AMPK/Sirt1/PGC-1α 신호 전달 경로의 조절을 통한 오미자 추출물의 비만 개선 효과 (Anti-Obesity Effect of Schizandrae Fructus Water Extract through Regulation of AMPK/Sirt1/PGC-1α signaling pathway)

  • Objectives : Although the anti-obesity effect of Schizandrae Fructus water extract has been demonstrated, its underlying mechanism is still unclear. Therefore, we aimed to evaluate the anti-obesity effect of Schizandrae Fructus water extract through the p-AMP-activated protein kinase (p-AMPK), sirtuin1 (Sirt1), and peroxisome proliferator-activated receptor-gamma coactivator 1α (PGC-1α) signaling in 60% high-fat diet (HFD)-induced obese mouse model. Methods : Male C57BL/6 mice were divided into four groups. The Normal group was fed a normal diet and the obese groups were fed 60% HFD. Except for the Control group, the GG group was supplemented with 0.5% Garcinia gummigutta and the SCW group was supplemented with 0.5% Schizandrae Fructus water extract. After 6 weeks, obesity-related biomarkers in serum were measured and the expressions of protein for lipid-related factors in liver tissue were analyzed by western blot. Results : Treatment with SCW significantly down-regulated body weight compared to the Control group. SCW down-regulated levels of triglyceride and total cholesterol in serum and significantly increased p-AMPK, Sirt1, and PGC-1α in liver tissue. In addition, the expressions of fatty acid oxidation-related proteins such as peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase 1A (CPT-1A), uncoupling protein 1 (UCP1), and uncoupling protein 3 (UCP3) were significantly up-regulated. However, fatty acid synthesis-related proteins including sterol regulatory element-binding protein-1 (SREBP-1), phospho-Acetyl-CoA Carboxylase (p-ACC), and fatty acid synthase (FAS) were significantly down-regulated. Conclusions : Taken together, SCW treatment showed anti-obesity effect by regulating both fatty acid oxidation-related and fatty acid synthesis-related proteins through AMPK/Sirt1/PGC-1α signaling in 60% HFD-induced obese mice.

금연.금주 남자대학생의 골밀도, 영양소 섭취, 혈액 성상 및 식습관 (Bone Density, Nutrient Intake, Blood Composition and Food Habits in Non-Smoking and Non-Alcohol Drinking Male University Students)

  • This study was conducted in order to investigate and compare anthropometric measurements, bone density, nutrient intake, blood composition and food habits between non-smoking, non-alcohol drinking and smoking, alcohol drinking male university students in Seoul, South Korea. The data for food habits and health-related behaviors were obtained by selfadministered questionnaires. The BQIs of the subjects were measured by Quantitative Ultrasound (QUS). The subjects were divided into two groups: NSND (non-smoking and non-alcohol drinking, n=62) group and General (smoking and alcohol drinking, n=160) group. The results were analyzed using the SPSS program and were as follows: The average heights, weights, and BMIs of the two groups were 173.3 cm, 66.5 kg and 22.1 and 173.4 cm, 68.7 kg and 22.9, respectively. There were no differences between the groups regarding height, weight or BMI. SBP and DBP, however, were significantly higher in the general group than in the NSND group (p<0.01). The BQIs, Z-scores and T-scores of the two groups were 99.83, -0.23, and -0.31 and 98.24, -0.27 and -0.39, respectively, producing no significant differences between the two groups. The percentages for normal bone status, osteopenia and osteoporosis were 83.88%, 16.12% and 0.0% and 74.37%, 25.62% and 0.01%, respectively. Mean intakes of animal protein (p<0.05), animal fat (p<0.05), fiber (p<0.05), animal Ca(p<0.05), animal Fe (p<0.001), Zn(p<0.05), vitamin B1 (p<0.05) and niacin (p<0.05) were significantly different between the two groups, and mean serum levels of SGOT (p<0.01), SGPT (p<0.001), ${\gamma}$-GTP (p<0.001), triglycerides (p<0.01), total cholesterol (p<0.05) and hematocrit (p<0.05) were also significantly different between the two groups. Overall, there were no differences in meal regularity, frequency of snacking, reasons for overeating, exercise and defecation between the groups. However, favorite foods (p<0.05) and night-time meals (p<0.05) were significantly different. In conclusion, the health status of the NSND group was superior compared to the general group. Thus, students who smoke and alcohol drink should receive a practical and systematically-organized education regarding the increased health benefits of quitting smoking and alcohol drinking.

