolic resorcylic acid lactone compound produced by several species of Fusarium. ZEA has toxic effects in the testes of domestic and laboratory animals. Korean red ginseng (KRG), the steamed root of Panax ginseng Meyer, has multiple pharmacological effects such as vasorelaxation, anti-thrombosis, anti-hypertension, etc. In this study, we investigated the effects of KRG extract on testicular toxicity induced by ZEA. Rats were treated with 300 mg/kg oral doses of KRG for 4 weeks every other day. The rats were then treated with a single dose of 5 mg/kg ZEA delivered intraperitoneally, whereas control rats received only doses of the vehicle. As a result, germ cell apoptosis induced by ZEA was decreased by KRG pre-treatment. In addition, Fas and Fas-L expression was reduced in rats that received KRG pre-treatment compared to ones treated with ZEA alone. In conclusion, impaired spermatogenesis resulting from ZEA treatment was prevented by KRG through Fas-Fas L modulating.