영양표시 제도에 대한 여성소비자 인식에 관한 연구 (A Study on the Awareness of Female-Consumers for Nutrition Labeling System)

  • This study had carried out to investigate the usage status of customer, the positive effects and problems, and the most important items of the nutrition labeling at purchasing the food etc. on the current nutrition labeling system in order to establish the customer-centric nutrition labeling system. Survey was carried out by questionnaire method that is targeted on adult female above 20 years old in Seoul and Kyeongnam area from May to June, 2004. For the experience of checking the nutrition label of the processed domestic and imported processed food, 82% and 75.4% of the respondents were replied 'have checked' respectively. For the positive effects due to enforcement of the nutrition labeling system, the respondents agreed highly with 'easy to compare with other products' and 'improve the products quality'. For the problems of the nutrition labeling system, the respondents agreed highly with 'different criteria for each product' and 'incendiary purchasing due to false or exaggerated labeling', and gave the higher scale for the positive effects than the problems relatively. For the necessity of the nutrition labeling system, 96.2% of the respondents were replied 'necessary', and it was indicated a significant difference on age and marital status(p<.01). For the price rising due to enforcement of the nutrition labeling system, 55.2% of respondents agreed, and it was indicated a very significant difference on age and monthly income(p<.001). For the most important nutrition labeling items at purchasing the food, the respondents were replied 'total calorie' on most of the food, and in addition, they checked carefully the lipid, cholesterol, protein, Ca, and Fe.

Association of CAPN10 gene (rs3842570) polymorphism with the type 2 diabetes mellitus among the population of Noakhali region in Bangladesh: a case-control study

  • Type 2 diabetes mellitus (T2DM) is a multifactorial, polygenic, and metabolically complicated disease. A large number of genes are responsible for the biogenesis of T2DM and calpain10 (CAPN10) is one of them. The association of numerous CAPN10 genetic polymorphisms in the development of T2DM has been widely studied in different populations and noticed inconclusive results. The present study is an attempt to evaluate the plausible association of CAPN10 polymorphism SNP-19 (rs3842570) with T2DM and T2DM-related anthropometric and metabolic traits in the Noakhali region of Bangladesh. This case-control study included 202 T2DM patients and 75 healthy individuals from different places in Noakhali. A significant association (p < 0.05) of SNP-19 with T2DM in co-dominant 2R/3R vs. 3R/3R (odds ratio [OR], 2.7; p=0.0014) and dominant (2R/3R) + (2R/2R) vs. 3R/3R (OR, 2.47; p=0.0011) genetic models was observed. High-risk allele 2R also showed a significant association with T2DM in the allelic model (OR, 1.67; p=0.0109). The genotypic frequency of SNP-19 variants showed consistency with Hardy-Weinberg equilibrium (p > 0.05). Additionally, SNP-19 genetic variants showed potential associations with the anthropometric and metabolic traits of T2DM patients in terms of body mass index, systolic blood pressure, diastolic blood pressure, total cholesterol, and triglycerides. Our approach identifies the 2R/3R genotype of SNP-19 as a significant risk factor for biogenesis of T2DM in the Noakhali population. Furthermore, a large-scale study could be instrumental to correlate this finding in overall Bangladeshi population.

  • Purpose : The objective of this study was to determine the major factors influencing the onset of diabetes and prediabetes and for collection of the basic data required to reduce the prevalence of diabetes and plan for administration of an effective health care system. By classifying the level of blood sugar management according to three categories: normal, prediabetes, and diabetes diagnosis, and determining the causes of diabetes in consideration of various variables, we will conduct an analysis of the main factors to be addressed for effective management of blood sugar and for preparation of basic data for use in early management. Methods : In this study, an analysis of raw data from the 8th National Health and Nutrition Examination Survey collected over a period of three years from 2019 to 2021, including 8,110 subjects in 2019, 7,359 subjects in 2020, and 7,090 subjects in 2021 was performed. A total of 22,559 subjects were aged 19 years or older, and 15,821 subjects were classified as subjects for inclusion in the final analysis. In the analysis, categorical variables were tested for difference, analysis of continuous variables using regression was performed, and analysis of influencing factors was performed using multinomial logistic analysis. Result : Significant factors related to the onset of diabetes and prediabetes included age (p<.001), marital status (p<.001), occupation (p<.001), hypertension (p<.001), dyslipidemia (p<.001), cardiovascular disease (p=.008), alcohol (p=.030) smoking (p=.005), systolic blood pressure (p<.001), diastolic blood pressure (p<.001), body mass index (p<.001) and waist circumference (p=.037), blood triglycerides (p<.001), and blood cholesterol (p<.001). Conclusion : Diabetes, a complex disease affected by a variety of diseases, requires active management from the prediabetes stage, and providing an appropriate level of medical information and services to elderly individuals without family support is considered a long-term health care system requirement in Korean society where the demographic structure is changing. In particular, determining the causes of prediabetes and development of a preventive approach to administering the health care system will be important for efficient management of diabetic patients.

Expanded IL-22+ Group 3 Innate Lymphoid Cells and Role of Oxidized LDL-C in the Pathogenesis of Axial Spondyloarthritis with Dyslipidaemia

  • Group 3 innate lymphoid cells (ILC3), which express IL-22 and IL-17A, has been introduced as one of pathologic cells in axial spondyloarthritis (axSpA). Dyslipidaemia should be managed in axSpA patients to reduce cardiovascular disease, and dyslipidaemia promotes inflammation. This study aimed to reveal the role of circulating ILC3 in axSpA and the impact of dyslipidaemia on axSpA pathogenesis. AxSpA patients with or without dyslipidaemia and healthy control were recruited. Peripheral blood samples were collected, and flow cytometry analysis of circulating ILC3 and CD4+ T cells was performed. The correlation between Ankylosing Spondylitis Disease Activity Score (ASDAS)-C-reactive protein (CRP) and circulating immune cells was evaluated. The effect of oxidized low-density lipoprotein cholesterol (oxLDL-C) on immune cell differentiation was confirmed. AxSpA human monocytes were cultured with with oxLDL-C, IL-22, or oxLDL-C plus IL-22 to evaluate osteoclastogenesis using tartrate-resistant acid phosphatase (TRAP) staining and real-time quantitative PCR of osteoclast-related gene expression. Total of 34 axSpA patients (13 with dyslipidaemia and 21 without) were included in the analysis. Circulating IL-22+ ILC3 and Th17 were significantly elevated in axSpA patients with dyslipidaemia (p=0.001 and p=0.034, respectively), and circulating IL-22+ ILC3 significantly correlated with ASDAS-CRP (Rho=0.4198 and p=0.0367). Stimulation with oxLDL-C significantly increased IL-22+ ILC3, NKp44- ILC3, and Th17 cells, and these were reversed by CD36 blocking agent. IL-22 and oxLDL-C increased TRAP+ cells and osteoclast-related gene expression. This study suggested potential role of circulating IL-22+ ILC3 as biomarker in axSpA. Furthermore, dyslipidaemia augmented IL-22+ ILC3 differentiation, and oxLDL-C and IL-22 markedly increased osteoclastogenesis of axSpA.

고지방·고단순당 식이 섭취 마우스에서 토종보리수 열매의 인슐린 저항성 및 고혈당 개선 효과 (Effects of autumn olive berry on insulin resistance and hyperglycemia in mice fed a high-fat, high-sucrose diet)

  • 토종보리수 열매의 항당뇨 효과를 규명하기 위하여, 마우스를 네 군으로 나누어, 기본 식이, 고지방·고단순당 식이, 토종보리수 열매 추출물을 저농도 (0.5%) 및 고농도 (1.0%)로 첨가한 고지방·고단순당 식이를 12주간 제공하였다. 고지방·고단순당 식이를 섭취한 군은 대조군에 비해 체중 및 체중 증가량, 부고환 지방무게가 유의적으로 증가하였으나, 고농도 보리수 열매 추출물의 급여는 부고환 지방무게를 유의적으로 감소시켰다. 고지방·고단순당군은 대조군에 비해 혈당, 인슐린, HOMA-IR값이 유의적으로 증가하였으나, 고농도 보리수 열매 추출물은 인슐린 농도를 감소시켰고, 저농도 및 고농도 추출물은 혈당 및 HOMA-IR값을 감소시켰다. 고지방·고단순당군은 대조군에 비해 IRS-2 및 AMPK 단백질 발현도가 유의적으로 감소하였으나, 고농도 보리수 열매 추출물의 급여는 IRS-2 발현도를 증가시켰고, 저농도 및 고농도 보리수 열매 추출물의 급여는 AMPK 발현도를 증가시켰다. 따라서, 토종보리수 열매는 제2형 당뇨병 동물에서 인슐린 저항성을 개선시켜 고혈당 개선효과를 나타낸 것으로 나타났다. 고지방·고단순당 식이를 섭취한 마우스에서 저농도 및 고농도 토종보리수 열매 추출물의 급여는 혈청 중성지방 농도 및 간조직의 총 지질과 중성지방 함량을 감소시켰고, 고농도추출물의 급여는 혈청 콜레스테롤 농도를 감소시켜, 토종보리수 열매는 지방간과 이상지질혈증 개선효과를 나타내었다.

중강도 운동을 실시한 비만 쥐에서 정향 투여가 지질과 근육 대사에 미치는 영향 (The effects of Syzygium aromaticum L. administration on lipid and muscle metabolism in obese rats subjected to moderate-intensity exercise)

  • 본 연구에서는 중강도 운동을 한 비만 흰쥐를 대상으로 정향 투여에 따른 염증 관련 단백질, 근육 합성 및 분해 관련 단백질에 미치는 영향을 평가하였다. 규칙적인 중강도 운동은 혈청 내 산화 스트레스와 leptin 수준을 억제시켰다. 또한, 염증 관련 단백질을 감소시켰으며, 근육 합성 관련 단백질을 활성화 및 근육 분해 관련 단백질 억제를 통해 근육 대사를 조절하였다. 여기에 정향의 투여는 혈청 내 염증성 사이토카인 IL-1β를 유의적으로 감소시켜 염증을 효과적으로 개선하는 효과를 발휘하였다.

  • 본 연구에서는 정향과 마가목 복합물 (SS)의 항비만 효과를 알아보기 위해 실험을 진행하였다. SS 투여는 3T3-L1 세포 내 TG와 TC가 유의적으로 감소하는 효과를 나타냈으며, 지질 합성 관련 유전자와 지방산 산화 관련 유전자 발현을 조절하는 효과를 보여주었다. 비만이 유도된 C57BL/6 mice에서 SS 투여는 혈청 내 leptin 호르몬 수치를 감소시켰으며, AMPK/ACC/SREBP-1 경로를 경유하여 TG 합성을 억제하였다. 또한, 조직병리학적 분석을 통해 지질 축적과 지방세포의 크기가 감소된 것을 확인하였다. 따라서 SS는 비만의 예방과 치료를 위한 잠재력을 갖춘 소재로 사료된다.

국내 옥수수 품종에 따른 기능성 성분의 자연 변이 분석 (Natural variation of functional components between Korean maize types)

  • 옥수수는 전세계적으로 다양한 형태로 소비되는 3대 식량작물로, 영양성분은 대부분은 전분으로 구성되어 있지만, 다량의 기능성 성분도 포함되어 있다. 옥수수는 형태적으로 찰옥수수, 단옥수수, 종실옥수수로 나뉘는데, 각 품종마다 쓰임이 다르기 때문에, 영양성분에서도 차이가 나타난다. 본 연구는 옥수수의 기능성 성분 함량을 분석하고 환경적 요인을 고려하여 자연변이 데이터를 생산하기 위해 수행하였다. 국내 3개지역(수원, 대구, 홍천)에서 재배한 옥수수 3가지 형태의(찰옥수수, 단옥수수, 종실옥수수) 시료를 수집하고, HPLC 및 GC-TOF-MS를 이용하여 성분을 분석하였다. ANOVA 및 다변량 통계분석, Pearson 상관분석을 통해, 형태별 그룹으로 비교하였을 때, 총 32개의 성분 항목 중 28개의 성분 항목이 유의적 차이를 보였으며(p<0.05), 지역별 그룹으로 비교한 결과, 15개의 성분이 유의적 차이를 보였다. 다변량 분석결과, 지역별보다 형태별로 군집 구분이 뚜렷하게 형성되었는데, 형태별 군집 형성 주요하게 기여한 성분은 α-carotene, lutein과 δ-tocopherol이었다. 본 연구에 따르면, 옥수수 기능성 성분은 환경적 요인보다 유전적 요인에 의해 영향을 더 많이 받는 것으로 사료된다. 자연변이 데이터는 옥수수 기능성 소재 연구분야 및 생명공학작물의 식품학적 안전성 평가시 기초자료로 활용될 수 있다.
